Experiments on granular flow in a hexagonal silo: a design that minimizes dynamic stresses

  • In this paper, an experimental study of the rheological behavior of granular flow in a new type of storage silo is presented. The main characteristic of the new design is a hexagonal shape chosen with the objective of minimizing the stresses applied to the stored grains, and to reduce grain damage during the filling and emptying processes. Measurements of stress distribution and flow patterns are shown for a variety of granular materials. Because of the design of the silo, the granular material adopts its natural rest angle at all times eliminating collisional stresses and impacts between grains. A homogeneous, low friction flow is naturally achieved which provides a controlled stress distribution throughout the silo during filling and emptying. Secondary dynamic stresses, which are responsible for wall failure in conventional silos of the vertical type, are completely eliminated. A comparison between the two geometries is presented with data obtained for these silos and a number of granular materials. The discharge pattern inhibits powder formation in the silo and the filling system virtually eliminates unwanted material packing. Finally, notwithstanding the rheological advantages of this new design, the hexagonal cells that constitute the silo have many other advantages, such as the possible use of solar energy to control the humidity inside them. The cell type design allows for versatile storage capabilities and the elevation above the ground provides unlimited transportation facilities during emptying.

Isolation of Hydrogen-producing Bacteria from Granular Sludge of an Upflow Anaerobic-Sludge Blanket Reactor

  • H$_2$-producing bacteria were isolated from anaerobic granular sludge. Out of 72 colonies (36 grown under aerobic conditions and 36 under anaerobic conditions) arbitrarily chosen from the agar plate cultures of a Suspended sludge, 34 colonies (15 under aerobic conditions and 19 under anaerobic conditions) produced H2 under anaerobic conditions. Based on various biochemical tests and microscopic observations, they were classified into 13 groups and tentatively identified as follows: From aerobic isolates, Aeromonar spp. (7 strains), Pseudomonas spp. (3 strains), and Vibrio spp. (5 strains); from anaerobic isolates, Actinomyces spp. (11 Strains), Clostridium 5pp. (7 strains). and Porphyromonas sp. When glucose was used as the carbon substrate, all isolates showed a similar cell density and a H$_2$ production yield in the batch cultivations after 12 h (2.24-2.74 OD at 600 nm and 1.02-1.22 mol H$_2$/mol glucose, respectively). The major fermentation by-products were ethanol and acetate for the aerobic isolates, and ethanol, acetate and propionate for the anaerobic isolates. This study demonstrated that several H$_2$ producers in an anaerobic granular sludge exist En large proportions and their performance in terms of H$_2$ production is quite similar.

  • The present study was carried out light and electron microscopic analysis of the structures of the tritocerebral neurons in butterfly (Pieris rapae Linne) from Insecta. We have summarized our morphological analyses as follows: In insecta, tritocerebrum of butterfly shows tentatively classified 6 neuronal types without any arrangement of cell layers. Type I cells with long oval or spindle shape are the largest out of 6 kinds. The nucleus contains a small amount of chromatin. In the large volume of cytoplasm there contained rich cell organelles except granular endoplasmic reticulum. Type II cells are smaller than type I neurons, but their ultrastructural features are similar to type I neuron. They have well-developed granular endoplasmic reticulum. Type III cells are spherical or triangular and smaller than type I neuron with spherical nuclei. Cell organelles are mostly prominent, esp., mitochondria, ribosomes and fine fibrils which are arranged in periphery of the cytoplasm.

Mechanism of Ethanol-induced Purkinje Cell Death in Developing Rat Cerebellum: Its Implication in Apoptosis and Oxidative Damage

  • Ethanol treatment during the brain growth spurt period has been known to induce the death of Purkinje cells. The underlying molecular mechanisms and the role of reactive oxygen species (ROS) in triggering ethanol-induced Purkinje cell death are, however, largely unresolved. We undertook TUNEL staining, western blotting assay and immunohistochemistry for the cleaved forms of caspase-3 and -9, with calbindin D28K double immunostaining to identify apoptotic Purkinje cells. The possibility of ROS-induced Purkinje cell death was immunohistochemically determined by using anti-8-hydroxy-2'deoxyguanosine (8-OHdG), a specific cellular marker for oxidative damage. The results show that Purkinje cell death of PD 5 rat cerebellum following ethanol administration is mediated by the activation of caspase-3 and -9. However, unexpectedly, TUNEL staining did not reveal any positive Purkinje cells while there were some TUNEL-positive cells in the internal and external granular layer. 8-OHdG was detected in the Purkinje cell layers at 8 h, peaked at 12-24 h, but not at 30 h post-ethanol treatment. No 8-0HdG immunoreactive cells were detected in the internal and external granular layer. The lobule specific 8-OHdG staining patterns following ethanol exposure are consistent with that of ethanol-induced Purkinje cell loss. Thus, we suggest that ethanol-induced Purkinje cell death may not occur by the classical apoptotic pathway and oxidative damage is involved in ethanol-induced Purkinje cell death in the developing cerebellum.

Granular Cell Tumor of the Axillary Accessory Breast: A Case Report (액와부 부유방에 발생한 과립 세포 종양: 증례 보고)

  • Granular cell tumors (GCTs) are rare benign soft tissue tumors that can occur throughout the body, particularly the head and neck; only 5%-8% of GCTs occur in the breast. We report a case of a GCT of the axillary accessory breast, which is a rare location of this tumor. A 50-year-old woman had a 2-month history of a palpable mass in the left axilla. Physical examination, as well as mammographic and ultrasonographic findings suggested a breast malignancy. Histopathological examination showed a benign GCT, and wide local excision was performed. The patient has remained disease-free over 2 years postoperatively. Although most GCTs are benign, wide complete resection of the tumor and follow-up are required considering the possibility of recurrence. The radiologist should know the characteristics of GCTs as a differential diagnosis of breast and axillary lesions to prevent unnecessary treatment.

Development and Verification of Large Triaxial Testing System for Dynamic Properties of Granular Materials (조립재료 동적물성 산정을 위한 대형삼축압축시험장비 구축 및 검증)

  • Coarse granular material is used as important fill material in most of large embankments such as railway, road, dam and so on. Therefore, the accurate design parameters of the coarse granular material are necessarily required in design and construction. The behavior of the coarse granular material was not well understood because of the lack of large testing equipment capable of coarse granular material. A large triaxial testing system was developed in this research, capable of large specimens of 500 mm, 300 mm and 150 mm in diameter. In the new large triaxial testing system, the load cell is installed inside the triaxial cell and axial displacement is measured locally on a specimen in order to improve control and measurement in small strain level. Urethane specimens of 300 mm and 50 mm in diameter were prepared. The large triaxial tests were performed on the 300 mm diameter urethane specimens while RC/TS and impact echo tests on the 50 mm diameter urethane specimens to verify this testing system. In this verification test results, we could ascertain the reasonable test results of the KRRI large triaxial testing system.

The Effects of EGEE on the Morphometry in the Thickness and Histogenesis of Rat Cerebral Cortex During Developmental Phase (발생기 흰쥐 대뇌 피질의 형태 구조에 미치는 Ethylene Glycol Monoethyl Ether의 영향)

  • This study attempts to investigate the developmental alterations of rat cerebral cortex, and the effects of EGEE on the developmental cerebral cortex in the prenatal, postnatal and adults were examined by morphological methods and H-E staining was used for the histological changes. In the case of injection of EGEE, at 14 day of fetal phase, parietal cortex was thickest $(95{\pm}12.7\;{\mu}m)$ but, it was thinner than in the control group $(102{\pm}14.0\;{\mu}m)$ and, occipital cortex $(57{\pm}10.5\;{\mu}m)$ compared with other cortexes was the thinnest in fetal phase. In the suckling phase, each cortex grew thick quickly but, after weanning phase, the growth of the cortex slowed and the thickness of cortex was similar to that of cortex in the adult phase. At 105 day after birth, the parietal cortex was thickest $(934{\pm}21.6\;{\mu}m)$ but, decreased compared with control group $(1113{\pm}19.0\;{\mu}m)$. When EGEE was injected in intraperitoneal of rat, the number of neuroblasts per unit area was largest $(207.7{\pm}11.4/10^{-2}\;mm$ at the mantle layer of parietal cortex at 14 day of fetal phase but, decreased compared with control group $(224.2{\pm}13.8/10^{-2}\;mm$ , and the size was largest $(7.5{\pm}1.3\;{\mu}m)$ at the ependymal cell layer of occipital cortex at 3 day after birth but, decreased compared with control group $(9.0{\pm}1.2\;{\mu}m)$. Simillar to control group, the number of granular cells and pyramidal cells were largest at the II and III layer of parietal cortex, but decreased during developmental phase. The size was largest at the IV and V layer of occipital cortex but it was decreased compared with control group. When EGEE was injected in intraperitoneal of rat, the cerebral cortex from fetal phase to 3 day after birth has differentiated into the 3 layers; ependymal, mantle and marginal layer, but empty cisternaes or vacoules in the cerebral cortexes and the condensed phases of neuroblasts were appeared. From 5 day after birth, it has differentiated into the 4 layers; molecular, external granular, mixed layer of internal granular, external and internal pyramidal cells and multiformal layer but, empty cisternaes or vacoules in the granular and pyramidal cell layers were appeared and the number per unit area of neuron was decreased. In the cerebral cortex of the weaning and adult phases, division of cell layers was not clear and empty cisternae was formed in the cortex with the cells in external granular and pyramidal cell layers, was magnified or condensed around blood vessels of neurons.

  • An electron microscopical study on hemocytes of last instar larvae of Orthoptera (Acrida cinerea Thunberg, Paratenodera sinensis De Saussure, Atractomorpha lata Hotschulsk was performed to observe cell types, structures and their origins. Prohemocytes are the smallest round cells with large, nucleus, poorly developed cytoplasmic organelles and lower composition ratio, accouting for less than 1% of all cell types. Little remarked differences were revealed among different species. Plasmatocytes are relatively large cells with higher composition ratio, accounting for 5 to 20%. The majority of the plasmatocytes are oval forms with numerous external cytophismic projections., relatively well developed cytoplasmic organelles. Granular cells are characterized by various granules in cytoplasm. Some considerable remarked differences were revealed among different species. Furthermore the granules themselves differ in fine structures. Adipohemocytes were found only in Acrida cinerea Thunberg.

  • Shrimp has efficient non-specific defense mechanisms based on activities of the hemocytes against pathogens. Up to now, it has been known that one of the non-specific immune reactions is related to mutual association among types of hemocyte, granular cell, semi-granular cell, and hyaline cell. In this study, we tried to know the effectiveness of feed-additives such as 5-aminolevulinic acid(ALA), chitosan, and hot-water extract of herb on immunity of shrimp (Penaeus chinensis) by hemocytic observation and SDS-PAGE analysis. Finally, we suggest a principle of the examination system for effects of various feed-additives.

Ultrastructural study of mouse ovary under X-ray irradiation (방사선 조사선량에 따른 생쥐 난소의 미세구조적 연구)

  • This study investigated the structural changes of folliculus ovarii according to the dose of the X-rays when mice were exposed to X-rays from 6MeV LINAC. The minute structural changes of folliculus ovarii were observed through an electron microscope with high magnification. Nuclei and protoplasm of granular cells in growing folliculus ovarii abruptly underwent minute structural changes according to the increase of dose of X-rays. Cell residue, by-product of cell decease, neutrophil and macrophage around follicular antrum were observed. The minute structural changes in granular cells showed typical characteristics of apoptosis: the increase of electronic density due to nuclear condensation, fragmentation of nuclei, and atrophy of protoplasm. Necrosis of cells was identified, but it was not so remarkable. Macrophage scattered with apoptotic bodies.
