Development of a Novel Spawn (Block Spawn) of an Edible Mushroom, Pleurotus ostreatus, in Liquid Culture and its Cultivation Evaluation

  • Mushroom cultivation has gained increased attention in recent years. Currently, only four types of spawn, including sawdust spawn, grain spawn, liquid spawn, and stick spawn, are commonly available for mushroom cultivation. This limited spawn diversity has led to difficulty in selecting suitable inoculum materials in some cultivation. In this study, three small blocks of lignocellulosic agro-wastes and one block of a synthetic matrix were prepared as support for growing Pleurotus ostreatus in liquid medium. Mycelium-adsorbed blocks were then evaluated for their potential as block spawn for fructification. Our results indicated that the edible fungus was adsorbed and abundantly grew internally and externally on loofah sponge and synthetic polyurethane foam (PUF) supports and also has the ability to attach and grow on the surface of sugarcane bagasse and corncob supports. The mycelia of P. ostreatus adhered on corncob exhibited the highest metabolic activity, while those on the PUF showed the least activity. Mycelial extension rates of block spawns made of agro-waste materials were comparable to that of sawdust spawn, but the block spawn of PUF showed a significantly lower rate. No significant differences in cropping time and yield were observed among cultivations between experimental block spawns and sawdust spawns. Moreover, the corncob block spawn maintained its fruiting potential during an examined period of 6-month storage. The developed block spawn could be practically applied in mushroom cultivation.

Characteristics and suitability of various cereal grains in spawn production of button mushroom

  • Spawn is the vegetative growth of the mycelium and serves as the inoculum or seed for cultivating mushrooms. Various cereal grains, such as wheat, millet, barley, sorghum, brown rice, rye, and oat were assessed to compare their characteristics and suitability for spawn production of button mushroom (Agaricus bisporus). The mycelial growth rates, density, and the number of completely colonized grains were measured from the twentieth day of inoculation. Wheat grains showed fastest mycelial growth with 8.4 cm followed by rye, oat, barley with 8.2, 7.5 and 7.3 cm, respectively. In the mycelial density, foxtail millet, barley, and sorghum were best compared with that of wheat grains. Especially, the number of grains which were completely colonized by mycelia were greatest in foxtail millet with 5,123 grains followed by proso millet, and wheat with 3,052 and 914, respectively. Based on the results obtained, barley, foxtail millet, and sorghum grains would be appropriate substituting for wheat grain in spawn production of button mushroom.

Response of Barley Genotypes to Fusarium Head Blight under Natural Infection and Artificial Inoculation Conditions

  • Forty-eight spring barley genotypes were evaluated for deoxynivalenol (DON) concentration under natural infection across 5 years at Harrington, Prince Edward Island. These genotypes were also evaluated for Fusarium head blight (FHB) severity and DON concentration under field nurseries with artificial inoculation of Fusarium graminearum by the grain spawn method across 2 years at Ottawa, Ontario, and one year at Hangzhou, China. Additionally, these genotypes were also evaluated for FHB severity under greenhouse conditions with artificial inoculation of F. graminearum by conidial suspension spray method across 3 years at Ottawa, Ontario. The objective of the study was to investigate if reactions of barley genotypes to artificial FHB inoculation correlate with reactions to natural FHB infection. DON concentration under natural infection was positively correlated with DON concentration (r = 0.47, P < 0.01) and FHB incidence (r = 0.56, P < 0.01) in the artificially inoculated nursery with grain spawn method. Therefore, the grain spawn method can be used to effectively screen for low DON. FHB severity, generated from greenhouse spray, however, was not correlated with DON concentration (r = 0.12, P > 0.05) under natural infection and it was not correlated with DON concentration (r = -0.23, P > 0.05) and FHB incidence (r = 0.19, P > 0.05) in the artificially inoculated nursery with grain spawn method. FHB severity, DON concentration, and yield were affected by year, genotype, and the genotype × year interaction. The effectiveness of greenhouse spray inoculation for indirect selection for low DON concentration requires further studies. Nine of the 48 genotypes were found to contain low DON under natural infection. Island barley had low DON and also had high yield.

아까시재목버섯이 옻나무 껍질의 urushiol 제거에 미치는 영향 (Effect of Perenniporia fraxinea on Eliminating Urushiol from Rhus verniciflua Stokes Stem Bark)

  • 본 연구에서는 urushiol 제거에 적합한 장수버섯 종균용 원료에 대하여 평가하였다. 세 가지 곡물조, 현미, 통밀에서의 장수버섯 균사체 증식속도는 각각 4.92±0.05, 2.20±0.03, 1.93±0.03 mm/day 이었다. 장수버섯이 배양된 곡물종균의 laccase의 활성은 각각 0.86±0.02, 0.04±0.01, 0.01±0.00 U/mL 이었다. 증식속도와 효소활성 측면에서 장수버섯 종균용 곡물로는 조를 사용하는 것이 적합하였다. 조에 배양된 장수버섯 종균을 옻나무에 배양하였더니 3일차에 urushiol이 최대 86.6% 감소하였고 7일차에 98.5%까지 감소하였다. Urushiol 제거를 위한 적정 배양기간은 3일이었으며 이때 flavonoid 잔존량은 68%, phenolic 성분의 잔존량은 42% 이었다. 액체종균을 사용하여 배양한 것과 비교했을 때 urushiol을 제거하기 위한 증식기간이 10일에서 3일로 단축되었다.

  • 이 연구는, 느타리버섯과 풀버섯만 재배하던 캄보디아에서 영지버섯, 목이버섯 그리고 표고버섯 등과 같은 여러 종류의 고온성 버섯들의 종균을 생산하고, 캄보디아에서 생산되는 경제적이고 효율적인 새로운 버섯배지를 찾기 위해 시도되었다. 종균 및 버섯재배 배지로, 여러 종류의 유기물찌꺼기, 곡물, 그리고 톱밥이 사용되었다. 고무나무 톱밥배지에서 자란 균사 (10.9 cm/15일)는 곡물배지에서 자란 균사 (11.2 cm/15일)에 비해 성장이 조금 느렸지만, 고무나무 톱밥은 낮은 가격 및 취급의 용이성이 있어, 비싼 곡물종균을 대치할 수 있는 매우 적절한 배지인 것으로 나타났다. 버섯생산을 위해서, 고무나무톱밥(60.8% BE), 사탕수수박(60% BE), 그리고 아카시아나무 톱밥 등에 미강과 탄산칼슘을 혼합하여 사용하였는데, 사탕수수박과 고무나무 톱밥은 매우 비슷한 높은 생물학적 효율(BE)을 보여주었고, 아카시아나무 톱밥은 비교적 낮은 22.4%의 생물학적 효율을 보여주었다. 따라서 낮은 가격에 구입할 수 있는 아카시아톱밥이 현재 사용되고 있는 높은 가격의 배지들과 혼합하여 사용된다면 매우 가능성이 높은 새로운 배지가 될 수 있을 것으로 보였다. 현재, 사용되고 있는 톱밥의 가격 (20 kg 고무나무톱밥=6500 Riel 혹은 1.6 USD)과 비교했을 때, 아카시아톱밥의 가격(20 kg=1000Riel 혹은 0.25USD)이 매우 낮아 버섯의 생산에 투자되는 비용을 크게 줄여 경제성을 높일 수 있을 것으로 사료된다.

Characteristics of sawdust cultivation of Lentinula edodes with different methods of spawn inoculation

  • This study was carried out to investigate the management characteristics and growth performance of L. edodes from the cooling stage to incubation. Bags of different heights and weights are available for bagging. When the medium size of $17{\times}13cm$ was used and the size of the inoculation hole was changed from 1/3 to 2/3, the browning period was shortened to 30 days. Mycelial growth was evaluated according to the cooling temperature after sterilization. It was observed to be the highest at 122 mm/15 days at $10^{\circ}C$ and 114 mm/15 days and 117 mm/15 days at $15^{\circ}C$ and $20^{\circ}C$, respectively. The contamination rate of the sawdust media before inoculation was measured as 0, $4.5{\times}10$, $1.3{\times}10^2$, $4.0{\times}10^3cfu$ at $5^{\circ}C$, $10^{\circ}C$, $15^{\circ}C$, and $24^{\circ}C$ respectively. The average of $1.6{\times}10^8$ colony forming units (cfu) of microorganisms was observed in the sawdust that had been piled for six months outdoors. In summer, the sawdust has to be used immediately after mixing. The sterilized medium had an average of $4{\times}10^3cfu$ of microorganisms at $24^{\circ}C$ and $1.3{\times}10^2cfu$ at $15^{\circ}C$. After 15 days of inoculation in vitro, the growth conditions of the sawdust was the best at 132 mm, followed by grain and liquid. When inoculated with liquid spawn, the moisture content of the substrate should be adjusted between 50% and 55% in advance.

Availability of Sikhae Factory Wastewater as a Submerged Culture Medium for Lentinula edodes

  • Sikhae is a Korean traditional beverage of saccharified rice. Its factory waste(SFW) is usually thrown away instead of being used. We developed a cheap substrate of SFW for use in liquid spawn that is known for its higher fruit body yields than grain spawn in sawdust cultivation. Mycelia of Lentinula edodes ASI 3046, which is regarded as the most suitable strain for sawdust cultivation, were cultured on six kinds of previous known media and SFW. As the seven kinds of media were applied, a Sikhae Factory Waste(SFW) was most excellent in growth. The dried mycelial weight in SFW was almost four times as much as that in the other media. In the flask culture, optimum culture conditions for the mycelial growth were obtained after 13 days of cultivation at media volume of 100 ml, 100 rpm, initial pH 4.5, and $25^{\circ}C$. The best mycelial growth was observed when $MgSO_4{\cdot}7H_2O$ and D-sucrose were added as a supplement in SFW. SWM must be a remarkable medium for L. edodes because of its simple preparation and low cost.

먹물버섯속균(Coprinus spp.)의 형태적 특성에 관한 연구 (Studies on the Morphological Characteristics of Coprinus species)

  • 식용과 약용버섯으로서 이용가치가 높은 것으로 알려진 먹물버섯(C. comatus)에 대하여 보다 우수한 인공재배법을 개발하고 자실체의 형태적 특성에 대한 연구 결과는 다음과 같다. 먹물버섯속균 5개 균주 중에서 C. comatus균인 CM 980301 균주를 우량균주로 선발하였으며, 먹물버섯(C. comatus: CM 980301)자실체를 생산하기 위한 우수 배지는 종균용으로 밀곡립배지를 사용하고, 먹물버섯을 볏짚 발효배지에 인공재배 하였을 때 $24{\sim}28^{\circ}C$에서 균배양은 종균 접종 후 15일, 복토 후 13일이 소요되었으며, 원기형성 및 초발이는 $20{\sim}24^{\circ}C$에서 $10{\sim}11$일이 소요되었고, 버섯생육은 $20{\sim}24^{\circ}C$에서 $7{\sim}8$일이 소요되었다. 먹물버섯 (CM 980301)의 수확적기는 갓이 피기 전으로 자실체 경도는 갓 $102{\sim}169\;g/cm^2$, 대 $126{\sim}182\;g/cm^2$으로 단단하였고, 인공재배한 수량성은 $37.7kg/3.3m^2$, 개체중 17.9 g이었다.

  • 본(本) 시험(試驗)은 양송이 복토중(覆土中) 미생물(微生物)의 Mycogone perniciosa Magn.에 대한 항균작용(抗菌作用)및 M. perniciosa 와 Agaricus bisporus (Lange) Sing. 사이의 상호관계(相互關係)를 구명(究明)하기 위하여 실시(實施)되었으며 시험(試驗)한 결과(結果), 1. In vitro test에서 M. perniciosa는 복토(覆土)에서 분리(分離)된 미동정(未同定)의 미생물(微生物)들에 의해서 생장(生長)이 억제(抑制)되었으며 재배시험(栽培試驗)에서 M. perniciosa의 포자현탁액(胞子懸濁液)을 복토표면(覆土表面)에 국소접종(局所接種)하였을 때 라병(羅病)버섯은 접종(接種)된 부분(部分)에서만 발생(發生)되었고 접종부분(接種部分) 주변(周邊)에서는 발생(發生)되지 않았다. 2. M. perniciosa와 A. bisporus사이의 길항적(拮抗的) 관계(關係)는 in vitro test에서 배양기(培養基)의 종류(種類)에 따라 다소(多少) 차이(差異)가 있었으나 A. bisporus의 균사(菌絲)는 M. perniciosa 균총(菌總) 위를 덮어서 자랐다. 재배시험(栽培試驗)에서 M. perniciosa의 포자현탁액(胞子懸濁液)을 A. bisporus의 곡립종균(穀粒種菌) 표면(表面) 혹은 복토층(覆土層)의 중간부위(中間部位)에 분무접종(噴霧接種)하였을 때에는 라병(罹病)버섯이 발생(發生)되지 않았으나, 복토표면(覆土表面)에 접종(接種)하였을 때에만 리병(罹病)버섯이 발생(發生)되었다.
