  • Five soybean varieties of two early maturing Karikei73 and SS 79168, and three late maturing Tohoku76, Baegunkong and Jangbaegkong were used and evaluated in the study. Of the varieties examined, Karikei73 was characterized by the delayed leaf senescence. The varieties were planted in the pots of 1/3500 a filled with volcanic ash soil at the experimental fields of the National Institute of Agrobiological Resources in Japan. Major agronomic characteristics including the activity of nitrogen fixation for root nodules during the grain filling period were measured. Measurements during the stages were followed by the stage of development descriptions for soybeans made by Fehr and Carviness (1977). The acetylene reducing activity (ARA) per dry weight of root nodule measured using acetylene reduction assays was the highest at R4-R4.5 with decreasing trends thereafter for the early matruing varieties, while it continuously increased up to R6 but decreased rapidly thereafter for the late maturing varieties. The dry weights of root nodules and all parts of the host plant at each stage checked were greater in the late maturing varieties being the same in ARA per pot.

Comparison of Rice Stripe Disease Occurrence and Yield under Different Rice (Oryza sativa L.) Cultivars (이앙시기에 따른 벼 품종별 줄무늬잎마름병 발생 및 수량 비교)

  • The purpose of this study is to investigate an occurrence rate of rice stripe virus disease under different rice transplanting time and different rice cultivars. After final harvest, the yield of rice infected by rice stripe virus was obtained on Wangchal cultivar compared to uninfected rice. The results obtained as following: 1) Yield of rice infected by rice stripe virus was decreased by 64% with reduction of culm length, pancile length, number of spike, number of spikelet, and grain filling compared to uninfected rice, 2) An occurrence rate of rice stripe virus disease under different rice planting season was high as order of May 30 > June 15 > July 1. In additional, to reduce a rice stripe virus disease on higher disease occurrence region and susceptible cultivars, rice planting season will be accepted on and after June 15, and 3) Eighteen cultivars were resistance cultivars such as Hwayeongbyeo, Onnuri, Ilmibyeo, Nampyongbyeo, Dongjin2ho, Hopumbyeo, Hwangkeumnuri, Malgeumi, Saenuri, Pungmi1ho, Haechanmulgyeo, Hwangkeumnodeul, Chilbobyeo, Dongjinbyeo, Younganbyeo, Junambyeo, Samkwangbyeo, and Nakdongbyeo. Twenty-one cultivars were susceptible cultivars such as Odaebyeo, Unkwangbyeo, Shinungbong1ho, Manabyeo, Hopyongbyeo, Borami, Damibyeo, Hwangchalbyeo, Sulgengbyeo, Nongrim6ho, Sasanishigi, Yunishigari, Chungmubyeo, Dongjin1ho, Huknambyeo, Guromy, Shanghehanghulna, Heukchalbyeo, Heukhwangbyeo, and Aranghwangchalbyeo.

Effect of Paclobutrazol and Flurprimido Application on Characteristics Related with Lodging of Paddy Rice Plant (벼 도복(倒伏) 관련(關連) 형질(形質)에 미치는 Paclobutrazol과 Flurprimido의 처리(處理) 효과(效果))

  • This study was conducted to find out the effect of paclobutrazol 0.6G and flurprimido 50wp application at 40 days before heading on the characters related with lodging and the growth and yield of paddy rice plants in 1983. The culm length were shortened by paclobutrazol and flurprimido applications and high level of suppression was observed in the 3rd to 5th internodes. Paclobutrazol caused thiner internode and thicker wall of internode than the untreated control, however flurprimido caused thicker wall of internode without change of internode diameter. The breaking weight of lower internode was higher by flurprimido application than those by the untreated control and paclobutrazol application. The lodging indexes effected by both chemicals were remarkably decreased, and also the field lodging was not observed. The leaf area indexes effected by paclobutrazol and flurprimido applications were increased because of the increasing of tillers and green leaves per stem in spite of shortening of lead blades. Paclobutrazol and flurprimido application out-yielded significantly the untreated control lodged at grain filling stage. However in conditions of untreated with no lodging, the yields of these treatments were significantly less than that of untreated control.

  • This experiment was carried out to investigate the stomatal aperture of wheat variety 'Chokwang' grown in the two different nitrogen level. Stomatal aperture was measured at the maximal tillering, shooting, booting, flowering and grain filling stages. Stomatal aperture in leaf blades gradually increased in accordance with the successive leaf growth. The maximal opening of stomata was observed at flowering stage and around noon in a day, but stomata closed around six o'clock in the afternoon. Stomata opened wider in the high nitrogen application than in the low nitrogen and their effects were the highest at the booting and flowering stage. Diurnal course at stomatal aperture of upper leaves was wider than that of lower leaves regardless of growth stages. Positive correlation (r=0.66$^{**}$) appeared between nitrogen content in leaf blades and stomatal aperture. The leaves of low position and the developing leaves showed smaller stomatal aperture than the full expanded top leaves irrespective of leaf stages. Water content of leaf, root weight and root activity were increased by the nitrogen application and thus considered as factors increase the stomatal aperture.ication and thus considered as factors increase the stomatal aperture.

  • This study was carried out to identify the ecological characters easily changeable with the conditions of temperature and day-length and to ascertain the managable means which can optimize the barley plants to secure the maximae yield and early heading under controlled condition. Yield and yield components were greatly varied with temperature. Especially, effective tiller formation and grain filling of sub-tillers were more favorably affected by thermal treatment than those of main tiller. In thermic response, 'Kangbori' was less susceptible than others, while 'Sacheon #6' showed particular susceptability. The varietal differences of photoperiodic response were recognized on all investigated traits and their degree of response were similar to those with temperature. Among varieties, 'Kangbori' was comparably sensitive on several characters while 'Sacheon #6' displaying low light-sensitivity. The turning point at which heading might be radically stimulated was estimated as about 10.5 hour day-length in 3 tested varieties and above 18 hours of irradiance was considered ineffective for more rapid heading. The condition under which barley varieties may obtain maximal yield and resonably early heading was 15$^{\circ}C$ in temperature and 12 to 15 hours of day-length, regardless of varieties. of day-length, regardless of varieties.

  • Five soybean varieties of two early maturing; Karikei 73 and SS79168, and three late maturing; Tohoku 76, Baegunkong and Jangbaegkong, were used and evaluated in the study, Of the varieties examined, Karikei 73 was characterized by the delayed leaf senescence, To investigate the periodical trends of sugars in plant organs and their roles to the nitrogen fixing activity of root nodules, the concentrations of reducing and non-reducing sugar in root and nodules during the grain filling period were measured. The concentration of non-reducing sugar in roots was not changed up to the stage of R6 for all of the varieties but it decreased rapidly thereafter for the varieties except Karikei 73. No such rapid decrease in the concentration of non-reducing sugar in the roots was observed for Karikei 73 having the characteristics of delayed leaf senescence. The concentration of reducing sugar in the root nodules was not greatly changed for all of the varieties up to the stage of R6 regardless of the earliness of varieties but increased temporarily at R6.5 when there was a rapid decrease in ARA. The phenomenon explained the fact that nitrogen fixing activities were controlled not only by supplying sugars as the source of energy for nitrogen fixation, but also by the need for fixed nitrogen of the plant. The concentration of non-reducing sugar in root nodules also increased up to the stage of R6-R6.5 but decreased at R7, which could apply the same explanation as in the concentration of reducing sugar of the root nodules.

Application and perspectives of proteomics in crop science fields (작물학 분야 프로테오믹스의 응용과 전망)

  • Thanks to spectacular advances in the techniques for identifying proteins separated by two-dimensional electrophoresis and in methods for large-scale analysis of proteome variations, proteomics is becoming an essential methodology in various fields of plant sciences. Plant proteomics would be most useful when combined with other functional genomics tools and approaches. A combination of microarray and proteomics analysis will indicate whether gene regulation is controlled at the level of transcription or translation and protein accumulation. In this review, we described the catalogues of the rice proteome which were constructed in our program, and functional characterization of some of these proteins was discussed. Mass-spectrometry is a most prevalent technique to identify rapidly a large of proteins in proteome analysis. However, the conventional Western blotting/sequencing technique us still used in many laboratories. As a first step to efficiently construct protein data-file in proteome analysis of major cereals, we have analyzed the N-terminal sequences of 100 rice embryo proteins and 70 wheat spike proteins separated by two-dimensional electrophoresis. Edman degradation revealed the N-terminal peptide sequences of only 31 rice proteins and 47 wheat proteins, suggesting that the rest of separated protein spots are N-terminally blocked. To efficiently determine the internal sequence of blocked proteins, we have developed a modified Cleveland peptide mapping method. Using this above method, the internal sequences of all blocked rice proteins (i. e., 69 proteins) were determined. Among these 100 rice proteins, thirty were proteins for which homologous sequence in the rice genome database could be identified. However, the rest of the proteins lacked homologous proteins. This appears to be consistent with the fact that about 30% of total rice cDNA have been deposited in the database. Also, the major proteins involved in the growth and development of rice can be identified using the proteome approach. Some of these proteins, including a calcium-binding protein that fumed out to be calreticulin, gibberellin-binding protein, which is ribulose-1,5-bisphosphate carboxylase/oxygenase activate in rice, and leginsulin-binding protein in soybean have functions in the signal transduction pathway. Proteomics is well suited not only to determine interaction between pairs of proteins, but also to identify multisubunit complexes. Currently, a protein-protein interaction database for plant proteins (http://genome .c .kanazawa-u.ac.jp/Y2H)could be a very useful tool for the plant research community. Recently, we are separated proteins from grain filling and seed maturation in rice to perform ESI-Q-TOF/MS and MALDI-TOF/MS. This experiment shows a possibility to easily and rapidly identify a number of 2-DE separated proteins of rice by ESI-Q-TOF/MS and MALDI-TOF/MS. Therefore, the Information thus obtained from the plant proteome would be helpful in predicting the function of the unknown proteins and would be useful in the plant molecular breeding. Also, information from our study could provide a venue to plant breeder and molecular biologist to design their research strategies precisely.

  • To study the effect of cold water irrigation at the reproductive growth stage of rice plants on several growth characters related to source and sink and nutrient uptake, the present experiment was carried out under the different conditions of water temperature and water depth. Deep irrigation with normal temperature water increased culm length, panicle length and panicle exsertion but with cold water resulted adversely. Most sensi-tive response in 5 cm water-depth appeared at reduction division stage and in 20 cm depth at panicle formation stage. Secondary branches and spikelets were increased in number by deep irrigation with normal temperature water, but decreased and degenerated by deep irrigation with cold water at panicle formation stage resulting in high spikelet sterility and low grain filling. Deep irrigation with normal temperature water increased the contents of total nitrogen, phosphate, potassium and silicate in leaf blades, branches and chaff. However, cold water irrigation reduced the uptake of phosphate, potassium and silicate except nitrogen particularly in deep irrigation. Ratios of phosphate, potassium and silicate to total nitrogen content were decreased by cold water irrigation. Branches seemed to have higher requirements for phosphate, potassium and silicate than leaf blades and chaff. Silicate-to-total nitrogen ratio in leaf blades, branches and chaff had significant correlations with yield showing closer relationship between yield and the ratio of silicate to total nitrogen in branches in particu-lar.

Changes in SPAD Value and Phothosynthetic Rate during Grain Filling of Oryza glaberrima Strains and Oryza sativa Cultivars (Oryza glaberrima 계통과 Oryza sativa 품종의 등숙기간중 SPAD치와 광합성속도의 변화)

  • The process of the senescence in Oryza glaberrima Steud. strains and Oryza sativa L. cultivars were examined in terms of SPAD value(chlorophyll content) and photosynthetic rate. SPAD values and photosynthetic rates of flag leaves for 10 genotypes of each species was measured at the 1, 3 and 5 weeks after heading. SPAD values and photosynthetic rates of O. glaberrima strains tended to decrease rapidly after heading. O. sativa cultivars showed slower decrease as compared to O. glaberrima strains, in particular during the period from 1 to 3 weeks after heading. Although there was no significant difference between the two species in the mean value of photosynthetic rate and SPAD value at 1 weeks after heading, O. glaberrima had lower values after 3 weeks after heading. There were significant positive correlation coefficients between the photosynthetic rate and the SPAD value at 1 and 3 weeks after heading for O. glaberrima strains, and at 1 and 5 weeks after heading for O. sativa cultivars. There were significant positive correlation between the decreasing rates of the photosynthetic rates and the decreasing rates of the SPAD values at the period from the 1 to 3 weeks after heading for both species. At the period from 3 to 5 weeks after heading, only O. glaberrima showed a significant correlation between two traits, indicating that rapid decrease in chlorophyll content would affect the photosynthesis in O. glaberrima.

  • Mesothermal gold deposits of the Heungdeok, Daewon and Ilsaeng mines in the Youngdong area occur in fault shear zones in Precambrian metamorphic rocks of central Sobaegsan Massif, Korea, and formed in single stage of massive quartz veins (0.3 to 3 m thick). Ore mineralogy is simple, consisting dominantly of pyrrhotite, sphalerite and galena with subordinate pyrite, chalcopyrite, electrum, tetrahedrite and native bismuth. Fluid inclusion data indicate that hydrothermal mineralization occurred at high temperatures (>240$^{\circ}$ to 400$^{\circ}$C) from $H_{2}O-CO_{2}(-CH_{4})$-NaCI fluids with salinities less than 12 wt. % equiv. NaC!. Fluid inclusions in vein quartz comprise two main types. These are, in decreasing order of abundance, type I (aqueous liquid-rich) and type II (carbonic). Volumetric proportion of the carbonic phase in type II inclusions varies widely in a single quartz grain. Estimated $CH_4$ contents in the carbonic phase of type II inclusions are 2 to 20 mole %. Relationship between homogenization temperature and salinity of fluid inclusions suggests a complex history of fluid evolution, comprising the early fluid's unmixing accompanying $CO_2$ effervescence and later cooling. Estimated pressures of vein filling are at least 2 kbars. The ore mineralization formed from a magmatic fluid with the ${\delta}^{34}S_{{\Sigma}S}$, ${\delta}^{18}O_{water}$ and ${\delta}D_{water}$ values of -2.1 to 2.2$\textperthousand$, 4.7 to 9.3$\textperthousand$ and -63 to -79$\textperthousand$, respectively. This study validates the application of a magmatic model for the genesis of mesothermal gold deposits in Youngdong area.
