Investigation of Pathogenic Microbial Contamination in Medicinal Herb Products on the Market (유통 한약재에 대한 병원성미생물 분포)

  • Background: The study was conducted to investigate the distributions of faecal bacteria in commercial oriental medicine herb products. Methods and Results: A survey was conducted on the microbial contamination levels and antimicrobial specificity of Bacillus cereus and other microbes using 106 oriental medicine herb products on sale in Seoul. Pouring and isolation methods such as standard plate counts were used to identify the bacteria. The isolated bacterias included coliforms, Bacillus spp., Enterococcus spp., Staphylococcus spp., Listeria spp.were identified by using gram staining and an API (analytical profile index) kit. Antimicrobial drugs discs were determined by CLSI (clinical and laboratory standards institute). Conclusions: The bacterial isolates present in the herbal medicines included 98 coliforms, 45 Bacillus spp., 29 Enterococcus spp., and 2 Listeria spp. Among these, there were nine Bacillus cereus strains, one Enterococcus faecium strain, and one Enterococcus faecalis strain present. The 9 Bacillus cereus strains were tested for susceptibility to 36 types of antibiotics products by the disc diffusion method. The strains showed resistance to 13 of these antibiotic products and semi-resistance to 5 antibiotic products. On the basis of these results, any oriental medicine herb product can be assumed to be contain resistant or semi-resistant bacterial strains. Therefore, we suggest prescribing guidelines and special management for the use of antibiotics in farms producing oriental medicine herb products.

Distribution of Foreign Mineral Materials and Heavy Metals Contained in Herbal Medicines, and Effect of Washing (한약재에 함유된 광물성이물과 중금속의 분포 및 수세효과)

  • This study was conducted to investigate the content of foreign mineral materials and heavy metals (Pb, As, Cd, Hg) contained in commercial herbal medicines (1504 samples) classified by parts used, and the effect of removing heavy metals by washing treatment (take 50 g of the sample and put 1000 ml of distilled water, shake it gentle by hand for 1 min). The 5% trimmed means of acid-insoluble ash value (%) were as follows; above ground parts (0.76) and underground parts (0.52). Those of the total amount of individual heavy metals value (mg/kg) were as follows; caulis (1.33), flos (1.23), herba (0.91), cortex (0.76), rhizoma (0.73), radix (0.67), semen (0.44) and fructus (0.39). Acid-insoluble ash content was correlated with Pb in herbal medicines of underground parts (r=0.446) (p<0.01). After washing, the removal rate of heavy metals contained in 6 radix herbal medicines shows 33-13% respectively.

  • The emission gas from meat grilled restaurants has unpleasant odor and sticky particulate matters (PM) in form of oil mist. This affects the residents living nearby. In oder to decrease the odor and PM, electrostatic precipitators (ESP) were installed at several restaurants by support of Seoul metropolitan government and emission characteristics of PM and odor were investigated. The removal efficiency of odor and PM at before- and after-ESP was that total hydrocarbon (THC), NH3, H2S, acetaldehyde, butyraldehyde were not much decreased. Total suspended particles (TSP) emitted from the vent was below the value of 7 mg/m3 regardless of the concentration of before-ESP and removal efficiency was in the range of 64 ~ 86%. The ratio of PM1.0 to PM10 of emission gas was over 0.9. So it was revealed that most PM of emission gas was PM1.0. The PM concentration in the air of the street near the vent was higher than another street due to the emission gas and the PM of indoor air of restaurant grilling meats was measured very high compared to outdoor air. So it needs more powerful ventilation of indoor air.

Investigation of stray cats Toxoplasmosis in Seoul area (서울지역 길고양이의 Toxoplasma 감염증 실태조사)

  • Toxoplasma gondii is one of the most common protozoa parasites of human and other warm-blooded animals. Cats and wild felidae play crucial roles in the epidemiology of toxoplasmosis. This study was performed to survey the prevalence of T. gondii infection among stray cats in the Seoul. A total of 422 stray cat blood samples were collected from Seoul area. Positive sera for T. gondii were identified in 56 samples (13.3%) exclusively via the latex agglutination test and the detection no antigen particles among seropositive samples by PCR. The overall infection rate of male stray cats (14.4%) presented as higher than that of female cats (10.7%). This study suggested that T. gondii is widespread in stray cats of Seoul area. It is needed to control urban stray cat population and to reduced the risk of zoonotic transmission of toxolasmosis to other animals and humans.

Characterization and isolation of shiga toxin-producing Escherichia coli from Bovine feces and Carcass (소의 분변과 도체에서 shiga toxin-producing Escherichia coli의 분리와 특성)

  • Shiga toxin (Stx)-producing Escherichia coli (STEC) strains can cause broad spectrum of human disease, including diarrhea, hemorrhagic colitis, and the life-threatening hemolytic uremic colitis (HUS). We examined 868 samples was taken from bovine feces and carcass from January to December 2008 in Seoul. Twenty two (9.5%) shiga toxin -producing Escherichia coli were isolated from the 230 of bovine feces, and two (0.31%) were isolated from the 638 of carcasses. Serotype of E. coli isolates were O157 (10, 41.6%), O26 (10, 41.6%), O111 (1, 4.2%) and UT (3, 12.6%). In PCR, the isolates displayed three different stx gene combination (stx1 [2, 8.4%]), stx2 [3, 12.6%] and stx1 and stx2 [19,87.5%]). The eaeA and hlyA gene were found in 11 (45.8%) of the 24 strains. Saa gene was present only one strains (4.2%). Toxin typing using reverse passive latex agglutination test showed the same result in VT 1. But it showed different result in VT 2. In antimicrobial susceptibility test, all isolates were sensitive to amikacin, amoxicillin/clavulanic acid, ciprofloxacin and colistin. Eighteen strains (75.0%) of 24 isolates showed the multi-resistant patterns with over 3 drugs. PFGE was performed after the genomic DNA of twenty four isolates was digested with Xba I. the 24 isolates showed 7 (A~G) PFGE type.

VOC Emission Characteristics of Dry Cleaned Wool Scarfs through Small Chamber Test (소형챔버를 이용한 드라이클리닝 모직물 목도리의 휘발성유기화합물 방출 특성)

  • Objectives: This study was conducted to evaluate the emission of VOCs from clothing that had been dry cleaned. Methods: In order to ensure the same conditions, f100% wool scarves were selected as the fabric type. Four identical tests were conducted on the option of either removing the plastic bags which came from the dry cleaning shop or not. The scarf was located inside a closet or room for one or two days. Small chamber tests were conducted to determine the VOC emission characteristics under the same conditions such as temperature, humidity, loading factor, and air exchange rates. Air from the chamber for VOCs was sampled by Tenax TA tube and analyzed by thermal desorption and GC/MSD. Results: Assuming that test represented dry cleaning and consumer's conditions well enough, we can conclude that immediate emissions after the dry cleaning of the scarfs caused elevated levels of TVOC, five VOCs (benzene, toluene, ethylbenzene, xylene, stylene), and decane group compounds. Conclusions: By removing the plastic bags which came from the dry cleaning shop or not, the storage conditions of dry cleaned scarfs by consumers during the storage time periods (one to three days) would be significant for reducing VOC emissions.

A Survey on Benzo(a)pyrene Contamination in Amomum Tsao-ko Fruit of Medicinal Herbs (유통 한약재 초과(草果) 중 벤조피렌 오염실태 조사)

  • Amomum Tsao-ko used as a traditional oriental herbal medicine, is indigenous to several Asia countries. This study was carried out to investigate the contamination by Benzo(a)pyrene in Amomum Tsao-ko Fruit of Medicinal Herbs. 20 samples of Amomum Tsao-ko Fruit were evaluated for the Benzo(a)pyrene contamination. They were analyzed for Benzo(a)pyrene using high-performance liquid chromatogrphy(HPLC)-fluorescence detection and the positive samples were confirmed using gas chromatography tandem mass spectrometry. The levels of Benzo(a)pyrene were from 9.2 to 95.5 ㎍/kg and the average was 40.6 ㎍/kg. There are no Benzo(a)pyrene standards for Amomum Tsao-ko Fruit of Medicinal Herbs. These data will be used as a basic data for the future legislation on the regulation and control of benzo(a)pyrene of Amomum Tsao-ko Fruit of Medicinal Herbs.

A survey of the microbial contamination level in butcher's shops in Seoul, Korea (서울지역 식육판매업소의 미생물 오염도 조사)

  • This survey was conducted to evaluate the microbial contamination level of butcher's shops in Seoul, Korea. For microbial inspections, a total of 584 samples (146 cotton work gloves, 146 utensils and equipments, 154 beef samples, 138 pork samples) were collected from butcher's shops. E. coli and pathogenic bacteria such as Staphylococcus aureus, Yersinia enterocolitica, Listeria monocytogenes, Salmonella spp., and E. coli O157:H7 were tested in the samples. As a result, the level of aerobic plate count (APC) ranged ${\leq}10^4\;CFU/cm^2$ from utensils and equipments, $10^1{\sim}10^9$ CFU/glove from cotton work gloves and ${\leq}10^6$ CFU/g from meat. The APC level of E. coli ranged ${\leq}10^1\;CFU/cm^2$ from utensils and equipments, ${\leq}10^5$ CFU/glove from cotton work gloves, and ${\leq}10^3$ CFU/g from meat, respectively. Staphylococcus aureus was detected in 2 beef samples, 1 pork sample, and 10 used cotton work gloves. Yersinia enterocolitica was detected in 3 beef samples, 1 pork sample, and 3 used cotton work gloves. Listeria monocytogenes was detected in 2 used cotton work gloves. In order to improve the sanitation status of butcher's shops, application of HACCP (Hazard Analysis Critical Control Point) or SSOP (Sanitation Standard Operating Procedure), regular hygiene education, and continuous monitoring for microorganisms will be required.

Molecular typing of Listeria monocytogenes using pulsed-field gel electrophoresis (PFGE) (Pulsed-field gel electrophoresis (PFGE)를 이용한 Listeria monocytogenes의 molecular typing)

  • A total of 1,354 samples was collected from bovine and porcine carcass from January 2005 to December 2006 in a slaughter house. Twenty five strains(1.8%) of Listeria monocytogenes were isolated from 1,354 samples using selective media. Ten(1.4%) L monocytogenes were isolated from the 677 of bovine carcasses, and 15(2.2%) were isolated from the 677 of porcine carcasses. Among 15 L mono-cytogenes from porcine, 11 siolates were serovars 1/2c, followed by 1/2b (3 strains, 20.0%) and 1/2a(1 strain) Out of 10 bovine samples, positive cases in 1/2a were 9 strains (90.0%), 1/2b were 1 strains(10.0%). PCR primers were selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene (hlyA) of L mono-cytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested. A total of 25 L monocytogenes strains were analysed by PFGE after digestion with Apa I. PFGE analysis of genomic DNA showed the $14{\sim}18$ fragments ranging in size from 30 to 550 kb, resulting in 14 patterns.

Analysis of Cinnamic Acid, Cinnamaldehyde and 2-Methoxycinnamaldehyde in Cinnamomi Ramulus on the Market in Seoul by HPLC (서울시내 유통 중인 계지의 지표성분 함량분석)

  • For the quality control of traditional herbal medicine, Cinnamomi Ramulus, simultaneous determination of cinnamic acid, cinnamaldehyde, and 2-methoxycinnamaldehyde was established by using a high performance liquid chromatographic (HPLC) method with diode array detector. To separate three constituents, Eclipse XDB C18 ($5{\mu}m$, $4.6{\times}250mm$) was used with 0.1% acetic acid and acetonitrile. Validation of the chromatography method was evaluated by linearity, recovery, and precision test. Calibration curve of standard components showed excellent linearity ($R^2$ >0.9999). A simple and efficient method by HPLC was developed to evaluate the quality of traditional herbal medicines made from Cinnami Ramulus. Three major bioactive ingredients in 30 samples that are from China(8) and Vietnam(22) were separated and quantified.
