  • Studies were made by measuring number of bacteria, water content, acidity and sugar content on spoiling at different kinds of containers in Summer season, to determine better kinds of containers adoptable for storing cooked rice being the most important food for Korean. It was found that brass, aluminum and porcelain containeres are not recommendable, whereas wooden containers such as a bamboo case (Sokori), a wooden box and a gourd pitcher (Bakaji) are quite useful for at least 24 hour storage even in hot Summer time.

  • Various types of the seeding of bottle gourd widely used as a rootstock of watermelon has been required to satisfy the farmers need. The study was done to determine the effect of light quality of blue, red and far-red lights treated with light emitting diodes on growth and morphology of bottle goured seeding and chlorophyll content of its cotyledons. The lights were treated in the growth chamber for 7 days to the seeding elapsed 8 days after sowing under natural condition, and 64 hole trays with commercial bedsoil. Plant height, length and diameter of hypocotyl, leaf area of cotyledon and first true leaf, its leaf length, number of true leaves, fraction and total dry weight were measured. Red light shortened and slenderized the hypocotyl, which lengthened by far-red light and thickened by blue light. Plant height was declined in order of far-red light treatment, blue and red lights mainly due to difference of hypocotyl length, Area and length of the first true leaf became smaller and shorter under far-red light than under the other lights. However, blue light increased leaf area of cotyledons. Two cultivars cv. Yongjadaemok and cv. Kunghap had different response to the light treatments in total seedling dry weight(dw); far-red and red light treatments showed the greatest and the least dw of hypocotyl, respectively, while blue and red lights did the greatest dw of the other organs. Among the ratio of each organ dw to total dw, those of hypocotyl and true leaves were different between the light treatments; the highest ratio of hypocotyl dw to total dw was observed in far-red light treatment but the lowest was in red light treatment. Those of the true leaves were the lowest in far-red light and similar response in blue or red light treatment. Chlorophyll content of cotyledons was decreased in order of red light treatment, blue and far-red lights, meaning that short period light treatment may influence photosynthesis of seeding and afterward its growth.

  • The adult of the Cotton caterpillar, Palpita indica (Saunder) laid eggs more on mature leaves than on aged and developing leaves. They laid more on leaves than on petiole and stem, and more on the adaxial than on the abaxial surface side of the leaves. Larvae of the Cotton caterpillar showed their preference in the order of the cucumber (Cucumis sativus), gourd (Lagernaria siceraria), watermelon (Citrullus lanatus) > oriental melon (Cucumis melon L. var makuwa), wax gourd (Benincasa hispida), melon (Cucumis melo), star cucumber (Sicyos angulatus) > sponge cucumber (Lufa cylindrica), cotton (Gossypium indicum). There were no distinct differences among varieties and lines that were collected and hybridized at the Gochang Watermelon Experiment Station in Korea. There was a positive relationship between the leaf area and the degree of damage by the Cotton caterpillar. The feeding amounts of the Cotton caterpillar excrement were gradually increased to 16-18 days after hatching, after that the amounts of excrement were rapidly decreased.

  • A multi-rapid immunofilter paper assay (multi-RIPA) system was prepared for simultaneous diagnosis of three Tobamoviruses, Cucumber green mottle mosaic virus (CGMMV), Kyuri green mottle mosaic virus (KGMMV), and Zucchini green mottle mosaic virus (ZGMMV) in cucurbitaceous crops. Each of these viruses was specifically detected by the multi-RIPA from cucumber, watermelon, zucchini, and bottle gourd inoculated with the three Tobamoviruses singly or in combination. The three viruses could infect cucumber, watermelon, and bottle gourd ; however, CGMMV could not infect zucchini as the latex-coated CGMMV antibody showed a negative reaction in the multi-RIPA of the virus-infected plant extract. When the minimum detection level of multi-RIPA was compared with that of double antibody sandwich-enzyme-linked immunosorbent assay (DAS-ELISA) using CGMMV, the latter was 10 times more sensitive than the former. The detection limit of the multi-RIPA for the purified CGMMV was 50 ng/ml. In a survey of the threeviruses in cucurbits growing in commercial fields in 1999 and 2000, CGMMV was detected in watermelon and cucumber, and ZGMMV was detected only in zucchini growing in plastic houses at the suburbs of Chonju, Korea. However, KGMMV was not found in the commercially growing cucurbit crops in our study, The results suggest that the multi-RIPA can be a simple, rapid, specific and convenient tool to detect simultaneously the Tobamoviruses.

  • The field experiment was conducted in vegetable area, Institute of Horticultural Sciences in University of Agriculture, Faisalabad in order to recognize morphological and fruit growth pattern and yield of Tinda (Praecitrullus fistulosus) gourd germplasm lines. Sixteen germplasm lines in which one line is used as check are included were sown and grown on flat beds in field. The field experiment was arranged as randomized complete block design (RCBD) with three replications. Data on days to maturity, fruit per vine, fruit weight in grams, fruit diameter and total yield obtained were recorded. The fruit texture is marked as phenotype parameter. All parameters were collected and then analyzed statistically. All lines and replications showed different results among each other according to parameter. The maximum DTM (days to maturity), F/P (fruit per plant), FW (fruit weight), FD (fruit diameter) and Yield were recorded was $L_0$ (68.66), $L_3$ (1.66), $L_{10}$ (248.33), $L_{13}$ (8.50) and $L_0$ (599.33) and the minimum were recorded was $L_1$ (56), $L_{11}$ (0.33), $L_0$ (198), $L_9$ (7) and $L_4$ (421) grams respectively. All lines showed smooth texture of fruits with no hairs when mature at harvesting stage.

  • To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. • Objectives: This study aimed to establish and provide reliable information for general public, based on expert consensus, on the risks of misuse of medicinal herbs for food and pure food ingredients for the fetus during pregnancy. Methods: A panelist of seven traditional Korean medicine (TKM) gynecologists responded to a questionnaire summarizing the fetal safety literature for twenty-five medicinal herbs for food and pure food ingredients derived from medicated diet (藥膳, Yaksun) recipes during three online Delphi rounds anonymously. Results: Ginkgonis Semen (Ginkgo nut), Illici Veri Fructus (Star anise), lavender, bitter gourd, and parsley were agreed at the level 1 of "Do not consume". These five ingredients were recognized as having significant risks both in the literature evidence and in expert opinion. Rosemary, Citri Unshius Pericarpium, Discoreae Rhizoma, lemongrass, Schisandrae Fructus, Cassiae Semen, Foeniculi Fructus, Mori Fructus, Cinnamomi Cortex, and Astragali Radix were agreed at the level 2 of "consultation with TKM practitioner is required". Conclusion: Based on the consensus of a seven-member expert panel of TKM gynecologists, consumption of Ginkgonis Semen (Ginkgo nut), Illici Veri Fructus (Star anise), lavender, bitter gourd, and parsley should be avoided by pregnant women. For Rosemary, Citri Unshius Pericarpium, Discoreae Rhizoma, lemongrass, Schisandrae Fructus, Cassiae Semen, Foeniculi Fructus, Mori Fructus, Cinnamomi Cortex, and Astragali Radix, the level 2 advisory may be recommended to use with caution and to consult a TKM practitioner for advice on consumption, dose, and duration.
