Impression cytology를 이용한 건성안의 결막 세포변화에 관한 연구 (A study of Conjunctival Cellular Changes in Dry Eye Patients by Impression Cytology)

  • Impression cytology는 결막상피의 표층을 떼어내기 위해 안구 표면에 cellulose acetate filter 재질을 같이 이용한다. 이 방법은 비 침습적이고 쉽게 시행할 수 있으며 환자에게 최소한의 불편감을 주며 결막표면에 반복해서 시행하여 변화를 관찰하기 위해 사용될 수 있다. 이 방법으로 결막 표면의 형태학적 연구와 squmaous metaplasia의 정도를 평가할 수 있다. 본 연구는 건성안 환자에서 impression cytology를 이용하여 결막표면을 평가하기 위해 수행되었다. 70명의 콘택트렌즈 비착용 학생을 대상으로 조사하였는데 먼저 조사 대상자들에게 McMonnies dry eye symptom questionnaire을 작성하도록 하였다. 그리고 비침습성 tear thinning time(TTT) 검사를 시행하고 다음에 침습성 검사인 Schirmer tear test(SIT), tear film break-up time(TBUT) tests 그리고 Rose-bengal staining올 시행하여 건성안 의심자를 분류하였다. 하 안구결막으로부터 결막 상피세포는 impression cytology기법으로 수집하여 PAS(Periodic Acid Schift)-haematoxylin 염색을 시행하였다. 배상세포와 결막상피세포는 광학현미경 400배에서 관찰하여 squamous metaplasia의 정도에 근거한 Nelson Grading scale에 따라 분류하였는데 이는 배상세포 밀도, 세포의 크기와 형태, 핵:세포질 비 등을 포함한다. 건성안 의심자는 상피세포의 형태적 변화, 핵의 변화, 배상세포 밀도 감소를 보였다. 세포학적 변화 정도는 건성안 조건의 정도에 관련이 있었다. 상피세포 형태를 Nelson의 분류시스템으로 단계를 나눈 결과 대조군의 91.43%가 grade 0 이었고 8.57%가 약간의 이상형태인 grade 1이었다. 반면에 건성안 의심자에서는 20%가 grade 0, 42.86%가 grade 1, 34.29%가 grade 2 그리고 2.86%가 심한 형태적 변화를 보인 grade 3를 나타냈다. Impression cytology는 부작용이나 거부반응이 없는 안구 표면 상피의 비 침습적 또는 최소 침습적 생검법으로 생각되며 안구표면을 포함하는 광범위한 과정을 위한 유용한 진단 기법으로 안전하고 간단하며 건성안 의심자의 다양한 안구 표면 변화의 이해를 증진시키는데 도움을 줄 수 있을 것으로 사료된다.

Effect of Erythromycin on Basal and Stimulated Mucin Release from Cultured Hamster Tracheal Surface Epthelial Cells

  • In the present study, we investigated whether lipopolysaccharide induce mucin release and erythro-mycin affect basal and adenosine triphosphate-induced (stimulated mucin release, from airway goblet cells. Confluent primary hamster tracheal surface epithelial cells were metabolically radiolabeled and chased for 30 min or 24 hr in the presence of varying concentrations of lipopolysaccharide or erythromycin to assess the effects on $^3H$-mucin release. The results were as follows : 1) Lipopolysaccharide failed to induce mucin release, 2) Erythromycin showed no effect on both basal and stimulated mucin release during 30 min of 24 hr treatment period. We conclude that lipopolysaccharide and erythromycin can not affect mucin release by direct acting on airway mucin-secreting cells.

  • Surface epithelial cells isolated from hamster tracheas and grown on a thick collagen gel become a highly enriched population of mucus-secreting cells. Epithelial cells from tracheas of hamsters were collected using enzymatic procedures and cultured under various conditions. The medium used consisted of a 1:1 mixture of medium 199 and Dulbecco's modified Eagle's (DME) medium which was conditioned before use. Insulin, transferrin, hydrocortisone, epidermal growth factor, and extract from bovine hypothalamus were used as supplement. Due to relatively low basal rates of min secretion from in vitro cultures, cultures are generally radiolabeled using $^3$H-glucosamine as a metabolic precursor. The radiolabeled mucinsreleased are quantitated by precipitation with TCA/PTA. Using this cell culture system, we investigated mucin release of goblet cells by altering the media bathing the apical surface of hamster tracheal surface epithelial(HTSE) cells. Acidic media added sulfuric acid caused sigcificant increases in mucin relesse (155${\pm}$20% at pH 4 and 146${\pm}$16% at, pH 5). Ammonium hydroxide also increased mucin release at pH 9.0(156${\pm}$17%) and pH 10(295${\pm}$9%) respectively. This additional mucin release seems to be associated with cell membrane damage as indicated by release of cellular LDH. SP stimulates secretion of mucin in cultured HTSE cells(154${\pm}$16% at 1${\times}$10$\^$-6/M and 165${\pm}$25% at 1${\times}$10$\^$-5/M. PAF at 5${\times}$10$\^$-6/M and 5${\times}$10$\^$-5/M enhanced by HTSE cells in vitro 168${\pm}$34% and 259${\pm}$30% of mucin secretion, respectively. The increase in mucin release by PAF and SP was not secondary to cell damage or necrosis. SP and PAF may be in mediating mucous secretion induced by inflammation irritantion and infection.

  • In this study. we tried to investigate whether polymerized basic amino acid e.g. poly-L-lysine(PLL) which has the molecular weight under 10 kD significantly affects the physiological and stimulated mucin release from cultured hamster tracheal surface epithelial cells. Confluent primary hamster tracheal surface epithelial(HTSE) cells were metabolically radiolabeled with 3H-glucosamine for 24 hr and chased for 30 min in the presence of either PLLs or adenosine triphosphate(ATP) and PLL to assess the effects on basic or ATP-stimulated 3H-mucin release. 오소리에서 Uncinaria의 감염 증례 (Uncinaria infection in a Badger, A case report)

  • A male badger which showed depression and bloody diarrhea was submitted to Iksan Branch of Chonbuk Veterinary Service Laboratory for necropsy on May 1998. Grossly, paleness of the mucous membranes was observed and the small intestinal contents were hamorrhagic. The numerous tiny hookworms, sized in 5-10 mm and greyish-white in color, attached to the intestinal mucosa. Male bursa was well developed. Histologically, intestinal lumen was filled with hemorrhagic contents, which contained worms. The epithelial cells of the villi were underwent degeneration and lamina propria was infiltrated by lymphocytes and plasma cells, and goblet cells were hyperplastic. There were rounded cutting plates in the funnel-shaped buccal capsule and transverse striation on sheath in hookworm, ultrastructurally, which were consistent with Uncinaria sp. The shape of eggs were ellipsoidal and morula, and some eggs contained a mobile larva. It was concluded that this badger was infected with Uncinaria.

Effect of Yam Yogurt on Colon Mucosal Tissue of Rats with Loperamide-induced Constipation

  • The effects of lactic acid fermented yam yogurt (Yam/YG) on colon mucosal tissue were investigated in a loperamide-induced constipation rat models. Sprague-Dawley rats were fed for 6 weeks with 3 types of diets (normal, supplemented with lactic acid bacteria, and supplemented with Yam/YG), and were then administered loperamide intraperitoneally twice daily for 5 days. Administration of loperamide decreased fecal excretion and the moisture content of feces with increasing of numbers of pellets in the colon. On the histopathologic findings from hematoxylin and eosin (H& E) and alcian blue stainings, supplementation with Yam/YG resulted in the recovery of depleted goblet cells and mucin, and increased the numbers of Ki-67 positive cells, indicating restoration of colonic mucosa through cell proliferation and crypt regeneration against damages observed in crypt epithelial cells of loperamide-induced rats. These results indicate that Yam/YG improves evacuation and mucus production in the gastrointestinal tracts of constipated-induced rats.

닭 태자의 십이지장에 대한 조직학적 및 면역조직화학적 연구 (Histological and immunohistochemical studies on the duodenum of the chicken embryos)

  • With histological changes, ontogeny and relative frequencies of bovine Sp-1/chromogranin(bCG)-, serotonin-, gastrin-, cholecystokinin-8(CCK-8)-, somatostatin-, S-100 protein-, polypeptide YY(PYY)- and glucagon-immunoreactive cells were investigated in the duodenum of the chicken embryos from 10 days of incubation to hatching. Histologically, pseudostraitified columnar epithelium were observed from 10 days of incubation to 14 days of incubation, thereafter these epithelium were differentiated to simple columnar epithelium. $Liberk{\ddot{u}}hn$ glands were observed from 18 days of incubation and goblet cells were detected from hatching. In the duodenum, bCG-immunoreactive cells were detected from 14 days of incubation and increased to 18 days of incubation, thereafter decreased with ages. Serotonin-immunorecative cells were detected from 14 days of incubation and increased with ages. Somatostatin-immunoreactive cells were detected from 14 days of incubation and CCK-immunoreactive cells were detected from 19 days of incubation. No gastrin-, S-100 protein-, PYY-, glucagon-immunoreactive cells were detected in this study.

Possible Protective Effects of Quercetin and Sodium Gluconate Against Colon Cancer Induction by Dimethylhydrazine in Mice

  • Micronutrients in food have been found to have chemopreventive effects, supporting the conclusions from epidemiologie studies that consumption of fresh fruits and vegetables reduces cancer risk. The present study was carried out to evaluate the role of querctin (Q) and sodium gluconate (GNA) supplementation separately or in combination in ameliorating promotion of colon tumor development by dimethyl-hydrazine (DMH) in mice. Histopathological observation of colons in mice treated with DMH showed goblet cell dysplasia with inflammatory cell infiltration. This pathological finding was associated with significant alteration in oxidative stress markers in colon tissues and carcinoembryonic antigen (CEA) levels in plasma. Mice co-treated with GNA and Q showed mild changes of absorptive and goblet cells and inflammatory cell infiltration in lamina properia, with improvement in oxidative stress markers. In conclusion, findings of the present study indicate significant roles for reactive oxygen species (ROS) in pathogenesis of DMH-induced colon toxicity and initiation of colon cancer. Also, they suggest that Q, GNA or the combination of both have a positive beneficial effect against DMH induced colonic cancer induction in mice.

Effect of combined sodium hyaluronate and carboxymethyl cellulose on ocular surface in rat dry eye model

  • This study was conducted to evaluate three different mixed formulations of sodium hyaluronate (SH) and carboxymethyl cellulose (CMC) using a low-humidity air flow-induced rat dry eye model and determine the most suitable mixture. The total thickness of the cornea, corneal epithelial thickness, corneal stroma thickness, damaged corneal epithelium percentage region, thickness of the bulbar conjunctiva epithelium, number of goblet cells, goblet cell occupation percentage region, and damaged bulbar conjunctiva epithelium percentage region were measured by histomorphological evaluation. After 5 h exposure to drying airflow, the thickness of the cornea and conjunctiva was decreased with desquamation of the corneal and conjunctiva epithelium. However, these dry eye symptoms were markedly inhibited by treatment with the reference and test formulations. More favorable effects on decreased thickness were detected in response to the CMC than the SH. However, SH had a greater protective effect against corneal and conjunctiva epithelial damage. The application of a mixture of 0.1% SH and 0.2% CMC showed more favorable effects on the corneal and conjunctival damage and the stabilization of the ocular surface than SH or CMC alone.
