상품 사료에 첨가한 감귤발효액이 치어기 넙치, Paralichthys olivaceus의 성장에 미치는 영향 (Effect of EM-fermented Orange in Commercial Diet on Growth of Juvenile Flounder, Paralichthys olivaceus)

  • 제주산 온주 밀감을 EM (Effective Microorganism)에 의해 발효시킨 감귤발효액을 넙치 사료에 첨가하여 공급하였을 때 넙치의 성장 및 소화기관 활성 그리고 혈액조성에 미치는 영향을 분석하고, 감귤발효액이 넙치 사료의 사료첨가제로 이용가능성을 검토하였다. 감귤발효액을 사료량의 10.00%, 0.20%, 0.10% 그리고0.02% 첨가한 처리구와, 감귤발효액을 첨가하지 않고 사료만을 공급한 대조구로 하였다 성장실험에 있어서 전장은 감귤발효액 0.20%처리구에서 대조구보다 높았다(P<0.05). 체중 감귤발효액 0.20%와 0.10% 처리구에서 대조구보다 양호한 성장을 하였다(P<0.05). 총 증중량은 감귤발효액 0.20%와 0.10% 처리구에서 대조구보다 26.24∼29.35% 증중 효과가 있었다. 사료계수 그리고 일간먹이섭취율에서 감귤발효액 0.20% 와 0.10% 처리구가 대조구보다 향상된 결과를 나타내었다(P<0.05). 전장(anterior intestine)과 중장(mid intestine)의 점막 주름에 존재하는 배상세포(goblet cell)의 수는 처리구가 대조구보다 많았고, 전장 보다 중장으로 갈수록 많았다 (P<0.05). 혈장내 total cholesterol의 값은 감귤발효액 0.02%처리구에서 대조구보다 낮았다(P<0.05). 감귤발효액을 넙치사료에 첨가하였을 때, 넙치의 사료효율 증가와 장 활성으로 넙치의 성장과 생리활성에 보조하는 사료첨가제로서 이용가치가 높은 것으로 생각된다.

행소탕(杏蘇湯) 및 가미팔미환(加味八味丸)이 호흡기(呼吸器) 배상세포(杯狀細胞)로부터의 뮤신 분비(分泌)에 미치는 영향 (Effects of HaengSoTang(HST), Gami-PalMiHwan(GPMH) on mucin secretion from airway goblet cells)

  • The intent of this study is to investigate whether two oriental medical prescriptions named haengsotang(HST) and gami-palmihwan(GPMH) significantly effect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells, Confluent HTSE cells were metabolically radiolabeled with $^3H-glucosamine$ for 24 hrs and chased for 30 min in the presence of HST or GPMH to assess the effect of each agent on $^3H-mucin$ release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of HST and GPMH on contractility of isolated tracheal smooth muscle were investigated. The results are consistant with the following assertions: (1) HST significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) GPMH did not effect mucin release without cytotoxicity; (3) HST and GPMH did not effect contractility of isolated tracheal smooth muscle. These results suggest a need for further investigation of HST and its components, for its potential in oriental medicine prescriptions and novel agents that effectively regulate (inhibit) mucin secretion from airway goblet cells.

The Bacillus subtilis and Lactic Acid Bacteria Probiotics Influences Intestinal Mucin Gene Expression, Histomorphology and Growth Performance in Broilers

  • The aim of the present study was to evaluate the effect of commercial monostrain and multistrain probiotics in diets on growth performance, intestinal morphology and mucin gene (MUC2) expression in broiler chicks. Three hundred seventy-eight 1-d-old male Arian broiler chicks were allocated in 3 experimental groups for 6 wk. The birds were fed on a corn-soybean based diet and depending on the addition were labeled as follows: control-unsupplemented (C), birds supplemented with Bacillus subtilis (BS) and lactic acid bacteria (LAB) based probiotics. Each treatment had 6 replicates of 21 broilers each. Treatment effects on body weight, feed intake, feed conversion ratio and biomarkers such as intestinal goblet cell density, villus length, villus width, and mucin gene expression were determined. Total feed intake did not differ significantly between control birds and those fed a diet with probiotics (p>0.05). However, significant differences in growth performance were found. Final body weight at 42 d of age was higher in birds fed a diet with probiotics compared to those fed a diet without probiotic (p<0.05). Inclusion of Bacillus subtilis based probiotic in the diets also significantly affected feed conversion rate (FCR) compared with control birds (p<0.05). No differences in growth performance were observed in birds fed different types of probiotic supplemented diets. Inclusion of lactic acid bacteria based probiotic in the diets significantly increased goblet cell number and villus length (p<0.05). Furthermore, diets with Bacillus subtilis based probiotics significantly increased gene expression (p<0.05), with higher intestinal MUC2 mRNA in birds fed diet with probiotics compared to those fed the control diet. In BS and LAB probiotic fed chicks, higher growth performance may be related to higher expression of the MUC2 gene in goblet cells and/or morphological change of small intestinal tract. The higher synthesis of the mucin gene after probiotic administration may positively affect bacterial interactions in the intestinal digestive tract, intestinal mucosal cell proliferation and consequently efficient nutrient absorption.

능성어 (Epinephelus septemfasciatus) 자어의 소화기관 구조 및 조직학적 특징 (The Structure of Digestive Tract and Histological Features of the Larvae in Sevenband Grouper, Epinephelus septemfasciatus)

  • 이 연구는 능성어 (Epinephelus septemfasciatus)의 소화관 발달과정을 형태학적과 조직학적인 방법으로 부화 후부터 60일 동안 관찰하였다. 먹이 급이는 부화 후 2일부터 20일 동안 Rotifer (Brachionus rotundiformis)와 클로렐라 (Chlorella ellipsoidea)를 급이 하였고, 20일부터 Rotifer와 brine shrimp (Artemia salina)를 급이 하였으며, 23일부터 Rotifer와 Artemia, 배합사료를 급이 하였다. 자치어 10마리를 임의로 선택하여 10% 중성포르말린에 고정한 후 형태학적 관찰과 파라핀포매법에 의한 조직학적인 관찰을 하였다. 능성어의 RLG는 평균 0.87으로 육식성어류의 특성을 보여주었다. 부화 직후의 자어는 입과 항문은 열려있지 않았고, 소화관은 난황을 따라 일직선상으로 관찰되었다. 부화 후 5일째 후기 자어는 입과 항문이 열리면서 첫 섭식 활동이 관찰되었고, 부화 후 8일째 변태가 시작되었다. 부화 후 11일째 후기 자어 식도는 네 개의 층이 구분 되었으며, 식도와 중장, 직장에서는 배상세포가 관찰되었다. 부화 후 19일째 후기 자어는 공식이 시작되었으며, 그로인해 개체의 성장차이가 관찰되었다. 위는 내부가 팽창되면서 분문위, 위체부 및 유문위의 구분이 가능해졌고, 유문수가 분화되었으며, 장 전체에서는 배상세포를 관찰할 수 있었다. 부화 후 28일째 위내부에 위선이 분화되었다. 부화 후 38일째 후기 자어는 위에서 위액이 분비되는 것을 확인할 수 있었고, 유문수에 점막주름이 확인되었다. 부화 후 38일째에서 치어기로 이행하는 시기 동안 위 내강은 넓어지고, 위선의 숫자와 장내점액세포, 점막주름은 지속적으로 증가하거나 길어졌으나 조직화학적인 변화는 관찰되지 않았다. 따라서 본 연구는 형태학적 및 조직학적 방법을 이용하여 능성어의 소화관 분화 및 발달에 관한 정보를 얻고자 하였으며, 초기 종묘생산과정에서 적절한 먹이투여시기를 확립함으로써 성장 및 생존율 향상에 기초자료를 제공하고자 하였다.

서울주걱흡충 감염 BALB/c 및 C3H 마우스에서 장점막 비만세포 및 배세포의 증식 (Intestinal mastocytosis and goblet cell hyperplasia in BALB/e and C3H mice infected with Neodiplostomum seoulLense)

  • 서울주걱흡충 (Weodiplostomum seoulense)에 감염된 BALB/c 및 C3H 마우스의 소장에서 비만 세포 및 배세포 반응 양상을 관찰하고, 이들 반응이 숙주 방어기전 및 충체 배출에 영향을 줄 수 있는지 알아보았다. 마우스 1마리당 피낭유충 200개씩을 감염시킨 후 28일까지 충체회수율을 관 찰한 바 BALB/c 마우스가 C3H 마우스에 비해 월등히 높은 충체회수율을 보였다 그러나, 마우스 주에 관계없이 충체의 주 기생부위인 십이지장에서 감염 7일에 비만세포수가 최고에 달하였고 곧 바로 감소하였다. 이러한 비만세포수의 반응 양상은 공장 및 회장에서도 비슷하게 나타났으며, 반응의 강도는 십이지장이나 공장이 회장에 비해 높았다. 또한, 비만세포수의 최고치는 BALB/c 마우스에서 C3H 마우스보다 오히려 높게 나타났다. 배세포 증식은 BALB/c 마우스의 경우 감염 14 일을 제외하고는 십이지장에서 뚜렷이 관찰되었고, C3H 마우스의 경우에는 감염 7일에 공장 및 회장에서 관찰되었다. 배세포의 점액 (mucin) 활성은 BAkBlc 및 C3H 마우스 모두에서 인정되었으나, 그 강도는 BALBlc 마우스에서 더욱 뚜렷하였다. 이상의 결과를 종합할 때, 이들 마우스의 장점막 비만세포 및 배세포 반응은 서울주걱흡충 감염에 대한 국소적인 면역반응으로 나타나지만 숙주 방어나 충체 배출에의 관여도는 낮을 것으로 추측되었다.

백서 기관 점막의$SO_2$ 노출 후 회복과정의 조직학적 관찰 및 laminin의 발현에 관한 연구 (The Histologic Findings and the Expression of Laminin in the Mucosa of the Rat Trachea During)

  • Background and Objectives : Sulfur dioxide gas is one of the major airborne Pollutants noxious to human in industrialized countries. The most vulnerable areas in the human respiratory system were the trachea and main bronchi and a gradient of decreasing damage was observed in the peripheral tracheobronchial tree. Induced functional alteration was increased mucosal permeability, and morphological changes were epithelial sloughing, intracellular edema, mitochondrial swelling, widened intercellular spaces, and ciliary cytoplamic extrusions. The laminins are a family of extracellular matrix glycoproteins localized in the basement membrane. Their primary role is cell-matrix attachment, but many additional biologic activities, including Promoting cell growth and migration, tumor growth and metastasis, wound repair, and graft survival, have been demonstrated. Materials and Methods : Histologic changes and expression of laminin in tracheal mucosa sacrificed at 1 day, 2 day, 3 day, 1 week, 2 weeks, and 3 weeks after continued SO2 exposure of 250 ppm for 30 minutes a day(to 7week) were studied in rats. In this study, mild immune reaction for laminin was noted at the apical cytoplasm of epithelial cells and basement membrane one day after a 7 week $SO_2$ exposure. The cilia and nucleoi of epithelial cells were normal and no immune reaction was noted in Goblet cells. The lamina propria of the tracheal tissue was infiltrated by monocytes and lymphocytes. Results : At 24 hours after exposure, all tracheal cells except Goblet cells revealed a mild immune reaction for laminin. No immune reactions were noted in the basement membrane. At 72 hours after exposure, mild or moderate immune reactions for laminin was seen in the tracheal cell cytoplasm. Irregular faint immune reaction for laminin was noted in the basement membrane. At 1 week after exposure, strong immune reaction for laminin was detected over all tracheal cells, and the basement membrane was seen clearly. At 2~3 weeks after exposure, strong immune reaction for laminin was seen in all tracheal epithelial cells except Goblet cells and a mild immune reaction was partly revealed in the basement membrane. Conclusion : Our study suggests that 502 produces histologic damage on the tracheal mucosa. Longer duration after exposure of $SO_2$ makes more progressive healing on the tracheal mucosa and increased immunoreactivity for laminin.

사삼맥문동탕이 Indomethacin으로 유발된 mouse의 대장 점막 손상에 미치는 영향 (The Effects of Sasammaickmoondong-tang against Colonic Mucosal Lesions)

  • Objectives: This study was carried out to investigate 1he effects of Sasammaickmoondong-tang(SME) on colonic mucosal lesions induced by indomethacin in mouse. Methods: The normal group is 1hat no inflammation elicitated mouse. Control group is that gastro-inflammation elicitated mouse. Sample group is that SME administered mouse after gastro-inflammation elicitation. Results: In the common morphology and histochemical change, control group was observed various injury-mucous surface cell, micro-villi, paneth cell, surface epithelial cell, goblet cell-by hemorrhagic erosion, while sample group was as same as normal group. In the immunohistochemical change, 1he distributions of COX-1, Bcl-2, and BrdU treated with SME noticeably increased than control group(P<0.05). The distributions of TUNEL, $NF-{\kappa}B$, COX-2, $IL-2R-\alpha$, NK-1.1, ICAM-1, and CD11b/18 treated with SME noticeably decreased than control group(P<0.05). And the distribution of SBA was as same as normal group. Conclusions: According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to colonic mucosal lesions.

Mouse의 위점막 염증에 대한 사삼맥문동탕의 면역반응 연구 (The Effects of Sasammaickmoondong-tang against Gastric Mucosal Lesions)

Conclusions: According to the above results, it is supposed that Sasammaickmoondong-tang is applicable to gastric mucosal lesions.

  • The tentacular epithelial cells of Cipangopaludina chinensis were studied with electron microscope. The tentacular epithelium consists of columnar supporting cells, numerous glandular cells and ciliated cells. Glandular cells are classified into three types; type I (mucous metachromasia cell), type II(mucous goblet cell) and type III. Ciliated cells are subdivided into two types; type I ciliated cell has cilia with typical axoneme(9+2), and type II ciliated cell has cilia with unusual axoneme.

The Role of Proprotein Convertases in Upper Airway Remodeling

  • Chronic rhinosinusitis (CRS) is a multifactorial, heterogeneous disease characterized by persistent inflammation of the sinonasal mucosa and tissue remodeling, which can include basal/progenitor cell hyperplasia, goblet cell hyperplasia, squamous cell metaplasia, loss or dysfunction of ciliated cells, and increased matrix deposition. Repeated injuries can stimulate airway epithelial cells to produce inflammatory mediators that activate epithelial cells, immune cells, or the epithelial-mesenchymal trophic unit. This persistent inflammation can consequently induce aberrant tissue remodeling. However, the molecular mechanisms driving disease within the different molecular CRS subtypes remain inadequately characterized. Numerous secreted and cell surface proteins relevant to airway inflammation and remodeling are initially synthesized as inactive precursor proteins, including growth/differentiation factors and their associated receptors, enzymes, adhesion molecules, neuropeptides, and peptide hormones. Therefore, these precursor proteins require post-translational cleavage by proprotein convertases (PCs) to become fully functional. In this review, we summarize the roles of PCs in CRS-associated tissue remodeling and discuss the therapeutic potential of targeting PCs for CRS treatment.
