A Study for Clinical Efficacy of GnRH Antagonist (Cetrorelix) Minimal Stimulation Protocol in Assisted Reproductive Techniques for Polycystic Ovaian Syndrome (다낭성 난소증후군의 과배란유도시 GnRH Antagonist (Cetrorelix)를 병합한 Minimal Stimulation Protocol의 임상적 유용성에 관한 연구)

  • Objective : The aim of this study was to evaluate the outcomes of the GnRH antagonist (Cetrotide) minimal stimulation protocol comparing with GnRH agonist combined long step down stimulation protocol in PCOS patients. Materials and Method: From Apr 2001 to May 2002, 22 patients (22 cycles) were performed in controlled ovarian hyperstimulation using by GnRH antagonist and GnRH agonist for PCOS patients. GnRH antagonist (Cetrotide) combined minimal stimulation protocol was administered in 10 patients (10 cycles, Study Group) and GnRH agonist long step down stimulation protocol was administered in 12 patients (12 cycles, Control Group). We compared the pregnancy rate/cycle, total FSH (A)/cycle, Retrieved oocyte/cycle, the incidence of ovarian hyperstimulation syndrome, multiple pregnancy rate between the two groups. Student-t test were used to determine statistical significance. Statistical significance was defined as p<0.05. Results: Group of GnRH antagonist (Cetrorelix) minimal stimulation protocol produced fewer oocytes (6.4 versus 16.3 oocytes/cycle) using a lower dose of FSH (22.2 versus 36.1 Ample/cycle) and none developed OHSS and multiple pregnancy. Although the trends were in favour of the GnRH antagonist (Cetrorelix) protocol, the differences did not reach statistical significance. This was probably due to small sample size. Conclusion: The use of GnRH antagonist reduce the risk of ovarian hyperstimulation and multiple pregnancy. We suggest that GnRH antagonist might be alternative controlled ovarian hyperstimulation method, especially in PCOS patients who will be ovarian high response.

Catecholaminergic Innervation of GnRH Neurons in the Rat Median Eminence (흰쥐 시상하부의 정중융기에서 카테콜아민 신경세포와 GnRH 신경세포와의 연접에 관한 연구)

  • The present study was carried out 1) to show the ontogenic development of CA-and GnRH-containing nerve fobres in the median eminence, 2) to simultaneously demonstrate the synaptic contact between these two nenre fibres in the rat median eminence at the ultrastructural level using light and electron microscopic doublelabel immunostainlngs. GnRH-and CA-nenre terminals were detectable in the median eminence at embryonic day 19.5. The CA-newe terminals were obsenred in the entire legion of the extern31 lavers, while GnRH-newe terminals only in the lateral portion. At the 14th postnatal daw, both %ropes of nerve terminals showed a very similar distribution to those of adult one. In the median eminence of adult rats, a substantial overlap existed in the distribution of GnRH fibres with CA-containing nerve fibres. This overlap was most intense throughout the external palisade zone. Furthermore, an electron microscopic double label immunostaining showed that there was a close apposition of CA- and GnRH-nenre fobres. These axo-axonic contacts occurred frequently in the internal and palisade zones, i.e. at the level of the fobre preterminals. These morphological results suggest that the CA-mediated GnRH secretion may occur via sxo-axonic interaction in the median eminence.

β3GnT8 Regulates Laryngeal Carcinoma Cell Proliferation Via Targeting MMPs/TIMPs and TGF-β1

  • Previous evidence showed ${\beta}1$, 3-N-acetylglucosaminyltransferase 8 (${\beta}3GnT8$), which can extend polylactosamine on N-glycans, to be highly expressed in some cancer cell lines and tissues, indicating roles in tumorigenesis. However, so far, the function of ${\beta}3GnT8$ in laryngeal carcinoma has not been characterized. To test any contribution, Hep-2 cells were stably transfected with sense or interference vectors to establish cell lines that overexpressed or were deficient in ${\beta}3GnT8$. Here we showed that cell proliferation was increased in ${\beta}3GnT8$ overexpressed cells but decreased in ${\beta}3GnT8$ knockdown cells using MTT. Furthermore, we demonstrated that change in ${\beta}3GnT8$ expression had significant effects on tumor growth in nude mice.We further provided data suggesting that overexpression of ${\beta}3GnT8$ enhanced the expression of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) at both the mRNA and protein levels, associated with shedding of tissue inhibitors of metalloproteinase TIMP-2. In addition, it caused increased production of transforming growth factor beta 1 (TGF-${\beta}1$), whereas ${\beta}3GnT8$ gene knockdown caused the reverse effect. The results may indicate a novel mechanism by which effects of ${\beta}3GnT8$ in regulating cellular proliferation are mediated, at least in partvia targeting MMPs/TIMPs and TGF-${\beta}1$ in laryngeal carcinoma Hep-2 cells. The finding may lay a foundation for further investigations into the ${\beta}3GnT8$ as a potential target for therapy of laryngeal carcinoma.

  • Objective: This study was performed to compare the clinical outcomes of GnRH antagonist (Cetrorelix) single dose and multiple dose protocols for controlled ovarian hyperstimulation with GnRH agonist long protocol. Materials and Method: From September 2001 to March 2002, 48 patients (55 cycles) were performed controlled ovarian hyperstimulation for ART using by either GnRH antagonist and GnRH agonist. Single dose of 3 mg GnRH antagonist was administered in 15 patients (17 cycles, single dose group) at MCD #8 and multiple dose of 0.25 mg of GnRH antagonist was administered in 15 patients (18 cycles, multiple dose group) from MCD #7 to hCG injection day. GnRH agonist was administered in 18 patients (20 cycles, control group) by conventional GnRH agonist long protocol. We compared the implantation rate, number of embryos, and clinical pregnancy rate among three groups. Student-t test and Chi-square were used to determine statistical significance. Statistical significance was defined as p<0.05. Results: There were no significant differences in ampules of used gonadotropins, number of mature oocytes, obtained embryos between single and multiple dose group, but compared with control group, ampules of used gonadotropins, number of mature oocytes, obtained embryos were decreased significantly in both groups. Clinical pregnancy rate and implantation rate were not different in three groups. There were no premature LH surge and ovarian hyperstimulation syndrome in three groups. Multiple pregnancy were occurred 1 case in multiple dose group and 2 case in control group. Conclusions: GnRH antagonist is a safe, effective, and alternative method in the controlled ovarian hyperstimulation compared with GnRH agonist. Clinical outcomes and efficacy of both single and multiple dose protocol are similar between two groups.

Hypophyseal and Gonadal Response to GnRH in Buffalo Heifers (Bubalus bubalis)

  • The objective of this study was to investigate the responsiveness of hypophysis and gonads to synthetic GnRH among noncycling Murrah buffalo heifers at 24 months of age. The plasma FSH, LH, estradiol and progesterone levels were measured in blood samples collected at 1 hour before and upto 18th day subsequent to the administration of GnRH ($(200 {\mu}g)$) or saline (2 ml). The pretreatment levels of plasma FSH, LH estradiol and progesterone among GnRH treated heifers (N = 6) were $11.55{\pm}0.57ng/ml$, $0.68{\pm}0.06ng/ml$, $19.84{\pm}0.82pg/ml$ and $0.45{\pm}0.07ng/ml$ respectively. A quick elevation of FSH (p < 0.01) and LH (p < 0.05) within 5 min of GnRH administration was observed in all the heifers. The peak FSH ($74.97{\pm}18.63ng/ml$) and LH ($3.09{\pm}0.54ng/ml$) level was obtained at 30 min of GnRH administration. The elevated level of plasma estradiol on 5th to 18th day, FSH on 7th to 9th day (n = 3) and the progesterone on 13th to 18th day (n = 2) of GnRH injection was obtained. The study indicates that gonads of buffalo heifers at 24 months of age are responsive of GnRH induced gonadotropin release for folliculogenesis and luteal tissue formation

Flexural properties, interlaminar shear strength and morphology of phenolic matrix composites reinforced with xGnP-coated carbon fibers

  • In the present study, exfoliated graphite nanoplatelets (xGnP) with different particle sizes were coated onto polyacrylonitrile-based carbon fibers by a direct coating method. The flexural properties, interlaminar shear strength, and the morphology of the xGnP-coated carbon fiber/phenolic matrix composites were investigated in terms of their longitudinal flexural strength and modulus, interlaminar shear strength, and by optical and scanning electron microscopic observations. The results were compared with a phenolic matrix composite counterpart prepared without xGnP. The flexural properties and interlaminar shear strength of the xGnP-coated carbon fiber/phenolic matrix composites were found to be higher than those of the uncoated composite. The flexural and interlaminar shear strengths were affected by the particle size of the xGnP, while the particle size had no significant effect on the flexural modulus. It seems that the interfacial contacts between the xGnP-coated carbon fibers and the phenolic matrix play a role in enhancing the flexural strength as well as the interlaminar shear strength of the composites.

Effect of Gonadotropin on the Expression of GnRH and GnRH mRNA in Rat Ovary (성선자극호르몬이 흰쥐 난소의 GnRH와 GnRH mRNA의 발현에 미치는 영향)

  • Expression of gonadotropin releasing hormone(GnRH) has been described in the rat ovary. It remains, however, unkown whether GnRH is synthesized as a prohormone. Therefore, this study was performed to verify the expression of pro-GnRH by in situ hybridization and further to investigate the effect of gonadotropin on GnRH or GnRH mRNA in rat ovary by immunohistochemical and in situ hybridization techniques. Adult female Sprague-Dawely rats were used and the estrous cycle was synchronized by intraperitoneal injection of pregnant mare's serum gonadotropin(PMSG). Ovaries were fixed with 4% paraformaldehyde and embedded with G.C.T. compound and cut by cryostat. For immunohistochemistry, avidin-biotin peroxidase complex(ABS) method was employed and for in situ hybridization, $^{35}S$-end labeled oligonucleotide was used and followed by autoradiography. By in situ hybridization using GnRH oligomer and GAP(GnRH associated protein) oligomer, GnRH mRNA and GAP mRNA were co-localized in the fullicular cells, luteal cells, interstitial cells and theca cells. GnRH or GnRH mRNA signals in the ovary increased by human chorionic gonadotropin(hCG) injection. At the 3 and 6 hrs after hCG injection, the number of GnRH and GnRH mRNA containing cells increased rapidly and the density of GnRH and GnRH mRHA culminated at 9 hrs after heG injection. With the follicular development, the high expression of GnRH and GnRH mRNA was also observed within the follicles. After ovulation, the density of GnRH or GnRH mRNA decreased in the follicles but increased in the corpus lutea.

  • Dendritic carbosilanes based on hyperbranched polycarbosilanes as core molecule have been prepared The core molecules were obtained by the use of hydrosilation of $HSiMe_{3-n}$$(CH_2CH=CH_2)_n$(n=2; $AB_2$,3;$AB_3$type). The hyperbranched core $AB_2\;and\; AB_3$ type polymers were generated to higher molecular dendritic carbosilanes Gn+1 by the use of hydrosilation and alkenylation sequence. The Gn+2P generations were not obtained as unified molecules by the use of hydrosilation with $HSiMeCl_2$. Gn and Gn+1 type polymers were produced to polysilol by the reaction of 9-BBN and alkali medium oxidation of hydroborated compounds. The degree for reaction has been controlled by the NMR spectroscopy.

Evaluation of lines of NERICA 1 introgressed with Gn1a and WFP for yield and yield components as affected by nitrogen fertilization in Kenya

  • In many sub-Saharan African countries, boosting rice production is a pressing food security issue. To contribute to the increase in rice production, we have developed lines of NERICA 1 introgressed with the gene for spikelet number, Gn1a, and the gene for primary rachis-branch number, WFP by cross breeding. The performance of rice lines introgressed with the genes for yield related traits can be affected by cultivation environment and management. Thus, in this study, we aimed to evaluate the lines of NERICA 1 introgressed with Gn1a or/and WFP for yield and yield components under different nitrogen fertilization conditions in Kenya. A field trial was conducted at a paddy field in Kenya Agricultural and Livestock Research Organization-Mwea, Kirinyaga County ($0^{\circ}39^{\prime}S$, $0^{\circ}20^{\prime}E$) from August 2016 to January 2017. Eight lines of NERICA 1 introgressed with Gn1a and/or WFP, and their parents, NERICA 1 and ST12, were grown under 0 (NF) and $75(SF)kg\;N\;ha^{-1}$. At maturity, five hills per plot were harvested to determine the yield and yield components. The number of primary and secondary rachis-branches per panicle was measured on the longest panicle in each hill. Under SF, the introgression of WFP to NERICA 1 increased the number of primary and secondary rachis-branches by 27 and 25%, respectively. On the other hand, Gn1a did not increase the number of primary rachis-branches, whereas the number of secondary rachis-branches was increased by 38% on average. The number of primary and secondary rachis-branches of the lines introgressed with both genes increased by 25 and 56%, respectively. Although grain number per panicle increased 33% by Gn1a, 34% by WFP, and 43% by Gn1a+WFP, the yield increase by Gn1a, WFP, and Gn1a+WFP was only 14, 7, and 14%, respectively. The suppression of the yield increase was mainly attributed to the decline in the filled grain ratio. Under NF, WFP increased the number of primary and secondary rachis-branches by 20 and 19%, respectively. The introgression of both genes increased the former and the later by 19 and 35%, respectively. However, Gn1a did not change them under NF. Thus, even under NF, grain yield increased 11% by WFP and 24% by Gn1a+WFP due to the increased grain number although filled grain ratio declined. Our findings suggest that the introgression of Gn1a and WFP could contribute to the rice productivity improvement in sub-Saharan Africa even under low fertility conditions. Improving filled grain ratio of the lines introgressed with these genes by further breeding and fertilization management will be the focus of subsequent work.

  • Objective: To investigate new signal transduction cascade through integrin, FAK and ERK in the suppressed cell proliferation by GnRH-I and -II. Method: Human endometrial cancer cells (HEC1A) were cultured under the following condition: DMEM/F12 (10% FBS). GnRH-I and -II were treated time (0, 5, 10, 15, 20, 30 min; 100 nM) and dose (10 nM or 100 nM; 20 min) dependent manner according to experimental purposes. Cell proliferation was measured using [$^3H$] thymidine incorporation assay. Immunoblotting was utilized to detect proteins. Results: GnRH-I and -II inhibited proliferation of HEC1A cells and induced expression of integrin ${\beta}3$. Phosphorylation of FAK and ERK were induced by GnRH-I and -II. Conclusion: GnRH inhibited cell proliferation via the expression of integrin and FAK, ERK phosphorylation.
