The aim of this study was to study the quantitative analysis of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Paecilomyces japonica, Ganoderma lucidum, honey or Nuruk. The amounts of dry on loss were measured and the quantitative analysis of glycyrrhizic acid was performed by high performance liquid chromatographic (HPLC). HPLC method was performed on C18 column ($250\;mm\;{\times}\;4.6\;mm$, $5\;{\mu}m$, RS tech) using gradient solvent mixtures of water-acetonitrile with photodiode array detector (254 nm). The flow rate was $1.0\;m{\ell}/min$. Retention time of glycyrrhizic acid was about 23.96 min and linearity of calibration was $R^2$=0.9998. Contents of glycyrrhizic acid in Glycyrrhizae Radix extract (control) was $5.048\;{\pm}\;0.14$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Paecilomyces japonica (SDT) was $1.975\;{\pm}\;0.07$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Ganoderma lucidum (SYT) was $2.676 \;{\pm}\;0.07$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with honey (SST) was $5.191\;{\pm}\;0.06$; Contents of glycyrrhizic acid in Glycyrrhizae Radix extract fermented with Nuruk (SNT) was $5.305\;{\pm}\;0.34$, respectively. Contents of glycyrrhizic acid in SDT and SYT were decreased but that in SST and SNT was increased when compared to control.
The Pyrolytic behavior of glycyrrhizic acid and glycyrrhetini c acid, which are natural flavorants for manufactured cigarettes was observed to find its contribution to the smoke composition. Pyrolyzates of them at 800t were identified using a gas chromatography and a mass spectrometer. According to the analysis of the pyrolytic Products, 43 different compounds were identified Among them the aromatic hydrocarbon compounds were found to be the major products.
The extraction of glycyrrhizic acid from licorice using supercritical carbon dioxide (SCCO$_2$) was investigated with respect to the effects of extraction parameters such as the kind and amount of modifier, temperature, pressure, and extraction time. The conventional organic solvent extraction was also conducted for a quantitative comparison. The content of glycyrrhizic acid in crude extracts was analyzed by HPLC and the yield of glycyrrhizic acid was computed as a weight percent recovery. The optimal pressure and temperature for SCCO$_2$ extraction were found to be 40 MPa and 80$^{\circ}C$, respectively, when SCCO$_2$ was modified with 70% aqueous ethanol. Under the same pressure and temperature, the highest recovery was attained to be 104.57% in the first 60 min when the concentration of 60%, aqueous ethanol in SCCO$_2$ was 15%.

This study investigated the glycyrrhizic acid content and anti-diabetic activities of Glycyrrhiza uralensis (GU) depending on the cultivation region (Jecheon, Youngju, Gokseong, China, and Uzbekistan). Glycyrrhizic acid accuracy recovery and intra- and inter-day precisions (RSD%) of the method were calculated at 99.10~107.07% and 3.92 and 6.31% for GU samples, respectively, whereas the limits of detection and quantitation were 0.14 and $0.20{\mu}g/mL$. Anti-diabetic activity was measured by ${\alpha}-glucosidase$ and glucose uptake. GU (20 g) was extracted with 70% ethanol at $70^{\circ}C$ for 6 h. The Jecheon and Gokseong GU showed good inhibitory activity compared to the control. The Jecheon, Youngju, and Uzbekistan GU ethanol extracts ($100{\mu}g/mL$) showed glucose uptakes (in $C_2C_{12}$ myotube) of 124.19, 127.18, and 126.92%, respectively, compared to the positive control. In conclusion, these methods were validated for detection of glycyrrhizic acid in GU, and the results indicate that GU might have potential anti-diabetic activities.
This study aims to elucidate the effect of glycyrrhizic acid on smooth muscle contraction and to determine the detailed mechanism incorporated. We hypothesized that glycyrrhizic acid played a role in the agonist-sensitive management of smooth muscle contraction. Stripped smooth muscles of Sprague-Dawley rats were prepared in organ baths and isometric tensions were converted, stored and analyzed by using isometric transducers, a physiograph and one way ANOVA. Interestingly, glycyrrhizic acid attenuated the thick filament regulating agonist (fluoride or thromboxane mimetic)-sensitive contraction (p=0.113, 0.008, 0.004 (Student's t-test), p=0.113, 0.008, 0.004 (One way ANOVA) at 0.01, 0.03, 0.1 mM fluoride, and p=0.156, 0.004, 0.003 (Student's t-test), p=0.156, 0.004, 0.003 (One way ANOVA) at 0.01, 0.03, 0.1 mM thromboxane mimetic) and did not attenuate the thin filament regulating agonist (phorbol ester)-induced contraction (p=0.392, 0.086, 0.065 (Student's t-test), p=0.392, 0.086, 0.065 (One way ANOVA) at 0.01, 0.03, 0.1 mM phorbol ester). It is suggesting that endothelial EDRF (NO) synthesis and accessory pathways besides endothelial EDRF (NO) synthesis such as ROCK restriction might be incorporated in the glycyrrhizic acid-induced modulation of smooth muscle contraction inhibiting acto-myosin interaction.

Here we present how a supramolecular complex of Glycyrrhizic acid (GA) with Menthol (Mt) affects the blood glucose levels and glycogen synthesis in the liver of rats in in-vivo experiments with diabetes caused by alloxan. We have shown that Menthol, Glycyrrhizic acid and GA:Mt supramolecular complexes can restore functional dysfunction of the liver mitochondria in alloxan diabetes, i.e., inhibit lipid peroxidation. The hypoglycemic activity and mitochondrial membrane stabilizing properties of the supramolecular complex GA:Mt (4:1) in alloxan diabetes were more pronounced than those of menthol, GA and its GA:Mt (2:1) and GA: Mt (9:1) supramolecular complexes.
A high performance liquid chromatographic (HPLC) method for the determination of glycyrrhizic acid was developed for the quality control of traditional herbal medicinal preparation Bu-Zhong-Yi-Qi-Tang (BZYQT), which is well-known herbal medicine used as tonic. RP-HPLC analysis was carried out using Capcell pak $C_{18}$ MG column $(5\;{\mu},\;150{\times}4.6\;mm)$ and a mobile phase consisting of acetonitrile and water containing 0.03% phosphoric acid (pH 2.46) at a flow rate of 1.0 ml/min. The optimum wavelength for the detection of the glycyrrhizic acid was found at 250 nm using diode-array UV/VIS detector. The glycyrrhizic acid in BZYQT shows good linearity $(r^2>0.999)$ in the range of $15\;{\mu}g/ml$ to 500 ${\mu}g/ml$. The limit of detection (LOD) was less than 5 ng and R.S.D for intra-day and inter-day reproducibility was less than 7%. The mean recovery of the glycyrrhizic acid was $97.3{\sim}113.0%$. These results suggest that the developed HPLC method is simple and efficient, and could be contributed for the quality control of commercial BZYQT products.
Reversed-phase high performance liquid chromatography (RP-HPLC) was used for the simultaneous determination of liquiritin (LQ), glycyrrhizic acid (GA) and glabridin from licorice. An optimized run condition was selected with a binary gradient elution of methanol-water which ramped 35/65 to 80/20 (vol. %) in 0.0-8.0 min and a flow rate of 1.0 mL/min. A good linearity was obtained between 0.2 mg/mL and 1.0 mg/mL for LQ and GA, and 0.01 mg/mL-0.2 mg/mL for glabridin with the relative standard deviations less than 0.90% (n=5). The developed method was successfully applied to determination of the three components from licorice samples. The mean recoveries of three components are 80.79% for liquiritin, 89.71% for glycyrrhizic acid and 72.50% for glabridin.
Menthol (M) is a cyclic monoterpenode and is a major component of essential oils. Menthol, along with menthol, isomenton, etc., gives taste and odor of the mint plant, and when it comes to menthol in general, L- or (-) -menthol is usually used. Included in pharmaceuticals, cosmetics and pesticides. It has antimicrobial, antibacterial, antioxidant properties. It is also known that the licorice plant (Glycyrrhiza Glabra L.) differs from other types of plants by its medicinal properties. For many years it has been used in folk medicine. Extraction of licorice root revealed up to 25% glycyrrhizinic acid (GA). Its aglycone - glycyrrhizic acid is notable for its structural similarity to the adrenal cortex hormones. Currently, GA and glycyrrhizic acid are widely used in medicine as a remedy for colds, allergies, viral diseases, tumors. The biological activity of menthol and GA-based supramolecular compounds has been poorly studied, and their effect on the functional parameters of rat liver mitochondria has been studied little. For this purpose, in our experiments, the effect of menthol (M), glycyrrhizinic acid (GA) and their supramolecular complexes obtained in different proportions on in vitro and in vivo studies on rat liver mitochondria was studied.

Background: The present study optimized ultrasound-assisted extraction conditions to maximize extraction yields of glycyrrhizic acid from licorice. Methods: The optimal extraction temperature ($X_1$), extraction time ($X_2$), and methanol concentration ($X_3$) were identified using response surface methodology (RSM). A central composite design (CCD) was used for experimental design and analysis of the results to obtain the optimal processing parameters. Results: Statistical analyses revealed that three variables and the quadratic of $X_1$, $X_2$, and $X_3$ had significant effects on the yields and were followed by significant interaction effects between the variables of $X_2$ and $X_3$ (p<0.01). A 3D response surface plot and contour plots derived from the mathematical models were applied to determine the optimal conditions. The optimum ultrasound-assisted extraction conditions were as follows: extraction temperature, $69^{\circ}C$; extraction time, 34?min; and methanol concentration, 57%. Under these conditions, the experimental yield of glycyrrhizic acid was 3.414%, which agreed closely with the predicted value (3.406%). Conclusion: The experimental values agreed with those predicted by RSM models, thus indicating the suitability of the model employed and the success of RSM in optimizing the extraction conditions.
