  • Objectives : The study was designed to observe the effect of Polygalae Radix Preparata Cum Glycyrrhizae Radix on 4-Hydroxynonenal (4-HNE)-induced apoptosis in PC-12 cell. Methods : A cytotoxic test on Polygalae Radix Preparata Cum Glycyrrhizae Radix (PG) was conducted and another MTT assay was conducted to observe the cytoprotective effect against 4-HNE that cause oxidative stress. In addition, in order to observe the expression of Bax, Bcl-2, Caspase-3 and TNF-α protein involved with apoptosis, western blot was conducted. Results : The groups treated with 25 ㎍, 50 ㎍ and 100 ㎍ of PG water extract had no toxicity for PC-12 cell. The groups treated with 25 ㎍, 50 ㎍ and 100 ㎍ of PG water extract showed a significant increase of cell survival rate in comparison with the control group injected by only 4-HNE. The groups treated with 25 ㎍ and 50 ㎍ of PG water extract showed a significant supression on increase of Bax protein expression in the control group. The group treated with 100 ㎍ of PG water extract showed a significant promotion on decrease of Bcl-2 protein expression in the control group. The group treated with 50 ㎍ of PG water extract showed a significant supression on increase of Caspase-3 protein expression in the control group. The group treated with 25 ㎍ of PG water extract showed a significant supression on increase of TNF-α protein expression in the control group. Conclusions : These results suggest that Polygalae Radix Preparata Cum Glycyrrhizae Radix is effective in reducing apoptosis by 4-HNE-dameged cell.

  • The purpose of this research was to investigate the color characteristic and the dyeing properties of Glycyrrhizae Radix extract on cotton fabrics. The highest absorbance of Glycyrrhizae Radix extract was obtained in 75 v/v% of ethyl alcohol in water. Pre-mordanted cotton fabrics dyed at $30^{\circ}C$ and pH 3 for 80 minutes showed the highest K/S value. According to the dyeing conditions, the colors of the fabrics varied from yellow(Y) and yellowish red(YR) to greenish yellow(GY). The color fastness of the fabrics in dry-cleaning and water was 4~5 rating. The deodorization capacity and UV-cut effect of the dyed fabrics were higher than those of undyed ones.

  • This study was carried out to investigate the effect of Glycyrrhizae Radix on serum corticosterone and blood histamine content by immobilization stress in mice. Corticosterone secretion and blood histamine level was significantly increased in mice by subjecting the animals to immobilization stress 1 hr. after intraperitoneal injection of Glycyrrhizae Radix extract (150 mg/kg) and glycyrrhizinic and (15 mg/kg). whereas, administration of cortisol $(7.5\;{\mu}g/kg)$ provoked a decrease in corticosterone secretion and histamine levels. These results suggested that glycyrrhizinic acid was effective on cortiosterone release provoked by immobilization stress and this release was mediated in part by histamine.

  • Second derivative (D2) spectrometry using ion-pair extraction technique was development for the determination of total glycyrrhetic acid (GA) in Glycyrrhizae Radix, Glycyr-rhizin (G) obtained from Glycyrrhizae Radix was hydrolyzed into GA in 2 N-HCI and methanol (1:1) and extracted from aqueous phase in the form of an ion-pair complex with tetrapentylammonium bromide (TPA) as a counter ion. Maximum D2 amplitude (Z value) was obtained when 1000-fold or greater molar ratio of TPA was used at pH 11. Reaction an effective extraction solvent of the ion-pair complex. The linearity of standard curve of ion-pair GA was obtained in the range of 4.120 $\mu$g/ml as GA. Assayed contents of GA in dry powder by D2 UV spectrometry and HPLC method were 5.31 $\pm$ 0.04% and 5.20 $\pm$ 0.008%, respectively.

  • Background and objective: Methamphetamine (Meth) is a widely abused psychostimulant that produces hyperlocomotion in rodents. Radix of Glycyrrhizae uralensis comprises a variety of bioactive components that have neuroprotective effects. In a previous study, we have demonstrated methanol extracts from radix of Glycyrrhizae uralensis (MEGR) suppress acute cocaine-induced extracellular dopamine release in the nucleus accumbens. In the present study, we investigated the effect of MEGR on acute Meth-induced hyperlocomotion. Methods: Male Sprague-Dawley rats were orally administered with MEGR (60 mg/kg and 180 mg/kg) 60 min prior to an intraperitoneal injection of Meth (1.0 mg/kg). Results: Behavioral analysis showed acute Meth greatly increased locomotor activities, while pretreatment with MEGR dose dependently inhibited the hyperlocomotion. In parallel, there were markedly increased levels of dopamine and its metabolite 3, 4-dihydroxyphenylacetic acid in the nucleus accumbens tissues in Meth-treated rats, which were also almost completely reversed by 180 mg/kg MEGR. Conclusions: These results showed that radix of Glycyrrhizae uralensis attenuates Meth-induced hyperlocomotion by inhibiting dopamine synthesis and utilization, suggesting that radix of Glycyrrhizae uralensis might be effective in blocking the rewarding effect of Meth.

  • This study was investigated to determine the quality monitoring of distributed materials from Glycyrrhizae Radix (26 samples), Atractylodis Rhizoma Alba (24 samples) according to storage period after $1{\sim}3$ year. We have estimated by identification, purity, loss on drying, ash, acid insoluble ash, extract content, essential oil content, assay and microbial contamination. As a result, Glycyrrhizae Radix (26 samples) were satisfied with the standard of K.P. (Korean Pharmacopoeia) and WHO's microbial contamination limit standard. In the Atractylodis Rhizoma Alba (24 samples), 2 samples were not satisfied with the standard of K.P.(Korean Pharmacopoeia) and WHO's microbial contamination limit standard. The results make practical application of the basic data for the quality control of herbal medicine in storage.

  • This study was carried out to investigate the antimicrobial and anti-inflammatory activities of ethanol extract from Glycyrrhizae radix (GR) and ethanol extract from Glycyrrhizae radix cultured with Paecilomyces japonica mycelium (GRPM). Antimicrobial activity was measured by paper disc diffusion assay and minimum inhibitory concentration (MIC). Anti-inflammatory activity was evaluated by measurement of NO production in LPS-stimulated RAW264.7 cells. For the results of the paper disc diffusion assay, GRPM showed high antimicrobial activities against Bacillus cereus, Listeria monocytogenes, and Staphylococcus aureus. In addition, the MIC of GRPM (100 ppm) was lower than that of GR (200 ppm) against L. monocytogenes. When the morphology of L. monocytogenes treated with GRPM was observed using a FE-SEM, the surface of cells treated with GRPM were damaged, and some parts of the cell wall were destroyed. The inhibitory effect on NO production in LPS-stimulated RAW264.7 cells was significantly increased by GRPM treatment. In conclusion, GRPM is superior to GR in terms of antimicrobial and anti-inflammatory activities.
