계피 및 감초를 첨가한 초콜릿의 생리활성 및 관능적 특성 (Physiological and Sensory Characteristics of Chocolate with Cinnamomi ramulus and Glycyrrhiza glabra L.)

  • This study was conducted investigate the effect of medicinal plant extracts on physiological and sensory characteristics. The medicinal plant extracts were added to chocolate at a weight percentage of 0, 3 and 6%. Color values(L-value, redness, and yellowness), total flavonoids, DPPH radical scavenging activity, and sensory characteristics made with varying various medicinal plants concentrations of the additives were measured. In sensory evaluation, significant differences(p<0.05 and p<0.01) were shown in taste, texture, and overall acceptability depending on the addition of plant extracts, but there were no significant differences in chocolate aroma and bitterness properties.

  • Intestinal epithelial cells can produce cytokines and chemokines that play an important role in the mucosal immune response. Regulation of this production is important to prevent inflammatory tissue damage. Glycyrrhiza glabra has been shown to inhibit inflammation. The aim of this study was to examine the inhibitory effect of 18- beta-glycyrrhetinic acid, a triterpenoid saponin of Glycyrrhiza glabra, on IL-S production via mitogen-activated protein kinases (MAPKs) and nuclear factor-kappa B (NF-kB) in TNF-alpha-stimulated human colon epithelial cells. The inhibitory effects of glabridin on human platelet aggregation and thrombus formation

  • Glycyrrhiza glabra is a well-known medicinal herb that grows in various parts of the world and glabridin is a major chemical compound that is found in the root extract of Glycyrrhiza glabra. Glabridin is a natural compound known to have antioxidant, anti-inflammatory, anti-atherogenic, anti-osteoporotic and skin-whitening. In this study, we investigated if glabridin inhibited platelet aggregation and thrombus formation. We observed that glabridin inhibited collagen-induced platelet aggregation and suppressed signal transduction molecules such as phosphatidylinositol-3 kinase (PI3K), Akt, glycogen synthase kinase-3α/β (GSK-3α/β), SYK, cytosolic phospholipase A2, and p38 expression. In addition, glabridin suppressed thromboxane A2 generation and thrombin-induced clot retraction. Taken together, glabridin showed strong antiplatelet effects and may be used to block thrombosis- and platelet-mediated cardiovascular diseases.

감초 신품종 및 약전 수재감초의 면역조절 효과 비교 연구 (The Comparative Study of Immunomodulatory Effect by Glycyrrhiza New Varieties and Official Compendia)

  • Objective : The genus Glycyrrhiza has been used in food and traditional herbal medicine. Glycyrrhiza new varieties Wongam and Sinwongam have been developed by Korea Rural Development Administration and investigated to register on Korean Pharmacopoeia of the Ministry of Food and Drug Safety. The aim of this study is to investigate the immunomodulatory effect of Wongam and Sinwongam comparing with listed Glycyrrhiza species (Glycyrrhiza uralensis Fischer and G. glabra Linne) for evaluations about pharmacological effect of Glycyrrhiza new varieties. Methods : We studied the immunomodulatory effect of Wongam and Sinwongam compared with G. uralensis and G. glabra using THP-1 cell in vitro model. The cells were treated with phorbol 12-myristate 13-acetate (PMA) for differentiation and stimulated with lipopolysaccharides (LPS) to induce immune activation. We analyzed and compared the effects Glycyrrhiza new varieties and listed Glycyrrhiza species using nitric oxide (NO) assay, western blot, and reverse transcription-quantitative polymerase chain reaction analysis. 1) Results : Wongam and Sinwongam showed no cytotoxicity in THP-1 cells. Wongam and Sinwongam, and listed Glycyrrhiza species increased NO production, and cyclooxygenase (COX)-2 expression with or without LPS in differentiated THP-1 macrophages. Furthermore, Wongam and Sinwongam and listed Glycyrrhiza species upregulated the mRNA expressions of T helper type 1 (Th 1)-associated cytokines in LPS-stimulated THP-1 macrophages. Conclusion : These results indicated that Wongam and Sinwongam would have effect of enhancing immune response through the increase of NO and COX-2 expression, and activate Th1-associated cytokines. The findings of this study suggest the wide applicability of Glycyrrhiza new varieties.

Effect of ethanolic extract of some anti-asthmatic herbs on clonidine and haloperidol-induced catalepsy in mice

  • The ethanolic extract of some medicinal plants having anti-asthmatic activity such as Solanum xanthocarpum, Curcuma longa, Glycyrrhiza glabra, Piper longum, A. vasica, A. lebbeck, and Tinospora cordifolia was evaluated for antihistaminic and anti-cataleptic activity. The aqueous solution of ethanolic extract of S. xanthocarpum and G. glabra potentiated histamine-induced tracheal chain contractions. Whereas, C. longa, P. longum, and T. cordifolia, and A. lebbeck were without any significant effect on histamine. Only A. vasica inhibited histamine-induced tracheal chain contraction. G. glabra per se produced contraction of the tracheal chain, which was blocked by pretreatment with atropine. Single dose of S. xanthocarpum potentiated clonidine-induced catalepsy but on repeated doses (once in a day for 3 days) inhibited catalepsy. Pretreatment with ethanolic extract of C. longa, P. longum, T. cordifolia inhibited catalepsy whereas G. glabra and A. lebbeck significantly potentiated clonidine-induced catalepsy. None of the extracts inhibited haloperidol-induced catalepsy. Thus the extracts having antihistaminic activity or mast cell stabilizing activity inhibited clonidineinduced catalepsy.

감초 추출물로 재배한 콩나물의 품질특성 (Quality Characteristics of Soybean Sprout Cultivated with Extract of Korean Glycyrrhiza glabra)

  • 감초추출물의 농도를 100 ppm, 200 ppm, 300 ppm and 400 ppm로 조절하여 콩을 발아시켜 재배한 콩나물의 아미노산, 유기산 및 유리당 함량을 재배 2일, 3일 및 4일째에 분석하였다. 총 아미노산 함량은 100 ppm에서 3일간 재배하였을 때가 가장 높았고, 또한 재배일수가 길어질수록 aspartic acid 함량은 증가하지만, glutamic acid 함량은 오히려 감소하는 경향을 나타내었다. 총 유리당은 GGE 콩나물의 경우 대조군에 비하여 낮은 함량을 나타내었으며, 그러나 재배일수가 증가할수록 그 함량은 많아졌다 Sucrose함량은 재배일수가 길어질수록 감소하였으나, 다른 유리당은 오히려 증가하였다 총 유기산은 재배일수가 증가할수록 많은 함량을 나타내었으며, 200 ppm으로 재배한 콩나물이 가장 많은 함량을 나타내었다. 결론적으로 감초추출물은 콩나물의 품질증진을 위한 아미노산 및 유기산 함량을 증가시키는 발아 및 재배수로서 효과적일 것으로 판단된다.

升麻葛根湯加味方이 消炎작용에 미치는 영향 (Study on the Anti-inflammatory Effects of Seungmagalgeuntanggamibang(SMG))

  • This study was carried out to investigate the effects of SMG on the in vitro and in vivo inflammatory reactions. In experiment I, in vitro tests, ethanol extract of SMG showed potent radical scavenging activity tested by DPPH(I,1-diphenyl-2-picryl-hyrazyl) method and inhibited the lipopolysaccharide-induced gene expressions of interleukin-1${\beta}$, interleukin-6 and tumor necrosis factor-${\alpha}$ (above 50$\%$ at a concentration of 50㎍/㎖) by the macrophage RAW 246.7 cells. Among the herbal ingredients of SMG, ethanol extracts of Scutellaria baicalensis, Paeonia lactiflora, Glycyrrhiza glabra showed potent radical scavenging activity. And Glycyrrhiza glabra and Scutellaria baicalensis showed potent inhibitory activity of nitric oxide production. Especially, ethanol extract of Scutellaria baicalensis inhibited the gene expression of IL-1${\beta}$, IL-6 and TNF-${\alpha}$. In cyclooxygenase-2 assay, Scutellaria baicalensis and Glycyrrhiza glabra showed the potent inhibition of prostaglandin E2 generation. In experiment 2, in vivo tests, SMG showed inhibitory effects on vascular permeability (28.7$\%$) and leukocyte migration (11.5$\%$). These results mean that SMG has a anti-inflammatory effects by it's inhibitory effects of leukocyte migration and vascular permeability as well as it's inhibitory effects of lipopolysaccharide-induced gene expression of IL-1${\beta}$, IL-6 and TNF-${\alpha}$, and radical scavenging activity. Therefore, I expect that SMG may be used as a effective drug for treatment on inflammation.

감초 중 리퀴리티게닌 분석법 개발 및 함량분석 (Analysis of Liquiritigenin, an Aglycone of Liquiritin in Licorice by High Performance Liquid Chromatography)

  • Licorice(Glycyrrhizae Radix et Rhizoma) is recorded as the root of Glycyrrhiza uralensis Fischer or Glycyrrhiza glabra Linne or Glycyrrhiza inflata Bat.(Leguminosae) in Korean Pharmacopoeia $9^{th}$ edition (KP9) and Chinese Pharmacopoeia 2005(CP2005), Glycyrrhiza uralensis Fischer or Glycyrrhiza glabra Linne in Japanese Pharmacopoeia 2005(JP2005). It is established the content standard of Glycyrrhizin 2.5 % and liquiritin 1% in KP9 and CP2005. But, according to the reports, all Licorice species were not sufficient for content standard of liquiritin 1.0% for licorice in KP9 and CP2005. It shows different content of liquiritin among G. uralensis, G. glabra and G. inflata. Also, it was reported liquiritin, liquiritin apioside are transformed into liquiritigenin by human internal flora. Therefore, we have studied for the pre-treatment condition and analytical method of liquiritigenin; It was good efficinet in 2M HCl reflux(1 hr) for hydrolysis and in methylene chloride for solvent fractionation. And 1% acetic acid in DW(A) and acetonitrile(B) with gradient condition as a mobile phase was most effective in HPLC analytical condition. According to these experimental methods, we have anlayzed content of liquiritigenin about 77 Licorice sample. In this research, it was also examined the content of liquiritin and liquiritigenin for Glycyrrhizae Radix related growing area. According to the results, we suggested the content standard of glycyrrhizin more than 2.5%, liquiritigenin more than 0.7%(after hydrolysis) of licorice.

Differentiation of Roots of Glycyrrhiza Species by 1H Nuclear Magnetic Resonance Spectroscopy and Multivariate Statistical Analysis

  • To classify Glycyrrhiza species, samples of different species were analyzed by $^1H$ NMR-based metabolomics technique. Partial least squares discriminant analysis (PLS-DA) was used as the multivariate statistical analysis of the 1H NMR data sets. There was a clear separation between various Glycyrrhiza species in the PLS-DA derived score plots. The PLS-DA model was validated, and the key metabolites contributing to the separation in the score plots of various Glycyrrhiza species were lactic acid, alanine, arginine, proline, malic acid, asparagine, choline, glycine, glucose, sucrose, 4-hydroxy-phenylacetic acid, and formic acid. The compounds present at relatively high levels were glucose, and 4-hydroxyphenylacetic acid in G. glabra; lactic acid, alanine, and proline in G. inflata; and arginine, malic acid, and sucrose in G. uralensis. This is the first study to perform the global metabolomic profiling and differentiation of Glycyrrhiza species using $^1H$ NMR and multivariate statistical analysis.

원산지별 감초 추출물의 항균 활성 비교 및 천연방부제로써의 효능 연구 (Comparative Study on the Antimicrobial Activity of Glycyrrhiza uralensis and Glycyrrhiza glabra Extracts with Various Countries of Origin as Natural Antiseptics)

  • 본 연구에서는 원산지별 감초 추출물의 항균활성을 측정하여 비교 평가하였고, challenge test를 통해 화장품방부제인 메틸 파라벤과 감초 추출물의 방부력을 비교하였다. 1. 원산지별 감초 추출물의 수득률의 경우 50% 에탄올 추출물의 수득률은 한국 감초(19.24%), 중국 감초(18.34%). 우즈베키스탄 감초(17.33%)이고, 에틸아세테이트 분획의 수득률은 한국 감초(2.22%), 중국 감초(2.43%), 우즈베키스탄 감초(2.07%)이었다. 2. 원산지별 감초 추출물의 항균활성은 50% 에탄올 추출물 보다 에틸아세테이트 분획에서 항균활성이 우수하였으며 B. subtilis, P. acnes, E. coli, P. aeruginosa 균을 저해하였다. 최소저해농도(MIC) 측정을 통해 원산지별 감초 추출물 에틸아세테이트 분획의 항균활성을 비교한 결과, B. subtilis에 대한 최소저해농도는 한국(625 ${\mu}g/ml$), 중국(625 ${\mu}g/ml$), 우즈베키스탄(625 ${\mu}g/ml$)로 동일한 항균활성을 나타내었고, P. acnes에 대한 최소저해농도는 한국(156 ${\mu}g/ml$), 중국 (625 ${\mu}g/ml$), 우즈베키스탄(2,500 ${\mu}g/ml$)로 한국 감초 추출물의 에틸아세테이트 분획이 중국 감초 추출물의 에틸아세테이트 분획 보다는 약 4배, 우즈베키스탄 감초 추출물의 에틸아세테이트 분획 보다는 약 16배 더 우수한 항균활성을 나타내었다. E. coli에 대한 최소저해농도는 한국(5,000 ${\mu}g/ml$), 중국(10,000 ${\mu}g/ml$), 우즈베키스탄(10,000 ${\mu}g/ml$)로 한국 감초 추출물의 에틸아세테이트 분획이 중국, 우즈베키스탄 감초 추출물의 에틸아세테이트 분획 보다 E. coli에 대해서 2배 좋은 항균활성을 나타냄을 확인하였다. P. aeruginosa에 대한 최소저해농도는 한국(78 ${\mu}g/ml$), 중국(156 ${\mu}g/ml$), 우즈베키스탄(5,000 ${\mu}g/ml$)로, 한국 감초 추출물의 에틸아세테이트 분획의 P. aeruginosa에 대한 항균활성이 중국 감초 추출물의 에틸아세테이트 분획 보다 2배, 우즈베키스탄 감초 추출물의 에틸아세테이트 분획 보다는 약 64배 뛰어난 항균활성을 나타냄을 확인하였다. 한국 감초 추출물이 중국, 우즈베키스탄 감초 추출물보다 뛰어난 항균활성을 보이며, 화장품방부제로 쓰이는 파라벤류 보다 높은 항균활성을 가지고 있음을 확인하였다. 3. B. subtilis, P. acnes, E. coli, P. aeruginosa에 대한 challenge test를 통해 메틸 파라벤 보다 한국 감초 추출물이 우수한 방부력을 나타냄을 확인하였다. 이상의 본 연구 결과로부터 한국 감초추출물이 중국, 우즈베키스탄 감초 추출물보다 항균활성이 우수함을 확인하였다. 한국 감초 추출물이 여드름균인 P. acnes에 대한 우수한 항균활성을 나타내므로 한국 감초 추출물이 여드름치료제의 기능성원료로서 이용가능성과 화장품방부제로 쓰이는 파라벤류를 대체할 수 있는 천연방부제로써 응용 가능성이 크다고 사료된다.
