황련추출물에 의한 HIV-1 복제 저해 (Inhibition of HIV-1 Replication by Extract of Coptis chinensis)

  • Natural products, total number of 175, were screened to test for their effect on the replication of human immunodeficiency virus type 1 (HIV-1). Five of them, such as Eriobotrya japonica, Eugenia caryphyllata, Cuscuta chinensis, Glycyrrhiza uralensis, and Coptis chinensis were shown to be effective in inhibiting the replication of HIV-1 in tissue culture and their selectivity indexes were 42, 40, 14, 18 and 65, respectively. To further fractionate Coptis chinensis, which is shown to be highest anti-HIV-1 activity, methanol extracts of Coptis chinensis were fractionated into methylene chloride at pH3, pH10 and water residue. The selectivity Indexes of CH$_2$C1$_2$(pH 3), CH$_2$C1$_2$(pH 10) and water residue were 50, 22 and 98 respectively. Our results show that the water residue of Coptis chinensis was the most effective for anti-HIV-1 activity.

계피 및 감초를 첨가한 초콜릿의 생리활성 및 관능적 특성 (Physiological and Sensory Characteristics of Chocolate with Cinnamomi ramulus and Glycyrrhiza glabra L.)

  • This study was conducted investigate the effect of medicinal plant extracts on physiological and sensory characteristics. The medicinal plant extracts were added to chocolate at a weight percentage of 0, 3 and 6%. Color values(L-value, redness, and yellowness), total flavonoids, DPPH radical scavenging activity, and sensory characteristics made with varying various medicinal plants concentrations of the additives were measured. In sensory evaluation, significant differences(p<0.05 and p<0.01) were shown in taste, texture, and overall acceptability depending on the addition of plant extracts, but there were no significant differences in chocolate aroma and bitterness properties.

히드로클로로치아지드와 감초의 병용에 의한 랫트의 혈청중 칼륨농도 변화 (Effect of Coadministration of Hydrochlorothiazide and Glycyrrhiza Extract on Serum Potassium Levels in Rats)

  • Effect of hydrochlorothiazide on serum potassium level was studied in the hypokalemia-induced rats by the oral administration of glycyrrhiza extract (GE) for 4 weeks. According to the concentration of GE, serum potassium levels were found to be $5.8{\pm}0.2,\;5.4{\pm}0.2\;and\;5.5{\pm}0.2\;mM/l$ after oral administration of 0.1, 0.2 and 0.5% GE solutions for 4 weeks, respectively, comparing $6.4{\pm}0.2\;mM/l$ in normal rats. The i.p. administration of hydrochlorothiazide (100mg/kg) showed no significant difference $(5.1{\pm}0.3-5.2{\pm}0.3\;mM/l)$ in the decrease of serum potassium levels between these hypokalemia-induced rats and normal rats, regardless of the concentration of pretreated GE solutions. Therefore it was considered that the administration of hydrochlorothiazide did not worsen the hypokalemia induced by the long term administration of GE in rats.

Licochalcone A, a Major Phenolic Constituent of Glycyrrhiza inflata, Suppresses Angiogenin Expression in Colon Cancer Cells

  • Tumor angiogenesis, which is essential for tumor growth and tumor metastasis, depends on angiogenic factors produced by tumor cells and/or infiltrating cells such as endothelial cells and immune cells in tumor tissue. Previously, we reported that licochalcone A (LicA), an important bioactive compound of Glycyrrhiza inflate, suppresses angiogenesis, tumor growth and metastasis. In this study, we evaluated the effect of LicA on angiogenin production in colon cancer cells because angiogenin is an essential factor to regulate angiogenesis and tumor progression. When we examined the angiogenin levels in three human colon cancer cells, HT-29, SW480 and Caco-2, LicA treatment significantly reduced the amounts of angiogenin among three cancer cell lines. In an in vivo study in which mice were implanted with HT-29 cells, oral administration of LicA reduced angiogenin in tumor tissues when compared with vehicle-administered mice. These results suggest that reduced angiogenin in response to LicA treatment may play essential role to inhibit tumor growth, angiogenesis as well as metastasis.

First Total Synthesis of Highly Anti-Inflammatory Active Licochalcone D Through Water-Accelerated [3,3]-Sigmatropic Rearrangement

  • Licochalcones, derived from the dried roots of Glycyrrhiza inflata, have been reported to show various biological activities including antitumor, antiparasitic, antileishmanial, antioxidative, superoxide scavenging, antibacterial, and PTP1B activity. Licochalcone D has an allyl group on ring A instead of ring B, however, most other natural licochalcones possess the group on ring B. Total synthesis of licochalcone D has not been reported even possessing the strongest anti-inflammatory activity. Therefore, the first total synthesis of licochalcone D has been developed by using water-accelerated [3,3]-sigmatropic rearrangement method.

  • Intestinal epithelial cells can produce cytokines and chemokines that play an important role in the mucosal immune response. Regulation of this production is important to prevent inflammatory tissue damage. Glycyrrhiza glabra has been shown to inhibit inflammation. The aim of this study was to examine the inhibitory effect of 18- beta-glycyrrhetinic acid, a triterpenoid saponin of Glycyrrhiza glabra, on IL-S production via mitogen-activated protein kinases (MAPKs) and nuclear factor-kappa B (NF-kB) in TNF-alpha-stimulated human colon epithelial cells. The inhibitory effects of glabridin on human platelet aggregation and thrombus formation

  • Glycyrrhiza glabra is a well-known medicinal herb that grows in various parts of the world and glabridin is a major chemical compound that is found in the root extract of Glycyrrhiza glabra. Glabridin is a natural compound known to have antioxidant, anti-inflammatory, anti-atherogenic, anti-osteoporotic and skin-whitening. In this study, we investigated if glabridin inhibited platelet aggregation and thrombus formation. We observed that glabridin inhibited collagen-induced platelet aggregation and suppressed signal transduction molecules such as phosphatidylinositol-3 kinase (PI3K), Akt, glycogen synthase kinase-3α/β (GSK-3α/β), SYK, cytosolic phospholipase A2, and p38 expression. In addition, glabridin suppressed thromboxane A2 generation and thrombin-induced clot retraction. Taken together, glabridin showed strong antiplatelet effects and may be used to block thrombosis- and platelet-mediated cardiovascular diseases.

Antioxidant Activity of Glycyrrhiza uralensis Fisch Extracts on Hydrogen Peroxide-induced DNA Damage in Human Leucocytes and Cell Death in PC12 Cells

  • In this study, antioxidant activity of methanol extract of Glycyrrhiza uralensis Fisch (GUE) against $H_2O_2$-induced DNA damage in human leucocytcs and cell death in PC12 cells was determined. The effect of GUE on $H_2O_2$-induced DNA damage in human leucocytcs was evaluated by the comet assay, where GUE ($1-50\;{\mu}g/mL$) was a dose dependent inhibitor of DNA damage induced by $H_2O_2$. The protective effect of GUE against $H_2O_2$-induced damage on PC12 cells was investigated by MTT reduction assay and lactate dehydrogenase release assay. A marked reduction in cell survival induced by $H_2O_2$ was significantly prevented by $1-50\;{\mu}g/mL$ of GUE. The enzyme activity of caspase-3 was elevated in $H_2O_2$-treated PC12 cells, while preincubation with GUE for 30 min inhibited $H_2O_2$-induced caspase-3 activation in a dose-dependent manner. In conclusion, GUE ameliorates $H_2O_2$-induced DNA damage in human leucocytes and has neuroprotective effect by preventing cell death in PC12 cell, suggesting that GU may be a potential candidate for novel therapeutic agents for neuronal diseases associated with oxidative stress.

감초 추출물 투여가 Ovalbumin으로 유발된 마우스 알레르기성 천식에 미치는 영향 (Effects of Glycyrrhiza uralensis Fisch (GUF) Extract on the Ovalbumin-Induced Allergid Asthma in Mice)

  • Objective : Glycyrrhiza uralensis Fisch (GUF) has been used as remedy of allergic diseases for a long time in Korea. In the present study, we investigated the anti-allergic effects of GUF on experimental allergic asthma mouse model using ovalbumin (OVA). Methods : BALB/c mice were sensitized and challenged with 100 ug of OVA and 1 mg of aluminum potassium sulfate of 0.2 ml phosphate-buffered saline (PBS) intraperitoneally on day 1 and 15. Mice were challenged on 3 consecutive days with 5% OVA and AHR was assessed 24 hrs after the last challenge. We examined total inflammatory cell number in bronchoaveloar lavage fluid (BALF), Th2-associated cytokine productions and lung histology. Results : GUF potently inhibited the development of airway hypersensitivity and also reduced the number of BAL cells during OVA-induced allergic asthma. GUF also inhibited cytokine productions such as IL-4, IL-13 in lung tissue. Furthermore, GUF treatment inhibited allergic airway inflammation. Conclusion : These results suggest that GUF may inhibit the production of IL-4, IL-13 and infiltration of inflammatory cell and be beneficial oriental medicine for allergic asthma.
