The chloroplast genome of Glycyrrhiza uralensis Fisch was sequenced to investigate intraspecific variations on the chloroplast genome. Its length is 127,689 bp long (34.3% GC ratio) with atypical structure of chloroplast genome, which is congruent to those of Glycyrrhiza genus. It includes 110 genes (76 protein-coding genes, four rRNAs, and 30 tRNAs). Intronic region of ndhA presented the highest nucleotide diversity based on the six G. uralenesis chloroplast genomes. A total of 150 single nucleotide polymorphisms and 10 insertion and deletion (INDEL) regions were identified from the six G. uralensis chloroplast genomes. Phylogenetic trees show that the six chloroplast genomes of G. uralensis formed the two clades, requiring additional studies to understand it.
Colorectal cancer is one of the most common lethal cancer types worldwide. In recent years, widespread and large-scale studies have been done on medicinal plants for anti-cancer effects, including Glycyrrhiza glabra. The aim of this study was to evaluate the effects of an ethanol extract Glycyrrhiza glabra on the expression of HSP90, growth and apoptosis in the HT-29 colon cancer cell line. HT-29 cells were treated with different concentrations of extract (50,100,150, and $200{\mu}g/ml$). For evaluation of cell proliferation and apoptosis, we used MTT assay and flow cytometry technique, respectively. RT-PCR was also carried out to evaluate the expression levels of HSP90 genes. Results showed that Glycyrrhiza glabra inhibited proliferation of the HT-29 cell line at a concentration of $200{\mu}g/ml$ and this was confirmed by the highest rate of cell death as measured by trypan blue and MTT assays. RT-PCR results showed down-regulation of HSP90 gene expression which implied an ability of Glycyrrhiza glabra to induce apoptosis in HT-29 cells and confirmed its anticancer property. Further studies are required to evaluate effects of the extract on other genes and also it is necessary to make an extensive in vivo biological evaluation and subsequently proceed with clinical evaluations.
The aim of this study was to investigate the physiological functionalities of Codonopsis lanceolata, Glycyrrhiza uralensis, Chaenomeles sinensis, Crataegus pinnafida, and their mixtures. We also determined their antioxidative, fibrinolytic, and ${\alpha}-glucosidase$ inhibitory activities. The antioxidative activities of Codonopsis lanceolata, Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida were 79%, 88.3%, 89.9%, and 89.3% respectively. Their fibrinolytic activities were 0.80plasmin unit/mi, 0.57 plasmin unit/mi, 0.52 plasmin unit/mi, and 0.53 plasmin unit/mi respectively. The ${\alpha}-glucosidase$ inhibitory activity of Codonopsis lanceolata was 25%. The 10-fold diluents of Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida showed ${\alpha}-glucosidase$ inhibitory activities of 93.6%, 65.3%, and 61.3% respectively. In antioxidative activity tests of the medicinal plants mixtures at various ratios, the mixtures of Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida with Codonopsis lanceolata showed antioxidative activities of approximately 90%. In fibrinolytic activity tests mixtures(1:1) of Codonopsis lanceolata with Chaenomeles sinensis and Crataegus pinnafida exhibited increases of 23% and 24% in activity respectively. In ${\alpha}-glucosidase$ inhibitory activity tests, a mixture (4:1) of Codonopsis lanceolata and 10-fold diluted Glycyrrhiza uralensis showed an inhibitory activity of 98%, a mixture (3:1) of Codonopsis lanceolata and 10-fold diluted Chaenomeles sinensis showed an inhibitory activity of 69.6%, and a mixture (1:1) of Codonopsis lanceolata and 10-fold diluted Crataegus pinnafida showed an inhibitory activity of 50.2%. In conclusion, the mixtures of Glycyrrhiza uralensis, Chaenomeles sinensis, and Crataegus pinnafida with Codonopsis lanceolata will be used as a material for the development of biofunctional foods.

Objectives This study is conducted to evaluate Th2 skewed condition control through skin fat barrier formation from the treatment using Coptidis Rhizoma and Glycyrrhiza uralensis extract. Methods The 6-week-old NC/Nga mice were divided into 4 groups: Control group (Ctrl), lipid barrier eliminate treatment group (LBET), Coptidis Rhizoma and Glycyrrhiza uralensis feeding treatment after lipid barrier elimination group (CGFT), dexamethasone feeding treatment after lipid barrier elimination group (DxFT). After 3 days, differences in skin condition, improvement of skin fat barrier, and control of Th2 skewed condition of each group were observed. Results Pathologic skin damage and tissue changes were less in the CGFT group than in the LBET and DxFT groups, and Transepidermal water loss (TEWL) and pH were also significantly decreased (p < 0.05). The filaggrin intensity and positive response also increased significantly in the CGFT group (p < 0.05). Kallikrein-related peptidase (KLK) 7, Protease activated receptor (PAR)-2, Thymic stromal lymphopoietin (TSLP), Interleukin (IL)-4, and the products of the Th2 differentiation process also showed a significant decrease compared to the LBET and DxFT groups (all p < 0.05). Conclusions The Coptidis Rhizoma and Glycyrrhiza uralensis extract causes skin barrier recovery and function recovery through the formation of skin fat barrier. This leads to the conclusion that Coptidis Rhizoma and Glycyrrhiza uralensis extract can control Th2 differentiation through the formation of skin fat barrier.
Background: Glycyrrhiza radix (licorice root) have been used as an oriental medicine material for long time, and its protective effects on oxidative stress, inflammation and cognition deficit have been recently reported. However, the cultivation of Glycyrrhiza species as medicinal crops is associated with some problems such as low productivity and early leaf fall, etc. To resolve this problems, Glycyrrhiza cultivars have been developed by direct hybridization of each Glycyrrhiza species by Korean researchers. The present study was conducted to compare the Glycyrrhiza cultivar radix (Dagam, Sinwongam and Wongam) for their anti-oxidation, anti-inflammation, and cognition improvement effects and levels of liquiritin, isoliquiritigenin and licochalcone in order to select an excellent cultivar as a material resource. Methods and Results: For evaluating the inhibitory efficacies of the Glycyrrhiza cultivar extracts on oxidative stress and inflammation in BV2 cells, we measured their reactive oxygen species (ROS) production and nitric oxide (NO) release after treating them with lipopolysccharide. The scavenging activities on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and peroxynitrite ($NOO^-$) radicals were evaluated. Cell proliferation and N-methyl-D-aspartate receptor (NMDAR) inhibition were analyzed. The total phenol, liquiritin, isoliquiritigenin and licochalcone A content in the extracts of the three culivars were quantified. Furthermore, the correlation coefficient between the activities and contents of total phenol, liquiritin, isoliquiritigenin and licochalcone A were also calculated. The results indicated that Sinwongam exhibited potent anti-oxidant, anti-inflammatory and NMDAR inhibititory activities. Sinwongam also showed higher total phenol and licochalcone A contents than the other cultivars. Among the three cultivars, Dagam exhibited a positive effects on NO release inhibition, cell proliferation and contents of liquiritin and isoliquiritigenin. Conclusions: Sinwongam is expected to be the most useful resource as a functional material for anti-oxidation/anti-inflammation and cognition improvement among the three studied licorice cultivars.
Objectives : This study was conducted to evaluate the antioxidative and anticancer activity of the water and ethanol extracts from medicinal herbs. Methods : Three kinds of medicinal herbs(Corni fructus, Astragalus membranaceus and Glycyrrhiza uralensis) were extracted with distilled water and 70% ethanol, and the extracts were tested for their antioxidative and anticancer activities. Results : The highest polyphenol contents of the water and ethanol extracts from medicinal herbs were 342.14 mg and 435.62 mg per 100 g of Cornus officinalis, respectively. The highest electron donating abilities (EDA) of the water and ethanol extracts from Glycyrrhiza uralensis were 88% and 91% at 1,000 ppm, respectively. The water and ethanol extracts from Astragalus membranaceus had the highest nitrite scavenging abilities (NSA) at 1,000 ppm. The highest anticancer activity of the extracts were from Glycyrrhiza uralensis against both of MDA and A549 cells. Conclusions : These results suggest that the medicinal herbs can be used as natural antioxidant to prevent oxidative damage in normal cells probably because of their antioxidant characteristics.

The purpose of this research was to investigate the effects of the extracts of Glycyrrhiza uralensis (GE) and the combined extracts of Glycine max Merr. and Glycyrrhiza uralensis (GGE) on the activity of murine splenocytes and macrophages. GE and GGE were administered orally twice a day for 7 days at the dose of 500 mg/kg. GE decreased the viability of T- and B-lymphocytes in splenocytes, but GGE increased the viability of B-lymphocytes in splenocytes. GE increased the population of B-lymphocytes in splenocytes, but decreased the population of T-lymphocytes and splenic $CD4^+$ cells. Also, GGE decreased the population of B-lymphocytes in splenocytes, but increased the population of T-lymphocytes and splenic $CD4^+$ cells. Furthermore, GE and GGE enhanced the phagocytic activity of peritoneal macrophages and the production of nitric oxide. These results suggest that the regulative action of immune response of GGE is more potent than their of GE.
Objectives The purpose of this study is to understand the correlations between lung, large intestine, and skin of 3-week-old mice in which ulcerative colitis was induced, up on administration of Coptidis rhizome and Glycyrrhiza uralensis mixed extract. Methods Mice were divided into 4 groups as follows; no treatment group (Ctrl group), ulcerative colitis-induced mice group (UE group), ulcerative colitis-induced mice group after administering Pentasa (PT group), ulcerative colitis-induced mice group after administering Coptidis rhizoma and Glycyrrhiza uralensis mixed extract (CGT group). Mice were induced ulcerative colitis by Dextran sulfate sodium (DSS). After 5 days of administration, We obvserved anti-inflammatory effect, alveolar formation, and skin barrier control in the colon mucosa. Results The CGT group was observed arrangement of normal intestinal cells, Infiltration of less inflammatory cells. The CGT significantly decreased positive rseponse of $TNF-{\alpha}$, p-IkB, Caspase 3 in large intestine, and significantly increased positive rseponse of EGF, IGF, catalase, Filaggrin, involucrin, loricrin. Conclusions The results of this study show the correlation of Lung-Large intestine-Skin by administering Coptidis rhizoma and Glycyrrhiza uralensis mixed extract to ulcerative colitis-induced mice.

Platelets are activated at the sites of vascular injury by a number of molecules, including adenosine diphosphate, collagen and thrombin. The full platelet aggregation is absolutely essential for the normal hemostasis. Glycyrrhiza glabra is a well-known medicinal herb that grows in various parts of the world and is known to have various effects such as antioxidant, anti-inflammatory, anti-atherogenic, anti-osteoporotic and skin-whitening. However, the platelet inhibitory effect of Glycyrrhiza glabra extract (GGE) has not been identified. In this study, we investigated if GGE inhibited platelet aggregation. We observed that GGE inhibited collagen-induced platelet aggregation, Ca2+ mobilization, and thromboxane A2 generation. In addition, GGE suppressed phosphorylation of phosphatidylinositol-3 kinase (PI3K), Akt and elevated phosphorylation of inositol 1,4,5-trisphosphate receptor (IP3R), vasodilator stimulated phosphoprotein (VASP). Taken together, GGE showed strong antiplatelet effects and may be used to block platelet-mediated cardiovascular diseases.

This study was investigated to elucidate the growth characteristics and the contents of glycyrrhizin and free sugars of Glycyrrhiza plants, i. e. G. glabra var. glandulifera, G. uralensis, and G. pallidiflora. The growth of G. glabra var. glandulifera and G. pallidiflora was better in plots planted with stolon and stem over 2cm in diameter compared to that of below 1cm and $1{\sim}2cm$. G. glabra var. glandulifera didn't bloom in Suwon but produced stolon, while G. pallidiflora bloomed but did not produce stolon. Total root weight of six-year-old G. glabra var. glandulifera consisted of stolon (73.2%) and primary root (26. 8 %). The contents of glycyrrhizin and free sugars were highest in stolon, followed by tap root and leaf. The glycyrrhizin contents in Glycyrrhiza plants were 7.38% in G. glabra var. glandulifera, 5.29% in G. uralensis and 0. 15% in G. pallidiflora.
