Cerebrosides and Terpene Glycosides from the Root of Aster scaber

  • Three cerebrosides 2, 3, and 5 and two terpene glycosides 1 and 4 have been isolated from the methanol extract of the root of Aster scaber. Their structures were determined as 3-Ο-$\beta$-D-glucuronopyranosyl-oleanolic acid methyl ester (1), (2S, 3S, 4R, 2 R, 8Z, 15 Z)-N-2 -hydroxy-15 -tetracosenoyl-1-Ο-$\beta$-D-glucopyranosyl-4-hydroxy-8-sphingenine (2), (2S, 3S, 4R, 8Z)-N-octadecanoyl-1-Ο-$\beta$-D-glucopyranosyl-4-hydroxy-8-sphingenine (3), 1$\alpha$-hydroxy-6$\beta$-Ο-$\beta$-D-glucosyl-eudesm-3-ene (4), and (2S, 3S, 4R, 2 R, 8Z)-N-2 -hydroxy-hexadecanoyl-1-Ο-$\beta$-D-glucopyranosyl-4-hydroxy-8-sphingenine (5) on the basis of spectroscopic methods.

복분자 딸기 잎의 플라보노이드 (Flavonoids from the Leaves of Rubus Coreanum)

  • Rubus coreantim (Rosaceae), which has been used in oriental traditional medicine as the remedies for impotence, pollution and premature ejaculation, bet-wetting and frequ ency of urination. is a fruit drug cultivating throughout southern Korea. The leaves of Rubus coreanum were extracted with 80% acetone. The extracts were separated by Ambellite XAD-2, Sephadex LH-20. MCI-gel CHP 20P and Cosmosil 75 C$_{18}$ OPN. And three flavonoids were isolated. The structures of these compounds were elucidated as quercetin-3-O-${\beta}$-D-glueuronopyranoside(l). quercetin-3-O-${\beta}$-D-glueuronopyranosyl methyl ester(2), quercetin-3-O-${\beta}$-D-xylopyranosyl-(2${\rightarrow}$1)-${\beta}$-glucopyranoside(3) on the basis of physicochemical and spectroscopic evidences.

Flavonoids of Crotalaria sessiliflora

  • Phytochemical investigation of the whole plants of Grata/aria sessiliflora L. led to the isolation of four flavonoids. The structures of these compounds were identified as 2',4',5,7-tetrahydroxy-isoflavone (1), 2',4',7-trihydroxyisoflavone (2), 4',7-dihydroxyflavone (3), and isovitexin (4) using spectroscopic analysis. Among these, compounds 2, and 3 have not been reported from Crotalaria species, whereas compounds 1, and 4 were reported from this plant for the first time.

  • To use onion(Allium cepa L.) residue as raw materials of food product, yields of onion-juice according to various extracting methods and optimal extracting condition of quercetin and its related glycosides were carried out. Onion juices gained by the methods of pressing, rotary crushing, freeze pressing and enzyme treatment. The yield by the method of enzyme treatment was higher than others. The yields of juice from fresh onion and heat-treated onion(8$0^{\circ}C$/10min) by pressing were 66% and 83%, respectively. Ethanol extraction of onion was efficient at 75$^{\circ}C$ and acetic acid extraction was proper at 3% concentration for 3 hours at 11$0^{\circ}C$. The onion extract was fractionated in the order of hexane, chloroform, ethylacetate and butanol to test flavonoids. The highest abundant flavonoids were found in ethylacetate and butanol fraction.

Determination of the Structure for Polysubstituted Flavonoid and 6-C-Glucosyl Flavonoids using $^{13}C-^{1}H$ Long Range Couplings

  • A flavone glycoside was isolated from the leaves of Betula platyphylla var. latifolia and characterized as $4, 6-Dimethoxy-5-hydroxyflavone-7-O-{\beta}-D-glucoside(pectolinarigenin-7-O-{\beta}-D-glucopy-ranoside)$ by method of chemical and NMR spectral analysis. $^13C-^1H$ long range coupling was confirmative for determination of its substituted position. In connection with this study, 6-C-Glucosylnalingenin and 6-C-Glucosylaromadendrin were confirmed its structures using this technique.

담쟁이덩굴엽의 플라보놀 배당체 (Flavonol Glycosides from Parthenocissus tricuspidata Leaves)

  • For the investigation of medicinal resources the studies were carried out to evaluated the pharmaco-constituents in the Leaves of Parthenocissus tricuspidata(Vitaceae), of which leaves have been used in Korea as folk remedies for the treatments of arthritis, jaundice, toothache, neuralgia, and etc. From 1-butanol fraction of the MeOH extract, Compound I ($C_{21}H_{18}O_{13}$, Quercetin-3-O-$\beta$-D-glucuronopyranoside), Compound II ($C_{21}H_{20}O_{12}$, Quercetin-3-O-$\beta$-D-glucopyranoside) and Compound III ($C_{25}H_{28}O_{12}$, Quercetin-3-O-(6"-n-butyl)-$\beta$-D-glucuronopyranoside) were isolated by column chromatographic separation using Sephadex LH-20 and ODS gel. Their structures were elucidated through instrumental analyses, such as $^{1}H$-NMR, $^{13}C$-NMR, IR, UV, El-Mass, FAB-Mass and GC. Especially compound III was Flavonol glycoside and named parthenosin.

  • Optimal medium for growth and glycosidases production of Bacteroides JY-6, an human intestinal bacterium, was general anaerobic medium or tryptic soy broth containing sod ium thioglycolate and ascorbic acid. By cocultivation of Staphylococcus R-48, Bacteroides JY-6 could be cultured in LB broth unable to culture JY-6. Heated Staphylococcus R-48 was also the inducer of the production of Bacteroides JY-6 glycosidases. These glycosidases were induced well by natural flavonoid glycosides, such as poncirin, naringin and rutin, but were not by synthetic substrates, p-nitrophenyl ${\beta$-D-glucopyranoside and p-nitrophenyl ${\alpha}$-L-rhanmopyranoside.

Bifidobacterium longum KCTC 5734를 이용한 비배당체 이소플라본 생산 (Production of Aglycone Isoflavones by Bifidobacterium longum KCTC 5734)

  • This study was performed to investigate the possibility of using three commercial bifidobacteria as a starter for soybean paste fermentation. In order to determine susceptibility to inhibition by high concentrations of salt in soybean paste, cell growth of three strains in sterilized soybean paste was analyzed. Bifidobacterium breve KCTC 5081 was the most resistant to salt, whereas Bifidobacterium bifidum KCTC 5082 showed low cell viability. Conversion efficiencies from glycoside isoflavone to aglycon isoflavone in soybean paste ranged from 11.3~28.6%, with Bifidobacterium longum KCTC 5734 the best strain. Therefore, B. longum KCTC 5734 may be used as a starter for Cheonggukjang fermentation, which is low-salt fermented soybean paste.

Inhibition of Monamine Oxidase by a Flavone and Its Glycoside from Ixeris dentata Nakai

  • Ixeris dentata Nakai (Compositae) is a perennial herb which has been used as a folk medicine for treating diabetes and gastroenteric troubles in Korea. Active compounds were isolated from the aerial parts of Ixeris dentata through the bioassay-guided fractionation and isolation method evaluated for inhibition of monoamine oxidase (MAO) in vitro. The compounds were identified as 5,7,3',4'-tetrahydroxyflavone (1) and 5,7,3',4'- tetrahydroxyflavone 7-glucoside (2), based on physical and spectroscopic characteristics. Compounds 1 and 2 showed a selective inhibition of type B MAO (MAO-B) activity, with IC/sub 50/ values of 15.3 μM and 36.4 μM, respectively, but did not inhibit type A MAO (MAO-A) activity.

서울귀룽나무 잎의 배당체에 관한 연구 제1보

  • The leaves of prunus padus L. var. Seoulensis Nakai distributed widely in Korea, were extracted with boiling alcohol(95%) after being mixed with calcium carbonate. The aqueous solution distilled off alcohol from the above filtered extract under the reduced pressure was evaporated to the dryness. The residue was extracted with acetic ether on the water-bath and the acetic ether solution extract was allowed to stand for a weck after removing out of acetic ether almostly, then a colorless necdles were Crystalized out which has the following characteristics : $C_{14}$ $H_{17}$ $O_{6}$ N, began to melt at 138-139.deg. bitter taste [.alpha.]$_$ $D^{20}$ ]=-26.99.deg. soluble in water, alcohol acetone acetic ether etc. slightly soluble in chloroform. It was proved to be identical with prunasine due to above characteristics and determination of Benzal-dehyde HCN and glucose respectively which was obtained on hydrolysis of this glycoside with emulsine.
