A total of 2,765 accessions were used as the initial set having both seed coat color and 100-seed weight data. As a result of molecular profiling using six SSR markers followed by stratification based on their usages, 335 accessions(12.1%) were selected by clustering based on UPGMA. Since 75 out of 335 accessions were mixed in phenotypic traits as a result of characterization, 260 accessions were finally set as a core set. This core set revealed nearly the same diversity compared with the other results on morphological traits of Korean soybean landraces. In total, 115 alleles(19.2 alleles per locus) were detected in the initial set and 79 alleles(13.2 alleles per locus) were detected in the core set. All 30 major alleles were present in the initial set and in the core set as well. In allele coverage, the core set was 71.4% of the initial set. These comparisons of number of alleles, gene diversity and coverage indicated that the core set represented the entire set well.
After a mixed carbohydrate diet, inhibition of ${\alpha}$-amylase and ${\alpha}$-glucosidase involved in the digestion and absorption of carbohydrates can significantly decrease the postprandial increase of blood glucose level. In the course of screening these useful enzyme inhibitors, we selected five kinds of bean, using an in-vitro enzyme inhibition assay method. To evaluate the effect of germination process on the functionality of the bean, we investigated the inhibitory activities of the water extracts of non-germinated bean and germinated bean against ${\alpha}$-amylase and ${\alpha}$-glucosidase, relevant to postprandial hyperglycemia. We also investigated the oxygen radical absorbance capacity(ORAC), total phenolics content, and postprandial blood glucose lowering effect in rats(Sprague-Dawley rat model). Most germinated beans showed significantly higher ${\alpha}$-glucosidase inhibitory activity, compared with non-germinated beans. Among germinated beans, Glycine max had the highest ${\alpha}$-glucosidase inhibitory activity(53.3%). The water extract of germinated Phaseolus vulgaris L. had the highest ${\alpha}$-amylase inhibitory activity(95.1%), followed by Glycine max(58.7%), and Glycine max L. Merr(54.1%). Furthermore, the five germinated beans also showed high antioxidant activities in ORAC assay. Results suggested that the germination process may improve and enhance the anti-hyperglycemia potential and antioxidant activity of the bean.
Genetic improvement of the cultivated soybean [Glycine max (L.) Merr] may be possible through hybridization with its wild progenitor, G. soja Sieb. & Zucc. Interspecific cross between G. max (Hwangkeumkong) and G. soja (IT.182932) was made in the summer of 1997. In F$_2$ the percentage of plant height, nodes per plant, and pods per plant were high but gradually reduced from F$_2$ to F$_4$. In contrast pod length, seeds per pod, and 100-seeds weight were increased gradually through generations advanced. Wild variation as evident in F$_2$ in plant height, number of branches, pods per plant, and 100-seeds weight. Twenty six percent of the F$_2$, 44 % of the F$_3$ and 60% of the F$_4$ segregants showed more G. max traits. The combination of useful traits from both species is possible through interspecific hybridization. The characters that could be transferred from wild species to cultivated species are more pod number, better capacity, and resistance to disease and insects. The interspecific derivatives offer scope for selection for high grain yield. Therefore, introducing genes from G. soja to G. max could be contribute to greater genetic diversity of future cultivars. And semicultivated soybean had some desired characteristics including tolerance to adverse environments and multi-seed characters. It means the infusing of semicultivated germplasm to the cultivated soybean could increase number of seeds and pods per plant significantly, and consequently could enhance selecting potential on yield.
The mobilization of proteins in the cotyledons of germinating soybean seeds (Glycine mar [L.] Merr.) and seedlings was studied by using light microscopy and transmission electron microscopy. The cotyledon tissues of soybean. were packed with protein bodies(diameter $0.1-15{\mu}m$) where storage protein of soybean is deposited. Degradation of protein bodies started in the epidermis and vascular tissues. After swelling of the protein bodies, autolysis of storage proteins began while the external membrane remained unbroken. Hydrolysis of proteins could be internal or peripheral and fusion might begin before complete protein degradation. Possible instances of vacuolar fusion were encountered in some cells. In all cases, the result of degradation was the same; the central vacuole of the cell. At the late stages of seedling growth, breakdown of tonoplast was observed in some cells.

A field experiment was conducted to assess the growth characteristics, seed and oil yield of two cultivars of soybean (G max (L.) Merr.) cv. PK-416 ($V_1$) and cv. PK-1024 ($V_2$) in relation to sulphur and nitrogen nutrition. Six combinations ($T_1-T_6$) of two levels of sulphur (0 and 40 kg $ha^{-1}$) and two levels of nitrogen (23.5 and 43.5 kg $ha^{-1}$) were applied to the two soybean cultivars as nutrients. Results indicated significant effect of sulphur and nitrogen, when applied together, on the growth characteristics, yield components, and seed and oil yield. Maximum response was observed with treatment $T_6$ (having 40 kg S and 43.5 kg N $ha^{-1}$). Seed and Oil yields were increased 90 and $102\%$ in $V_1$> and 104 and $123\%$ in $V_2$, respectively as compared to the control i.e. $T_1$ (having 0 kg S and 23.5 kg N $ha^{-1}$). Positive responses of S and N interaction on leaf area index, leaf area duration, crop growth rate and biomass production were also observed. The results obtained in these experiments clearly suggest that balanced and judicious application of nitrogen and sulphur can improve both seed and oil yield of soybean cultivars by enhancing their growth.
Glycine soja Sieb. et Zucc., a predominantly selfing annual, has been served as a reservoir of germplasm for soybean, G. max (L.) Merr., cultivar improvement. This study describes the levels and distribution of genetic variation within and among 22 Korean populations of G. soja using starch gel electrophoresis. The species maintains very similar levels of genetic variability within populations observed in most other annuals. At the population level, the mean percent of polymorphic loci (P) was 32.6%, mean number of allele per locus (A) was 1.32, and mean expected heterozygosity (He) was 0.112. In addition, total genetic diversity (HT) calculated only for polymorphic loci was 0.347. However, significant differences in allele frequencies among populations were found for all loci (P<0.001 in each case) and, on average, about 70% of the total variation in the species is common to all populations. Indirects estimate of the number of migrants per generation (Nm=0.58, calculated from mean GST) indicates that gene flow is low among Korean populations of the species. In addition, analysis of fixation indices revealed a substantial heterozygote deficiency in most populations and at all loci. This indicates that most populations sampled may have been substructed largely due to inbreeding (predominantly selfing) and restricted gene flow, coupled with founder effect and genetic drift. Considering a high genetic divergence among populations, it is recommended that several Korean populations of the species should be preserved, especially such as populations in the eastern and southeastern Korean peninsula with high variation.
A series of aqueous extracts and residues from leaves, stems, roots, pods and seeds of Korean black soybean (Glycine max (L.) Merr.) were assayed against alfalfa (Medicago sativa) and barnyard grass (Echinochloa crus-galli) to determine their allelopathic activities through petri-dish and greenhouse experiments, and the results showed highest inhibition in the extracts or residues from the seeds, and followed by pods. The extracts of 40g dry tissue $\textrm{L}^{-1}$ applied on filter paper in petri-dish bioassay significantly inhibited root growth of alfalfa, and especially extracts from seeds and pods reduced root length of alfalfa more than those from leaves, stems, or roots. Plant height, root length, shoot and root dry weights of barnyard grass were reduced significantly by residue incorporation of seeds and pods as the incorporated amount increased. These results suggest that black soybean plants had herbicidal potential, and their activities were exhibited differently depending on plant parts.
Complex of mulberry extract (CME) is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). In this study, we investigated prevention effects of CME on degenerative arthritis. The $SC_{50}$ value of DPPH radical scavenging by CME was $158.49{\pm}11.35{\mu}/mL$. We found that CME significantly reduced the production of nitric oxide (NO) and protein expression of cyclooxygenase-2 (COX-2) in RAW 264.7 cells which were activated by LPS. Experiments using animal model of degenerative arthritis showed that CME (400 mg/kg body weight) inhibited the production of TNF-${\alpha}$ (77.5%) and IL-$1{\beta}$ (95.0%). Furthermore, it was observed that CME reduced to 85.9% of paw edema induced by carrageenan. These results suggest that CME could improve degenerative arthritis.
Study on effectiveness of Bradyrhizobium japonicum on soybean [Glycine max (L.) Merr.], local cultivar 'Sathiya' were carried out in the garden soil of Trib-huvan University. Different parameters like nodulation, chlorophyll content in fresh leaves and growth of plant in inoculated and uninoculated plant was studied. Pot experiment was conducted in the green house to evaluate the effectiveness of B. japonicum on soybean. It was observed that B. japonicum inoculation increased the number of nodules, shoot length of plant and total chlorophyll content in fresh leaves of soybean plant. However, root length was decreased in all inoculated plants.
