포공영약침액의 발암과정 blocking agent로서의 활성 (Taraxacum mongolieum Hand-Mass Aqua-acupuncture Solution as the Blocking Agent of Carcinogenesis)

  • 포공영약침액의 암억제 효과를 살펴보기 위하여 quinone reductase (QR) 생성유도를 생쥐의 간암세포인 Hepa Iclc7를 이용하여 측정한 결과, 포공영약침액을 처리한 Hepa Iclc7에서 QR생성이 유도되었다. 또한 포공영약침액은 phase II detoxification 효소인 glutathione S-transfer-ase (CST) 활성을 증가시키고 glutathione 생성도 증가시켰다 QR과 CST는 간세포에서 주로 생성되는 phase II효소로 발암물질에 의해 일어나는 돌연변이와 종양화 효과를 막아주고 발암물질을 무독화 시키는 것으로 알려져 있다. 따라서 본 실험에서 포공영약침액은 QR 효소 생성 유도, glutathione 함량 증가 및 CST 효소활성 증대등 phase II detoxification의 기작을 통한 발암과정의 blocking agent로서 효과가 있음을 알 수 있었다.

Enhancement of Anticancer Activities of Kimchi by Manipulating Ingredients

  • To enhance the antitumor activity of Chinese cabbage kimchi, four kinds of kimchi, which ere differently prepared in kinds and levels of sub-ingredients, were fermented at 15$^{\circ}C$ for 1 day and then at 5$^{\circ}C$ up to pH 4.3. The solid tumor formation, hepatic glutathione S-transferase activity and glutathione contents in the liver, and natural killer (NK) cell activity of spleen were determined from the sarcoma-180 cell injected Balb/c mice that were treated with methanol extracts of the kimchi samples. Kimchi IV, prepared with organically cultivated Chinese cabbage, red pepper powder, garlic, Chinese pepper powder mustard leaf and heat processed salt (Gueun salt), reduced the tumor formation by 39.3% compared to the sarcoma-180 cell treated group, resulting in the smallest tumor weight. Methanol extracts of the kimchi III and kimchi IV recovered the activities of hepatic glutathione S-transferase(GST) that was decreased by the transplantation of the sarcoma-180 cells to th mice. The injections of methanol extracts of kimchi II and kimchi IV increased glutathione contents in sarcoma-180 cells treated mice. The methanol extract of kimchi IV increased the natural killer (NK) cell activity of spleen lymphocytes a more effectively (p<0.05) than those the other kimchi samples. These results suggest that the anticancer activities of kimchi can be increased by changing the kinds and levels of sub-ingredients.

Hepatoprotective effect of kasni against aflatoxin B1 induced hepatic damage in mice

  • The efficacy of alcoholic extract of Kasni (Cichorium intybus L.) to control hepatic damage induced by aflatoxin $B_1$ was explored in Swiss albino mice. Aflatoxin $B_1$ was administered orally to the mice with a daily dose of $66.6{\mu}g/kg$ body weight till three months. A signifi-cant increased in thio barbituric acid reactive substances (TBARS) levels with concomitant reduction in enzymatic (glutathione-s-transferase, glutathione peroxidase, superoxide dismutase, and catalase) and nonenzymatic (reduced glutathione) antioxidants were shown in aflatoxin treated group of mice. However, there was a significant reduction in increased TBARS levels and elevation in enzymatic. and non enzymatic antioxidant levels in group of mice which received alcoholic extract of kasni (300 mg/kg bw / day) along with aflatoxin. Histopathological analysis of liver samples also confirmed the hepatoprotective effect of kasni extract. These results suggest that kasni shows hepatoprotective effect against aflatoxin $B_1$ induced hepatic damage in mice.

녹용대보탕(鹿茸大補湯)이 AAPH로 유발된 산화병태(酸化病態)모델 흰 쥐의 항산화(抗酸化) 작용(作用)에 미치는 영향 (The Anti-oxidative Effect of Oral Administration of NYD (Nocyongdaebo-tang) in Oxidized Rats induced by AAPH)

  • 1. Objectives This study aimed to investigate the anti-oxidative effect of Nocyongdaebo-tang (NYD) decoction in oxidized rats induced by AAPH. 2. Methods AAPH (2.2-azobis-2 aminodinopropane-hydrochloride) was injected intraperitoneally to induce oxidization and NYD was orally administered to the rats. To observe the anti-oxidative effect of NYD, we performed blood chemistry analysis, histological analysis, and evaluated the levels of SOD, catalase, glutathione, NO and MDA in liver. 3. Results & Conclusions 1) Serum Albumin level was increased significantly in the NYD group as compared with the control group and the saline group. 2) Serum LDL-cholesterol level was reduced significantly in the NYD group as compared with the control group. 3) GPT level in liver was reduced significantly in the NYD group as compared with the control group. 4) SOD, catalase activity and glutathione levels were increased significantly in the NYD group as compared with the control group and the saline group. 5) The levels of NO and MDA were reduced significantly in the NYD group as compared with the control group. 6) The degenerative change of liver tissue of NYD group was decreased in comparison with the control group. In conclusion, NYD decoction is considered to have an anti-oxidative effect in rats.

천궁이 Phase II 효소 유도와 Ornithine Decarboxylase 활성에 미치는 영향 (Effect of Cnidii Rhizoma on Phase II Enzyme and Ornithine Decarboxylase Activities)

  • Water extract from Cnidii Rhizoma (CRW) was tested for colon cancer chemopreventive activity by measuring the induction of phase II detoxification enzyme activity [quinone reductase (QR) and glutathione S-transferase (GST)] and glutathion (GSH) levels and ornithine decarboxylase (ODC) activity in cultured human colorectal adenocarcinoma HT-29 cells. CRW inhibited cell proliferation in cultured HT-29 cells. CRW induced QR activity in a dose-dependent manner in a concentration range of 0.1${\sim}$5.0 $mg/m{\ell}$. GST activity was also induced with the treatment of CRW in HT-29 cells. In addition GSH levels was increased with CRW. CRW inhibited ODC activity, a key enzyme of polyamine biosynthesis, which is enhanced in tumor promotion. These results suggest that CRW has colon cancer chemopreventive activity by increasing phase II enzyme activity and GSH levels and inhibiting ODC activity in vitro.

고지방식이에서 서로 다른 불포화 지방산 섭취가 쥐의 혈청과 조직의 Tocopherol함량과 지질과산화물 형성에 미치는 영향 (Effects of Dietary Polyunsaturated Fatty Acids on Tocopherol Contents and Lipid Peroxidation of Plasma and Tissues in Rats Fed High Fat Diet)

  • This study was observed the effect of n6 and n3 polyunsaturated fatty acids of dietary corn oil and fish oil which was supplemented with similar levels of tocopherol in high fat diet on the levels of tocopherol, malondialdehyde ( MDA) productions of plasma and tissues of rats. Also RBC hemolysis, superoxide dismutase (SOD) and glutathione peroxidase (GSH Px) activities In liver were determined. Male Sprague Dawley rats were fed high fat (40%Cal) diet which was different only In fatty acid composition for 6 weeks. Dietary (arts were beef tallow(BT) as a source of saturated fatty acid, corn oil(CO) for n6 linoleic acid (LA) and fish oil(FO) for n3 eicosapentaenoic acid(EPA) + docosahexaenoic acid (DHA). Plasma and liver tocopherol levels were lowered by n3 PUy4 but there was no difference in malondialdehyde(MDA) level by different dietary PUFA. However, MDA content of RBC and hemolysis were increased by n3 PUFA. MDA content, superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities in liver were increased in more unsaturated dietary fat groups. Especially, SOD activity was increased in proportion to the degree of fat unsaturation.

Fumonisin B1-Induced Toxicity Was Not Exacerbated in Glutathione Peroxidase-1/Catalase Double Knock Out Mice

  • Fumonisin B1 (FB1) structurally resembles sphingolipids and interferes with their metabolism leading to sphingolipid dysregulation. We questioned if FB1 could exacerbate liver or kidney toxicities in glutathione peroxidase 1 (Gpx1) and catalase (Cat) knockout mice. While higher serum levels of thiobarbituric acid reactive substances (TBARS) and sphinganine (Sa) were measured in Gpx1/Cat knockout mice (Gpx1/Cat KO) than wild type mice after 5 days of FB1 treatment, serum levels of alanine aminotransferase (ALT), sphingosine-1 phosphate (So-1-P), and sphinganine-1 phosphate (Sa-1-P) were found to be relatively low. Although Sa was highly elevated in Gpx1/Cat KO mice and wild mice, lower levels of So and Sa were found in both the kidney and liver tissues of Gpx/Cat KO mice than wild type mice after FB1 treatment. Paradoxically, FB1-induced cellular apoptosis and necrosis were hastened under oxidative stress in Gpx1/Cat KO mice.

식이성 Tungstate가 사염화탄소 투여에 의한 흰쥐 간 손상에 미치는 영향 (Effect of Dietary Tungstate on the Liver Damage in $CCl_4$-treated Rats)

  • To evaluate the role of xanthine oxidase in liver damage by CCl4, a group of rats were fed tungstate for a month, which suppressed the activities of xanthine oxidase in serum and liver. Control group of rats were fed standard diet without tungstate. Liver damage was induced both in tungstate fed and control groups by two intraperitoneal injections of CCl4 at the level of 0.1ml/100g body weight at intervals of 24 hours. Increases in the levels of serum alanine aminotransferase by CCl4 were significantly smaller in tungstate fed rats than in control rats. Concomitantly, histopathologic changes were less in tungstate fed rats than in control ones. In rats either treated with CCl4 or not, hepatic type O xanthine oxidase activities were remarkably reduced by tungstate feeding. Hepatic aniline hydroxylase activities were higher in rats fed tungstate than control rats when animals were not treated with CCl4, but the enzyme activities were lower in tungstate fed rats than control when they were treated with CCl4. Neither tungstate feeding nor CCl4 treatment caused any significant changes in hepatic glutathione contents, and activities of hepatic glutathione S-transferase, glutathione peroxidase and superoxide dismutase. It is concluded xanthine oxidase reaction augment CCl4 induced liver damage via oxygen free radical system.

사염화탄소로 독성을 유발시킨 일차배양 간세포에 미치는 G009의 효과 (Hepatoprotective Effect of G009 on CCl4-induced Hepatotoxicity in Primary Cultured Rat Hepatocytes)

  • G009, a polysaccharide isolated from the mycelia of Ganoderma lucidum IYO09, showed a hepatoprotective activity against $CCl_4$ induced cytotoxicity in primary cu ltured rat hepatocytes. Incubation of $CCl_4$-intoxicated hepatocytes with G009 significantly reduced the levels of glutamic pyruvic transaminase and sorbitol dehydrogenase released from hepatocytes in the medium. G009 showed antioxidative effect by elevating the activities of glutathione reductase and superoxide dismutase, and the content of glutathione in $CCl_4$-intoxidcated primary cultured rat hepatocytes. Furthermore, G009 significantly elevated glutathione-S-transferase activity in $CCl_4$-intoxicated primary cultured rat hepatocytes. G009 also reduced the production of malondialdehyde, a byproduct of lipid peroxidation. From these results, it could be concluded that G009 exerted hepatoprotective activity against $CCl_4$-induced cytotoxicity through antioxidation.
