Aerobic and Graduated Treadmill Exercise Decreases Blood Glucose Levels, Lipid Levels and Oxidative Stress in an Animal Model of Type 1 Diabetes Mellitus

  • Purpose: Exercise has been shown to be a simple and economical therapeutic modality that may be considered as an effective aid for diabetic mellitus. For example, exercise training increases insulin sensitivity in type 2 diabetes. But we found no reported of how exercise affect type 1 diabetes. This study investigated the impact of aerobic and graduated treadmill exercise regimens on body weight, glucose and insulin concentrations, lipid profiles, and oxidative stress indicators in rats with streptozotocin (STZ) induced diabetes. Glycosylated hemoglobin ($HbA_{1c}$) was determined as an indicator of glucose control during exercise. Methods: In our study, a total of 40 rats were used. Three groups of 10 rats each were given STZ to induce diabetes. The remaining 10 rats became the normal group. After 28 days we determined biochemical parameters such as glucose, glycosylated hemoglobin ($HbA_{1c}$), insulin concentration, serum total cholesterol (TC), triglycerides (TG), and high-density lipoprotein (HDL). Superoxide dismutase (SOD) and catalase activities were also measured. Results: Concentrations of blood glucose and $HbA_{1c}$ in the moderated exercise groups were significantly decreased after 28 days compared with the control group (p<0.05). There was a significant reduction in serum TC and TG in the experimental groups. The activity of SOD increased significantly by 17.70% and 48.25% respectively. Conclusion: These results indicate that physical training and exercise training affects body weight, fasting blood glucose, $HbA_{1c}$, insulin, lipid profiles, and antioxidant status in rats with streptozotocin-induced diabetes. We suggest that graduated treadmill exercise may have therapeutic, preventative, and protective effects against diabetes mellitusby improving glycemic control, oxidant defenses, and lipid metabolism.

Effects of Preoperative Epidural Block with Low Dose Bupivacaine and Morphine on Postoperative Pain, Plasma Cortisol and Serum Glucose in Total Abdominal Hysterectomy (복식전자궁적출술에서 술전 저용량 국소마취제와 Morphine을 이용한 경막외차단이 술후통증, 혈장 Cortisol, 혈당에 미치는 영향)

  • Background: Preoperative blocking of surgical nociceptive inputs may prevent sensitization of central nervous system (CNS) and reduce postoperative pain. The stress responses to surgical trauma consist of increase in catabolic hormones and decrease in anabolic hormones. We studied whether preoperative low dose epidural bupivacaine and morphine could affect postoperative pain, changes plasma cortisol, and serum glucose. Methods: Thirty patients undergoing total abdominal hysterectomy were randomly assigned to one of three groups. General anesthesia was induced in all patients and after that, epidural blocks were done except the control group (n=10) patients. Preoperative block group (n=10) received 0.5% bupivacaine 50 mg and morphine 2 mg epidurally as a bolus before operation and followed by 0.1% bupivacaine $5\;mghr^{-1}$ and morphine $0.2\;mghr^{-1}$ for 10 hours. Postoperative block group (n=10) received the same doses of bupivacaine and morphine under the same method postoperatively. Postoperative pain relief was provided with i.v. fentanyl through Patient-Controlled-Analgesia Pump. Postoperative pain by visual analogue scores (VAS), analgesic requirement (first requirement time, total amounts used), side effects, plasma cortisol level and serum glucose level were compared. Results: Until postoperative 6 hrs, VAS of control group was higher than those of the epidural groups. No difference was observed in VAS between the two epidural groups. First analgesics requirement time and total amounts of used analgesics were not different between the two epidural groups, but first analgesic requirement time of preoperative block group was significantly prolonged compared with control group. Plasma cortisol and serum glucose levels were not different among groups. Conclusions: Low dose preoperative epidural bupivacaine and morphine could not reduce postoperative pain, plasma cortisol level and serum glucose level compared with postoperative block group.

  • Chum salmon (Oncorhynchus keta) is a species of anadromous salmonid inhabiting coastal rivers in the North Pacific and the Arctic in the Bering and is the most widely distributed among Pacific salmon species. It is an important fish species in Korea as the salmon releasing project is being actively carried out. This study was conducted to investigate changes in the physiological activity of O. keta according to water temperature. Three experiments were conducted according to the water temperature and period, and the plasma concentrations of cortisol and glucose were analyzed from the blood samples of the experimental groups. Experiment I is a short-term water temperature experiment, in which water temperature stimulation was given for 4 hours at water temperatures of 12℃, 16℃ (control), 20℃, and 24℃, and a recovery period was given for 4 hours. Experiment II is an experiment in which water temperature stimulation was given for 24 hours, 48 hours, and 72 hours at a high temperature of 24℃, and a recovery period was given for 12 hours, respectively. Experiment III is a long-term water temperature experiment, in which the water temperature was 12℃, 16℃ (control), 20℃, and 24℃ for 8 weeks. As a result of the experiment, in Experiment I, there was no significant difference in the survival rate between the experimental groups, but the concentration of cortisol and glucose in the plasma according to the set water temperature showed a significant difference. In Experiment II, there was no significant trend according to the maintenance time of the high-temperature state, but as the temperature increased, the plasma cortisol and glucose levels significantly increased compared to the control group. In Experiment III, all of the experimental group C (24℃) died in the 1st week, and there was no significant difference in the plasma glucose at the 1st and 8th weeks among the remaining experimental groups.

A Study on the Glucose Level Control and Chronic Complication Rate in Diabetic Compliance Groups with Health Care Regimen (당뇨병 환자의 치료지시 이행군과 비이행군의 혈당과 만성 합병증 발생비교)

  • The purpose of this study is to examine to control of glucose level and the occurrence of chronic complications of diabetes by compliance groups with health care regimen The subjects were consisted of 300 out patients with type 2 diabetes mellitus from beginning of March through the end of April in 2001, who visited at the endocrinology department at Kangnam St. Mary's Hospital of Catholic University in Seoul. The patient's compliance level with health care regimen was assessed at questionnaire. However, the blood glucose level and the occurrence of chronic complications of diabetes were assessed at medical record review. The data were analyzed by SAS program for chi square test and t-test. The results were as follows. 1. Significant differences were found among the factors such as duration of diabetes, the number of participation of diabetes educational program, job, smoking, self monitoring of blood glucose and the methods of diabetes therapy between high and low compliance groups. High compliance group patients had a diabetes longer than low compliance group patients. High compliance group patients more frequently attended the educational program and checked themselves monitoring blood glucose than low compliance group patients. Also, they did not work recently, smoked less and got more insulin injection therapy compared to low compliance group patients. 2. No significant differences were found among the result of fasting blood glucose. 2-hour postprandial blood glucose, and $HbA_{l}c}$ between high and low compliance groups. 3. The occurrence rate of macrovascular complications of chronic complications of diabetes were lower, however, the occurrence rate of microvascular complications were higher in high compliance group than in low compliance group with health care regimen.

Effect of Buckwheat Supplementation on Blood Glucose Levels and Blood Pressure in Rats (메밀 보충급여가 백서의 혈당 및 혈압에 미치는 영향)

  • To investigate possible effects of buckwheat on blood glucose level and blood pressure, Sprague-Dawley rats were divided into two groups and fed either AIN-76 diet or modified AIN-76 diet with buckwheat flour for 4 weeks. At the end of experiment, determinations of blood pressure and blood glucose level(glucose tolerance test) were performed. In order to determine insulin levels in both fasted and glucose loaded serum, the animals in both groups were subdivided into two groups. Blood pressure was slightly decreased in the group fed buckwheat( 6mmHg) but not significantly. Glucose tolerance curve of the animal fed buckwheat flour showed clearly different pattern from that of the control group with decreasing tendency. Mobilization of serum insulin was 1.5 times faster in buckwheat group than in control group 1 hour after glucose loading. Further research is needed to find what substance(s) in buckwheat do this role.

  • UDP-glucose regeneration system using metabolic engineeringis unique and efficient strategy for oligosaccharide synthesis. To exploit the efficient UDP-glucose regeneration system, we introduced four enzymes, which would be important in partitioning the flux of UDP regenerationsuch as UDP-glucose pyrophosphorylase, UDP-Kinase gene, UDP-galactose 4-epimerase, and $\beta$-1, 4-galactasyltrasnsferase, into E. coli AD202. To determine the optimal expression level for UDP-regeneration, LacNAc concentration was compared depending on IPTG concentration. 0.5 mM IPTG induction showed the higher oligosaccharides synthesis. Using metabolic engineering under optimal IPTG induction, LacNAc synthesis of AD202/pQNGLU increased until 16 h and showed the 1.34 mM. This concentration is 10 times higher than that of control strain at same reaction time. Lactose of AD202/pQNGLU showed the maximum synthesis of 0.39 mM at 16 h and showed the 2.6 times higher than that of control strain.

Effect of fructose or sucrose feeding with different levels on oral glucose tolerance test in normal and type 2 diabetic rats

  • This study was designed to determine whether acute fructose or sucrose administration at different levels (0.05 g/kg, 0.1 g/kg or 0.4 g/kg body weight) might affect oral glucose tolerance test (OGTT) in normal and type 2 diabetic rats. In OGTT, there were no significant differences in glucose responses between acute fructose- and sucrose-administered groups. However, in normal rats, the AUCs of the blood glucose response for the fructose-administered groups tended to be lower than those of the control and sucrose-administered groups. The AUCs of the lower levels fructose- or sucrose-administered groups tended to be smaller than those of higher levels fructose- or sucrose-administered groups. In type 2 diabetic rats, only the AUC of the lowest level of fructose-administered (0.05 g/kg body weight) group was slightly smaller than that of the control group. The AUCs of fructose-administered groups tended to be smaller than those of the sucrose-administered groups, and the AUCs of lower levels fructose-administered groups tended to be smaller than those fed higher levels of fructose. We concluded from this experiment that fructose has tendency to be more effective in blood glucose regulation than sucrose, and moreover, that smaller amount of fructose is preferred to larger amount. Specifically, our experiments indicated that the fructose level of 0.05 g/kg body weight as dietary supplement was the most effective amount for blood glucose regulation from the pool of 0.05 g/kg, 0.1 g/kg and 0.4 g/kg body weights. Therefore, our results suggest the use of fructose as the substitute sweetener for sucrose, which may be beneficial for blood glucose regulation.

Effects of Modified Sedang-Hwan added Hog Pancreas on the Experimental Diabetes of Rat induced by Streptozotocin (저췌(猪膵)를 가미한 세당환변방(世糖丸變方)이 Streptozotocin으로 유발된 흰쥐의 실험적 당뇨에 미치는 영향)

  • Objectives We performed this study in order to investigate the effects of Sedang-Hwan(世糖丸) and modified Sedang-Hwan(世糖丸變方)on the diabetes mellitus. Methods We injected a vein with 65mg/kg of streptozotocin(STZ) on the rats. And then administered Sedang-Hwan(Sample 1 group); 18.7mg/kg/day, modified Sedang-Hwan; 16.5mg/kg/day(Sample 2 group) to Sample groups and observed the body weight, glucose and insulin levels. Results 1. The Sample 1, 2 groups showed a high suppressive effect of body weight loss compared to Control group. 2. The Sample 1, 2 groups’ glucose level showed a effective in lowering level compared to Control group. 3. The sample 1, 2 groups showed a higher insulin level than Control group. Conclusions Conclusively, modified Sedang-hwan was recognized to have decrease effect of serum glucose of the diabetic rats induced by streptozotocin. It is also required to study on the further detailed mechanism of decrease effect of serum glucose by modified Sedang-hwan.

On-line Monitoring of a Glucose Concentration on a Fermentation Process of Wine for an Automatic Control of a Fermentation Process (발효공정 자동제어를 위한 포도주 발효 중 포도당 농도 온라인 측정)

  • A flow injection analysis method (FIA), which analyzes sample conditions after injecting a sample and reagents into a continuous stream, are recognized as the most adequate analyzing method according to the increase of sampling frequency, the decrease of measuring time and the diversity of measuring targets. Specially, the FIA is considered to be used effectively for the control of a fermentation process to produce fermentation food and useful microbial production by activation of a fermentation industry for development of biological materials. In this study, a flow injection analysis sensor unit was developed for on-line monitoring of the fermentation process. The performance was verified by on-line measuring the concentration of glucose of the fermentation process of wine. The glucose concentrations of the samples were measured every 12 hours during the whole fermentation process and compared with those by a HPLC. The concentration relative errors of glucose on the fermentation process of wine showed below 30% within 72 hours and over 50% after the 72 hours. The sensor unit had potential to on-line monitoring of the fermentation process but some problems to overcome for an commercial application.

The Effect of Anesthetic Information on Pre-operative Anxiety, Glucose, Cortisol and Epinephrine of Patients Undergoing Tonsillectomy (마취관련 정보제공이 편도선 절제술 환자의 불안, 혈당, 코티졸 및 에피네프린에 미치는 효과)

  • Purpose: The purpose of this study was to ascertain the effects of anesthetic information on preoperative state anxiety, plasma glucose, cortisol and epinephrine for patients under tonsillectomy and to provide generic data with anesthetic nursing intervention. Method: Data were collected from 60 patients who had tonsillectomy from November 1, 2003 to April 30, 2004. The group were divided into experimental group and control group: an experiment group of 30 patients was provided with an anesthetic information prepared by the researchers; a control group of another 30 patients was provided with general information. Data were analyzed through chi-squared test, t-test, repeated measure ANOVA using SPSS Program(version 12.0). Result: There were no significant difference between the experimental group and the control group in pre-operative state anxiety, glucose and cortisol. However, there were statistically significant difference between the above two groups in pre-operative pre-operative blood pressure(p=.001), heart beat(p=.000), and epinephrine(p=.035). Conclusion: The authors, with the results of this study, concluded that the preoperative anesthetic information for patients under tonsillectomy had direct effect on lessening pre-operative anxiety.
