Biological oxidation of D-sorbitol to L-sorbose using permeated and immobilized cells of Gluconobacter suboxydans was carried out to investigate the optimum reaction condition. The stabilization effect of cross-linking agents such as glutaraldehyde, tannic acid, and polyethylene imine to prevent the leakage of enzymes from beads containing permeated and immobilized cells of G. suboxydans was examined by the production of L-sorbose from the mixture of D-sorbitol and gluconic acid. The protein concentration effused from immobilized beads treated with only glutaraldehyde was $5.2\mug/m\ell$ after 20 h. The beads of G. suboxydans immobilized with alginate and cross-linked with 0.3% glutaraldehyde was the most useful for the oxidation of D-sorbitol to L-sorbose.
The D-xylulokinase from Candida sp. L-16 was purified through a sequence of ammonium sulfate fractionation, DEAE-cellulose chromatography, Sephadex G-100 and Sephadex G-200 gel filtration. The specific activity of the purified Dxylulokinase was increased to 23.2 fold and the yield was 11.2%. The enzyme was showed to be a single protein band by SDS-PAGE. The molecular weight of the enzyme was 150,000 dalton, this enzyme was identified to be a dimer with two subunits. The optimum conditions of the enzyme were pH 8.0 and 40$\^{C}$, respectively. The enzyme was relatively stable between pH 7.0 to pH 9.0, but it was unstable over 30$\^{C}$. The enzyme showed substrate specificity on D-xylulose, D-arabinose and D-ribose, Km value and Vmax for D-xylulose were 0.042 mM and 117 units/ml, respectively. The activation energy of the enzyme was 4.75 Kcal/mol. The one was inhibited by metabolic intermediates such as 6-phosphogluconic acid, 2-keto-gluconic acid. The enzyme was activated by EDTA and thiol compounds such as cysteine-HCI, DTT and glutathione.

This study investigated the culture characteristics of Cryptoporus volvatus, whichis grow naturally in Korea, to determine the suitable environmental conditions for its cultivation. The physiological characteristics of the mycelia were assessed according to the cultivation conditions, to determine the optimal conditions for artificial cultivation. The visual characteristics of the hyphae of Cryptoporus volvatus KACC52303 included an irregular and uneven surface and a fuzzy or cotton-like texture. Under the microscope, its microstructure showed pre-chlamydospore formation, but no clamps were seen. The appropriate culture temperature was found to be a medium/high temperature of approximately 25-30℃, and the optimal pH was found to have a wide range from weakly acidic (pH 4) to neutral (pH 7). In the optimal nutrient source experiment, hyphal growth was shown to be fair in a mixed medium with 2.5% dextrin as the carbon source and 0.1% yeast extract as the organic nitrogen source. Among the various amino acids, organic acids, and inorganic salts tested, the fastest hyphal growth was observed in the presence of leucine, acetic acid or gluconic acid, and KCl or KH2PO4, respectively. The column test showed that the best mycelial growth occurred in a mixed medium of 80% pine sawdust, 10% rice bran, and 10% corncob sawdust.
When hot weather concrete is utilized, the cooling methods of cooling pipe, liquid nitrogen, ice, etc., are used to prevent the poor consistency and cold joint due to high temperature. These methods, however, spike the production cost and energy consumption, and make quality control difficult. Among these methods is one that involves the use of a retarder. Although economical, retarder is caused difficulty of retarded hardening and setting time control due to inaccurate weighing and poor working condition. Therefore, how to make a tablet for hot weather concrete, as with the existing pharmacy and foods, is discussed in this study, including the following items: mortar setting time, flow test by elapsed time, physical and mechanical properties of concrete. As a result, gluconic acid is superior to lignosulfonic acid and the possibility of using them for such purpose without quality degradation was confirmed in this study, when retarder is tabletting.
Well-aligned Zinc oxide (ZnO) nanowires were synthesized on silicon substrates by a carbothermal evaporation method using a mixture of ZnO and graphite powder with Au thin film was used as a catalyst. The XRD results showed that as-prepared product is the hexagonal wurzite ZnO nanostructure and SEM images demonstrated that ZnO nanowires had been grown along the [0001] direction with hexagonal cross section. As-grown ZnO nanowires were coated with glucose oxidase (GOx) for glucose sensing. Glucose converted into gluconic acid by reaction with GOx and two electrons are generated. They transfer into ZnO nanowires due to the electric force between electrons and the positively charged ZnO nanostructures in PBS. Photoluminescence (PL) spectroscopy was employed for investigating the movements of electrons, and the peak PL intensity increased with the glucose concentration and became saturated when the glucose concentration is above 10 mM. These results demonstrate that ZnO nanostructures have potential applications in biosensors.
미생물학적 물질변환에 대해서는 인류 초기에서부터 효모를 이용하여 빵, 유제품, 알코올, 음료 등의 생산에 이용하여 왔으며, 주로 농업분야 또는 식품 분야에 국한되어 왔다. 1862년 Pasteur에 의해 bacterium xylinum의 순수 배양균주를 사용하여 알코올로부터 초산을 만드는데 응용한 것이 본격적인 시발점으로 보아 무방하겠다. 그 후 acetobacter aceti에 의한 포도당으로부터 gluconic acid 생산과 acetobacter sp.에서의 sorbitol로부터 sorbose 생산 등이 이루어졌고 정통적인 유기합성 방법에 의해 쉽게 만들 수 없는 반응들에 응용되기 시작하였다. 인류 초기의 혼합배양에 의한 물질변환에의 응용과는 달리 순수배양으로 미생물, 식물세포, 혹은 정제된 효소들에 의해 반응이 이루어지게 되었고 순수한 특정 물질에 대한 새로운 순수물질로의 선별적인 수식도 가능해지게 되었다. 특히 발효와 bioconversion의 차이는 racemates의 분리, 비슷한 반응성을 갖는 여러 기들로부터 특정기능을 갖는 기만의 선별적인 수식, 입체 이성체(chiral center)의 제작, 특정 비활성화된 탄소의 기능 등이 bioconversion에서만 수행할 수 있는 독특한 영역으로서 정밀화학분야, energy 분야, 환경오염분야에서의 특히 미래의 관심 기술로 대두되고 있다.
We investigated the anti-proliferative effects of lxeris sonchifolia H. (godulbaegi) root extracts. luteolin(3'. 4', 5. 7-Q-glucoside and 3'. 4', 5, 7-tetrahydoxyflavone) and apigenin (3', 4'. 5. 7-O-gluconic acid) on HepG2 (p53 wild type) cells. Hep3B (p53 null) cells, and Chang liver cells. In MTT assay 3', 4'.5. 7-tetrahydoxyflavone showed the most efficient anti-proliferative effects on these three cell lines. However, there was no significant anti-proliferative effect on Chang liver cell line in MTT results. (omitted)
Penicillium oxalicum strain CBPS-3F-Tsa, an efficient phosphate solubilizing fungus, was evaluated for its production of organic acid in vitro and effect of inoculation on the growth promotion of Maize under greenhouse conditions. The fungus solubilized 129.1, 118.8, and 54.1 mg P/1 of tricalcium phosphate [$Ca_{3}(PO_{4})_{2}$], aluminum phosphate ($A1PO_{4}$),and ferric phosphate ($FePO_{4}$), respectively, after 72 h of incubation. Malic acid, gluconic acid, and oxalic acid were detected in the flasks supplemented with various phosphate sources [240, 146, 145 mM/1 $A1PO_{4},\;FePO_{4},\;and\;Ca_{3}(PO_{4})_{2}$, respectively] together with a large amount of malic acid followed by the other two. The effects of inoculation of P. oxalicum CBPS-3F-Tsa on maize plants were studied under pot culture conditions. P. oxalicum CBPS-3F-Tsa was inoculated to maize plants alone or together with inorganic phosphates in the form of fused phosphates (FP) and rock phosphates (RP). Inoculation of P. oxalicum CBPS-3F-Tsa increased the plant growth and N and P accumulation in plants, compared with control plants, and also had positive effects when applied with RP. The results of this study show that the fungus P. oxalicum strain CBPS-3F-Tsa could solubilize different insoluble phosphates by producing organic acids, particularly malic acid, and also improved the efficiency of RP applied to maize plants.
Poly(3-hydroxybutyrate)(P(3HB)) accumulation under nutrient-rich condition with various amounts of $(NH_4)_2 SO_4$ was systematically investigated. The results of the electron-microscopy and the solvent extraction showed that the P(3HB) accumulation is unavoidable even under nutrient-rich condition. This indicates that in a two-step culture of Alcaligenes eutrophus H16, the researches should be careful in interpreting the data of polyhydroxyalkanoates(PHAs) accumulation in terms of the carbon-source fed in the second step because the two-step culture product contains the P(3HB) produced under nutrient-rich condition. The polyester production capability in a two-step batch culture of A. eutrophus H16(ATCC 17699) was also investigated using various saccharides and their derivatives such as glucose, fructose, gluconic acid, glucaric acid, sorbitol, lactose, galactose, and mannose. The polyesters synthesized were characterized by 500 MHz$^{1}H-NMR$ spectroscopy, intrinsic viscosity$[\eta]$ measurement in chloroform and differential scanning calorimetry(DSC). 500 MHz $^{1}H-NMR$ analysis showed that all polyesters synthesized generally contained 1~2 mol% of 3HV. Another finding is that the glucose utilization can be increased by changing the autoclaving procedure of the substrate to enhance the P(3HB) production yield up to 46 wt% of P(3HB) in dry cells.
Pantoea agglomerans DSM3493 was isolated from green house soils collected from Chungchugnam-do province, Gongju-Gun area in South Korea and phosphate solubilization and organic acid production of the strain were assessed using three types of insoluble phosphate sources (Ca phosphate, Fe phosphate and Al phosphate) under three different pH conditions (7, 8 and 9). The highest Ca phosphate solubilization ($651{\mu}g\;mL^{-1}$) was recorded at pH 7 followed by pH 8 and 9 (428 and $424{\mu}g\;mL^{-1}$ respectively). The solubilization rate was found to be 80.4, 98.1 and $88.7{\mu}g\;mL^{-1}$ (for Fe phosphate containing medium) and 9.3, 12.1 and $29.8{\mu}g\;mL^{-1}$ (for the Al phosphate containing medium) respectively at pH 7, 8 and 9. Though increasing pH of the medium caused reduction in the rate of solubilization of Ca phosphate, solubilization of Fe and Al phosphates enhanced with increasing pH. By contrast, the highest amount of organic acid was produced with Ca phosphate while the lowest was recorded with the presence of Al phosphate. Among the organic acids, gluconic acid production was found to be the highest, followed by oxalic acid and citric acid regardless the source of phosphate. Results can thus be concluded that the production of organic acids appears to play a significant role in the inorganic phosphate solubilization.
