Comparison of Ginsenoside Contents in Different Parts of Korean Ginseng (Panax ginseng C.A. Meyer)

  • The present study was conducted to investigate the ginsenoside profiles of the main root, root hair, and leaf of ginseng in order to demonstrate their possible application in medicine. The total ginsenoside content of the leaf was up to 12 times than that in the main root, and the content of protopanaxadiol groups was higher than that of protopanaxatriol groups in all the samples. The leaf was shown to contain high amounts of ginsenosides Rb3 and Rh1, whereas the main root contained large amounts of ginsenosides Rb1 and Rc. Moreover, Rb2, Rb3, and Rg1 were only detected in the root hair, leaf, and main root, respectively. The ginsenoside Re content of Panax ginseng leaf and root hair was 2.6~4 times higher than that of the main root. Therefore, the results indicate that the ginsenoside content of Panax ginseng is higher in the leaf and root hair, and lower in the main root.

  • New methods have been developed to transform Panax ginseng with Ri plasmids of Agrobacterium rhizogenes 15834 and A. rhizogenes A4. Modified leaf disc method was made feasible to establish hairy root culture even when an axonic plantlet was not available as in the case of P. ginseng. The contents of ginsenosides (Rgl, Rf, Rc, Rbl, and Rb2) in hairy roots. were determined by HPLC. Hairy root cultures, established as liquid culture in MS medium, was produced 0.34~1.19% ginsenosides on dry weight basis, and this result is significantly higher level than that of normal P. ginseng.

The Quality Characteristic of Ginseng Cultured in Bioreactor System (생물반응기에서 배양된 인삼의 품질 특성)

  • In the present study, we investigated the quality characteristic of ginseng cultured in bioreactor system and the optimum recipe condition of the liquid tea using cultured ginseng. The contents of soluble solid and crude saponin in cultured ginseng were 31.8% and 1.94%, respectively, which were lower than commercial ginseng. In the concentrated extract, crude saponin content was 4.77% and the contents of ginsenoside Rc, Re and Rg$_1$were 7.36, 4.40 and 1.75 mg/g, respectively. The ginsenoside Rb$_1$and Rb$_2$, main contents of commercial ginseng, were not detected. The optimum ranges of recipe on organoleptic properties of ginseng liquid tea were estimated on 9.0~10.4% of the extract, 6.8~8.1% of apple vinegar and 40% of fructose. The liquid tea using commercial ginseng showed higher scores of sensory lest than the liquid tea using cultured ginseng in bioreactor system at the given condition, 10% of the extract, 7% ofapple vinegar and 40% of fructose, with the same recipe condition ranges.

Studies of Ginseng on the Antistress Effects (인삼(人蔘)의 항(抗)스트레스작용(作用)에 관(關)한 연구(硏究))

  • Two pure saponin components, Panax saponin C (protopanaxatriol derivative, ginsenoside Re) and Panax saponin E (protopanaxadiol derivative, ginsenoside $Rb_l$) were isolated from Panax ginseng root and their acute toxicities in mice and antistress effects in rats were investigated. Average lethal doses $(LD_{50})$ of ginsenoside Re were 130mg/kg (i.v.), more than 1,000mg/kg (i.p.) and more than 1,500mg/kg (s.c.), respectively. Average lethal dose of ginsenoside $Rb_{1}$ was 243mg/kg intravenously. Adrenal ascorbic acid and cholesterol contents were significantly decreased when normal rats were exposed to heat $(40^{\circ}C)$ for 30 min. The reduction of the adrenal ascorbic acid and cholesterol contents in rats was partially prevented when the rats received the ginseng saponins prior to exposure to heat stress and most pronounced effects were observed in rats received ginsenoside Re. However, it was found that administration of ginseng alone, without stress, did not significantly change the ascorbic acid and cholesterol contents in adrenal glands. Eosinophil counts in the blood of the rats were elevated when the rats were exposed to the heat stress, and the elevation of the eosinophil counts were prevented with the ginseng saponins under the stress, but the changes were all insignificant statistically.

  • The analytical conditions of ginsenosides were optimized using high performance liquid chromatography (HPLC). The optimal analysis conditions were set up from the experiments using different gradient conditions, and the ginsenosides contents of red ginseng products and their raw materials were analysed. Red ginseng contained 0.29% Rg1, 0.82% Rb1, 0.38% Rc, 0.32% Rb2, 0.11% Rd, showing the highest ginsenoside contents. Among the red ginseng products, red ginseng extract showed the highest content. The ginsenoside contents were varied according to the raw material type and product type. Re and Rb1 contents were the highest in most raw materials and products.

Saponin and Ginsenoside Content in Korean Red Ginseng Products (고려홍삼 제품류의 사포닌 및 진세노사이드 함량)

  • Red ginseng products manufactured by the Korea Ginseng and Tobacco Corporation were analyzed to determine the crude saponin , total saponin and ginsenoside contentents by gravimetry, spertrometry and HPLC, respectively, to see if effective quality control of the components in the products can be achieved. Medicinal powders, powders, tablets and capsules which were made from ginseng powder showed similarity in saponin content, the ratio of PD to PT saponin, and the ginsenoside content and composition, while extract powder, extract, extract tea, extract pills and tea, which were made of ginseng extract, showed difference in saponin content, ratio of PD to PT saponin, and the content and composition of ginsengside. It is, accordingly, believed that ginseng products which are uniform in contents and saponin composition can be produced by carrying out strict quality control throughout the processes of making raw red ginseng into final products.

Production of Ginsenoside-Rg3 Enriched Yeast Biomass Using Ginseng Steaming Effluent (수삼 증자 시 생성되는 유출액을 이용한 ginsenoside-Rg3 강화 효모 제조)

  • To produce ginsenoside-Rg$_3$ enriched edible yeast, ginseng steaming effluent (GSE) was used for yeast cultivation in this study. Four kinds of edible yeasts were cultured in sterilized GSE (2% w/v, pH 6.5), without any nutrient, for 48 h at 30$^{\circ}C$, and their growth and ginsenoside compositions were determined. Among the yeasts, Saccharomyces cerevisiae showed the highest growth in the GSE medium. 267.1 mg of Saccharomyces cerevisiae biomass was produced from 1 g of GSE solid and ginsenoside-Rg$_3$ contents was determined with 0.033 mg. Saccharomyces cerevisiae also showed the best overall acceptability, with a herbal and fermentative flavor and a slightly bitter taste. From these data, we conclude that Saccharomyces cerevisiae is the excellent strain for production of ginsenoside-Rg$_3$ enriched edible yeast using GSE.

Effect of Nitrogen Phosphorus and Potassium on Ginsenoside Composition of Panax Ginseng Root Grown with Nutrient Solution (영양액재배 인삼근의 진세노사이드 조성에 미치는 N.P.K.의 영향)

  • Panax ginseng seedlings were grown in vermiculite with nutrient solution different in nitrogen, phosphorus ana potassium level. Ginsenoside contents of root were investigated by high performance liquid chromatogram. Elimination or increase of one of N.P.K. increased or decreased total saponin content. Nitrogen was most effective (15.5% for-N to 8.9% for 3N) and potassium least. Similar trend was shown in each ginsenoside. According to coefficient of variation in one nutrient treatment or among all nutrient treatments ginsenoside Re was most insensitive to nutrient change and also other environmental factors and Rd most sensitive. Diol content (PD) was more variable than triol (PT) and variation of PT/PD was about half of them. Variation of ginsenoside content by nutrient change had no relation with the ginsenoside content. Similarity of ginsenoside pattern slightly decreased with the difference of saponin content by nutrient change. Root weight was significantly small only in tap water plot.

The Changes of Physicochemical Characteristics and Quality Stability of Korean Red Ginseng (Panax ginseng C.A. Meyer) Stored over 20 Years (20년 이상 장기저장된 홍삼의 이화학적 특성변화 및 품질안정성)

  • This study was investigated the changes of quality stability and physicochemical characteristics of the Korean red ginsengs stored for a long times over 20 years. The Korean red ginsengs were stored for 4 to 22 years in canned packaging with polypropylene film and wooden box at room temperatures. The unusal phenomena such as discoloration and pin hole in packaging were not observed. General bacteria showed the vlaues of below 100 CFU/g, coliform groups and molds were not found in any samples stored for 22 year. Any samples also were not detected in mycotoxins. The contents of moisture, ash and crude saponin were the levels of 10.6~11.1%, 3.8~4.2% and 4.1~4.7% during the whole storage periods, respectively. The contents of maltol, which has been known as characteristic flavour and antioxidant of Korean red ginseng, showed remarkably increasing tendency from 0.10 mg/g for 4 years to 2.53 mg/g for 22 years during the storage. The contents of AFG (arginyl-fructosyl-glucose), arginine and free sugar were slightly decreased. Acidic polysaccharide and ginsenoside were not changed significantly during the storage periods. The contents of acidic polysaccharide and total ginsenosides were the 75.1~76.3 mg/g and 15.1~16.6 mg/g, respectively. The sums of ginsenoside-Rg1,-Rb1 and -Rg3s were the ranges of 9.3~9.9 mg/g and PD (ginsenoside-Rb1, -Rb2,-Rc,-Rd,-Rg3s,-Rg3r)/PT (ginsenoside-Rg1,-Rg2,-Re,-Rf,-Rh1) saponin ratios were the levels of 1.4~1.5. These results suggest that Korean red ginsengs stored for long periods show relatively stable quaility stabilities and not significantly changed the contents of ginsenoside and polysaccharide during the storage up to 22 years.

Effect of Variety and Shading Material on Growth Characteristics and Ginsenoside Contents of 2-Year-Old Ginseng (Panax ginseng C. A. Meyer) Grown in Imperfectly Drained Paddy Soil (배수약간불량 논토양에서 품종 및 해가림 피복물 종류가 2년생 인삼의 생육과 진세노사이드 함량에 미치는 영향)

  • To selection of optimal shading material, two-year-old ginseng (Panax ginseng C. A. Meyer) of new cultivar, 'Cheonpoong' (CP), and native species 'Hwangsookjong' (HS) were cultured under three kinds of shading materials such as three-layered blue and a one-layered black PE (polyethylene) net (TBSB), blue PE sheet (BS), and aluminium coated PE sheet (AS) in imperfectly drained paddy soil. Growth characteristics, yield and ginsenoside contents were investigated under three shading materials. Yield and ginsenoside contents of ginseng were distinctly affected by intensity and quality of sunlight penetrated through shading materials. Light transmission ratio, air and soil temperature according to shading materials were higher in order of BS, AS, and TBSB. However, ratio of aerial phase and porosity of the soil were higher in order of AS, BS, and TBSB, respectively. There was no significantly difference in the ratio of rusty colored root by shading materials. CP showed higher stem length, leaf area, and root weight than that of HS, while the former showed distinctly lower discolored leaf ratio than that the other. Eight kinds of ginsenosides content of CP were higher than that of HS in $Rg_1$, Re, Rf, $Rb_1$ and Rc except $Rg_2$, $Rb_2$, and $Rb_3$. Total ginsenoside contents of CP was distinctly higher than that of HS. Total ginsenoside contents as affected by shading materials was higher in order of BS, TBSB, and AS in CP, while TBSB, BS, and AS in HS.
