A Study on the Amendment Scheme of Ginsenoside Content Standard Regulation for Red Ginseng Products in Korea (홍삼가공품의 Ginsenoside 함량 규격기준 개선방안 연구)

  • Red ginseng is a widely used dietary supplement and medicinal herb, and there are so many forms of ginseng products including tea, extract, capsule and jelly. The purpose of the present study was to propose some amendments on ginsenoside content of red ginseng products in Korea. For this purpose, we analyzed red ginseng products for simultaneous determination of 26 ginsenosides by ultra performance liquid chromatography with diode array detector. Some developmental aspects of Korea's ginsenoside content standard regulations for red ginseng products are needed to be examined as follows : Firstly, we proposed that four ginsenosides ($Rb_1$, $Rg_1$, Rf and $Rg_3$) would be detected in red ginseng products. Secondly, in case of red ginseng extracts, the sum of $Rb_1$, $Rg_1$ and $Rg_3$ would be 4.0 mg/g. The two proposals are helpful to comprehensive evaluation of quality of red ginseng products. In conclusion, the scientific studies on amendment scheme of ginsenoside content standard regulation of red ginseng product are very important to fortify quality control.

Identification of Saponin and Sapogenin in Root, Leaf and Stem of Ginseng by Thin Layer Chromatography (얇은막 크로마토그래피에 의한 인삼(人蔘)의 근(根) 엽(葉) 및 경(莖)의 saponin 및 sapogenin화합물(化合物) 동정(同定))

  • Saponins of ginseng root, leaf and stem were identified by TLC. Eleven unknown spots were detected in ginseng leaf and ten unknown spots in ginseng stem on TLC besides seven ginsenosides such as $ginsenoside-Rg_1,\;-Rf,\;-Re,\;-Rd,\;-Rc,\;-Rb_2,\;and\;-Rb_1$ which are contained in ginseng root. $Ginsenoside-Rg_3\;and\;-Rg_2$ were identified on TLC from mild hydrolysates with 50% acetic acid of total saponins from ginseng root, leaf and stem. Meanwhile, panaxadiol, panaxatriol and oleanolic acid were identified from hydrolysates with 7% ethanolic sulfuric acid of total saponin of ginseng root, while panaxadiol and panaxatriol from those of total saponins of ginseng leaf and stem.

  • The stability of red ginseng saponin in aqueous solution was studied with the acceleration test method. The degradation rate constant of ginsenoside Rb1, an index component of red ginseng saponin, was $2.371{\times}10^{-4}\;day^{-1}$ at $20^{\circ}C$, and the shelf-life was about 570 days. The pH-rate profile demonstrated that the most stable range was pH 6-8. Mannitol and benzyl alcohol, common excipients for injection, exerted no influence on the degradation reaction of ginsenoside Rb1.

Influence of Ginsenoside Rb1 on Brain Neurosteroid during Acute Immobilization Stress

  • This study examined whether or not acute stress is linked to increases in the neurosteroid levels, which is a well-known neurotransmitters associated with stress stimuli. The ginsenoside, Rb1, was tested in order to better understand its potential effects on altering the neurosteroid levels and ultimately attenuating stress. The optimal stressed condition was checked by measuring the 5a-dihydroprogesterone (DHP) and allopregnanolone (THP) levels in the brain after immobilization stress at various times. Based on this result, an acute stress model was set up to give 30 min of immobilization stress. The DHP and THP brain levels of the stressed mice were then investigated after administering Rb1 orally (10 mg/kg). These results were compared with the neurosteroid level in the stressed mice not given Rb1. Saline was administered orally to the nonstressed mice to check the placebo effect. Acute immobilization stress induced an increase in the THP and DHP concentration in the frontal cortex and cerebellum. When Rb1 was administered orally prior to immobilization stress, the THP level in the frontal cortex and cerebellum was significantly lower than that in the stressed animals not given Rb1. On the other hand, the DHP level was lower in the cerebellum only. This suggests that the metabolism of the brain neurosteroids is linked to psychological stress, and Rb1 attenuates the stressinduced increase in neurosteroids.

The Ginsenoside-Rb2 lowers cholesterol and triacylglycerol levels in 3T3-L1 adipocytes cultured under high cholesterol or fatty acids conditions

  • The effects of the ginsenoside Rb2 (Rb2) on lipid metabolism were characterized in 3T3-L1 adipocytes to evaluate their utility for treating obesity. While the amounts of total cholesterol and triacylglycerol (TAG) were markedly increased in the adipocytes treated with high amounts of cholesterol and fetal bovine serum (FBS), the test groups treated with Rb2 showed levels that were close to normal. The effect of Rb2 on these cells was comparable to that of lovastatin. Rb2 enhanced the expression of the sterol regulated element binding protein (SREBP) mRNA whereas treatment with cholesterol and FBS led to a reduction in the abundance of this transcript. The activity of fatty acid synthetase (FAS) was lower in the cholesterol group compared to the Rb2 treatment group suggesting that the observed decrease in cholesterol levels and activated SREBP was mediated by Rb2. Treatment with Rb2 also resulted in a decrease in TAG levels in adipocytes cultured under high fatty acid conditions. This effect was mediated by stimulating the expression of SREBP and Leptin mRNA, suggesting that Rb2 might be a valuable component capable of lowering the levels of lipids.

Ginsenoside Content of North American Ginseng (Panax quinquefolius L. Araliaceae) in Relation to Plant Development and Growing Locations

  • North American ginseng (Panax quinquefolius L.) was analysed for total ginsenosides and ten major ginsenosides (R$_{0}$ , Rb$_1$, Rb$_2$, Rc, Rd, Re, Rf, Rg$_1$, pseudoginsenoside F$_{11}$ and gypenoside XVII), and variations in ginsenoside content with age of plant (over a four-year-period) and geographic location (Ontario versus British Columbia) were investigated. In the roots the total ginsenoside content increased with age up to 58-100 mgㆍg$^{-1}$ dry weights in the fourth year, but in leaves it remained constant over time. Roots and leaves, moreover, had different proportions of individual ginsenosides. The most abundant ginsenosides were Rb$_1$ (56mgㆍg$^{-1}$ for Ontario; 37mgㆍg$^{-1}$ for British Columbia) and Re (21mgㆍg$^{-1}$ for Ontario; 15 mgㆍg$^{-1}$ for British Columbia) in roots, and Rd (28-38 mgㆍg$^{-1}$ ), Re (20-25 mgㆍg$^{-1}$ ), and Rb$_2$ (13-19 mgㆍg$^{-1}$ ) in leaves. Measurable quantities of Rf were found in leaves (0.4-1.8 mgㆍg$^{-1}$ ) but not in roots or stems. Our results show that ginsenoside profiles in general, and Rf in particular, could be used for chemical fingerprinting to distinguish the different parts of the ginseng plant, and that ginseng leaves could be valuable sources of the ginsenosides Rd, Re, and Rb$_2$.

Ginsenoside Production by Hairy Root Cultures of Panax ginseng Transformed With Agrobacterium rhizogenes (Agrobacterium rhisogense에 의해 형질 전환된 인삼의 모상근 배양에서 Ginsenoside의 생산)

  • New methods have been developed to transform Panax ginseng with Ri plasmids of Agrobacterium rhizogenes 15834 and A. rhizogenes A4. Modified leaf disc method was made feasible to establish hairy root culture even when an axonic plantlet was not available as in the case of P. ginseng. The contents of ginsenosides (Rgl, Rf, Rc, Rbl, and Rb2) in hairy roots. were determined by HPLC. Hairy root cultures, established as liquid culture in MS medium, was produced 0.34~1.19% ginsenosides on dry weight basis, and this result is significantly higher level than that of normal P. ginseng.

  • 생약의 화학적 특성에 대한 계속적인 연구가 이루어짐에 따라 우리는 홍삼 및 백삼의 화학성분을 상대적으로 규명하였다. 홍삼은 극성이 약한 분획에서 5개의 새로운 배당체(20R-ginsenoside $Rg_{2},\;Rh_{1};20R$, 20S-ginsenoside $Rg_{3}; ginsenoside\;Rh_{2}$와 새로운 아세칠렌 화합물(Panaxytriol)을 함유하는 특징적인 성분들이 gins - enoside Rh1, Rg2와 함께 분리되었다. ginsenoside Rh2는 배양된 종양세포에 대해 세포독소 효과를 보여주었다. 백삼은 수용성 분획에서 특징적인 성분이 있는 것으로 밝혀졌으며, 여기에서 malonly-ginsenosides Rb1, Rb2, Rc 및 Rd로 명명된 새로운 배당체 성분이 분리되었다. Malona-ginsenosides는 백삼에서는 주요한 배당체이지만, 홍삼에서는 검출되지 않았다.

The Quality Characteristic of Ginseng Cultured in Bioreactor System (생물반응기에서 배양된 인삼의 품질 특성)

  • In the present study, we investigated the quality characteristic of ginseng cultured in bioreactor system and the optimum recipe condition of the liquid tea using cultured ginseng. The contents of soluble solid and crude saponin in cultured ginseng were 31.8% and 1.94%, respectively, which were lower than commercial ginseng. In the concentrated extract, crude saponin content was 4.77% and the contents of ginsenoside Rc, Re and Rg$_1$were 7.36, 4.40 and 1.75 mg/g, respectively. The ginsenoside Rb$_1$and Rb$_2$, main contents of commercial ginseng, were not detected. The optimum ranges of recipe on organoleptic properties of ginseng liquid tea were estimated on 9.0~10.4% of the extract, 6.8~8.1% of apple vinegar and 40% of fructose. The liquid tea using commercial ginseng showed higher scores of sensory lest than the liquid tea using cultured ginseng in bioreactor system at the given condition, 10% of the extract, 7% ofapple vinegar and 40% of fructose, with the same recipe condition ranges.

Relationship Between Ginsenoside Content and Stem Color Intensity of Panax ginseng (경색별인삼근(莖色別人蔘根)의 Ginsenoside 함량(含量))

  • Ginsenosides in epidermis·cortex(EC) and xylem-pith(XP) of main body of Panax ginseng(var. atropurpureacaulo) root were investigated in relation to dark purple area on stem. Pattern of ginsenosides, ratio of protopanaxatriol(PT) to diol(PD) and total ginsenoside content were significantly different between EC ana XP, and not related with stem color. The increasing trend of total ginsenosides with decreasing in purple area on stem needs to be tested with greater sample size. The order of ginsenoside content was $Rb_1>Rg_1>Re>Rc>Rg_2>Rb_2>Rf>Rd$ for EC, $Rg_1>Rb_1>Rg_2>Re>Rb_2>Rc>Rf>Rd$ for XP. PT/PD was 1.08 for EC,1.95 for XP. Since total ginsenoside content was 3 times higher in EC than in XP and weight of two parts was almost same, the content of ginsenosides of main body mostly depends on those of EC.
