Characteristics of New Cultivars in Panax ginseng C.A. Meyer (고려인삼 신품종 특성)

  • This paper reports the characteristics of 8 new cultivars for selected from Korean ginseng. The occurance of multi stems were the highest in Yunpoong(45%) and the lowest in Gumpoong(7%), but growth of aerial parts were the highest in Gumpoong and the lowest In Yunpoong among new cultiyars. The ratio of seeds harvest were the highest in Gumpoong(85.4%) and the lowest in Chunpoong(69.1%), but number of seeds per plant were the highest in Yunpoong(108.3ea) and the lowest in Chunpoong(77.5ea) among new cultivars. The ratio of leaf burning were the highest in Chunpoong but the lowest in Yunpoong among new cultivars. In weight distribution of the different parts of the ginseng roots, the ratio of main root were high in Jakyungjong(63.1%) but low in new cultivars(49%-55.9%), but the ratio of lateral root were high in new cultivars(19.3-23.3%), but low in Jakyungjong(13.2%), the ratio of fine root were not different. Root yield declined in the order of Yunpoong, Gumpoong, Gopoong, Chunpoong, Sunpoong, Jakyungjong. The length of main root were the longest in Chunpoong(8,0cm) but the shortest in Yunpoong(6,4cm), The ratio of rusty-root was low in new cultivars(0,2-9,5%), but high in Jakyungiong(16,3%). The grade of red ginseng roots decreased in the order of Chunpoong, Gumpoong, Gopoong, Sunpoong, Yunpoong, Cheongsun, Jakyungjong. The total ginsenoside contents per dry weight in main roots was high in Gumpoong(8.53mg), Yunpoong(8.13mg), Gopoong(7,47mg), but low in Chunpoong(5.73mg), Sunpoong(4.87mg).

Optimal Desiccation Condition and Moisture Content of Dehisced Seeds of Ginseng (Panax ginseng C.A. Meyer) for Cryopreservation (인삼 개갑종자의 초저온동결보존을 위한 최적 건조조건과 수분함량)

  • This study was conducted to establish the efficient protocols of the germination and cryopreservation of dehisced Korean ginseng seeds for long-term germ­plasm conservation. $GA_3$ and BA treated on seed for 24 hr facilitated germination at 5 and $10^{\circ}C$. Germination percentage of desiccated seeds was decreased under moisture content (MC) of below $7.2\%$. Dehisced ginseng seeds were dried under airflow of laminar floor cabinet and seed drying room. The high levels (more than $90\%$) of germination after cryogenic exposure were obtained after drying under vertical airflow of laminar floor for 12-30 hours (MC $10.6{\~}7.2\%$). Decrease in germination percentage of ginseng seeds due to desiccation damage and freezing injury was observed at MC of below $7.2\%$ and of above $12.1\%$, respectively. Therefore, MC of ginseng seeds need to be controlled with a range of $8{\~}10\%$ to avoid damages from both desiccation and freezing.

Effects of Sodium Hydroxide and Sulfuric Acid on the Embryo Growth of Ginseng Seed (Panax ginseng C. A. Meyer) (NaOH 및 $H_2SO_4$ 처리가 인삼 종자의 배생장에 미치는 영향)

  • Endorcarp inhibited the embryo growth of ginseng seeds. This inhibition is not due to impermeability to water, but is probably caused by mechanical resistance. The embryo growth rate was enhanced by endocarp injury by soaking for 10 to 30 minutes in 2.5% solution of sodium hydroxide. But sulfuric acid did not affect on the embryo growth of ginseng seed.

Production Procedures and Economics of the American Ginseng (미국 화기삼의 종류별 생산방법과 경제성분석)

  • The purpose of this study is classifying types of American ginseng and estimating their production cost and revenue by the types. Usually, the American ginseng can be classified as 4 different types; wild ginseng(WG), wild simulated ginseng(WSG), woods grown ginseng(WGG), and field cultivated ginseng(FCG). This paper estimates costs and benefits for FCG, WGG, and WSG per acre. The WGG & WSG are produced under the tree at mountain while the FCG is produced at large scale farm with machinery. Annual profit for the FCG is $2,222 while that of the WGG and the WSG are $2,759 and $3,799 per acre. Although quantity produced per acre for the WGG and WSG(600lbs and 160lbs) are much smaller than that of the FCG(3,000lbs), prices per pound for the WGG and WSG($125, 375$) are higher than that of the FCG($24). In addition, production costs for the WGG and WSG are lower than that of the FCG because of the costs for seeds, shadow facility, and chemicals are different by the types of production.

Plant Regeneration through Somatic Embryogenesis from Mature Zygotic Embryos of Ginseng(Panax ginseng C. A. Meyer) and Flowering of Plantlets (인삼(Panax ginseng C. A. Meyer)의 성숙한 배로부터 체세포 배발생을 통한 구분화 및 유식물체의 개화)

  • Mature zygotic embryos dissected from ginseng(Panax ginseng C. A. Meyer) seeds were cultured on Murashige and Skoog's (MS) medium containing various concentrations of 2, 4-dichlorophenoxyacetic acid(2, 4-D) and kinetin. Somatic embryos were induced directly from cotyledonary tissue or from intervening callus. The induction frequency of somatic embryos was up to 55%. Upon transfer to half-strength MS medium supplemented with 1 mg/1 6-benzyladenine(BA) and 1 mg/1 GA3, most somatic embryos developed into plantlets. Over 50% of the plantlets flowered after 4 weeks of culture and then a few bore immature fruits in vitro. Therefore, it is suggested that the juvenility of the ginseng tissue which give rise to somatic embryos does not interfere with in vitro flowering of their regenerated plantlets.

  • Ginseng(Panax ginseng C.A. Meyer) is a perennial herbaceous plant which grows very slowly. It takes about 3 to 4 years from seeding to collecting the ripe seeds and the ginseng propagation is very difficult. and so, it is very difficult to breed ginseng plant. Ginseng tissue culture was started from at 1960, and ginseng commercial product by in vitro callus culture was saled, however upto now, regenerants were not planted to soil normally. Recently, plant genetic engineering to produce transgenic plants by introducing useful genes has been advanced greatly. In a present paper, transformation of ginseng plants was achieved by co-cultivation with Agrobacterium harboring the binary vector coding Proteinase-II gene, which confer resistant or tolerant to insect pests, The binary vector for transformation was constructed with disarmed Ti-plasmid and with double 35S promoter. The NPT II gene and introduced genes of the transgenic ginseng plants were successfully identified by the PCR. Especially the transgenic ginseng plants were regenerated using new techniques such as repetitive single somatic embryogenesis.

Localization and Function of Cellulase in Endosperm Cells of Panax ginseng Seeds during Maturation and After-ripening (인삼 종자의 성숙과 후숙 과정에서 배유세포내 섬유소 가수분해효소의 분포 및 기능)

  • The active sites, intracellular transport, function of cellulase in association with the disintegration of the storage materials of the endosperm cells during seed maturation and after-ripening of Panax ginseng C.A. Meyer seeds were studied by electron microscopy. Cytochemical activities of the cellulase occurred in protein bodies and vesicles of endosperm cells in seed with red seed coat. In after-ripening seed, the activities were strongly found in the cell wall of endosperm near the umbiliform layer and on neighbouring vesicles, so it is assumed that these cells begin to be decomposed. Cellulase activities were initiated before the decomposition of storage materials. But, no activity was observed in the umbiliform layer, so it is suggested that cellulase lose its activity after the completion of lysis process.

Mass Production of Eleutherococcus senticosus Plants through in vitro Cell Culture (세포 배양을 통한 가시오갈피(Eleutherococcus senticosus) 묘목의 대량 생산 시스템 개발)

  • Immature zygotic embryos of Eleutherococcus senticosus seeds matured rapidly within one month when the seeds comprising zygotic embryos were pieced to small size and cultured on 1/2 MS medium. Frequency of somatic embryos formation was declined rapidly when the zygotic embryos germinated and grew to plantlets. Embryogenic cells were induced by consecutive subculture of somatic embryos on MS medium with 1.0mg/L2,4-D. After heart-shaped somatic embryos were induced by suspension culture, these embryos were plated onto petri dish to support maturation of embryos. Germination of embryos occurred on medium with 5mg/L GA$_3$and transferred to culture bowl to stimulate the further growth. Frequency of soil survival of plantlets was influenced by soil mixture (perlite and peatmoss). The suitable combination of perlite and peatmoss was 1:5, and the soil survival rate was 78% after 4 months. The soil transferred plantlets were over-wintered in field condition after defoliation. New year sprouting of plants was achieved successfully and they grew to adult plants. These results indicate that the systematic procecure of plant production in E. senticosus for micro propagation.

The Effect of Korean Ginseng on Growth and Harvest of the Rice Plant (고려인삼이 벼의 생장 및 수확에 미치는 영향)

  • When rice seeds were rinsed in If) Korean ginseng tea (KG 722, Korean Ginseng Product Co. Ltd. made) for 28 hours prior to seeding, the early growth was found relatively fast and their resistance against blight and harmful insects was increased. Rice yield showed that control group, rinsing group and spraying group were 514 kg/10a, 562 kg/ 10a and 571 kg/ 10a respectively. However, rinsing and spraying groups were 590 kg/ 10a (leaf spraying) and 605 kg/ 10a (spray immediate after seeding) respectively.
