토양 훈증 및 녹비작물 재배가 인삼의 연작장해 경감에 미치는 영향 (Effect of Soil Fumigation and Maize Cultivation on Reduction of Replant Failure in Ginseng)

  • Background: Dazomet are widely used as soil fumigant to solve soilborne problems, and the degradation intermediates are toxic to nematodes, fungi, bacteria, insects and weeds. Methods and Results: The effects of cultivation of green manure crop, maize before and after soil fumigation on the control of ginseng root rot disease were compared using soil where 6-years-old ginseng was harvested. Fumigant (dazomet) were used for soil fumigation in May and September, respectively. Maize was grown for soil management before and after soil fumigation. After May fumigation, the sowing date of maize was delayed by 15 days and thus its dry weight was decreased significantly. Maize cultivation after May fumigation increased pH but decreased EC, $NO_3$, $P_2O_5$, and K significantly. Maize cultivation after May fumigation decreased fungi population and the ratio of fungi and bacteria. Growth of 2-years-old ginseng was improved and the incidence of ginseng root rot was significantly decreased by maize cultivation after May fumigation. After harvesting 2-years-old ginseng, the population of Cylindrocarpon destructans was not different between treatment of May and September, but Fusarium solani showed a significant increase in September fumigation after maize cultivation. Conclusions: Maize cultivation after soil fumigation was effective in inhibiting ginseng root rot by the amendment of mineral composition and microorganism in fumigated soil.

고려인삼, 미국삼 및 죽절삼의 생육 및 형태적 특성 비교 (Comparison of Plant Growth and Morphological Characteristics Among the Korean Ginseng, the American Ginseng and the Bamboo Ginseng)

  • An investigation was conducted to ascertain the basic information on characteristics of growth and morphological characters among the Korean (Panax. ginseng), the American (Panax. quinquefolium) and the Bamboo (Panax. japonicus) ginseng. In aerial parts growth of the ginseng species by age, The Korean ginseng and American ginseng's stem and leaf growth was alike in 2-4 years old, but growth cycle changed in 6 years old. The Korean ginseng was more vigorous than the American ginseng. The Korean ginseng roots were highly observed in ratio of red skin roots among three species, whereas The American ginseng roots were highly infected by root rot. It seems to be variable depending on growing stage and species. The Korean ginseng flowered about the middle of May, the American ginseng early June, and the Bamboo ginseng was late of May, The berry color of the ginseng species was observed, The Korean and American ginseng's mature berry color was red, The Bamboo ginseng's berry was three type of color and shape. In root characteristics of the seedling, Korean (p. ginseng), American (p. quinquefolium) ginseng's root shape was similarity in type, the bamboo ginseng showed different type, which root length and root weight was smaller than those of ginseng. In morphological characters of Leaf surface, pollen, and stoma, the Korean ginseng and American ginseng had crystal rosette on epidermis cell, but the Bamboo ginseng didn't has crystal rosette. Pollen shape observed tricolpate pollen and size was media type among the ginseng species, and also guard cell was anomocytic type, which were observed by scanning electronic microscope.

인삼의 생리장해 (Physiological disorder of Panax ginseng)

  • 인삼의 생리장해를 산지중심으로 증상별로 종합검토하였다. 뿌리의 장해는 적피, 은피, 동해, 동해, 근부, 안삼, 출아불재, 달래삼, 소수가 있고 지상부에는 황엽, 조기낙엽, 소엽, 지엽, 백반엽, 동해, 풍해와 경열이 있다. 적피와 황엽이 인삼생산에 가장 크게 영향한다. 농약해, 붕소과 잉해 및 산업공해에 의한 피해 경우도 보고되었다. 원료수삼 가공한 후의 품질요인에 관계되는 생리장해들도 검토하였다.

Bacillus subtilis YB-70 as a Biocontrol Agent of Fusarium solani causing Plant Root-Rot

  • A bacterial strain YB-70 which has powerful biocontrol activity against Fusarium solani causing plant root-rot resulting in considerable losses of many economical crops was isolated and selected from over 500 isolates from a ginseng rhizosphere in suppressive soil, and identified as a strain of Bacillus subtilis. In several biochemical and in vitro antibiosis tests on F. solani with culture filterates from B. subtilis YB-70, our data strongly indicated metabolites which mediated inhibition of the fungal growth were presumed to be heat-stable, micromolecular, and ethyl alcohol solutable antifungal substances. Suppression of root-rot by B. subtilis YB-70 was demonstrated in pot trials with eggplant (Solanum melongena L) seedlings. Treatment of the seedling with the bacterial suspension (1.7~1.9$\times$$10^5$ CFU/g) in F. solani-infested soil significantly reduced disease incidences by 68 to 76% after 25 to 30 days. The results supported that B. subtilis YB-70 have excellent potentials as a biocontrol agent.

  • The 543 microbial strains were isolated from the rhizosphere in ginseng root rot-suppressive soil. From these, isolate KGM-100 was finally selected for the screening of powerful antagonistic bacterium for the soil-born Fusarium solani causing root rot of many important crops. The isolate KGM-100 was identified as Pseudomonas aeruginosa. Antibiotics produced by the P. aeruginosa KGM-100 were partially characterized as heat-stable and low-molecular weight antibiotics. The strain also produced siderophore, which was assumed to be pseudobactin. Pot test showed approximately 30-50% biological control effects when Fusarium solani-infected soil was treated with the P. aeruginosa KGM-100.

Biological characteristics of Paenibacillus polymyxa GBR-1 involved in root rot of stored Korean ginseng

  • Background: This study aims to describe the characterization of Paenibacillus polymyxa GBR-1 (GBR-1) with respect to its positive and negative effects on plants. Methods: The morphological characteristics of GBR-1 were identified with microscopy, and subjected to Biolog analysis for identification. Bacterial population and media optimization were determined by a growth curve. The potential for GBR-1 as a growth promoting agent, to have antagonistic activity, and to have hydrolytic activity at different temperatures was assessed. The coinoculation of GBR-1 with other microorganisms and its pathogenicity on various stored plants, including ginseng, were assessed. Results: Colony morphology, endospore-bearing cells, and cell division of GBR-1 were identified by microscopy; identification was performed by utilizing the Biolog system, gas chromatography of fatty acid methyl esters (GC-FAME). GBR-1 showed the strongest antagonistic activity against fungal and bacterial pathogens. GBR-1 cell numbers were relatively higher when the cells were cultured in brain heart infusion (BHI) medium when compared with other media. Furthermore, the starch-hydrolytic activity was influenced by GBR-1 at higher temperature compared to low temperatures. GBR-1 was pathogenic to some of the storage plants. Coinoculation of GBR-1 with other pathogens causes differences in rotting on ginseng roots. A significant growth promotion was observed in tobacco seedlings treated with GBR-1 suspensions under in vitro conditions, suggesting that its volatile organic compounds (VOCs) might play a role in growth promotion. Conclusion: The results of this study indicate that GBR-1 has both positive and negative effects on ginseng root and other stored plants as a potential biocontrol agent and eliciting in vitro growth promotion.

The Effect of Fungicides on Mycelial Growth and Conidial Germination of the Ginseng Root Rot Fungus, Cylindrocarpon destructans

  • Ginseng root rot caused by Cylindrocarpon destructans is the most destructive disease of ginseng. Six different fungicides (thiophanate-methyl, benomyl, prochloraz, mancozeb, azoxystrobin, and iprodione) were selected to evaluate the inhibitory effect on the mycelial growth and conidial germination of C. destructans isolates. Benomyl and prochloraz were found to be the most effective fungicides in inhibiting mycelial growth of all tested isolates, showing 64.7% to 100% inhibition at a concentration of $10{\mu}g/mL$, whereas thiophanate-methyl was the least effective fungicide, showing less than 50% inhibition even at a higher concentration of $100{\mu}g/mL$. The tested fungicides exhibited less than 20% inhibition of conidium germination at concentrations of 0.01, 0.1, and $1{\mu}g/mL$. However, the inhibition effect of mancozeb on condium germination of C. destructans was significantly increased to 92% to 99% at a higher concentration of $100{\mu}g/mL$, while the others still showed no higher than 30% inhibition.
