This study was conducted to evaluate the effects of natural bioactive products such as Manda enzyme (T1), Yangmyeongwon (T2), effective microorganisms (T3), and Kelpak (T4) on the growth and ginsenoside contents of Panax ginseng cultured in an aeroponic system using a two-layer vertical type of nutrient bath under natural light conditions. The growth of ginseng plants showed specific characteristics according to the positions in which they were cultured due to the difference of light transmittance and temperature in the upper and lower layers during aeroponic culture in a two-layer vertical type of system. The growth of the aerial part of the leaves and stems of ginseng plants cultured in the lower layer (4,000 to 6,000 lx, $23^{\circ}C$ to $26^{\circ}C$) of the nutrient bath was observed to be superior to that of the ginseng plants cultured in the upper layer (12,000 to 15,000 lx, $25^{\circ}C$ to $28^{\circ}C$). The leaf area was significantly larger in the treatment of T2 and T4 (46.70 $cm^2$) than with other treatments. Conversely, the values of the root weight and root diameter were higher in ginseng plants cultured in the upper layer of the nutrient bath. The root weight was significantly heavier in the treatment of T4 (6.46 g) and T3 (6.26 g) than with other treatments. The total ginsenoside content in the leaves and roots was highest in the ginseng plants cultured by the treatment of T1, at 16.20%, while the total ginsenoside content obtained by other treatments decreased in the order of T4, T5 (control), T2, and T3, at 13.21%, 12.30%, 14.84%, and 14.86%, respectively. The total ginsenoside content of the ginseng leaves was found to be significantly higher in the treatment of T1 in the lower layer of the nutrient bath, at 15.30%, while the content of the ginseng roots in the treatments of T3 and T4, at 1.27% and 1.23%, respectively, was significantly higher than in other treatments in the upper layer of the nutrient bath.
This study was conducted to develop a resistant tobarro against Potato virus Y (PVY) by transformation of the plants with genetically engineered viral genes. The complimentary DNAs (cDNAS) of potato virus Y-necrosis strain (PVY-Vn) replicase mutant genes (3'-deleted, 5'-deleted and ADD-mutant Nlbs) were synthesized through RT-PCR by using purified PVY-VN RNA and synthesized primers, and cloned in the sense orientation into a plant expression vector (pMBPI), The cDNAS of the genes were transferred into Agrobacterium tumefaciens LBA 4404, and then transformed into tobacco (Nicotiana tabacum cv. Burley 21) plants. Regenerated plants were tested for PVY resistance by inoculation test; 13 transgenic plants including 7 for 3'-deleted Nlb, 3 for 5'-deleted Nlb, and 3 for ADD-mutant Nlb appeared to be resistant at 4 weeks after inoculation with PVY-VN. Among the 13 transgenic tobacco plants, 8 plants had no symptom up to 14 weeks after inoculation. The progenies ($T_1$) from self-fertilization of the transgenic lines varied 0.0% to 81.2% in their resistance (% of resistant plants). The analysis of Nlb-31deleted, -5'deleted and -ADD mutant in the $T_1$ plants by polymerase chain reaction (PCR) showed that Nlb-3'deleted, -5'deleted and -ADD mutants were detected in all of the resistant plants. These results suggest that the PVY resistance was inherited in the $T_1$ generation.
Transformation of ginseng plants was achieved by biolistic system with cotyledon explants and callus using phosphinothricin acetyl-transferase (PAT) gene resisting to a herbicide of Bialaphos. The binary vector for transformation was constructed with disarmed Ti-plasmid and with double 355 promoter. The introduced NPT II and PAT genes of the transgenic ginseng plants were successfully identified by the PCR, and the survival test on the medium with basta. The transgenic ginseng plants were propagated using repetitive secondary embryogenesis. The transgenic ginseng plantlets had normal structures of roots and shoots, and dormant buds for new year sprouting. We transferred the transgenic plants to greenhouse and observed the continuing growth until a new year.
It is very important factors to control optimal light transmission ratio and soil moisture content in order to produce good quality of ginseng seedling. To study the effect of light transmission ratio (LTR) and soil moisture content(SMC) on growth characteristics and yield of ginseng seedling, LTR was controlled by three level such as 21.2, 24.8% and 30.3%, and SMC was plotted by four level of 11.0%, 12.5%, 15.3% and 18.9% at the greenhouse. Chlorophyll content was gradually decreased in the low LTR (21.2%), while it was distinctly decreased by the decrease of SMC in excessively high LTR (30.3%). The decrease of SMC in the high LTR increased heat injury ratio distinctly, while heat injury ratio in the low LTR was only increased when SMC was very low such as 11.0%. All of fresh root weight per Kan (3.3$m^2$), root weight per plant, and the number of usable seedling were distinctly decreased by the increase of LTR and the decrease of SMC. Excessive increase of LTR in optimal SMC (18.9%) hadn't a great effect on the decrease of root weight, while root weight in low SMC was distinctly decreased by the increase of LTR. Ratio of rusty root was distinctly increased in the condition that both of LTR and SMC were high. Ratio of rusty root in the excessively high LTR was gradually fallen off by decrease of SMC, but its ratio in low SMC didn't changed distinctly by the decrease of SMC.
An efficient in vitro protocol has been established for somatic embryogenesis and plantlet conversion of Korean wild ginseng (Panax ginseng Meyer). Wild-type and mutant adventitious roots derived from the ginseng produced calluses on Murashige and Skoog (MS) medium supplemented with 0.5 mg/L 2,4-dichlorophenoxyacetic acid and 0.3 mg/L kinetin; 53.3% of the explants formed callus. Embryogenic callus proliferation and somatic embryo induction occurred on MS medium containing 0.5 mg/L 2,4-dichlorophenoxyacetic acid. The induced somatic embryos further developed to maturity on MS medium with 5 mg/L gibberellic acid, and 85% of them germinated. The germinated embryos were developed to shoots and elongated on MS medium with 5 mg/L gibberellic acid. The shoots developed into plants with well-developed taproots on one-third strength Schenk and Hildebrandt basal medium supplemented with 0.25 mg/L 1-naphthaleneacetic acid. When the plants were transferred to soil, about 30% of the regenerated plants developed into normal plants.
Vesicular-arbuscular mycorrhizae (VAM) fungi naturally colonise American ginseng roots and this relationship is highly beneficial to enhance plant productivity. Our goal was to determine the effect of adding two Glomus species (Glomus etuticatum, G. intraradices) on survival, photosynthetic capacity, growth, morphology and root ginsenoside content of one-year-old American ginseng plants grown in a broadleaf forest. While our study revealed that VAM inoculations significantly affected root morphology and Re ginsenoside content, the survival, photosynthetic capacity and root growth of American ginseng plants were not significantly influenced by VAM inoculations. Surface area and volume of rootlets were 16-25% higher for ginseng grown in VAM-inoculated soil compared to those grown in the control plots. Also, Re ginsenoside content was 18 % higher in YAM-inoculated roots compared to controls.
American ginseng plants (Panax quinquefolium L.) of various ages were harvested every two weeks during each of three growing seasons and dry matter yield of components and root respiration determined. Shoot dry weight was about 0.5 g, 2.5 g and 4 g for 2, 3 and 4-year-old plants, respectively and fruit dry weight was as much as 50% of this in 3- and 4-year-.old plants. Root dry weight decrease by 30~50% as shoots emerged and at the end of the season was about 2 g, 3.5 g and 5 g in 2, 3, 4 and 5-year-old plants, respectively. Shoot and root dry weight were linearly related with an approximate 1:2 ratio. Root respiration rate at 2$0^{\circ}C$ in the dark was about 5 $\mu\textrm{g}$ CO2 g-1 DW(dry weight) min-1 in the early season, then doubled within 50 days as shoots emerged, and thereafter declined over the season to 2~5 $\mu\textrm{g}$ CO2 g-1 DW min-1. The Q10 for dark respiration over the interval from 10 to 2$0^{\circ}C$ was 1.58. Root respiration rate and shoot growth rate was positively linearly related in all ages of plants. This study was conducted to select and develop effect of eco-friendly organic materials for the eco-friendly prevention of Anthracnose occurred in the ginseng. Anthracnose on ginseng is occurred by Colletotrichum gloeosporioides and the crop damage is severe especially from July to August after rainy season. The test results showed that control effect by test products materials on the three years ginseng and four years ginseng field was lower in eco-friendly organic materials than that of chemical pesticide. However, the control effect of bordeaux mixture was higher with 71.3% and 73.8% levels than those of mineral matter, microbial agent, and developed plants extract mixtures (Eugenol, Curcumin, Wood vinegar, etc). On the other hand, three types of developed plants extract mixtures (3 types) showed control effect in a range of from 58.1% to 63.6% against Anthracnose which was higher as compared with plant extract alone and sodium silicate regardless of ages of ginseng. The results of this study would attribute in verifying the control effect of eco-friendly materials against Anthracnose for ginseng through investigating antimicrobial compounds contained in the plants body. Also, it would be used as control method against Anthracnose occurred in ginseng by judging the right control time through monitoring occurrence of disease.
