To isolate rhizobacteria exhibiting antifungal activities for for five pathogenic fungi (Sclerotinia sclerotiorum, Fusarium solani, Collectotricum gloeosporides, Fusarium oxysporum, and Botrytis cinerea) which cause damage to Ginseng root in Ginseng grown fields, four soils were collected from Cheorlwon gun, in Korea. From 4 soils, a total of 160 bacterial strains were isolated by dilution plate method. Among 160 strains, 32 strains showed antifungal activities for one or more pathogens. From 32 strains, three strains exhibited antifungal activities for all pathogens. These are two Burkholderia cepacia (ATCC 25416 and ET 13) and one Paenibacillus polymyxa (ATCC 842). These potent antifungal strains showed high identities (99% using 16S-rRNA sequencing).

We investigated the anti-microbial activity of propolis against the pathogenic bacteria and fungi on ginseng. We selected six microbials that caused postharvest root rots in ginseng. Propolis extracts were prepared by using the ethanol extraction method. We seeded the bacteria and fungi related to ginseng disease on a specific culture medium, and treated it with propolis extracts by using the paper disc method. Propolis extracts indicate the anti-microbial activity against Paenibacillus polymyxa, Fusarium solani, Rhizoctonia solani AG-1 and Pythium ultimum. However, the anti-fungal activity of propolis is weak on Pseudomonas fluorescens subsp. Cellulosa and Colletotrichum gloeosporioides. As a result, the antimicrobial effects of propolis against microbial that prevent ginseng growth were confirmed. The antimicrobial effects are shown according to the concentration of propolis against root rot. The fungi also showed antibacterial effects in a dose-dependent manner.
Background: Glomus intraradices is a species of arbuscular mycorrhizal fungi that, as an obligate endomycorrhiza, can form mutually beneficial associations with plants. Panax ginseng is a popular traditional Chinese medicine; however, problems associated with ginseng planting, such as pesticide residues, reduce the ginseng quality. Methods: In this experiment, we studied the effect of inoculating G. intraradices on several physiological properties and microbial communities of ginseng. UV-Visible Spectrum method was used to detect physical properties. Denaturing gradient gel electrophoresis method was used to analyze microbial communities. Results: The results indicated that inoculation with G. intraradices can improve the colonization rate of lateral ginseng roots, increase the levels of monomeric and total ginsenosides, and improve root activity as well as polyphenol oxidase and catalase activities. We also studied the bacterial and fungal communities in ginseng rhizospheric soil. In our study, G. intraradices inoculation improved the abundance and Shannon diversity of bacteria, whereas fungi showed a reciprocal effect. Furthermore, we found that G. intraradices inoculation might increase some beneficial bacterial species and decreased pathogenic fungi in rhizospheric soil of ginseng. Conclusion: Our results showed that G. intraradices can benefit ginseng planting which may have some instructive and practical significance for planting ginseng in farmland.
The soil-borne ascomycete fungus Cylindrocarpon destructans causes ginseng root rot disease and produces various secondary metabolites such as brefeldin A and radicicol. The slow growth of this fungus compared with other plant pathogenic and saprophytic fungi in soil disturbs isolation of this fungus from soil and infected ginseng. In this study, we developed a selective medium for C. destructans using radicicol produced by this fungus. Supplementing 50 mg/L of radicicol to medium inhibited the mycelia growth of other fungi including Botrytis cinerea, Rhizoctonia solani and Alternaria panax, but did not affect the growth of C. destructans. In addition, conidia germination of other fungal species except for C. destructans was inhibited in submerged culture supplemented with radicicol. This medium provides a very efficient tool for isolating C. destructans and also can be used as an enrichment medium for this fungus.
The production of Korean ginseng, one of the most important medicinal root crops, is limited by many factors including soil sickness, root rots in fields as well as during storage prior to consumption. Although much research has been conducted on the diseases in field condition, little information is available on the control of postharvest roots rots. To obtain better management strategy of postharvest root rots in ginseng, biological control using antagonistic bacteria was attempted. Of 208 bacteria obtained form suppressive soil samples, 4 were selected based on the inhibitory effect on mycelial growth of two major causal fungi for postharvest root rots in ginseng, Botrytis cinerea and Fusarium solani. The culture filtrates of these bacterial antagonists greatly inhibited the conidial germination of both pathogenic fungi and produced abnormal morphology such as swollen germ tubes in F. solani and vacuolation of nongerminated conidia in B. cinerea. The population levels of bacterial antagonists on the ginseng roots were gradually increased up to 8 days of incubation. Postharvest root rots of ginseng caused by f. solani and B. cinerea were controlled in dipping tests in the ranges of 60∼80% by antagonistic Bacillus spp. obtained from suppressive soil. These results suggest that biological control using these antagonistic bacteria would be an alternative strategy to control postharvest root rots in ginseng.
In a previous study, we identified a Streptomyces sp., A3265, as exhibiting potent antifungal activity against various plant pathogenic fungi, including Botrytis cinerea, Colletotrichum gloeosporioides, and Rhizoctonia solani. This strain also exhibited a biocontrolling effect against ginseng root rot and damping-off disease, common diseases of ginseng and other crops. In this study, we isolated two antifungal substances responsible for this biocontrolling effect via Diaion HP-20 and Sephadex LH-20 column chromatography, medium pressure liquid chromatography, and high-performance liquid chromatography. These compounds were identified as guanidylfungin A and methyl guanidylfungin A by spectroscopic methods. These compounds exhibited potent antimicrobial activity against various plant pathogenic fungi as well as against bacteria.
Background: Panax ginseng Meyer is one of the most valuable medicinal plants which is enriched in anti-microbe secondary metabolites and widely used in traditional medicine. Botrytis cinerea is a necrotrophic fungus that causes gray mold disease in a broad range of hosts. B. cinerea could overcome the ginseng defense and cause serious leaf and root diseases with unknown mechanism. Methods: We conducted simultaneous transcriptomic and metabolomic analysis of the host to investigate the defense response of ginseng affected by B. cinerea. The gene deletion and replacement were then performed to study the pathogenic gene in B. cinerea during ginseng - fungi interaction. Results: Upon B. cinerea infection, ginseng defense responses were switched from the activation to repression, thus the expression of many defense genes decreased and the biosynthesis of antifungal metabolites were reduced. Particularly, ginseng metabolites like kaempferol, quercetin and luteolin which could inhibit fungi growth were decreased after B. cinerea infection. B. cinerea quercetin dioxygenase (Qdo) involved in catalyzing flavonoids degradation and ∆BcQdo mutants showed increased substrates accumulation and reduced disease development. Conclusion: This work indicates the flavonoids play a role in ginseng defense and BcQdo involves in B. cinerea virulence towards the P. ginseng. B. cinerea promotes disease development in ginseng by suppressing of defense related genes expression and reduction of antifungal metabolites biosynthesis.
전국 각지에서 채집한 사멸곤충에서 분리한 곤충 기생성 진균이 생산하는 살충성 생리활성물질을 탐색한 결과 곰팡이 분리주 CNAB-63 균주가 목화진딧물과 점박이응애 모두에 대해 다른 분리균주들보다 비교적 높은 살충효과를 보인 바, 본 균주가 생산하는 살충성분을 각종 chromatography로 분리 정제하였다. 정제된 활성물질 CNAB는 점박이응애에서 66.59%, 목화진딧물에서 52.96%의 살충효과를 보였으며, 그 외에도 온실가루이에 살충효과를 보였으나 배추좀나방에는 미미한 살충효과를 보였다. 한편 활성물질 CNAB는 $100\;{\mu}g/disk$ 농도에서 곰팡이, 효모, 세균류에 전혀 항균활성을 보이지 않았다.
Disease is the major problem in ginseng cultivation from seed stratification, soil preparation prior to planting, right through to drying of the roots. There are many soil-borne disease pathogen in rhizosphere soil environment, furthermore occurrence of diseases by a diverse group of fungi and related organisms are closely related to various soil condition. Observable symptoms for soil-borne diseases include wilting, leaf death and leaf fall, death of branches and limbs and in severe cases death of the whole plant. The fungus Cylindrocarpon destructans is the cause of root rot characterized by a decay of the true root system in many ginseng production areas in Korea. Some pathogens are generally confined to the juvenile roots whilst others are capable of attacking older parts of the root system. However, the relation between the soil environmental characteristics and ginseng root rot by soil-borne disease pathogen is not clearly identified in ginseng field. In this paper, we reviewed soil-borne plant pathogen causing root rot disease of ginseng with respect to soil environment.
인삼의 안정적인 생산 및 합리적인 병해방제에 기여하기 위하여 연구를 실시한 결과 갈잎 퇴비에서 분리된 Streptomyces lauretii B8180과 Bacillus subtilis B8856, Burkholderia cepacia B7944 세 균주의 인삼병원균 5종에 대한 항균력이 인정되었으며, 특히 실내실험을 통하여 높은 역병 방제효과가 인정되었다. 그러나 이 실험에서와 같이 식물의 지상부에 길항균을 살포하는 방법을 통하여 역병 방제효과를 높일 수 있을 것으로 생각된다. 이 길항균들은 갈잎 퇴비에서 분리되었기 때문에, 갈잎 퇴비에서 증식시켜 인삼재배 예정지에 투입하면, 억제형 토양 (suppressive soil) 조성을 통한 주요병해 방제가능성도 클 것으로 생각된다.
