Background: Notoginseng stem-leaf (NGL) ginsenosides have not been well used. To improve their utilization, the biotransformation of NGL ginsenosides was studied using ginsenosidase type-I from Aspergillus niger g.848. Methods: NGL ginsenosides were reacted with a crude enzyme in the RAT-5D bioreactor, and the dynamic changes of multi-ginsenosides of NGL were recognized by HPLC. The reaction products were separated using a silica gel column and identified by HPLC and NMR. Results: All the NGL ginsenosides are protopanaxadiol-type ginsenosides; the main ginsenoside contents are 27.1% Rb3, 15.7% C-Mx1, 13.8% Rc, 11.1% Fc, 7.10% Fa, 6.44% C-Mc, 5.08% Rb2, and 4.31% Rb1. In the reaction of NGL ginsenosides with crude enzyme, the main reaction of Rb3 and C-Mx1 occurred through Rb3${\rightarrow}$C-Mx1${\rightarrow}$C-Mx; when reacted for 1 h, Rb3 decreased from 27.1% to 9.82 %, C-Mx1 increased from 15.5% to 32.3%, C-Mx was produced to 6.46%, finally into C-Mx and a small amount of C-K. When reacted for 1.5 h, all the Rb1, Rd, and Gyp17 were completely reacted, and the reaction intermediate F2 was produced to 8.25%, finally into C-K. The main reaction of Rc (13.8%) occurred through Rc${\rightarrow}$C-Mc1${\rightarrow}$C-Mc${\rightarrow}$C-K. The enzyme barely hydrolyzed the terminal xyloside on 3-O- or 20-O-sugar-moiety of the substrate; therefore, 9.43 g C-Mx, 6.85 g C-K, 4.50 g R7, and 4.71 g Fc (hardly separating from the substrate) were obtained from 50 g NGL ginsenosides by the crude enzyme reaction. Conclusion: Four monomer ginsenosides were successfully produced and separated from NGL ginsenosides by the enzyme reaction.

Background: Due to the interrupted blood supply in cerebral ischemic stroke (CIS), ischemic and hypoxia results in neuronal depolarization, insufficient NAD+, excessive levels of ROS, mitochondrial damages, and energy metabolism disorders, which triggers the ischemic cascades. Currently, improvement of mitochondrial functions and energy metabolism is as a vital therapeutic target and clinical strategy. Hence, it is greatly crucial to look for neuroprotective natural agents with mitochondria protection actions and explore the mediated targets for treating CIS. In the previous study, notoginseng leaf triterpenes (PNGL) from Panax notoginseng stems and leaves was demonstrated to have neuroprotective effects against cerebral ischemia/reperfusion injury. However, the potential mechanisms have been not completely elaborate. Methods: The model of middle cerebral artery occlusion and reperfusion (MCAO/R) was adopted to verify the neuroprotective effects and potential pharmacology mechanisms of PNGL in vivo. Antioxidant markers were evaluated by kit detection. Mitochondrial function was evaluated by ATP content measurement, ATPase, NAD and NADH kits. And the transmission electron microscopy (TEM) and pathological staining (H&E and Nissl) were used to detect cerebral morphological changes and mitochondrial structural damages. Western blotting, ELISA and immunofluorescence assay were utilized to explore the mitochondrial protection effects and its related mechanisms in vivo. Results: In vivo, treatment with PNGL markedly reduced excessive oxidative stress, inhibited mitochondrial injury, alleviated energy metabolism dysfunction, decreased neuronal loss and apoptosis, and thus notedly raised neuronal survival under ischemia and hypoxia. Meanwhile, PNGL significantly increased the expression of nicotinamide phosphoribosyltransferase (NAMPT) in the ischemic regions, and regulated its related downstream SIRT1/2/3-MnSOD/PGC-1α pathways. Conclusion: The study finds that the mitochondrial protective effects of PNGL are associated with the NAMPT-SIRT1/2/3-MnSOD/PGC-1α signal pathways. PNGL, as a novel candidate drug, has great application prospects for preventing and treating ischemic stroke.
Varietal response to cool temperature(18$^{\circ}C$) and short (8hr.)-day treatment were investigated under controlled conditions of Phytotron in the Suwon Agronomy Experiment Station. Five flue-cured and one burley tobacco varieties (Nicotiana tabacum L.) were used, and the length of treatment ranging from 8 to 32 days at 3-day intervals and also includes continuous treatment up to the plants were flowered. The days to flower decreased significantly than the no treatment when the length of treatment was 11 to 14 days for the varieties NC82, Br.21 and NC22NF, and 17 to 26 days for SPG-28, Mc.944 and TC499, respectively. And also the number of leaves decreased significantly than the no treatment when the length of treatment was 8 days for NC22NF, 11days for NC82 and Br.21, 14 days for Mc.944, and 17 days for SPG-28 and TC499, respectively. The maximum decreasing ratio of the leaf number by the cool temperature and short-day treatment were 47.7 to 58.5% for NC82, Br.21 and NC22NF, and 38.9% for Mc.944, 33.4% for SPG-28 and 29.0% for TC499, respectively.
This study was carried out to compare growth characteristics, and extract and crude saponin contents of 4-year-old ginseng cultured by direct seeding and transplanting cultivation at several farms of the main producing district, Geumsan and Eumsung. Though root weight per plant of direct seeding cultivation was lower than that of transplanting cultivation, but yield of the former was higher than that of the latter owing to high rate of survival plant and Leaf Area Index. Dry matter partitioning ratio of direct seeding cultivation was high in primary root and low in secondary root because direct seeding cultivation elongated the length of primary root, while it suppressed the growth of secondary root. Ratio of rusty root was decreased in condition of direct seeding cultivation, while the contents of extract and crude saponin were lower than that of transplanting cultivation.
This study was conducted to know the effects of light intensity and temperature on growth of V. fauriei plants. Photosynthesis of V. fauriei had highly significant relations to light intensity and temperature in a quadratic regression model, from which the optimum light intensity and temperature for the plant growth were estimated to be 40,000lux and $17.7^{\circ}C.$ Root was produced less by shading at Jinbu where is located in alpine region, but root yield is increased by shading at Umsong where is located in plane region. Roots were produced more in Jinbu than in Umsong. A highly significant quadratic regression was noted between temperature and leaf width or root weight of V. fauriei. It was estimated from the regression equation that the optimum temperature for root growth was $20.3^{\circ}C.$
The Siberian ginseng and Eucommia are a kind of medicinal plant with powerful anti-oxidant activity. An experiment was conducted to investigate the effect of Siberian ginseng leaf and Eucommia leaf at level of 0.5% and 1% per feed in Ross commercial broiler for 4 to 35 days of age on performance, organ weight, blood biochemical profiles and telomere quantity. Chickens consuming diets containing 1% Siberian ginseng had higher feed conversion ratio than the other treated chicken during experimental period whereas no significant differences were detected in body weight, weight gain and feed intake. The weight of bursa of fabricius was significantly increased in chickens with dietary supplementation compared with chickens fed control but this was not seen in liver, spleen and thymus. In blood biochemical profiles, chickens with dietary supplementation had higher concentration than chickens fed control in triglyceride, cholesterol and glucose. The concentration of aspartate aminotransferase, alanine aminotransferase, albumin and total protein, however, was not significantly different between dietary supplemented chickens and control chickens. The relative amount of telomeric DNA of lymphocytes in chickens with dietary supplementation was significantly higher than that of control chickens but the difference was not found in liver, heart and testis tissues. In conclusion, dietary supplementation of Siberian ginseng and Eucommia in broiler improved immune activity and telomere length without decreasing chicken growth performance.
This study was carried out to investigate the effect of seasonal changes on some of the morphological and physiological characteristics, including the photosynthetic abilities and dark respiration, of young ginseng plants due to planting location under shading. The results obtained are as follows: 1. Seedlings and 2-year old plants planted in the back rows appeared to have broader leaf area, and their leaf weight greatly increased in September. Chlorophyll content was significantly reduced in September rather than in June and the plants in the back rows had more chlorophyll content than those in the front rows. 2. There was no difference in the light compensation point between the front and back rows in June, while in September the light compensation point of 2-year old ginseng leaves was much lower for plants in the back rows compared with those in the front rows. A difference in the light saturation point was not noticeable between plants in the front and back rows in June and September. But the light saturation point of 2-year old ginseng leaves at $15^{\circ}C$ was high in June, while it was high at $20^{\circ}C$ high in September. 3. Maximum photosynthetic ability was attained at $15^{\circ}C$ in June and at $20^{\circ}C$ in September. During June no significant difference in photosynthetic ability was found between plants in the front and back rows, but in September the amount of photosynthesis was significantly increased at the leaves of seedlings as well as 2-year old plants planted in the back rows. 4. The optimum temperature for maximum photosynthesis in 2-year old plants ranged from $14.0^{\circ}C$ to $14.5^{\circ}C$ in June and from $19.5^{\circ}C$ to $20.5^{\circ}C$ in September. However, the optimum temperature for maximum photosynthesis in the seedlings was from $21.2^{\circ}C$ to $21.6^{\circ}C$ in September, but a significant difference in the optimum temperature for the maximum photosynthesis in seedlings and 2-year old plants was not noticeable between the front and back rows. 5. The respiration rate was rather high in seedlings compared with 2-year old plants. During September the respiration rate in seedlings was much lower in the back rows than in the front rows. The rate of increase in the respiration of 2-year old plants was higher at September than June. The increase in respiration rate due to temperature was more significant in seedlings than 2-year old plants. 6. In September, the level of $Q_{10}$ in 2-year old plants was much lower than that found in seedlings. During June, 2-year old plants showed lower $Q_{10}$ levels at a temperature difference between $15^{\circ}C$ and $25^{\circ}C$; but in September this occurred at a temperature difference between $20^{\circ}C$ and $30^{\circ}C$.
To select optimal shade material in paddy soil, growth characteristics and ginsenoside contents were investigated in new cultivar, 'Cheonpoong' of three-year-old ginseng cultured under three kinds of shade materials such as three-layered blue and one-layered black PE (polyethylene) net (TBPN), blue PE sheet (BPSS), and aluminium-coated PE sheet (APSS). The order of light transmission ratio and air temperature by shade materials were BPSS > APSS > TBSB among three shade materials. Average soil water tension in PDC and IDC was 64 mbar (absolute soil moisture, 25%) and 123 mbar (absolute soil moisture, 17%), respectively, and soil water tension in IDC was changed more distinctly than that of PDC by season and shade materials. Yield in PDC was distinctly decreased more than that in IDC because of the increase of discolored-leaf and rusted-root ratio. BPSS and TBPN among three shade materials were the most effective on the increase of yield in PDC and IDC, respectively. Ratio of rusty-colored root showed not significant difference by drainage class and shade materials. Contents of panaxatriol ginsenoside (Rg1, Re and Rf) were decreased in PDC, while it of panaxadiol ginsenoside (Rb1, Rc and Rd) were increased in IDC. Total ginsenoside contents of IDC was distinctly higher than that of PDC, and BPSS showed the highest contents among three shade materials regardless of poorly and imperfectly drainage class.
Photosynthetic inhibition to temperature were conducted with ginseng(4 year old) and tobacco(var. Bulgaria). The plants were kept under various temperature conditions from 1$0^{\circ}C$ to 4$0^{\circ}C$ and 440$\mu$E/$m^2$/sec for 3 and 6hrs, and net $CO_2$ uptake were measured after 2hrs at $25^{\circ}C$. Photosynthetic optimal leaf temperature of ginseng was 21$^{\circ}C$ and tobacco was $25^{\circ}C$. Stomatal resistance and mesophyll resistance increased at high temperature. Especially, stomatal resistance seemed to have a significant role in determining the temperature responses of photosynthesis. In tobacco photosynthetic capacity was not changed by temperature treatment for 3hrs. However, 6hrs exposure reduced 8% of net photosynthesis at 4$0^{\circ}C$ and 12% at 1$0^{\circ}C$. Ginseng plants exposed for 6hrs at 4$0^{\circ}C$ lost photosynthetic capacity by 83%. Temperature responses of ginseng were very sensitive at above-optimum temperature resulting greater thermal inhibition other than photoinhibition.
This study was carried out to obtained the basic information for seed coat dehiscence and seedling performance according to seed size of eight ginseng cultivars. Seed size was separated into four grade(upper 5.0, 5.0-4.7, 4.7-4.0, and under 4.0mm in diameter). Seed coat dehiscence and survival ratio of seedling were not affected by seed size. Aerial parts growth of seedling such as stem length, stem diameter, and leaf area of seedling were larger seed(upper 5.0 mm) better than those of small seeds(4.7-4.0, and under 4.0 mm in diameter). Also root part growths such as root diameter, and fresh weight were larger seed(upper 5.0 mm) higher than those of small seeds(4.7-4.0, and under 4.0 mm in diameter). However, root length was not affected by seed size. Growth status of aerial and root parts of 'unpoong' 'umpoong' and 'opoong' were better than those of other cultivars in each grade of seed sizes. The growth of characteristics of 'hunpoong' and 'hungsun' were less than those of other cultivars.
