Tolclofos-methyl applied as seed dipping at 1,000 ppm for 3 hrs before sowing and soil drenching at the rate of 300 g ai./10 a in the middle of April protected emerging seedlings of Panax ginseng from damping-off caused by Rhiiutonia solani(AG2-1) in Yangjik Soil artificially infested with the pathogen. Germination rates with tolclofos-methyl, pencycuron, and control were 53.7%, 45.8%, and 7.5%, respectively, while the rate of the seeds at non-infested soil was 62.6%. The effectiveness of Tolclofos-methyl against the pathogen in the soil lasted upto 32 days in vitro. However, the transpiratio of ginseng seedlings increased greatly with chemical treatment, showing 0.02, 0.12, and 0.24 m1/cm2 leaf area/day at 0, 1,2, and 4 ppm a.i. of the fungicide, respectively.

Recently, some of the previous studies reported that tolclofos-methyl is still exist in ginseng cultivated soil, even though it is has been banned for ginseng. Therefore, the current study was aimed to examine the levels of absorption and translocation of tolclofos-methyl from ginseng cultivated soil to ginseng root and leaf stem for the period of 1 year. For this study, ginseng plants were transplanted in pots and treated with $5.0mg\;kg^{-1}$ of tolclofos-methyl (50% WP). At the end of each interval periods (every three months) the samples (soil, roots and leaf stems) were collected and analyzed the absorption and translocation levels of tolclofos-methyl using gas chromatography and mass spectrometry (GC-MS). The limit of quantitation of tolclofos-methyl was found to be $0.02mg\;kg^{-1}$ and 70.0~120.0% recovery was obtained with coefficient of variation of less than 10% regardless of sample types. In this study, a considerable amount of translocation of tolclofos-methyl residues were found in soil (4.28 to $0.06mg\;kg^{-1}$), root (7.09 to $1.54mg\;kg^{-1}$) and leaf stem (0.79 to $0.69mg\;kg^{-1}$). The results show that the tolclofos-methyl was absorbted and translocated from ginseng cultivated soil to ginseng root and ginseng leaf stem and found to be decreased time-coursely. Secondly, we were also analyzed soil, root and leaf stems samples from Hongcheon, Cheorwon, Punggi and Geumsan by GC-MS/MS (172 pesticides), LC-MS/MS (74 pesticides). In this study, 43 different pesticides were detected ($0.01{\sim}7.56mg\;kg^{-1}$) in soil, root and leaf stem. Further, tolclofos-methyl was detected 4 times separately in root sample alone which is less ($0.01{\sim}0.05mg\;kg^{-1}$) than their maximum residual limit (MRL) in ginseng. Consequently, the results from both studies indicate the residues of tolclofos-methyl found in ginseng cultivated soil and ginseng ensuring their safety level. Moreover, long-term evaluations are needed in order to protect the soil as well as ginseng free from tolclofos-methyl residues.
Extraction properties were investigated on ginseng leaf tea as functions of the current ethylene oxide fumigation and gamma irradiation at 2.5 to 10 kGy. In the extraction of tea constituents at boiling conditions, water soluble matters were the highest (83% yield for 5 min) in the time dependence, followed by tannin (82%), sugar (73%) and saponin (66%). No significant difference was observed in the extraction yield between the 5 kGy-irradiated and control samples, whereas both EO fumigation and 10 kGy irradiation reduced the extraction time of soluble matter and tannin, leading to the decrease in Hunter L value of tea extracts.
Physico-chemical properties were investigated on ginseng leaf tea which was decontaminated by the current ethylene oxide fumigation and gamma irradiation, respectively. Chemical components such as soluble matter, saponin and polyphenols were found to be stable to both treatments, while ascorbic acid and chlorophylls were relatively labile to fumigation and irradiation more than 5 kGy. Instrumental analysis has shown that EO fumigation causes apparent changes in pH and color of extracts and volatile profile of the sample, as compared to negligible changes in the irradiated sample at a decontaminating dose, 5 kGy.
Effect of nitrogen rate with and without chance of Phosphorus and Potassium rate was investigated in a field experiment. Percentage of plant mortality after transplanting increased with N rates above 32.5kg/10a. The content of total nitrogen increased and postassium decreased slightly as the rates of applied N was increased, but the contents of total alkaloid and phosphorus were not affected at each growing stage. It was considered that the application of 17.5kg of P2O5 and 35.0kg of K2O Per 10a might be sufficient for high yield and good Quality. As the N rates being increased, the yield, value, contents of total alkaloid and total nitrogen of cured leaf increased However, the Brice per kg was not significantly different among 17.5 ~ 37.5kg / 10a of N rates. The application of 37.5kg/10a of N may be profitable for farm economy; but, the N application should be controlled 17.5~22.5kg/10a for the low nitrogen and alkaloid leaf.
Wilting and transpiration charactistics of Panax ginseng leaves were investigated at two temperature levels. Water potential and water absorption characteristics of leaf segments were also observed. Soybean leaves were compared. 1. Ginseng leaves were thiner, higher in water content and lower in dehydration rate. But time required to reach permanent wilting point (pwp) was less than half of that of soybean leaves because water content at pwp was about two times higher (80% of initial water for ginseng and 50% for soybean leaves). The time was shorter under high air temperature. 2. Transpiration rate was about a quater of soybean leaves and lower at $33^{\circ}C$ than $23^{\circ}C$, indicating that ginseng leaves are less tolorant to high air temperature. 3. Ginseng leaf segment showed smaller water free space but greater water deficit and little difference in was absorption rate. 4. Water potential of leaves measured by liquid immersion method was lower than that of soybean leaves. 5. Above results strongly suggest that ginseng plants are more susceptible to water stress. Thus greater light intensity during leaf growing stage (April to June) is recommendable to increase stomate frequency resulting greater transpiration rate and high temperature tolerance during July and August. Abundant water around roots seems to be beneficial as long as oxygen is not limited in rhizosphere.
Tobacco leaves cultivated in Korea, U.S A., Greece and Turkey were analyzed for their essential oils that are solanone, nor- solanadione, damascenone, dana scone and mesa stigmatrienone (4- isomer). Regardless of tobacco varieties and cultivated localities, solanone was the most abundant. Among them particularly, Basma has the highest amounts of the solanone than other varieties. Flue-cured tobacco leaf has much higher amounts of damascellone than burley and aromatic tobacco leaf do. The major flavonoid component of Ginseng leaf is trifolin, a glycoside of kaempferol. To evaluate the antioxidative properties of trifolin and kaempferol on cellular membranes, we compared them with the other flavonoids through the 102-Induced photohemolysis of rabbit erythrocytes. All the flavonoid aglycones including kaempferol, quercetin and baicalein protected effectively the cells from the 102-caused damage in a dose- dependent manner, by scavenging 102 and free radicals in the cellular membranes. The solubilization of the flavonoid aglycones into micelles or erythrocyte membranes was deduced from spectro-photometric and microscopic observations. The flavonoid glycosides were not protective or less protective than their corresponding aglycones, and trifolin was the only glycoside that exhibited a solubilization into the membranes and a significant protection against the photohemolysis. We also tested some phenolic compounds contained in Ginseng, and found that they did not prevent the photohemolysis so effectively as kaempferol or trifolin.
This experiment were conducted to investigate heterosis and combining ability for several mutant characters by analyzing dialled crosses of flue-cured tobacco. In a dialled cross of 3 flue-cured varieties and the mutant line 83H -5, the heterosis was somewhat higher in Fl than in F2. For growth character, the heterosis was 0.28-6.03% in plant height, leaf number, leaf shape index and yield, and was 43.2% for bacterial wilt disease index. The mutant line 83H-5 showed significantly negative GCA effect for plant height, leaf width and bacterial wilt disease index in Fl and F2, leaf length in F2, and positive GCA effect for total alkaloids, total nitrogen in Fl and days to flower in F2, respectively. Specific combining ability(SCA) in 83H-5 x Hicks was significant in negative effect for leaf length(F2), number of leaves(F2), leaf shape(F1, F2), bacterial wilt(F2) and alkaloids(F1), and in 83H-5 x NC 2326 in positive effect for leaf length(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in positive effect for plant height(F1, F2) and leaf width(F2), and for 83H-5 x NC 82 in Positive effect for Plant height(F1, F2), leaf length(F2) and yield(F1, F2).
This Study was conducted to estimate the degree of dominance and gene frequency of some sueful characters in tobacco. The eight parents and a set of 28 crosses of F'1s was F'2s were used as materials, and planted on oriental's and burley cultivated system as randomized block designs, respectively. The observed characters were six agronomic characters which were plant height, number of leaves per plant, leaf length, leaf width, days to flowering and yield, and the data obtained from the experiment were analyzed from methods by Hayman's and Jinks. The results obtained are summarized as follows: 1. In Vr-Wr graphical analysis, number of leaves per plant, leaf length, days to flowering and yield were found to be inherited in partial dominance, and plant height was over dominance to be similar to complete dominance, but leaf width was inherited with partial dominance close to complete dominance. 2. In the gene frequency, two varieties Xanthi-Basma and KA 102, for days to flowering and yield had larger number of dominant genes as those were situated near the point of origen. 3. Additive effects of genes(D) were greater than dominance effects of Genes(H) for six agronomic characters except plant height, and mean degree of dominance over all loci was lower than 1 for days to flowering yield, leaf length and number of leaves per plant.
