Proximate, Saponin and Amino Acid Compositions in Camellia (Camellia japonica L.) Seeds and Defatted Camellia Seeds (동백종실(Camellia japonica L.)과 유박의 일반성분, 사포닌 및 아미노산 조성)

  • To furnish basic data for the utilizatin of camellia(Camellia japonica L.) seeds as a raw material of industrial products, major chemical components of camellia and defatted camellia seeds were investigated. The moisture, crude ash, crude fat and crude protein contents of camellia seeds were 4.06%, 1.83%, 65.75% and 8.44%, respectively. Defatted camellia seeds indicates that it contains 10.31% moisture, 5.28% crude ash and 17.49% crude protein while no crude fat was detected. The curde ginseng saponin, crude saikosaponin and red ginseng saponin contents of camellia seeds were 108.0mg%, 0.4mg% and 40.3mg%, respectively. Total amino acid contents of camellia seeds and defatted camellia seeds were 7,851mg% and 13,002mg%, respectively, and the major amino acids were glutamic acid, arginine, aspartic acid and leucine. The ratio of essential/total amino acid of camellia seeds was 0.29.

  • Cultures of hairy root induced from ginseng(Panax C.A. Meyer) transformed with Agrobacterium rhizogenes (strain A4, ATCC 15834) were established and morphologically two different hairy root strains (HB1, HB2) were obtained. To determine the optimum growth rate, the hairy root (HB2) was cultured in various liquid medium supplemented with or without plant growth hormone. The growth rate of hairy root cultured on MS medium was 1.3-3.1 times higher than those cultured on other media, and the optimum sucrose concentration and pH were 3-6%, 5.5-6.5, respectively. Also, the growth rate of hairy root was increased when 0.02 M ammonium nitrate, 1.2 mM potassium phosphate (monobasic) and 0.5 mg/l IBA were supplied to liquid medium. The saponin patterns and contents of hairy root (HB2) were determined by TLC and HPLC. The crude saponin contents were 4.67% and the total saponin contents were 1.0%, on dry weight basis.

  • In an attempt to investigate the effects of white ginseng on the proliferation and the nitric oxide(NO) secretion of mouse peritoneal macrophages, which are the first mai or defense phagocytes in the immune system, the studies have been carried out. In the macrophage proliferation assay using the $^3$H-thymidine incorporation, the total saponin or Ginsenoside Rb$_2$ were added to the medium at the concentration of 0 to 256$\mu\textrm{g}$/$m\ell$. DNA synthesis of the macrophage was increased at 64$\mu\textrm{g}$/$m\ell$ of total saponin and either 16$\mu\textrm{g}$/$m\ell$ or 64$\mu\textrm{g}$/$m\ell$ of Ginsenoside Rb$_2$, respectively. Also, the effect o(white ginseng on the nitric oxide secretion of the macrophages was investigated. The addition of either total saponin or Ginsenoside Rb$_2$ at the concentration of 20$\mu\textrm{g}$/$m\ell$ significantly increased the secretion of NO from the macrophages. The nitric oxide synthase (NOS) gene expression which is responsible for the synthesis of the nitric oxide has been studied using reverse transcription polymerase chain reaction. In RT-PCR, the $\beta$-actin and nos gene expression have been analyzed. 20$\mu\textrm{g}$/$m\ell$ of either total saponin or Ginsenoside Rb$_2$ increased nos gene expression of the macrophages.

Antioxidant and Hepatoprotective Effects of Hydroponic-cultured Ginseng Folium by fermentation (발효에 의한 수경재배 인삼 잎의 항산화 및 간 보호 효과)

  • Objectives : Positive effects of Ginseng has great research attentions such as anticancer, anti-diabetic, antiaging, liver, immune function, CNS, etc. In this study, we investigated Hydroponic-cultured Ginseng Folium fermented byBacillus subtilisto establish fermentation conditions for enhancing functionality.Methods : Ginseng Folium were cultivated hydroponic-cultured and were extracted with methanol. We inoculateBacillus subtilisfor fermentation by adding to 0%, 3% and 5% sugar respectively and checked antioxidant activities, total phenolic content and total saponin content in 2 days intervals during 11 days. The antioxidant activities were studied by the 1,1-diphenyl-2-picryl hydrazyl(DPPH) radical, 2, 2'-Azino-bis(3-ethylbenzothiazoline-6 sulfonic acid) diammonium salt(ABTS) radical scavenging assay and Reducing power assay. We analyzed the Total phenol content, crude saponin content and ginsenoside content. Moreever, Hepatoprotective effects by Glutamic oxaloacetic transaminase(GOT) and Glutamic pyruvic transaminase(GPT) in Sprague-Dawley rat.Results : The results of DPPH and ABTS were 66.89% and 96.72%, respectively. The reducing power was resulted in optical density of 0.7312 with 3% sugar after 9 days of fermentation. and the concentration at 200 ㎍/㎖. Total phenol content was 36.92㎎/g with 3% sugar after 9 days of fermentation, in which crude saponin content wasn't changed, and ginsenoside content such as Rg3, Re and Rb was increased. Activities of GOT and GPT concentration were decreased in rat.Conclusions : This study suggests that hydroponic-cultured Ginseng Folium fermented byBacillus subtilisin 9 days showed significant efficacy of hepato-protection as well as antioxidant compared to the others. In addition, it shows not only improved value but also utilized hydroponic-cultured Ginseng Folium by fermentation.

Quantitative aspects of the hydrolysis of ginseng saponins: Application in HPLC-MS analysis of herbal products

  • Background: Ginseng is one of the most valuable herbal supplements. It is challenging to perform quality control of ginseng products due to the diversity of bioactive saponins in their composition. Acid or alkaline hydrolysis is often used for the structural elucidation of these saponins and sugars in their side chains. Complete transformation of the original ginsenosides into their aglycones during the hydrolysis is one of the ways to determine a total saponin group content. The main hurdle of this approach is the formation of various by-products that was reported by many authors. Methods: Separate HPLC assessment of the total protopanaxadiol, protopanaxatriol and ocotillol ginsenoside contents is a viable alternative to the determination of characteristic biomarkers of these saponin groups, such as ginsenoside Rf and pseudoginsenoside F11, which are commonly used for authentication of P. ginseng Meyer and P. quinquefolius L. samples respectively. Moreover, total ginsenoside content is an ideal aggregated parameter for standardization and quality control of ginseng-based medicines, because it can be directly applied for saponin dosage calculation. Results: Different hydrolysis conditions were tested to develop accurate quantification method for the elucidation of total ginsenoside contents in herbal products. Linearity, limits of quantification, limits of detection, accuracy and precision were evaluated for the developed HPLC-MS method. Conclusion: Alkaline hydrolysis results in fewer by-products than sugar elimination in acidic conditions. An equimolar response, as a key parameter for quantification, was established for several major ginsenosides. The developed approach has shown acceptable results in the analysis of several different herbal products.

Effect of Alcohol Treatment on Growth of Microorganisms Contaminated in Ginseng Powders (알콜처리가 인삼분말에 오염된 미생물의 성장에 미치는 영향)

  • Alcohol treatment was applied to ginseng powder for the improving hygienic quality of ginseng powder. A bacterial strain designated as GT5 was isolated from ginseng powder contaminated and was identified as Escherichia coli species by IMVIC test method. Ethanol used as alcohol, inhibited strongly the growth of coliforms in ginseng powder at the concentrations of 50 to 90%. Ethanol treatment also decreased numbers of total bacteria at the same concentrations. There was not significant changes in saponin of ginseng powder after treated with ethanol. However, ethanol treatment caused a decrease in Hunter's color L value and an increase in a and b values of ginseng powder. As a hygienic quality control of ginseng powder, ethanol treatment could be cosidered as an effective means for decontaminating microorganisms in ginseng powder.

Effect of Extraction Temperature and Time on Saponin Composition of Red Ginseng Extract (추출온도(抽出溫度) 및 시간(時間)이 홍삼(紅蔘)엑기스의 사포닌 조성(組成)에 미치는 영향)

  • The saponin yield and its compositional changes of red ginseng extract (RG-EXT) was investigated during extraction at various temperature for 5 times of 8 hours. The higher temperature resulted an increase in solids yield while the total saponin recovered was decreased, particularly at $100^{\circ}C$. A relatively lower thermal stability was found for protopanaxadiol saponin, one of the saponin fractions, than protopanaxatriol saponin. The compositional ratio of saponin at ginsenoside level was little affected by extraction time. The yields data showed that more than 94% of total saponin was recovered by 3$\sim$4 times of 8 hours extraction. Extraction at $80^{\circ}C$ for 4 times of 8 hours were suggested for preparation of RG-EXT from the result of this work.

Effect of Saponin Fraction from Platycodon grandiflorum on Clinical Chemical Changes in TCDD (2,3,7,8-Tetrachlorodibenzo-ρ-dioxin)-induced Rat Toxicity

  • This study was carried out to investigate the protective effect of crude saponin from Platycodon grandiflorum on Clinical chemical parameters in male rats acutely exposed to 2,3,7,8-tetrachlorodibenzo-ρ-dioxin (TCDD). Crude saponin was prepared from Korean Platycodon grandiflorum with Diaion HP-20 adsorption chromatography after extraction of 80% ethanol at 75℃. The crude saponin was confirmed by thin layer chrmatography. When compared with ginseng saponins, the crude saponin had both a few number of saponins and a broad distribution. Forty male rats (200±20 g) were divided into 4 groups. Normal control (NC) group received vehicle and saline; TCDD-treated (TT) group received TCDD (40 ㎍/kg, single dose) intraperitoneally; Platycodon grandiflorum saponin (PG5 and PG10) groups received crude saponin 5 mg/kg and 10 mg/kg (p.o), respectively, for 2 weeks before 1 week of TCDD-exposure. Increase of body weight was retarded greatly by TCDD-exposure. Body weight of animals in TT group was significantly decrease after 2 days of TCDD-exposure. However, body weights of animals in PG groups increased through the experimental perimental period, although the increasing rate was slower than that of NC group. Increases in contents of blood glucose, total cholesterol and triglyceride (TG) and activities of amylase, lipase, AST, ALT and LDH by toxic action of TCDD were significantly attenuated by crude saponin from Platycodon grandiflorum (P<0.05). In conclusion, these results suggest that crude saponin prepared from Korean Platycodon grandiflorum might be a member of useful protective agents against TCDD, which is one of the environmental hormones.

  • Free fatty acid and total saponin, which were extracted from the spent media and colony of the strain Phizopus delemar Rh-1, were qualitatively examined by GLC and spectroscopic methods and compared with these of intact ginseng media. The 18, 18, and 14 free fatty acids were respectively identified from the control, spent media and colony. Among them, $iC_{12:0},\;C_{16:2},\;C_{17:0},\;C_{18:3}\; and\;C_{20:0}$ acids were not detectable from the colony. In add ition, $C_{16:2}\;and\;C_{20:0}$ peaks were rot found in the spent media and control, respectively. The isible absorption spectra of free fatty acids and total saponins in the control, spent media and colony were invariable. In contrast, however, the infrared and ultraviolet absorption spectra showed significant spectral variations, particularly in the it finger-print region, demonstrating that the ginseng components were considerably utilized by the strain Rhizopus delemar Rh-1.

Inhibitory Effect of Ginseng Total Saponins on the DEvelopement of Tolerance to U-50,488H-Induced Antinociception is Dependent on Serotonergic Mechanisms (U-50,488H 진통성 내성형성에 대한 인삼 사포닌의 억제효과는 Serotonin 기전에 의존적이다)

  • We have previously reported that the antagonism of U-50,488H-induced antinociception in mice pretreated with ginseng total saponins (GTS) Ivas abolished by pretreatment with a serotonin precursor, 5-hydroxytryptophan (5-HTP), but not by a noradrenaline precursor, L-dihydroxyphenylalanine (L-DOPA) in the tail flick test. In the present experiments, the effect of the same GTS on the development of tolerance to U-50,488H-induced antinociception was determined. GTS inhibited the development of tolerance to U-50,488H-induced antinociception. The inhibitory effect of GTS on the development of tolerance to U-50,488H-induced antinociception was reversed by 5-HTP, but not by L-DOPA. These findings suggest that the inhibitory effect of GTS on the development of tolerance to U-50,488H-induced antinociception is dependent on serotonergic mechanisms.
