Effects of Antioxidants on the Photosynsthesis and Carbohydrates/Saponin Contents in Panax ginseng Leaves (인삼잎의 광합성과 탄수화물.사포닌 함량에 미치는 항산화제의 효과)

  • We studied the folilar wiping effects of antioxidants (ascorbate, glutathione and sodium azide), which effectively inhibited the chlorophyll bleaching or completely recorved the early stage of photosynthesis of Panax ginseng C.A. Meyer, on photosynthesis, stomatal resistance, free sugar, starch, and total saponin contents of ginseng under the excess light intensity (45 kLux) during 6 days. Ascorbate and glutathione, endogenous antioxidant, recovered photosynehtsis and stomatal resistance, and reduced the photoinhibition by the excess light intensity (45 kLux) on free sugar, starch and total saponin contents. But sodium azide, exogenous $^{1}O_2$ quencher, showed negative effect. Therefore, we assumed that carbohydrates and saponin metabolisms of ginseng by antioxidants (ascorbate, glutathione) were normal. For the reduction of inhibition by excess light in ginseng a program for the higher activation of antioxidants and antioxidative enzymes in ginseng leaf will be desirable.   • To investigate the stability of ginseng saponin, various physical and chemical treatments for red ginseng alcohol extract (70% ethyl alcohol) were carried out, and then the variations of ginseng saponin in extract were investigated by high performance liquid chromatography (HPLC) method. Irradiation of ${\gamma}$-ray, and ultraviolet ray, sonocatalysis by ultrasonicator, treatment of electronic range, catalytic ozonation did not or slightly affect degradation of ginseng saponins, but they were degraded by heat treatment.

  • Saprophagous fungi which were isolated from ginseng products were investigated the change of mycellial weight, saponin pattern and saponin contents according to culture periods at different of saponin concentration. Aspergillus sp. showed the greatest mycellial weight in 9 days at 0.3% saponin concentration as well as Penicillium specise A and B. Mycellial weight of all Saprophagous fungi was decresed than control group at 1.0% concentration of crude saponin. Saponin pattern were changed in 6th days of culture by Aspergillus sp. at 0.3% and deteriorated diol ginsenoside respectively. The amount of diol saponins was decreased all the duration of culture by Aspergillus sp. and Penicillium sp. B. whereas Pencillium sp. A was not any change. The amont of saponin in the fresh ginseng and white ginseng medium was decreased gradually according to culture periods by the saprophagous fungi.

  • It was attempted to observe the effects of ginseng saponin, one of the major components of the roots of Panax ginseng, on androgen biosynthesis from cholesterol in vitro as well as in vivo in rat testis. Ginseng saponin was administered by stomach tubing prior to intraperitoneal injection of cholesterol containing (4-14C)-cholesteroll into adult male rats and the liver, testis and blood serum were analyzed. The first high radioactivity of the liver and blood serum of test animal was observed at 6 hours after radioactive cholesterol injection, while that of control appeared at 12 hours after the injection. In the case of testis, the first high radioactivity of test group appeared between 4 and 6 hours after the radioactive cholesterol injection, while that of control appeared at 10-14 hours. Analysis of radioactivity distribution of cholesterol, androstenedione and testosterone in the testis of rats fed with/without ginseng saponin piror to (4-14C)-cholesterol injection showed that the saponin stmulated the synthesis of androgens from cholesterol. This was confirmed again by in vitro experiment using testis homogenate as an enzyme source. From the above experimental results, it was suggested that the ginseng saponin stimulates both cholesterol transport and the biosynthesis of androgens from cholesterol in rat testis.

  • It was attempted in this experiment to observe the effect of ginseng saponin on the growth and recovery rate of Saccharomyces rouxii which was shocked by heat-stress. The growth of S. rouxii was found most active when the saponin concentration in the malt extract broth was $10^{-3}%$ And the growth rate of S. rouxii grown on medium containing $10^{-3}%$ ginseng saponin was faster thanh that of the yeast grown on medium without saponin from the early log phase of the yeast. The growth recovery rate of S. rouxii which was heat-shockied by $80^{\circ}C$ - 30 minites was accelerated in the medium added ginseng saponin.

  • A promoting effect of ginseng saponin on the growth recovery rate of Saccharomyces rouxii which was treated by heat was confirmed in previous report(22). In order to deduce the promoting effect on the growth recovery of the heat stressed yeast, the effect of ginseng saponin on the activity and the heat stability of the amylase produced by Sacchirromyces rouxii were observed. The amylase showed the highst activity at 0.01% of saponin. At this concentration, the activity increased about 23% compared to the control. Furthermore, the ginseng saponin showed a protective effect against thermal inactivation of the amylase produced by Saccharomyces rouxii.

  • Effects of Panax ginseng on allergic reactions were studied uslng various in vivo and in vitro experimental models such as 48-hr passive cutaneous anaphylaxis, mediators-induced skin reactions, histamine release from rat peritoneal mast cells, hexosaminidase release from RBL-2H3 cells, and lipoxygenase assay . In all of anti-allergic experiments we conducted, ginseng components (50% ethanol extract or ginseng total saponin or ginsenosides) extracted from Korean red ginseng, did not show significant anti-allergic actions. In 48-hr passive cutaneous anaphylaxis and mediators-induced skin reactions, 50% ethanol extract did not suppress hypersensitivity reactions. Total saponin, 50% ethanol extract, and 8 major ginsenosides did not show inhibitory effects on lipoxygeanse activity. Ginseng total saponin did not inhibit histamine release from rat peritoneal mast cells. All of the ginseng components mentioned above were also tested on RBL-2H3 cells, but none of them inhibited hexosaminidase release from this cell line. These results suggest that Panax ginseng does not have effects on allergic reactions at the level of 50% ethanol extract or total saponin used. All of 8 major saponin components tested ($Rb_1$, $Rb_2$, Rc, Rd, Re, Rf, $Rg_1$, $Rg_2$), did not inhibit lipoxygenase activity and degranulation events.

  • In an attempt to elucidate the anti-inflammatory action of ginseng total saponin, protopanaxadiol and protopanaxatriol, the anti-inflammatory activity of three compounds was investigated under various acute and chronic inflammatory models. The blood vessel permeability was slightly inhibited by total saponin and protopanaxatriol treatments. Granuloma formation induced by 2% carrageenan was inhibited by total saponin and protopanaxatriol. The cotton-pellet granuloma formation was significantly inhibited by intraperitoneal injection of total saponin. Total saponin and protopanaxadiol inhibited leukocyte emigration and protein exudation in CMC-induced pouch but protopanaxauiol increased leukocyte emigration. The swelling of rat hind paw induced by 1% carrageenan was significantly inhibited by total saponin, protopanaxadiol and protopanaxatriol both single and 2 weeks treatments. Total saponin, protopanaxadiol and protopanaxatriol decreased the anti-inflammatory activity in adrenalectomized rat. Our results suggest that total saponin, protopanaxadiol and protopanaxatriol have potent anti-inflammatory activity, this may be mediated in part through stimulation of adrenal glands.

Enhanced Secondary Metabolite Biosynthesis by Abiotic Elicitor in Transformed Plant Root System

  • Plants generally produce secondary metabolites in nature as a defense mechanism against pathogenic and insect attack. In this study, we applied several abiotic elicitors in order to enhance growth and ginseng saponin biosynthesis in the hairy roots of P. ginseng. Generally, elicitor treatments were found to inhibit the growth of the hairy roots, although simultaneously enhancing ginseng saponin biosynthesis. The addition of selenium at inoculum time did not significantly affect ginseng saponin biosynthesis. However, when 0.5 mM selenium was added as an elicitor after 21 days of culture, ginseng saponin content and productivity increased to about 1.31 and 1.33 times control levels, respectively. These results suggest that processing time for the generation of ginseng saponin in a hairy root culture can be reduced via the application of an elicitor.

  • On the cAMP content in isolated gastric glands from rabbit stomach, the effect of ginseng components (total saponin, diol saponin, triol saponin) with pepsinogen secretion regulatory agents (cholecystokinin, isoproterenol, carbachol, propranolol, atropine, DECAMP, DBcGMP) were studied in vitro. According to the results, ginseng components may have the effect of stimulation or inhibition on cAMP production, and both dial saponin and triol saponin may be reciprocal effect to pepsinogen secretion regulatory agents. It seemed that the ginseng components may have the normalization action to pepsinogen regulatory agents on cAMP content in isolated rabbit gastric glands.
