Agrobacterium rhisogense에 의해 형질 전환된 인삼의 모상근 배양에서 Ginsenoside의 생산 (Ginsenoside Production by Hairy Root Cultures of Panax ginseng Transformed With Agrobacterium rhizogenes)

  • 인삼의 조직에 Agrobacterium rhizogenes strain 15834와 A4을 감염하여 형질전화체를 얻었다. 이는 인삼에서처럼 무균식물을 얻기어려운 경우 leaf disk 방법으로 모상근을 유도할 수 있었다. 모상근의 ginsenoside(Rg2,Rg1,Rf,Rd,Rc,Rbl, and Rb2)는 HPLC에 의해 定量하였으며, 진탕배양한 모상근의 ginsenoside의 함량은 0.34-1.19% 건량이었다. 이러한 결과는 재배 인삼과 배양 인삼의 ginsenoside의 함량에 비해 좋은 성과라고 사료된다.

Photosynthesis rates, growth, and ginsenoside contents of 2-yr-old Panax ginseng grown at different light transmission rates in a greenhouse

  • Background: Ginseng is a semishade perennial plant cultivated in sloping, sun-shaded areas in Korea. Recently, owing to air-environmental stress and various fungal diseases, greenhouse cultivation has been suggested as an alternative. However, the optimal light transmission rate (LTR) in the greenhouse has not been established. Methods: The effect of LTR on photosynthesis rate, growth, and ginsenoside content of ginseng was examined by growing ginseng at the greenhouse under 6%, 9%, 13%, and 17% of LTR. Results: The light-saturated net photosynthesis rate ($A_{sat}$) and stomatal conductance ($g_{s}$) of ginseng increased until the LTR reached 17% in the early stage of growth, whereas they dropped sharply owing to excessive leaf chlorosis at 17% LTR during the hottest summer period in August. Overall, 6-17% of LTR had no effect on the aerial part of plant length or diameter, whereas 17% and 13% of LRT induced the largest leaf area and the highest root weight, respectively. The total ginsenoside content of the ginseng leaves increased as the LTR increased, and the overall content of protopanaxatriol line ginsenosides was higher than that of protopanaxadiol line ginsenosides. The ginsenoside content of the ginseng roots also increased as the LTR increased, and the total ginsenoside content of ginseng grown at 17% LTR increased by 49.7% and 68.3% more than the ginseng grown at 6% LTR in August and final harvest, respectively. Conclusion: These results indicate that 13-17% of LTR should be recommended for greenhouse cultivation of ginseng.

인삼포의 환경조건과 인삼생육과의 관계 제1보 재식위치별 생산구조 (Relationship between Environmental Conditions and the Growth of Ginseng Plant in Field I. Productive Structures as Affected by Planting Positions and Ages)

  • 일복내에서의 인삼의 생육실태를 파악하기 위하여 3~5년근의 경 및 엽의 건물중, 엽면적과 동지수(LAI), 비엽중(SLW) 등의 수직분포 및 조도의 변화를 재식위치별로 조사하였다. 1. 3년근에서 조도는 지상 10cm 정도에서 가장 낮았고 후행일수록 낮았다. 2. 3년근 엽면적은 상의 가운데 $\frac{1}[3}$부분이 넓었고 비엽중은 수광량이 많은 곳일수록 높았으나 엽면적 지수는 행별로 큰 차이가 없었다. 3. 4년근에서는 3년근보다 광환경이 악화되었고 엽면적 및 동지수는 행별 차이가 현저했다. 4. 5년근에서는 통로부분의 엽량이 많아진 것 외에는 대체적인 경향이 4년근과 비슷하였다. 5. 경엽의 건물중은 고년근일수록 현저히 증대되었고 잎의 건물중은 보다 상층에 분포하였다.

연초 III형 Grey엽의 발생양상과 화학성분 특성 (An Aspect of Occurrence and Chemical Properties of Grey Leaf Tobacco (Type III))

  • The occurrence and chemical properties of grey tobacco leaves (Type III) found in 1995 crop of flue-cured tobacco cultivated in paddy field were investigated to compare with those of normal leaves. Cured leaves of NC82 were separated by visual characters into 3 classes of slight, fair and severe symptoms of grey leaf by the percentage of grey parts to whole leaf area. Number of samples classified with discoloring portion was the order of greyish brown > reddish brown > greyish yellow, respectively. Grey leaves of this type were mostly found among the leaves harvested from upper stalk position, and it was estimated that growth rate of upper leaves also influenced on the occurrence of grey leaves. Grey leaves showed remarkably lower b* and L* values than those of normal leaves, while a* value was mostly higher in grey leaves. These tendencies in chromatic aberration showed more remarkable difference in the degree of grey symptoms. Chemical analyses of grey leaf samples indicated that they contained less total nitrogen and nicotine, and more total sugar and starch than those of normal leaves. In chemical traits, these tendencies were accordance with the degree of grey symptoms, and within the same leaf, grey parts were decreased in total nitrogen and ether extract content compared with those of normal parts, but there was no difference in nicotine and Cl contents. 인삼(人蔘)의 잎(葉) 또는 줄기(莖) 첨가급식(添加給食)에 의(依)한 식이(食餌)의 영양효과(營養效果) 연구(硏究) (A Study on Nutritional effects of Rat diet by Supplementing with Leaf or Trunk of Panax Ginseng)

  • Korea has produced large quantities of Panax Ginseng roots which have a stimulating effect on the metabolisma of protein, lipid and nucleic acids in the body. Authors believe that the lear and trunk of Panax Ginseng might have some components possessing a similar activity to Panax Ginseng root although the quantity and quality of the functional components may be somewhat different. Therefore, this study was designed to observe the nutritional effects of diet supplemented with the leaves or trunks of Panax Ginseng. Weanling(body weight; $82{\pm}3g$) male albino rats were subjected to six different dietary groups as followings; A groups; dietary groups which were treated with steam for 30 min at $115^{\circ}C}$. B Groups; dietary groups which were not treated with steam. A-C (or B-C) dietary group; Control for A groups(or B groups) containing 99% wheat flour. A-1 (or B-1) dietary group; dietary group supplemented with 2% leaf of Panax Ginseng, which replaced 2% wheat flour of control diet. A-2 (or B-2) dietary group; dietary group supplemented with 2% trunk of Panax Ginseng, which replaced 2% wheat flour or control diet. Each group of rats was maintained with the corresponding diet for 40 days. And then they were sacrificed. The growth rate, protein efficiency ratio, and the contents of lipid and cholesterol in organs were determined. The results obtained are summarized as follows;1) The gained body weights of dietary group supplemented with 2% leaf(A-1 and B-1) or 2% trunk(A-2 and B-2) of panax Ginseng were more increased in comparison to the corresponding control group(A-C and B-C). 2) The gained body weight of each group in A-group(A-C, A-I and A-2) was higher than that or each corresponding dietary group in B-group(B-C, B-1 and B-2). 3) The protein efficiency ratios of A-1 and A-2 dietary group, and B-1 and B-2 dietary group were more improved in comparison to the corresponding control group(A-C and B­C). 4) The lipid contents in the liver of A-1 and B-1 dietary groups were lower than in that of A-C and. B-C dietary group, respectively. According to the above results, it could be suggested that the nutritional value of the wheat flour can be improved by supplement of 2% leaf or 2% trunk of Panax Ginseng.

인삼 신품종의 광합성 특성 (Characteristics of Photosynthesis among New Cultivars of Ginseng (Panax ginseng C.A. Meyer))

  • 고려인삼을 순계분리 육성하여 품종 등록한 인삼 신품종의 광도, 온도별 광합성 능력과 광합성 관련 형질인 기공, 엽록소, 비엽중 및 지상하부 형질들을 조사한 결과를 요약하면 다음과 같다. 신품종 및 자경종의 광포화점은 15,000 lux내외였고, 광합성 최적온도는 2$0^{\circ}C$였다. 광합성 능은 연풍>천풍>고풍>자경종 순으로 연풍이 가장 높았다. 호흡속도는 모든 품종이 온도가 높아짐에 따라 빨라지는 경향이었으며, 타품종에 비해 호흡량이 높았다. 비엽중은 천풍>고풍>연풍>자경종 순서로 높았고, 엽록소 함량은 품종간 차이가 근소하였다. 단위면적당 기공수는 연풍이 69.2개로 타계통의 40~47개보다 많았고, 기공의 크기는 연풍이 26.5 $\mu\textrm{m}$ 타품종 37.5~38.8 $\mu\textrm{m}$보다 적었다. 연풍은 경수가 1.8개, 장염수가 7.7개, 소염수가 41.0개, 엽면적이 12.3 dm$_2$로 타 품종보다 지상부의 생육이 양호하였다. 근중은 연풍>천풍>고풍>자경종 순으로 무거웠고, 동장은 천풍과 고풍이 각각 6.5, 6.8 cm로 자경종 5.6 cm보다 길어서 체형이 양호하였으나 연풍은 동장이 4.4 cm로 짧았다.

해가림 투광 정도에 다른 인삼의 생육 특성 (Growth Characteristics by Shading Rates in Panax ginseng C. A. Meyer)

  • 인삼포의 관행해가림 (볏짚피복, 투광율 3%)과 개량해가림(P.E. 차광망피복, 투광율 10%)의 해가림간 인삼 개체군의 군낙구조를 비교하기 위해서 년근별 (2, 4, 6년근), 재식위치별, 높이별로 엽면적, 비엽중(S.L.W,), 경엽의 건물중 및 조도의 분포를 조사한 결과는 다음과 같다. 1. 투광률(지상 40cm부위)은 2년근에서 관행해가림은 전행이 3%, 중간행이 2%, 후행이 1.5%였으며 평균 2.2%로 행간에 차이가 심했으나, 개량해가림에서는 전행이 12%, 중간행이 10%, 후행이 8%였으며 평균 10%로 투광률이 증가되어 행간의 차이가 근소하였다. 4, 6년근에서는 관행해가림의 광환경이 더욱 악화되었으나 개량해가림에서는 수광량이 행간의 차이가 근소하였다. 2. 2년근에서 재식위치별로 엽면적, 비엽중, 경 및 잎의 건물중 분포는 관행해가림과 개량해가림간에 차이가 근소하였다. 3. 4년근에서 재식위치별로 엽면적, 경, 엽의 건물중 분포가 관행해가림에서는 전행 및 중간행부분에 많이 분포되고 후행쪽은 적은 반면, 개량해가림에서는 행간에 차이가 적고 고루 분포되었으며 이러한 경향은 6년근시에 더욱 심하여 통로쪽의 지상부 건물중이 관행해가림은 개량해가림보다 현저히 증가하였다. 4. 근수량은 개량해가림이 관행해가림에 비해 28%증가되었으며, 재식위치별 근수량 지수는 관행해가림은 전행인 1∼2행이 높은 반면 개량해가림은 5행을 제외하고는 각행 고르게 분포되었다.

Identification of Phenolic Compounds and Quantification of Their Antioxidant Activities in Roasted Wild Ginseng (Panax ginseng C.A. Meyer) Leaves

  • The objectives of this study were to systemically identify phenolic compounds in roasted wild ginseng (Panax ginseng C.A. Meyer) leaves and investigate their radical scavenging activities. Seven phenolic compounds were identified by NMR (H, C, COSY, HMQC, HMBC) and mass (EI-MS, FAB-MS) analyses: 5-caffeoylquinic acid, kaempferol, quercetin, 3,4-dihydroxy-benzoic acid, 4-hydroxy-benzoic acid, 3-(3,4-dihydroxyphenyl)-2-propenoic acid, and 3-(4-hydroxy-3-methoxyphenyl)-2-propenoic acid. Their concentrations ranged from 0.4 (3,4-dihydroxy-benzoic acid) to 7.5 mg (kaempferol) per 100 g of roasted leaves. Among these compounds, 5-caffeoylquinic acid, kaempferol, and quercetin were found exclusively in the leaf portions of the ginseng plants. When their antioxidant activities were measured by DPPH and superoxide anion radical scavenging activity, quercetin, and kaempferol were most effective.

홍삼박을 활용한 토양미생물제제가 수박의 생육에 미치는 영향 -현장연구를 중심으로- (Effects of Soil Microbial Agent with Red Ginseng Marc on Growth of Watermelon -A Field Study-)

  • The aim of the present study was to determine the effects of soil microbial agent with red ginseng marc on growth of watermelon during 5 months. The three treatments were distributed in a completely randomized design with four replicates per plot. After 1 week in planting dates, the growth of watermelon (full length, stem thichness, leaf length and lead width) showed no significant difference in all treatments. During elongation stage (20 days), soil microbial agent with red ginseng marc was increased by 5% in leaf thickness (May 23) and 7~14% in leaf length (May 16 and 23) when compared to other treatments. For changes in fruit bearing thickness, there were no differences among treatments. Characteristics of watermelon in harvest season have an effect on harvest and length, stalk length, naval length, weight, sugar content and yield, except for harvest and width. In particular, yields increased with treatments with two soil microbial agent (7~12%), indicating that soil microbial agent with red ginseng marc showed higher yield than the other treatments. In conclusion, red ginseng marc-treated soil microbial agents have a positive effect on the harvest season of watermelon and can provide useful information for the selection of the functional microbial properties and the registration of microbial fertilizer.
