Optimization Study for the Production of 6-Shogaol-rich Ginger (Zingiber officinale Roscoe) under Conditions of Mild Pressure and High Temperature (가압조건에서 생강 유래 6-shogaol 변환을 위한 가열 조건 최적화)

  • Under optimized condition mild pressure in combination with specific temperature for heat treatment transform the 6-gingerol into 6-shogaol. The purpose of this study was to optimize the conditions used for heat treatment under pressure for increasing 6-shogaol content in ginger (Zingiber officinale Roscoe). A central composite experimental design was used to evaluate the effects of application temperature ($70-130^{\circ}C$) and temperature-holding time (95-265 min) on the transformation of 6-shogaol. The experimental values were shown to be in significantly good agreement with the predicted values (adjusted determination coefficient, $R^2{_{Adj}}=0.9857$). 6-Shogaol content increased as the application temperature and temperature-holding time increased. By analyzing the response surface plots, the optimum conditions of heat treatment (temperature and time) for increasing 6-shogaol content were found to be $127^{\circ}C$ and 109 min, respectively. Under these optimal conditions, the predicted 6-shogaol content was 3.98 mg/g dried ginger. The adequacy of the model equation for predicting the optimum response values was effectively verified by the validation data.

Isolation, Identification and Determination of Antioxidant in Ginger (Zingiber officinale) Rhizome

  • The antioxidative compounds and antioxidant contents of ginger (Zingiber officinale) rhizomes were determined. Substances reextracted using ethyl acetate from crude methanol extract of fresh ginger rhizome were separated through thin layer chromatography. Ten phenolic antioxidative bands were visualized through color reactions using ferric chloride-potassium ferricyanide and 1,1-diphenyl-2-picrylbydrazyl (DPPH). The antioxidative compounds were purified through preparative TLC and high performance liquid chromatography (HPLC), among which, five antioxidants were identified as 4-, 6-, 8-. and 10-gingerols and 6-shogaol on the basis of their molecular weights determined through LC-MS. As shown in experiments using DPPH free radicals, 6-Gingerol and PT4-HP8 (unknown) were revealed to be more efficient than BHT (butylated hydroxy toluene). Contents of gingerols were determined through reverse phase HPLC. Total gingerol contents (sum of 6-,8-, and 10-gingerols) in rhizomes of different ginger varieties varied significantly. The HG55 (collected at Wanju district in Korea) and the HG52 (imported from Brazil) showed the highest gingerol contents.

  • Ginger (Zingiber officinale Roscoe) is widely used as a common condiment for a variety of foods and beverages. In addition to its extensive utilization as a spice, the fresh or the processed rhizome is a useful crude drug in traditional Chinese medicine. It is considered to possess stomachic, carminative, stimulant, diuretic and antiemetic properties. Chemical studies on the pungent principles of ginger have been carried out by a number of investigators, and 6-gingerol and 6-shogaol as a major pungent substance have been isolated. In this study, the constituents inhibiting a drug metabolizing enzyme CYP3A4 from ginger were investigated. CYP3A4 is responsible for drug metabolism as heme-containing monooxygenases. As a result of experiment, 10-gingerol (lC$_{50}$ 5.75$\mu$M) isolated from EtOAc extract of ginger showed remarkable inhibitory activity compared to 6-gingerol ($IC_{50}$/ 14.56 $\mu$M) and zingerone ($IC_{50}$/ 379.63 $\mu$M). This paper describes the isolation, structure elucidation, and CYP3A4 inhibitory activity of these compounds. The structure of the compounds were identified by instrumental analysis such as LC-mass spectrometer and NMR.R.

Effects of Curcumin on the Microglial Activation (Curcumin이 microglia의 활성화에 미치는 영향)

  • Microglia, brain resident macrophages, play a central role in the inflammatory responses of the brain and are activated in brain injuries and several neurodegenerative diseases such as Alzheimer's and Parkinson's disease, thereby aggravating the course of these diseases. In this study, the effects of plantderived compounds such as curcumin or gingerol on the microglial activation were examined. Microglial cultures were prepared from 2~3 week mixed primary glial cultures obtained from the cerebral cortex of 1~2 day old rats and identified by immunocytochemistry using microglial-specific antibody OX-42. Microglia were activated by lipopolysaccharide (LPS) and interferon-${\gamma}$ (IFN-${\gamma}$) and the effect of curcumin or 6-gingerol on the microglial activation was examined. Specific parameters measured to monitor microglial activation were nitric oxide (NO), prostaglandin E$_2$(PGE$_2$) and tumor necrosis factor-$\alpha$ (TNF-$\alpha$) release. Curcumin (1~10 $\mu$M) inhibited NO release induced by LPS and IFN-${\gamma}$ in a dose-dependent manner whereas 6-gingerol (2~20 $\mu$M) did not have any effect on LPS/IFN-${\gamma}$-induced NO release. The levels of PGE$_2$and TNF-$\alpha$ induced by LPS and IFN-${\gamma}$ were also inhibited by 1~10 $\mu$M curcumin in a dose-dependent manner. These results showed that curcumin could modulate microglial activation.

Comparative antidiabetic activity of different fractions of methanolic extract of Zingiber officinale Roscoe in streptozotocin induced NIDDM rats

  • Earlier we have reported the antidiabetic activity of fresh juice of rhizomes of Zingiber officinale (Z. officinale) and its correlation with 5-HT receptor antagonism. Since 6-gingerol the marker compound of Z. officinale is reported to posses 5-HT anatgonistic activity, the present investigation, was undertaken to find out the concentration of 6-gingerol present in methanolic extract of Z. officinale and its different fractions (petroleum ether, toluene and chloroform). We also evaluated these fractions for antidiabetic activity in streptozotocin (STZ)-induced neonatal type 2 diabetic rats. Fasting glucose and insulin levels in non insulin dependent diabetes mellitus (NIDDM) rats were found to be significantly (P < 0.05) higher than control rats and these were significantly decreased by treatment with methanolic extract of Z. officinale and its fractions. The results of oral glucose tolerance test (OGTT) showed that methanolic extract and its fractions significantly (P < 0.05) decreased both STZ-induced increase in $AUC_{glucose}$ and $AUC_{insulin}$ values in NIDDM groups. Treatment with petroleum ether fraction produced a greater reduction in elevated glucose and $AUC_{glucose}$ levels as compared to treatment with other fractions. Treatment with methanolic extract of Z. officinale and its fractions also produced significant reduction in the elevated lipid, serum glutamate oxaloacetate transaminase (SGOT) and serum glutamate pyruvate transaminase (SGPT) levels in NIDDM rats. The effect of petroleum ether fraction on elevated lipid, SGOT and SGPT levels was significantly greater as compared to treatment with other fractions. The concentration of 6-gingerol was found to be maximum in petroleum ether fraction (11.430%) and minimum in chloroform fraction (0.973%). The methanolic extract and toluene fraction was found to contain 3.080% and 2.191 %, 6-gingerol respectively. In conclusion, our data suggest that methonolic extract and its fractions possess significant antidiabetic activity in NIDDM rats. The extent of activity appears to be dependent on the concentration of 6-gingerol present in the extract or its fractions.

Evaluation of Agronomic Characteristics Indigenous Germplasm for Development of High Quality and Yield of Ginger (Zingiber officinale Rosc.) (고품질 다수성 생강 품종육성을 위한 국내 유전자원의 농업적 특성 평가)

  • To obtained the basic information needed in the development of higher quality and yield of ginger, twenty-four indigenous colones of ginger were evaluated major agronomic characteristics under field and polyethylene house culture. There was highly significant differences in yield and 6-gingerol contents among the clones studied. Maximum yield was recorded in clone CG9579(3520kg/10a) followed by CG95134, CG9512, CG96109, CG9596, in field culture. The maximum 6-gingerol percentage was recorded in the clone CG9579 containing 4.44mg/g, and next was CG9583 4.35mg/g, CG95134 4.32mg/g DW. In addition, CG9503 and CG95105 clones showed more than 4,000kg/10a in polyethylene house culture.

생강엑기스의 제조에 관한 연구

  • 1) 본 연구에서 시료로 선정한 충남 서산산 건강(dry ginger)은 수분이 $9.4\%$, 회분이 $8.7\%$ 그리고 alcohol에 의한 추출량이 약 $9\%$이다. 이는 선진국에 채택사용하고 있는 건강의 규격기준에 의하면 양호하다. 2) Non- flavor물질의 추출을 최소화하고 특히 증류과정에서 유효성분 손실을 최소화 할 수 있고, 엑기스내의 용매 잔류량이 인체에 유해하지 않고 추출효율을 높일 수 있는 용매는 ethyl alcohol이다. 3) 널리 사용하고 있는 관류추출(percolation)의 성능을 분석하고 이의 개선방안을제시하였다. - 추출효율을 높이기 위하여 건강(dry ginger)의 입자를 작게하면 압력강하가 증대되어순환되는 용액의 유속을 제어하기가 힘들다. - 입자가 작을 시에는 유체의 흐름이chan-nelling현상을 나타낸다. - 위와 같은 조건에서는 물질 전달속도가 느리므로 추출효율을 증대시킬 수가 없다. - 따라서 percolation추출에 사용되는 건강의 입자크기는 30mesh크기 이상이어야 운전조작이 용이하나 추출효율이 낮으므로, 추출시간 6시간에 회수된 생강엑기스양은 약 $2.5\%$이다. 4) percolation추출의 단점을 보완하기 위하여 기계적교반 추출을 선택하여 다음과 같은 개선점을 찾았다. - 교반형 추출에서는 고 - 액분리시 cake 저항에서 문제가 야기되지 않는 범위까지 건강의 입자를 작게할 수 있으므로 추출효율을 크게 향상시킬 수 있었다. 즉, 작게 분쇄된 건강(30mesh통과$90\%$)을 대상으로 추출시간 3시간에 $7\%$의 회수율로 증대시켰다. 최적 운전조건은 다음과 같다. 건강시료:1kg 시료크기:-30mesh$90\%$ 용매:ethyl alcohol 3$\iota$ 교반속도:900r.p.m 추출온도:상온($15\~25^{\circ}C$) 추출시간:3시간 일차 추출조건과 동일하게 하여 얻어진 엑기스의 수율이 $2\~2.5\%$이므로 총엑기스의 수율은 건강(dry ginger)무게기준으로 $8.5\~9.5\%$이었다. 5) 교반추출의 효율이 개선되었다 하더라도 추출물의 분리가 용이하여야만 공정의 이용이 가능하다. 그러므로 교반추출후 고 - 액분리를 위하여 정압여과 장치를 이용하여 여과시 cake의 평균 비저항을 얻었으며, 이의 값은 $4.31\times10^8cm\;/\;gr$으로서 여과에는 어려움이 없다는 것을 의미한다. 따라서 추출속도와 효율이 상대적으로 우수한 교반형 추출기의 가능성을 예시할 수 있음을 알 수 있었다. 6) 추출물을 농축과정에서 휘발성 oil의 손실을 최대로 줄이기 위해서는 단순증류를 하지 말고 분별증류를 수행하여야 하며, gingerol과 같은 중요성분의 열분해 반응을 억제하기 위해서는 열전달 효율을 증대시켜 증류조작을 원활히 수행하여야 하므로, still내의 농축물을 계속 교반시켜야 하며 감압상태에서 증류온도는 $40\~50^{\circ}C$로 유지시키는 것이 가장 바람직하다. 7) Ethyl alcohol로 추출된 엑기스내의 수분이나 회분함량은 외국산 제품에 비하여 약간 낮고, 반면에 조지방 및 조단백 성분의 함량은 약간 높게 나타나고 있어 대체적으로 본 연구에서 얻어진 엑기스내의 비풍미성분(non- fla-vour component) 함량은 외국산에 비하여 많은 차이가 없다. 8) 수입 외국산에 비하여 국산엑기스(본 연구에서 ethyl alcohol로 추출)내의 무기성분등의 함량은 비교적 낮은 편이다. 9) 건강에서부터 oleoresin을 얻어 paradol을 제거시킨 후 순수한 gingerol을 분리하여 IR과 NMR로 확인한 결과, 국산건강의 엑기스에는 주로 6-gingerol이고 약간의 10-gingerol이 함유된 것으로 나타났다. 10) 순수하게 분리된 gingerol을 열분석(TGA와 DTG)한 결과 약 $75^{\circ}C$에서 gingerol의 열분해 반응이 일어남을 알수 있었다. 11) 건강 분말시료와 엑기스내의 미생물 검사 결과 건강분말에서는 세균수가 많이 존재하는 것으로 나타났으나, 이는 ethyl alcohol로 추출하는 공정 중 대부분의 균들이 사멸된 것으로 나타났다. 12) 관능적 측면에선, 본 연구에서 제조한 엑기스와 수입엑기스를 비교한 결과 생강 특유의 맛은 비슷했으나, 수입엑기스에서는 쓴맛과 톱밥냄새를 느낀다는 결과를 나타내었으며 전체적인 종합적 풍미는 국산 건강엑기스가 좋은 것으로 나타났다.

Physicochemical Properties of Repetitive Heat-treated Ginger and Its Quantitative Conversion of Gingerol to Shogaol

  • Ginger was steamed at $121^{\circ}C$ and $1.5lb/in^2$ for 30 min, dried at $60^{\circ}C$ for 12 h, and each step was repeated nine times. During processing, the lightness ($L^*$ value) and yellowness ($b^*$ value) decreased from $85.65{\pm}0.33$ and $26.99{\pm}0.20$ in the non-treated ginger to $56.91{\pm}0.25$ and $16.69{\pm}0.06$ in ginger treated for the ninth treatment. On the other hand, redness ($a^*$ value) increased from $-1.51{\pm}0.03$ to $7.34{\pm}0.08$ on the eight treatment and then decreased to $7.21{\pm}0.04$ on the ninth theatment. The contents of 6-gingerol decreased from $3.257{\pm}0.067mg/g$ in the non-treated ginger to $0.567{\pm}0.036mg/g$ on the theatment, whereas the contents of 6-shogaol increased from $1.299{\pm}0.050mg/g$ to $2.999{\pm}0.089mg/g$ on the sixth treatment and decreased to $2.099{\pm}0.039$ on the ninth treatment. The contents of 10-gingerol decreased slightly from $1.106{\pm}0.125mg/g$ to $0.806{\pm}0.026mg/g$. Unlike the 6- and 10-gingerol, the contents of 8-gingerol did not change greatly, with values between $0.916{\pm}0.005mg/g$ and $1.106{\pm}0.005mg/g$ being observed during processing. The tyrosinase inhibitory activities were increased from $43.42{\pm}11.45%$ in the non-treated ginger to 100% on the sixth treatment and then decreased to $51.98{\pm}7.36%$ on the theatment. The antioxidative activity was retained during processing.

CA Storage for Ginger Depending on CO2 Concentrations (탄산가스 농도에 따른 생강의 CA 저장효과)

  • In order to establish effectiveness of CA storage and adequate CO2 concentration, it was investigated the quality chanties of Singer during CA storage for 150 days at different CO2 concentrations ranging from 3% to 12% and 3% fixed oxygen concentration. Weight loss tend to decrease with increase of CO2 concentrations. Sprouting ratio and the loss of gingerol was shown to be less as CO2 concentration increase but to be more than control stored at 12$^{\circ}C$, 95% RH within the concentration less than 6% CO2.

Comparison of the Antioxidant Effects of Diallyl Sulfide, Capsaicin, Gingerol and Sulforaphane in $H_2O_2$-Stressed HepG2 Cells (산화스트레스가 유도된 인체 간암세포 (HepG2)에서 Sulforaphane과 Diallyl Sulfide, Capsaicin, Gingerol의 항산화효과 비교연구)

  • Oxygen is necessary to sustain life, yet cellular oxygen metabolism creates destructive elements called free radicals. Free radicals are chemically unbalanced and carrying free electrons that can damage molecules, potentially damaging the cell itself. For this reason, many antioxidant products, including supplements and functional foods, are being developed. In particular, natural products are rich sources of pharmacologically active compounds. The purpose of this study was to investigate the antioxidant effects of target biomaterials in Korean traditional spices such as diallyl sulfide (DAS), capsaicin (CAP), and gingerol (GGR), and to investigate the response of the antioxidant defense system to oxidative stress by hydrogen peroxide ($H_2O_2$) compared to sulforaphane (SFN) in HepG2 cells. After the analysis of the cell viability using Cell Counting kit-8 (CCK-8) assay, we determined that the optimum levels were $200{\mu}M$ DAS, $25{\mu}M$ CAP, $50{\mu}M$ GGR, and $12.5{\mu}M$ SFN. Antioxidant enzymes were measured and protein expression was detected by Western blotting. All treatments showed a significant decrease in antioxidant enzyme activity such as superoxide dismutase, catalse, and glutathione peroxidase in HepG2 cells. Additionally, DAS, CAP, GGR and SFN increased the antioxidant system-related transcription factor Nrf2 which was found to be regulated by the activation of MAPK-JNK in this study. In conclusion, these results indicate the protective effects of DAS CAP, GGR, and SFN against $H_2O_2$-induced oxidative stress.
