Silk Fibroin Microsphere and Its Characterization

  • Using gel filtration chromatography, high molecular silk fibroin with high purity was obtained and silk flbroin microsphere particles (SFMP) could be simply made by spray dryer method. Also, some of the physicochemical properties of SFMP and morphology were investigated. The average molecular weight of pure silk fibroin protein dissolved in calcium chloride is about 61,500g/㏖ as measured by gel permeation chromatography. SFMP was spherical in shape, and particles, sized average of 2 ${\pm}$ 10 ${\mu}$, were observed by SEM and particle analyzer, respectively. Obtaining microspheres particles by spray dryer method accelerated the transition from the random coil to the $\beta$-sheet structure during spray dryer treatment. It was identified by the basic fourier transform infrared spectroscopy of SFMP. The swelling ratio of SFMP is majorly dependent on the pH of the solution, not on the occurred gelation. The characteristic structure, which might be applicable to immobilization of drugs is superior to other matrix materials for the use of biomaterials with skin affinity.

  • Hydrogels are crosslinked hydrophilic matrices for a variety of biomedical applications. Silk fibroin (SF), one of typical natural biomaterials, has been explored as base material for hydrogel. Photocrosslinked SF hydrogel containing poly(ethylene glycol) (PEG) was formulated through visible light initiated thiol-acrylate photopolymerization. The morphological, structural and thermal properties of SF - PEG hydrogel was investigated through scanning electron microscopy, X-ray diffractometry, thermogravimetry, and differential scanning calorimetry. The morphology of SF hydrogel showed dot and uneven surface with network cross-section. X-ray diffraction curves showed that the specific diffraction peaks of PEG were not changed by the intensity of the peaks were affected by sonication. Thermo-degradation behavior of SF - PEG hydrogel sonicated was significantly affected and became complex pattern compared to unsonicated ones. However, the melting endothermic temperature of SF - PEG hydrogel was not changed but the crystalline enthalpy was decreased by gelation and sonication.

Histopathology of a microspridian infection in ayu, Plecoglossus altivelis, from southern Korea (양식 은어의 미포자충 감염예에 대한 병리조직학적 관찰)

  • On September in 1998, a microsporidian infection was recognized in ayu, Plecoglossus altivelis, farmed on Kyongnam province of South Korea. Cumulative mortality was around 10% in 10 days. Infected fish which were piping for air near the surface of the water or in the asphyxic, lethargic condition revealed darkening of body and abdominal distention. Numerous whitish nodules up to 3mm in size were observed throughout most of body organs and tissues including gill, operculum, peritoneal wall and organs. Xenomas were also histologically confirmed in multiple internal organs with the evidences suggesting circulatory disturbance. Based on the morphology of spore and xenoma, and the distribution of xenomas in organs and tissues, this disease was diagnosed to be a microsporidiosis caused by Glugea plecoglossi. The mortality might be deeply related to the local circulatory disturbance by xemonas rather than the mechano-chemical effect of xenomas on adjacent tissues.

Silk Fibroin Membrane as Guided Bone Regeneration in Rat Calvarial Defects

  • Silk fibroin membrane was prepared and examined to know the feasibility of SF membrane as guided bone regeneration. The morphology of silk membrane was flat and smooth surface. The conformation of silk fibroin was $\beta$-sheet structure. When the silk membrane was applied on the rat calvarial defect model, it showed significantly higher new bone formation than uncovered control in histomorphometric analysis. The silk membrane was covered by thin fibrotic tissue and there was not observed any inflammatory cells infiltration. In conclusion, silk fibroin membrane could be useful materials for guided bone regeneration.

  • Antheraea pernyi silk powder was prepared by treatment with HCl and NaOH. The degree of hydrolysis of Antheraea pernyi silk fiber was examined. The morphology and structural characteristics of Antheraea pernyi silk powder were investigated by using SEM, FTIR and X-ray diffractometer. As the concentration of HCl and NaOH and tratment temperature increased, in general, the degree of hydrolysis of Antheraea pernyi silk fiber increased. On the other hand, the degree of hydrolysis of Antheraea pernyi treated with 3 N NaOH at 120$^{\circ}C$ for 24 hr was 70 wt%, which was lower than that of 90$^{\circ}C$(83 wt%). The morphology of acid/alkali resistance fraction of Antheraea pernyi silk fibroin was transformed from fiber form to powered one with an increase of hydrolysis. The conformation of Antheraea pernyi silk powder characterized by FT-IR spectrometer and X-ray diffractometer ${\beta}$-sheet and ${\alpha}$-helix structure.

The long-term prognostic impact of sentinel lymph node biopsy in patients with primary cutaneous melanoma: a prospective study with 10-year follow-up

  • Purpose: Sentinel lymph node (SLN) biopsy (SLNB) is widely accepted for staging of melanoma patients. It has been shown that clinico-pathological features such as Breslow thickness, ulceration, age, and sex are better predictors of relapse and survival than SLN status alone. The aims of this study were to evaluate the long-term (10-year) prognostic impact of SLNB and to determine predictive factors associated with SLN metastasis, relapse, and melanoma specific mortality (MSM). Methods: This was a prospective observational study on 289 consecutive patients with primary cutaneous melanoma who underwent SLNB from January 2000 to December 2007, and followed until January 2014, at an Italian academic hospital. Results: SLN was positive in 64 patients (22.1%). The median follow-up was 116 months (79-147 months). Tenyear disease-free survival and melanoma specific survival were poor in patients with positive SLN (58.7% and 66.4%, respectively). Only the increasing Breslow thickness resulted independently associated to an increased risk of SLN metastasis. Cox regression analysis showed that a Breslow thickness >2 mm was an independent predictor of relapse, and male sex and Breslow thickness >2 mm was a predictor of MSM. At 10 years, SLN metastasis was not significantly associated to either relapse or MSM. Conclusion: After the fifth year of follow-up, SLN metastasis is not an independent predictive factor of relapse or mortality which are mainly influenced by the characteristics of the primary tumor and of the patient. Patients with a Breslow thickness >2 mm regardless of the SLN status should be considered at high risk for 10-year relapse and mortality.

Studies on the Features of Amyloodinium sp. Parasitized in Black Seabream, Acanthopagrus schlegeli (감성돔(Acanthopagrus schlegeli)에 기생한 Amyloodinium sp.의 성상에 관한 연구)

  • To know the features of Amyloodinium sp. parasitized in Black seabream, Acanthopagrus schlegeli, morphology and reproduction type of the parasite were investigated. Infection mode and histopathology of the parasites were also studied. In the developmental and morphological observation, the parasite passed through parasitic and non-parasitic phases with three developmental stages termed trophont, tomont, and dinospore. The trophont, 30-$80{\mu}m$ ovoidal or pyriform, for the vegetative stage had a spherical nucleus with $1.5{\mu}m$ in diameter, many food vacuoles and starch grains in cytoplasm. The tomont, 80-$90{\mu}m$ spherical, for the reproductive stage resorbed a stalk, secreted a cyst wall and reproduced within it. A dinospore, small 10-$15{\mu}m$ biflagellated, for the infestation stage had a stigma. The parasite was propagated for 15 days by serial passage in Korean rockfish (Sebastes schlegeli) at $25{\pm}1^{\circ}C$. Trophonts began to detach from the fish about 3 days after infection and was completed within 5 days at $25{\pm}1^{\circ}C$. It took from 3 to 5 days to reproduce at the same temperature. Reinfected fish showed that parasites penetrated under the epithelia of gill filament and gill lamellae causing hyperplasia and degeneration at infected area.

A TREMATODE GENUS BACCIGER PARASITIC IN BIVALVES (조개류에 기생하는 Bacciger속 흡충류에 관한 연구)

  • Cercaria larvae of Bacciger harengulae which is parasitic to Meretrix lusoria, rapes japonica and Solen strictus were studied in terms of their morphology and incidence of infection rate. The host bivalves were collected from Naechodo, Okgu-gun during the Period from January to November in 1980. The tail of the cercaria bears 27 pairs of tufts of setae, the tufts being composed of 6 fine rays. Oral sucker, intestine, ventral sucker and a pair of testes are distinctive. The excretory duct bears numerous granules of various size, and the flame-cell arrangement is represented by a formula 2[(3+3)+(3+3)]=24. The cercaria is somewhat similar to the previously known Cercaria pectinata . It is however conformed that present cercaria belongs to Bacciger harengulae. Infection rate showed seasonal variation and host selectivity. Higher infections were observed in Solen strictus, and lower in Meretrix lusoria. Generally, the larger bivalves over 2-3 years old showed the higher infection rate.

Identification of Lacrymaria velutina (Pers. Ex Fr.) Konrad & Maubl. from Micheon-myeon, Jinju-city, Korea

  • We identified Lacrymaria velutina of the Coprinaceae in Korea. The unusually large and sturdy fruiting body, fibrillose to fibrillose-scaly cap and stalk without a volva with an obscure superior hairy ring zone or hairy annulus, and blackish brown, warted spores distinguished this species from closely related Psathyrella species. An illustrated account of the microscopic traits is presented. Fruiting bodies with obtusely hemispherical caps, 2.5~6 cm, becoming convex with age; surface dry, densely fibrillose-scaly with split margin; stipe, 4.5~6 cm, equal, hollow, fibrillose, dry, whitish above the superior ring zone, light brown below; crowded gills, adnexed, dark black at maturity. Pileipellis typically cellular with the gill edge appearing white and beaded. Blackish brown basidiospores that discolor in concentrated sulfuric acid. Spores elliptical, warted, $9\sim11{\times}6\sim8{\mu}m$, with prominent snout-like germpores. Cheilocystidia abundant, $57\sim68{\times}19\sim25{\mu}m$, and narrowly elongated clavate, often clustered in threes or fours. Pleurocystidia rarely present, $45\sim47.5{\times}12\sim13{\mu}m$, and clavate to utriform. This trait distinguishes our sample as L. velutina from other Psathyrella spp. of the Coprinaceae, which have smooth spores. This taxon was clarified by the observation that Psathyrella spores fade in concentrated sulfuric acid. A molecular phylogenetic study revealed that our specimen was Lacrymria velutipes, which is closely related to Lacrymaria lacrymabunda. Moreover, those two species are clearly distinguishable from other Psathyrella species, which agreed with the morphologically distinctive traits described above. We believe that this is the first report of this taxon, which has not been described in Korea.

Heterojunction Quantum Dot Solar Cells Based on Vertically Growth TiO2 Anatase Nanorod Arrays with Improved Charge Collection Property

  • The Quantum dot (QD) solar cells have been under active research due to their high light harvesting efficiencies and low fabrication cost. In spite of these advantages, there have been some problems on the charge collection due to the limitation of the diffusion length. The modification of advanced nanostructure is capable of solving the charge collection problem by increasing diffusion length of electron. One dimensional nanomaterials such as nanorods, nanowires, and nanotubes may enhance charge collection efficiency in QD solar cells. In this study, we synthesized $TiO_2$ anatase nanorod arrays with length of 200 nm by two-step sol-gel method. The morphology and crystal structure for the nanorod were characterized by using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and X-ray diffraction (XRD). The anatase nanorods are single-crystalline and possess preferred orientation along with (001) direction. The photovoltaic properties for the heterojunction structure QD solar cells based on the anatase nanorod were also characterized. Compared with conventional $TiO_2$ nanoparticle based QD solar cells, these nanostructure solar cells exhibited better charge collection properties due to long life time measured by transient open circuit studies. Our findings demonstrate that the single crystalline anatase nanorod arrays are promising charge transport semiconductors for heterojunction QD solar cells.
