Heterochromatic Knob Number And Karyotype in Korean Indigenous Waxy Corn by Giemsa C-banding Pattern of Mitotic Chromosome (C-banding 패턴에 의한 한국 재래종 찰옥수수 염색체의 Heterochromatic knob 수와 핵형)

  • A Giemsa C-banding method was used for the identification of somatic chromosomes and heterochromatic knob position in Korean indigenous waxy com (Zea mays L.). 5 inbred stocks were examined and their heterochromatic knob numbers ranged from 6 to 12. In comparison of homologous chromosomes of two stocks of YS-1 and MY-1, knob numbers, knob positions, arm ratios and relative length of chromosomes were different between the genotypes. The length of homologous chromosomes in YS-1 were generally larger than those of MY-1. The Giemsa method was proved to be useful for the identification of somatic chromosome and a C-banded diagram showing knob positions, arm ratios and relative length of chromosome could be used as a good tool to compare the characteristics of chromosomes of Korean indigenous waxy corn stocks.

  • Giemsa C-banded karyotypes of A. thunbergii, A. deltoide-fistulosum, A. cyaneum and A. cyaneum var. deltoides were analyzed, and the interspecific relationships were investigated on the basis of the C-banding patterns. The banding pattern of each species was unique and made possible an easy and clear separation among them. A. thunbergii and A. deltoide-fistulosum revealed very close banding relationship but a. cyaneum and A. cyaneum var. deltoides showed significant difference in banding pattern in spite of their close plant systematic position.

  • 한국산 박새속 조류 Porus maior(박새), Parus ater(진박새), Parus palustris(쇠박새), Parus varius(곤줄박이)의 핵형을 C-banding 방법으로 분석한 결과 ZZ-ZW 염색체를 동정, 확인할 수 있었으며, 구조적 이질염색질의 부위가 일반염색 방법에 의한 핵형 비교에서 보다 종간에 더 많은 차이를 나타냈다. 그러나 일반염색 방법과 C-banding 방법 만으로는 4종간의 종분화 기작을 추측할 수 없었다. The chromosomal analysis of Paws motor, Paws ater, Pahs palustris and Pows varius of the genus Paws in Korea were performed by C-handing method. The identification of sex chromosones was ascertained and the part of constitutive heterochromatin was more different between interspecies than by conventional Giemsa staining method. However, conventional Giemsa staining and C-banding methods could not explain mechanisms of speciation between four species.

  • A Giemsa C-banding method was used for the identification of somatic chromosomes of Korean indigenous maize(Zea mays L.). Two Korean indigenous maize stocks and an American hybrid maize for comparison were examined. Ten deeply stained heterochromatic knobs whose position and size were different between the genotypes, two satellites and interstitial bands were observed. The length of homologous chromosomes compared by the relative lengths of chromosomes presented as a percentage of the length of chromosome 10 were different between the genotypes. The Giemsa method proved to be useful for the identification of somatic chromosomes and for the characterization of different stocks of Korean indigenous maize.

  • C-banded karyotypes of eight taxa of Korean Allium sect. Sacculiferum were examined by means of Giemsa techniques. Each of the taxa had their own characteristic bands. Most of the bands on the somatic metaphase chromosomes were mainly located on the short arm, intercalary region and medium sized chromosomes. In addition, the heterochromatic bands were usually facultative, recognized as two spots, and all investigated taxa had a pair of satellites of heterochromatin. The similarities of total length and number of bands revealed that three varieties of A. thunbergii G. Don have a closer cytological relationship than the others. The C-banding patterns of A. longistylum and A. linearifolium were examined for the first time in this study, and the former was clearly distinguished from the others by the distribution and number of its bands. The proportion of heterochromatic bands suggested that A. thunbergii var. thunbergii is the most primitive, and A. sacculiferum the most advonced, within the section.

  • Giemsa C-banded mitotic chromosome prepatations from White Leghorn, New Hampshire and Rhode Island Red inbred lines were compared for frequency of C-band regions on individual chromosomes. Except for autosomes 3, 6, 8 and 9 and W sex chromosomes, C-banding was extremely variable in other macrochromosomes. No divergence for C-band difference between homologous chromosomes of these lines was detected. Approximately 75% of the mitotic metaphase microchromosomes have recognizable C-band regions with the current technique.

  • On the basis of karyotypic analysis performed by conventional staining and Giemas C-banding technique, cytological relationship was inferred for 21 lines of Paeonia lactiflora Pal. cultivated in Korea. It was very difficult to infer their organized karyotypic classification system using the composition of somatic chromosomes involving sat-chromosomes, relative length of chromosomes, arm ratio and karyotypic formulae by conventional staining. From the distribution and number of Giemsa C-bands on the chromosomes b and c, 21 lines can be subclassified into 5 groups. It seems that the karyotypic polymorphism is observed in 21 lines of cultivated P. lactiflora because peony mainly propagates by outbreeding.

  • The karyotypes of Korean Sciunis vulgaris coreas and Tamias sibiricus asiaticus were analyzed by conventional Giemsa staining and C-banding method. The diploid chromosome number (2n) of Sciunis vulgaris coreae 40 consisting of 6 metacentric, 8 submetacentric, 3 subtelocentric and 2 telocentric autosome pairs, submetacentric X and acrocentric or subtelocentric Y chromosome. The arm number (NF) of this species was obtained as 72, excluding the gonosomal arms. Tamias sibiricus asiaticus has a 2n of 38. The karyotype was represented by 3 metacentric, 4 submetacentric, 5 subtelocentric and 6 telocentric autosome paits and 2 sex chromosome. The X chromosome was submetacentric chromosome and the Y was the smallest chromosome with a median. The NF was 60. In S. vulgaris coreae constitutive heterochromatins were observed at the centromeres and telomeres. Constitutive heterochnomatins of T sibiricus asiaticus were primarily observed at the centromeres. These results suggested that non-Robensonian reanagenents and distribution of constitutive heterochromatin played an imporiant role in karyological differentiation of these species.

  • The chromosomal analyses of Dendmcopos major hondoensis and D. leucotos Ieucotos(Picidae:Piciformes) in Korea were performed by conventional Giemsa staining and C-banding method. The diploid number of two species was 2n=90-92 and arm number was AN= 92-96.The conventional karyotypes were very similiar but distribution of constitutive heterochromatin were differ in the first chromosome. The second and several pairs of macro-telocentric chromosbmes have telomeric constitutive heterochromatin.
