Evaluation of Resistance to Rice Sheath Blight (Rhizoctonia solani Kühn) of Rice Germplasms at Seedling Stage (유묘접종에 의한 벼 유전자원의 벼잎집무늬마름병 저항성 평가)

  • Rice sheath blight (ShB), caused by Rhizoctonia solani K$\ddot{u}$hn, is one of the serious fungal disease resulting in severe yield loss in rice field worldwide. There are limited sources of genetic resistance and no commercial cultivar with high level of ShB resistance is currently available in Korea. In order to seek available resources with high level of resistance to ShB, 40 rice germplasms were used to evaluate disease reactions including ShB, leaf blast and bacterial leaf blight and these germplasms also examined agronomic traits such as days to heading, culm length, panicle length, No. of panicles, No. of spikelets per panicle and so on. There is wide variation in agronomic characters and disease reactions. Rice germplasms also showed considerably different ShB reaction caused by inoculation at seedling stage. Areumbyeo, Gayabyeo, IR579-Es44 and IR64 showed more strong reaction to ShB than the others. Especially, Gayabyeo is considerably available to develop a new variety with resistance to ShB in Korea.

Variation of Oil Contents and Fatty acid Compositions in Korean Soybean Germplasms (국내 콩 유전자원의 지방함량 및 지방산 조성변이)

  • The objectives of this study was to determine the oil contents and fatty acid compositions of soybean germplasms including major Korean cultivars, and to provide the basic information of fatty acid composition for development high quality soybean varieties. Among 46 Korean major cultivated soybean samples, the oil contents were varied from 15.8% to 20.3%, and the average oil content was 18.2%. Crude oil content of cv. Saebyolkong (20.3%) was the highest among that of other cultivated soybeans. Likely many other oils of plant origin, most fatty acids in soybean are unsaturated. The highest percentage of fatty acid in soybean was linoleic acid (18:2), followed in a decreasing order by oleic (C18:1), palmitic (C16:0), linolenic (C18:3), and stearic acid (C18:0). The composition of C18:1 and C18:2 fatty acids among 563 soybean germplasms and cultivars which were positively correlated with nutritional quality of edible vegetable oil, were ranged $13.29{\sim}43.30%\;and\;38.21{\sim}61.51%$, respectively, and the C18:3 which were negatively correlated with flavor quality were varied from 5.03% to 11.48%. Also, the composition of C16:0 and C18:0 which were known to raise total cholesterol levels in human serum were ranged $8.50{\sim}14.78%\;and\;1.86{\sim}4.78%$ respectively. On the other hand, the range of fatty acid composition among 46 Korean major soybean cultivars has been investigated to be: C16:0, $8.96{\sim}13.23%$; C18:0, $2.55{\sim}4.20%$; C18:1, $16.33{\sim}36.41%$; C18:2, $42.32{\sim}58.84%$; C18:3, $6.98{\sim}10.72%$. Among the 563 germplasms tested, there are significant correlations among unsaturated fatty acids: negative between C18:1 and C18:2 (r=-0.961**) or C18:3 (r=-0.756**), and positive between C18:2 and C18:3 (r=0.608**). Also, the C18:1 was negatively correlated with saturated fatty acids, C16:0 (r=-0.231**) and C18:0 (r=-0.479**).

Identification of Lettuce Germplasms and Commercial Cultivars Using SSR Markers Developed from EST (EST로부터 개발된 SSR 마커를 이용한 상추 유전자원 및 유통품종의 식별)

  • The objective of this study was to develop simple sequence repeat (SSR) markers from expressed sequence tags (EST) of lettuce (Lactuca sativa) and identify 9 germplasms from 3 wild species of lettuce and 61 commercial cultivars using the developed EST-SSR markers. A total of 81,330 lettuce ESTs from NCBI databases were used to search for SSR and 4,229 SSR loci were identified. The highest proportion (59.12%, 2500) was represented by trinucleotide, followed by dinucleotide (29.70%, 1256) and hexanucleotide (6.62%, 280) among SSR repeat motifs. Totally 474 EST-SSR primers were developed from EST and a random set of 267 primers was used to assess the genetic diversity among 9 germplasms and 61 cultivars. Out of 267 primers, 47 EST-SSR markers showed polymorphism between 7 cultivars. Twenty-six EST-SSR markers among 47 EST-SSR markers showed high polymorphism, reproducibility, and band clearance. The relationship between 26 markers genotypes and 70 accessions was analyzed. Totally 127 polymorphic amplified fragments were obtained by 26 EST-SSR markers and two to nine SSR alleles were detected for each locus with an average of 4.88 alleles per locus. Average polymorphism information content was 0.542, ranging from 0.269 to 0.768. Genetic distance of clusters ranged from 0.05 to 0.94 between 70 accessions and dendrogram at a similarity of 0.34 gave 7 main clusters. Analysis of genetic diversity revealed by these 26 EST-SSR markers showed that the 9 germplasms and 61 commercial cultivars were discriminated by marker genotypes. These newly developed EST-SSR markers will be useful for cultivar identification and distinctness, uniformity and stability test of lettuce.

Annual Analysis of the Agronomic Traits of Global Wheat Germplasms in the Korean Environment (국내환경에서 밀 유전자원의 연차간 농업특성 분석)

  • Securing a range of wheat resources is of particular importance with respect to wheat breeding, as it provides a broad genetic foundation. Although wheat breeders have used different wheat germplasms as material resources in current breeding systems, the traits of most germplasms collected from foreign countries differ from the unique traits that have evolved in the Korean environment. In this study, conducted over a 2-year period (2018 and 2019), we therefore evaluated the agricultural traits 1,967 wheat germplasms collected not only in Korea but also worldwide. During the period from sowing to February, the average temperature in 2019 was greater than 1℃, whereas from March to June, the average temperature was approximately 0.9℃ higher in 2018. Compared with the growth recorded in 2018, the stem length in 2019 increased by approximately 20 cm in 2019, and there were notable differences heading date and maturation between 2018 and 2019. In 2019, the heading dates of 973 and 713 wheat resources were earlier and later than those in 2018, respectively. Moreover, stem length was found to be highly correlated with the heading date and maturation. In Korea, where the rainy season and tine of rice transplantation overlap with the time of wheat harvest, early flowering time with high grain yield has been the most important selection target with respect to wheat breeding. We anticipate that the findings of this study will provide would use a foundation for the selection of elite materials and the development of resource core-sets for Korean wheat breeding programs.

  • The green marine algae, Capsosiphon fulvescens has been cultivated in south coast of southern Korea for many years on a commercial scale. This species is very popular in Korean as a food supplement because of its attractive flavor and flexcible taste. It is, therefore, necessary to isolate and utilize qualified germplasms for mass production of this economic seaweed. Several reports have been published on phycological applications of RAPDs including the characterization of interspecific genetic variation, the identification of isolates and hybrids, and the study of phylogenetic relationships. However few authors have used RAPDs to assess the genetic variability among populations of a seaweed species(van Oppen et al., 1994; Alberto et al., 1997). The present study was undertaken for characterizing the identities of Capsosiphon fulvescens populations cultivating in Korea through the analysis of PCR based random amplified polymorphic DNAs (Welsh and MacClelland, 1990; Willams et al., 1990) and chemical composition aimed to isolate the useful strains for aquaculture. Development of Black Shank Resistant Burley Tobacco Germplasm, KB 104 and KB 106, and Their Characteristics (버어리종 역병저항성 계통 KB 104와 KB 106의 육성경과 및 특성)

  • Black shank(Phytophthora parasitira roar. nicotianae) resistant burley tobacco (Nicotiana tabacum L.) germplasms, KB 104 and KB 106, were developed by Korea Ginseng & Tobacco Research Institute. KB 104 was developed from the single cross of Burley 21$\times$Newton 77, using a modified pedigree method. KB 104 was highly resistant to black shank, and its agronomic characteristics and chemical contents were comparable to those of Burley 21, and value per 10a was slightly higher than Burley 21, KB 106 is a maternally derived doubled haploid made by N. africana method from the single cross of Burley 21$\times$ Va 509. KB 106 was also highly resistant to black shank, had two more harvestable leaves per plant and flowered three days later than Burley 21 did. Total alkaloid and nicotine contents of KB 106 were significantly lower than those of Burley 21. But its nornicotine content was higher than Burley 21 5. Variation of Isoflavone Contents in Korean Soybean Germplasms (국내 콩 유전자원의 Isoflavone 함량 변이)

  • Isoflavone in soybean is known to have the important physiological functions such as antioxidative, estrogenic, and anti-tumor activities. Therefore, isoflavone content is considered as important aspect of soybean quality. The objectives of this study are to determine the isoflavone contents of soybean germplasms including major cultivars in Korea, and to provide the basic information of isoflavone far development soybean cultivar containing high isoflavone content. Among 43 Korean major cultivated soybean samples, the total isoflavone contents were varied from $277.0{\mu}g/g\;to\;2,002.0{\mu}g/g$, and the average total isoflavone content was $1,017.3{\pm}390.0{\mu}g/g$. Total isoflavone content of Seokryangputkong $(2,002{\mu}g/g)$ was the highest among those of the other cultivated soybeans. On the other side, Hwangkeumkong $(277{\mu}g/g)$ contained lower isoflavone than other cultivars. The total isoflavone content of vegetable use or early maturity soybean cultivar $(1,438{\pm}493.7{\mu}g/g)$ showed more higher total isoflavone content than others. The daidzein, glycitein, genistein and total isoflavone contents of 648 soybean germplasms collected in Korea, were ranged $16.9{\sim}2,019.8{\mu}g/g,\;0{\sim}294.9{\mu}g/g,\;38.5\sim1,337.7{\mu}g/g,\;and\;82.6{\sim}3,479.2{\mu}g/g$, respectively, and total isoflavone content of IT 180001 line $(3,479.2{\mu}g/g)$ was the highest among those of other soybean germplasm lines.

  • The tea has traditionally been used as a foodstuff by unique flavor, however recently not only the diversity of consumer demands but also the public interest in unique favorite and functional aspects have increased. It has been also reported that the main components contained in the leaves of tea (Camellia sinensis) include total nitrogen, free amino acids, polyphenols, and fiber, of which catechin has powerful bioactive effect such as anti-cancer, anti-aging, and anti-diabetic. (-)-Epigallocatechin gallate (EGCG) which is a major phenolic constituent of green tea extract has received considerable attention for a variety of important bioactivities. This study was carried out to obtain useful information for tea breeding programs, and to investigate the concentration of quality and functional related components in Korean indigenous tea germplasms. Korean indigenous tea lines were classified into three groups of sprout time, i.e, early, medium and late sprout time, and the ratio were 20%, 43% and 37%, respectively. There was a difference in characteristics among these Korean indigenous tea lines, leaf width of those ranged from 19.8 to 75 mm, leaf length was 35.5-160.0 mm, and leaf area was $660-8,400\;mm^2$. Experimental data on chlorophyll content (SPAD value) of Korean indigenous tea genetic resources ranged from 51.3 to 82.3. The concentrations of the total nitrogen, total free amino acids, and theanine were ranged 4.18-6.07%, 2.87-4.58%, and 1.64-2.66%, respectively. Also, catechin concentration showed from 11.54 to 15.07%, and concentration of caffeine was 2.82-4.23%. These results indicated indicated that it is possible to select elite lines with high concentration of quality related components and low concentration of caffeine from Korean domestic tea germplasms.

Variation of Protein Content and Amino Add Composition of Maize Germplasms (옥수수 종실의 단백질함량 변이와 아미노산 조성)

  • Corn proteins have been known as nutritionally poor, being deficient in the essential amino acids. lysine and tryptophan. Improving the quality of protein in the corn grain would be a great benefit to the farmer. This study was conducted to evaluate the variation of the protein content and the protein constitution of the maize germplasms in the Crop Experiment Station in 1989. The average protein content of 101 germplasms was 11.5% with range from 8.0% to 17.3%. Elite hybrid field corns and table corns possessed 9.1-13.9% protein for the dried whole kernel. Major amino acids were glutamic acid and leucine. Lysine and methionine were limited. Varietal differences were observed in the amino acid composition. Qpm, a modified opaque-2 mutant had 1.4-1.7 times higher lysine content than Suwon 19, a dent corn and Suwon SS-21, a sweet corn. Suwon SS-21 had high threonine content. Maize seed protein gave three fractions. an alchol-soluble fraction (zein), an alkali-soluble fraction (glutelin), and a salt-soluble fraction (globulin) by the Osborne method. The zein fraction accounted respectively for 50.7% and 41.7% of the total protein is Suwon 19 and Suwon SS-21. The nonzein fractions increased in percentage of total protein in Qpm kernels. The amino acid composition of zein fraction from three types maize endoperms of dent, sweet and opaque-2 was essentially identical. Zein contained the high contents of glutamic acid and leucine but low content of lysine. The glutelin fractions of three types maize endosperms were mainly similar in overall amino acid composition. The lysine content of glutelin was higher than that of zein. The amino acid composition of globulin fraction was some different from those of zein and glutelin In Qpm it had higher levels of histidine and lysine than both of zein and glutelin. The increased lysine content in Qpm was resulted from changing the proportions of proteins which contained different levels of lysine.
