Establishment of Seed Treatment for Healthy Production of Peanut Sprout (청정 땅콩나물 생산을 위한 종자처리기술 확립)

  • The present study was conducted to develop seed treatment for the production of healthy and clean peanut sprout. Dry heat treatment of peanut seeds reduced the incidence of the rot. The seed treatment condition at $52^{\circ}C$ for 10 h. was the most efficient without inhibiting seed viability significantly. Seeds were dark cultured at $27^{\circ}C$ for up to 9 days. The treatment of Indole-B and gibberellic acid influenced germination, T50, fresh, dry weight, hypocotyl length, hypocotyl length diameter, root length, number of lateral root and epicotyl of peanut sprout. There were no differences in the germinability of peanut seeds between gibberellic acid treatment methods but higher fresh weight was observed in the GA3 solution spray after 2 hour water soaking. The general growth and lateral root development of peanut sprouts were suppressed by Indole-B which is used for inhibiting root formation and promoting hypocotyls. The treatment of gibberellic acid promoted hypocotyl elongation, but it did not influence on the growth of hypocotyls and root system.

Effect of Freezing on Hydration Rate and Sprouting Characteristics of Soybeans (대두의 수화속도와 콩나물의 발아특성에 대한 동결처리 효과)

  • Effect of freezing on the hydration rate and growth characteristics of bean sprout of two domestic cultivars of soybean, Eunha beans and Taegwang beans, was investigated. Freezing of the soybeans at $-80^{\circ}C$ affected characteristics of the hydration and growth of soybean sprout of the beans. Hydration rate of the frozen beans depended on the hydration temperature. It decreased up to 50% at lower than $60^{\circ}C$ of hydration temperature, but it did not decrease at higher than $60^{\circ}C$. Activation energies of frozen Eunha and Taegwang beans were 39.79 and 39.25 kJ/mol, respectively. Kinetic compensation effect between activation energy values and the pre-exponential factor for the hydration of soybeans with or without freezing was observed. Germination rate and thickness of the bean sprout increased by freezing, however, yield and weight were not affected by freezing.

Breeding of four-leaf white clover (Trifolium repens L.) through 60Co gamma-ray irradiation

  • Four-leaf white clover is not found easily in nature due to its low appearance rate (1 in 10,000). Because people believe that it brings good luck and like to either keep it or present it to a loved one, it has commercial and ornamental value. To breed four-leaf clover, we exposed its flowers to ${\gamma}-rays$ at the pollination stage. The $M_1$ seeds produced following doses at 25-100 Gy showed an approximately 74% germination rate, with seedling survival at 46%. In the $M_1$ generation of plants irradiated within that dose range, we found an increased frequency of four leaflets. One of them, Jeju Lucky-1 (JL-1), had a frequency of about 60%. To see whether that mutation was somaclonal or genetic, we observed its $M_2$ generation and found that such a phenotype reappeared. Although our results demonstrated that the irradiation of fully mature flowers led to a higher frequency of 4-leaflets, we could not clearly explain the genetic mechanism involved. We suggest that JL-1 is valuable as a new variety, without further genetic fixation, because white clover can be propagated vegetatively by stolons.

Storability of Soybean Lines for Maintanance of Germplasm (대두유전자원의 안전보존을 위한 계통간 발아수명검정)

  • To develop a long term storage technique of soybean germplasm, fifty lines of soybean seeds were kept in storage room controlled by air-conditioner and dehumidifier at KAERI. The storage room was kept below 25$^{\circ}C$ of temperature and 60% relative humidity. Germinability of stored seeds was evaluated at every three or six months. On the other hand, the newly harvested seeds of the lines were accerelated aging at 40$^{\circ}C$ and 90% of relative humidity, and compared with non-accerelated seeds. The average germinability at the starting time of storage was 97% but the germinability was decreased to 62% at 42 months after storage in the storage room. The germinability seemed to be affected by seed size and seed coat color. The germinability of small seed lot tended to be higher than that of large seed lot, and the germination response in longer term of storage was greater than those of shorter term of storage. The accerelated aging test for 7 days proved to be a useful technique to pretest germinability of soybean seeds for assaying the relative storage potential.

Varietal Differences of Germinative Traits Related with Malting Barley Breeding (맥관맥의 발아관련형질들의 품종간 차이)

  • Sixteen Korean leading barley varieties were tested on the sixtieth day after harvest, in order to investigate differences for germinative traits related, and on the eightieth day to test optimum water level for germination test. The germinative energy(GE) and capacity(GC) in the 4.5cc water level were the highest individually. Varietal variations among GE, GC, promptness index(PI) and water sensitivity(WS) were highly significant in storage conditions and water levels. Correlation coefficient estimated were positive among GE, GC, PI, but negative between these traits and WS. Also the varietal difference of WS gets higher with the following order of malting barley<naked barley<covered barley. Heritabilities of broad sense for GE, GC, PI and WS were high, therefore, these traits could be considered in malting barley breeding.

Foeniculum vulgare essential oil nanoemulsion inhibits Fusarium oxysporum causing Panax notoginseng root-rot disease

  • Background: Fusarium oxysporum (F. oxysporum) is the primary pathogenic fungus that causes Panax notoginseng (P. notoginseng) root rot disease. To control the disease, safe and efficient antifungal pesticides must currently be developed. Methods: In this study, we prepared and characterized a nanoemulsion of Foeniculum vulgare essential oil (Ne-FvEO) using ultrasonic technology and evaluated its stability. Traditional Foeniculum vulgare essential oil (T-FvEO) was prepared simultaneously with 1/1000 Tween-80 and 20/1000 dimethyl sulfoxide (DMSO). The effects and inhibitory mechanism of Ne-FvEO and T-FvEO in F. oxysporum were investigated through combined transcriptome and metabolome analyses. Results: Results showed that the minimum inhibitory concentration (MIC) of Ne-FvEO decreased from 3.65 mg/mL to 0.35 mg/mL, and its bioavailability increased by 10-fold. The results of gas chromatography/mass spectrometry (GC/MS) showed that T-FvEO did not contain a high content of estragole compared to Foeniculum vulgare essential oil (FvEO) and Ne-FvEO. Combined metabolome and transcriptome analysis showed that both emulsions inhibited the growth and development of F. oxysporum through the synthesis of the cell wall and cell membrane, energy metabolism, and genetic information of F. oxysporum mycelium. Ne-FvEO also inhibited the expression of 2-oxoglutarate dehydrogenase and isocitrate dehydrogenase and reduced the content of 2-oxoglutarate, which inhibited the germination of spores. Conclusion: Our findings suggest that Ne-FvEO effectively inhibited the growth of F. oxysporum in P. notoginseng in vivo. The findings contribute to our comprehension of the antifungal mechanism of essential oils (EOs) and lay the groundwork for the creation of plant-derived antifungal medicines.

lonizing Radiation Hormesis in Crops (저선량 전리방사선에 의한 작물의 활성증진)

  • The most remarkable aspect in the hormesis law is that dose of harmful agents can produce effect that are diametrically opposite to the effect found with high doses of the same agent. Minute quantities of a harmful agent bring about very small change in the organism and control mechanisms appear to subjugate normal processes to place the organism in a state of albert and repair. The stimulated organism in more responsive to changes in environmental factors than it did before being alerted. Routine functions, including repair and defense, have priority for available energy and matetial. The alerted organism utilizes nutrients more efficiently, grows faster, shows improved defense, and lives longer. Accelerated germination, sprouting, growth, development, blooming and ripening, and increased crop yield and resistance to disease are found in plants. Another concept supported by the data in that low doses of ionizing radiation provide increased resistance to subsequent high doses of radiation. The hormesis varies with subject plant, variety, state of seed, environmental and cultural conditions, physiologic function measured, dose rate and total exposure. The results of hormesis are less consistently found, probably due to the great number of uncontrolled variables in the experiments. The general dosage for radiation homlesis in about 100 (10 to 1,000) times ambient or 100 (10 to 1,000) times less than a definitely harmful dose, but these must be modified to the occasion. Although little is known about most mechanisms of homzesis reaction, overcompensation of repair mechanism is offered as one mechanism.

A Comparative Study on Physiological Characters Between the Cultivar and Wild Plant in Amaranthus sp. (비름과(科)(Amaranthus sp.) 식물(植物)의 재배종(栽培種)과 야생종간(野生種間)의 생리학적(生理學的) 특성비교(特性比較))

  • The purpose of this comparative study is to investigate the differences of growth behaviors (height, weight, yield capacities etc.) and germination characters (induced photo-dormancy) among the three different species of Arnaranthus sp. The results obtained are summarized as follows. The growth of the cultivar of A.hypochondriacus was more vigor than that of the wild plant of A.retroflexus during the whole life cycle. The fresh weight, dry weight, yield capacity of A.hypochondriacus were much more higher than those of the others two species (A.retroflexus and A.Cruenthus). There are very different germinability formation and induced dormancy patterns between the cultivar and wild plant during seed formation periods. The cultivar of A.hypochondriacus was not induced primary dormancy in the any seed formation stages but wild plant of A.retroflexus was induced deep true dormancy. We suggested that these differences occured by the sum of different absorption of solar energy spectrum by the different seed colors between the cultivar (white color seed) and wild (black seed). And we confirmed the different absorption spectrum of the three different Arnaranthus sp. seeds.

Physical Properties and Apparent Thermal Diffusivity of the Soils where Soil Temperature is Measured Regularly (기상청(氣象廳) 지온(地溫) 측정(測定) 토양(土壤)의 물리적(物理的) 성질(性質)과 겉보기 열확산(熱擴散) 계수(係數) 산정(算定))

  • Soil temperature is one of the important environmental factors which control all the physical, chemical and biological processes in soil including germination and root growth of plants and other organisms living in the soil ecosystem. Soil water and nutrient availability and mobility are temperature dependent. Soil temperature change is depended primarily upon energy exchange in soil surface, meteorological variance and physical properties of the soils which are closely related to heat transfer mechanism. In this study physical properties including bulk density, soil texture and organic matter content were measured and thermal diffusivity on the soils was calculated. Soil samples from the 66 meteorological stations under the Korea Meteorology were collected and the physical parameters were measured. To obtain relationship between thermal diffusivity and soil water content a heat probe thermal diffusivity measurement apparatus was designed and used in this experiment. According to the survey on soil physicsal properties on the 66 meteorological stations, the 52% of the surface soil texture were sandy loam and laomy sand or sand, 38% were loam and silty loam, and 10% were clay loam and silty clay loam. The bulk density which was closely related with thermal properties showed average of $1.41g/cm^3$ for sandy soils, $1.33g/cm^3$ for loam and silty loam soils, and $1.21g/cm^3$ for clay loam and silty clay loam soils. The apparent thermal diffusivity of the upper layer from 0 to 30cm ranged from 1.16 to $8.40{\times}10^{-3}cm^3/sec$ with average of $3.53{\times}10^{-3}cm^3/sec$. The apparent thermal diffusivities of the Jeju soils of which organic matter contents were high and the bulk densities were low were near $2{\times}10^{-3}cm^3/sec$. The thermal diffusivity of snow measured in Chuncheon ranged from 0.822 to $2.237{\times}10^{-3}cm^3/sec$. The damping depth calculated from the thermal diffusivity ranged from 5.92 to 13.65cm for daily basis and 124 to 342cm for yearly basis. The significant regression equation to estimate thermal diffusivity with bulk density and soil water content was obtained by the heat probe in laboratory.

Development of New Mushroom Substrate of Pleurotus Ostreatus Using Foodwaste and Wormcast (남은 음식물과 지렁이 분변토를 이용한 느타리버섯의 대체 배지개발)

  • This study was investigated to develop an alternative medium for the cultivation of oyster mushrooms. The cultivation characteristics for major cultivated varieties of oyster mushrooms were campared with the food waste medium. In addition, water control & a gap formation of food waste is being pointed out as a problem of the medium material. Therefore, certain amounts of earthworm casting were supplied to the food waste medium to overcome the water and compaction phenomenon. This study investigated the use of the total 6 cultivars of oyster mushrooms, the Suhan(PL.1), Chiak No. 5(PL.2), Chiak No. 7(PL.3), Samgu No. 5(PL.4), Samgu No. 8(PL.5), Jangan No. 5(PL.6). In terms of medium, the optimum growth of the mycelium of the oyster mushroom was observed in the beet pulp medium but in food waste, the mycelium's growth was observed as insufficient in the bulk of cultivars. The Jangan No. 5(PL.6) was superior to other cultivars relative to the mycelium's growth. The optimum concentration of earthworm casting added the foodwaste medium was 30%. When Jangan No. 5(PL.6) was incubated in the foodwaste meidum containing 30% earthworm casting, the early days to germination were long, almost double the amount of time than that of the beet pulp medium. However, it was found that the fruit body size was larger than that of the beet pulp by up to 20.5%, and the length of the fruit body was up to 6.2% longer than that of the beet plup. The total yields increased by 60.5%. The analysis results of the oyster mushroom's nutritional contents cultured using the food waste medium, shows that the protein, vitamin A and vitamin E increased 70.6%, 2.4%, 0.8% respectively, the fat decreased by 12.6% and the oleic acid increased in a very small amount compared to that of the beet pulp medium.
