Comparison of Growth, Yield and Malting Quality Characters of Two Row and Six Row Barley (2조 및 6조대맥의 생육, 수량 및 품질특성 비교조사)

  • A comparison was made for growth and yield of two rowed and six rowed barley varieties to differentiate their fundamental characteris in relation with brewing quality. Heading and maturing time of two rowed barley varieties were similar to those of six rowed covered but were earlier than those of six rowed naked ones. Most of two rowed varieties were spring barley and low temperature susceptible, but they were relatively photoinsensitive compared to six rowed varieties. Eventhough malting barley has heavier stands and grains, they were quite similar in yielding ability with six rowed cultivars with decreased number of grains per spike. In quality characters, malting barley has better germination energy and rate, lower protein content than the common six rowed barley and decisively better for brewing that six rowed barley with higher malt yield, extract yield and quality and enzyme activity. In comparison of malting barley varieties produced in Korea and Japan indicated that those varieties produced in Japan were better in germination characters, extract yield and quality, Kolback index and enzyme activity than those produced in Korea.

Purification and properties of soybean ${\alpha}-galactosidase$ (대두 ${\alpha}-galactosidase$의 정제 및 성질)

  • To elucidate enzymatic properties of ${\alpha}-galactosidase$ (EC 3, 2, 1, 22) from germinated soybean, changes in the enzyme activities and oligosaccharide contents during germination of soybean were determined. ${\alpha}-Galactosidase$ from germinated soybean was purified by ammonium sulfate fractionation, ion exchange chromatography and gel filtration. Their chemical and enzymatic properties was investigated. ${\alpha}-galactosidase$ activity of sobeam was maximized when it was germinated at $25^{\circ}C$ for 120 hour. Raffinose and stachyose in soybean were decomposed completely after 96 hours and 120 hours of germination, respectively. Soybean ${\alpha}-galactosidase$ was purified by 6.6 fold by ammonium sulfate fractionation, ion exchange chromatography on DEAE-Cellulose and Sephadex A-50, and gel filtration on Sephadex G-150. Its specific activity was 825 Units/mg protein and the yield was 2.5% of the total activity of crude extracts. The purified ${\alpha}-galactosidase$ of soybean was found to be homogeneous by polyacrylamide gel electrophoresis and by HPLC. Isoelectric point of soybean ${\alpha}-galactosidase$ was determined analytical isoelectric focusing to be pH 4.8. The soybean ${\alpha}-galactosidase$ was monomeric and its molecular weight was estimated to be 30,000 by SDS-PAGE. The optimal temperature and pH for the soybeam ${\alpha}-galactosidase$ activity were $40^{\circ}C$ and pH 6.0 and 75% of its activity was lost by heating at $60^{\circ}C$ for 10 min. The enzyme was appeared to have higher affinity to raffinose than to stachyose. The Km value of soybean enzyme was 5.3 mM for ${\rho}-nitrophenyl-{\alpha}-D-galactopyranoside$ and the activation energy on PNPG was calculated to be 13.02 Kcal per mole.

  • The present study was conducted to development mass production methods for peanut sprouts that is considered as a field of blue ocean among the agricultural products. 'Jopyeong' was the best as a major cultivar for peanut sprouts production. The manual for the production of high-quality peanut sprouts is as following. Germination temperature appropriate for production of high-quality peanut sprouts was $27^{\circ}C$. Peanut sprouts at the growth stage of 8th day, and older plants with advanced growth showed deteriorated merchantable and eating quality. Resveratrol compound was not found in the seeds, but its highest amount was detected from 9-day old sprouts. The best water temperature applicable to high quality peanut sprout production was $25^{\circ}C$. The growth of peanut sprout was inhibited by the high temperatures above $35^{\circ}C$ and low temperatures below $15^{\circ}C$.

In-Bin Drying of Paddy with Ambient Air: Influence of Drying Parameters on Drying Time, Energy Requirements and Quality (상온통풍에 의한 벼의 In-Bin 건조 : 건조시간, 에너지 소요량 및 품질에 미치는 건조조건의 영향)

  • Low-temperature in-bin paddy drying has been examined to study the limitations of this drying method under Korean weather conditions, the initial moisture content of the paddy, the bulk depth and the airflow rate. The results are reported and discussed with regard to drying time, energy requirements and costs, uniformity in the moisture content of the dried kernels and, finally, the quality of the paddy. The tests carried out during the paddy-drying period in 1981 and 1982 have shown that under Korean weather conditions paddy can be dried to safe storage conditions by continuous aeration with ambient air. Depending upon the initial moisture content of the kernels(19.2%-25.5% w.b.), the bulk depth(1.1-3.5m) and the airflow $(3.0-6.9m^3\;air/m^3\;paddy/min)$ the paddy could be dried within 5 to 17 days. The energy requirements and energy costs are shown to be considerably lower than for conventional high-temperature drying. No significant changes in the quality in terms of milling yield, cracking ratio, acid value and germination were observed.

  • Quinoa (Chenopodium quinoa Willd.) is one of the ancient crops cultivated in the Andes region at an altitude of 3,500-4000m in Chile and Bolivia from 5000 BC. It contains a large amount of protein, minerals and vitamins in comparison with other crops. The cultivation area has been increasing worldwide because of its excellent resistance to various abiotic stress such as salinity, drought and low temperature. ${\gamma}$-Ray radiation of high dose is often used as a tool to induce mutations in plant breeding, but it has a deleterious effect on organisms. However, the radiation may have a positive stimulatory effect of 'hormesis' in the low dose range. This experiment was carried out to investigate the optimum dose range for creating the quinoa genetic resources and to investigate the hormesis effect at low dose on the quinoa. This experiment was performed for 120 days from November, 2016 to February, 2017 in the greenhouse of Gyeongsang National University. ${\gamma}$-Ray radiation was irradiated to seeds at 0 Gy, 50 Gy, 100 Gy, 200 Gy, 300 Gy, 400 Gy, 600 Gy, 800 Gy and 1000 Gy for 8 hours. (50 Gy) using the low level radiation facility ($Co^{60}$) of Cooperative Research Institute of Radiation Research Institute, KAERI. Fifty seeds were placed on each petri dish lined with wet filter paper and germination rate was measured at a time interval of 2 hours for 40 hrs. The length of the root length was measured one week after germination. Each treatment was carried out in 3 replicates. The growth of seedlings were investigated for 10 days after transplanting of 30 day-old seedlings. The plant height, NDVI, SPAD, Fv/Fm, and panicle weight were measured. The germination rate was highest at 50Gy and 0Gy and the rate of seeds treated with 400Gy or higher rate decreased to 25% of the seeds treated with 50Gy. The emergence rate of seedling in pot experiment was higher at the dose of 200 Gy, 300 Gy and 400 Gy than at 0 and 50Gy. However, the rate was lower at strong radiation higher than 600Gy at which $1^{st}$ leaf was not expanded fully and dead due to extreme overgrowth at 44 days after treatment (DAT). The highest value of panicle weight was observed at 50Gy (6.15g) and 100Gy (5.57g). On the other hand, the weight at high irradiated dose of 300Gy and 400Gy was decreased by about 55% compared to low dose (50 Gy). NDVI measurement also showed the highest value at 50 Gy as the growth progressed. SPAD was the highest at 400 Gy and showed positive correlation with irradiation dose except 0 Gy. Fv/Fm was high at 50 Gy up to 30 DAT and no difference between treatments was observed except for 400 Gy from 44 DAT. The plant height was the highest in 50Gy during the growing period and was higher in the order of 50Dy, 100Gy, 0Gy, 200Gy, 300Gy and 400Gy in 88 DAT. In this experiment, the optimal radiation dose for hormesis was 50Gy and 100Gy, and the optimal radiation dose for mutagenesis seems to be 400 Gy.

Effects of Herbicides on Growth and Reproductive Characters of Glycine max (대두(Glycine max)의 생장 및 번식 특성에 미치는 제초제의 영향)

  • Herbicides have been used to control weeds for decades. If detoxification upon exposure to herbicides requires considerable amounts of energy, it could affect the pattern of resource allocation to growth and reproduction of crops. We examined the effects of three levels of a herbicide (Control, Low, and High) on germination, growth and reproductive characters of Glycine max treated twice, i.e., before and after seed germination. Since flowering time of G. max was separated into two groups, flowering time was also considered as a variable in this study. The rate of seed germination tended to be higher at the low level of herbicide compared to other levels. Chlorosis and shape variation of leaves were apparent after the second herbicide treatment, but completely disappeared after six weeks of treatment. The herbicide effects on growth characters were somewhat different between early and late flowering plants, but plants treated with both low and high levels of herbicide reduced their growth compared to those in the control group regardless of flowering time. Plants at the high level of herbicide exhibited the highest growth rate later in the season, suggesting that plants compensated to some extent for reduced growth. However, growth reduction among plants at the high level of herbicide was persistent until the end of growing season. Among plants flowered late in the season, plants in the control level bore a higher number of nodules per plant than those in other levels; such a pattern did not exist among plants flowered early in the season. Plants treated with low and high levels of herbicide produced a lower number of flowers than those in the control. Thus, the herbicide examined affected not only the growth and reproductive characters of non-target crops but also the development and growth of root nodules.

Control of Diurnal Night Temperature on Watermelon(Citrullus vulgaris S.) for Energy Saving Greenhouse (시설내 수박재배시 에너지 절약을 위한 야간온도의 조절)

  • In winter, most of the energy for a greenhouse crop is supplied during the night. Since watermelon is grown under high night temperature, the experiments were set up to investigate night temperature influence on watermelon in order to obtain the best economic output. Day temperatures ranged from $25^{\circ}C$ and 3$0^{\circ}C$ ; night temperatures ranged from 2$0^{\circ}C$ to 3$0^{\circ}C$ at 5$^{\circ}C$ interval. Two cultivars of watermelon(Citrullus vulgaris S. ‘Binna’ and ‘Kamro’) treated with 30/3$0^{\circ}C$ yielded maximum leaf areas, flowers and leaf numbers. 30(14h)/25(10h)$^{\circ}C$ or 30(12h)/25(12h)$^{\circ}C$ grown plants had higher germination ratio and more dry weight and chlorophyll than those of 30/3$0^{\circ}C$ which were the highest temperature integral. Although 25/$25^{\circ}C$ and 30/2$0^{\circ}C$ regime are same average temperature, the growth of watermelons at 30/2$0^{\circ}C$ was significantly higher than 25/$25^{\circ}C$. ‘Binna’ was growing more than ‘Kamro’ at the same temperature. Leaf area ratio(LAR) was reduced with increasing DIF temperature from 30/3$0^{\circ}C$ to 30/2$0^{\circ}C$, but leaf weight ratio(LWR) was increased.

Proteomic Analysis and Extensive Protein Identification from Dry, Germinating Arabidopsis Seeds and Young Seedlings

  • Proteins accumulated in dry, stratified Arabidopsis seeds or young seedlings, totaled 1100 to 1300 depending on the time of sampling, were analyzed by using immobilized pH gradient 2-DE gel electrophoresis. The molecular identities of 437 polypeptides, encoded by 355 independent genes, were determined by MALDI-TOF or TOF-TOF mass spectrometry. In the sum, 293 were present at all stages and 95 were accumulated during the time of radicle protrusion while another 18 appeared in later stages. Further analysis showed that 226 of the identified polypeptides could be located in different metabolic pathways. Proteins involved in carbohydrate, energy and amino acid metabolism constituted to about 1/4, and those involved in metabolism of vitamins and cofactors constituted for about 3% of the total signal intensity in gels prepared from 72 h seedlings. Enzymes related to genetic information processing increased very quickly during early imbibition and reached highest level around 30 h of germination.

EARLY CROP ESTABLISHMENT OF RAINFED LOWLANG RICE BY SLIT SEEDING

  • The uncertainty of rains at the onset of wet season (WS) and the drought risk involved hinder growing more than one rainfed lwoland rice crop per year. Establishing transplanted rice well into the WS leaves insufficient moisture in the soil for subsequent crop. Rice establishment early in the season gives the farmer better opportunities to grow a crop after rice. An experiment was conducted startign in 1993 to evaluate dry seeding of rice through slit soil seeding. It is done utilizing the vertical metering slit seeder for conserving soil moisture coming from the first rains in the early WS to sustain germination and establishment of the seeding at least until the succeeding rains under therainfed lowland (RL) environment. The treatment consisted of slit-seeding the PSBRc 14 into the tilled and nontilled plots at 100kg/ha and at depths of <10 mm (shallow seeding) and 60-70m (deep seeding). The control treatment was broadcast seeded on tilled soil and harrow to cover the seeds The superior crop establishment observed in 1995 WS experiment on nontilled, slit-seeded plots confirmed the results observed in 1993 WS and 1994 WS experiments. Emergence in deep seeding was not significantly different from shallow seeding in nontilled plots giving an average yield of 2.1 t/ha in all slit-seeded plots. This offers an advantage of reduced energy in put in nontilled shallow seeding. However, heavy weed infestation has to be addressed at the early stage of rice in nontilled soil to get the full advantage from slit seeding. The consistently better crop establishment observed in slit seeding over that of broadcast seeding in the WS of 1993, 1994 and 1995 also demonstrates that the slit seeding technology can be adopted with confidence in the rainfed lowland field condition to reduce the risks involved in broadcast seeding.

Effect of Germination on the Nutritive Value of Barley in Rats (흰쥐에 있어서의 발아(發芽)보리의 영향효과(營養效果))

  • This study was conducted to investigate the nutritional value of germinated barley with animal experiment. Fourthyeight weanling albino rats, which were divided into 8 groups, were fed during 4 weeks. The feed intake, energy intake, FER (feed efficiency ratio) and growth rate were slightly high in the casein supplemented groups and were significantly high in the rice and casein supplemented groups. But there was no a significant difference between the barley and germinated barley groups. A higher PER (protein efficiency ratio) was shown in the rice and casein supplemented groups. It was induced that mixed cereal protein increase the PER compared to simple cereal protein. Low apparent digestibility of the diet and of the protein were found in barley and germinated barley groups compared to standard and rice groups. But apparent digestibility of protein in germinated barley groups were markedly increased by the supplementation of 5% casein or supplementation of rice and casein. From this result it was concluded that germinated barley has a nutritional effect on increasing protein digestibility when added enough amount of animal protein.
