비탈면녹화용 몇가지 자생식물의 종자발아특성 (Seed Germination Characteristics of Korean Native Plants for Slope Restoration and Revegetation)

  • The differences in seed germination rates of Korean Native Plants were observed depending on the harvest years, the seed companies, the storage periods, the drying methods and the plant morphological types. The seed germination rates were changed significantly by the harvest years and the seed companies in all 4 species tested. The seed germination rates were decreased after 1 year storage at $4^{\circ}C$ in all 8 species tested and three of them did not germinate at all. A rapid drying method at $35^{\circ}C$ with a convection oven was not suitable for Chrysanthemum spp., but there were no significant changes of germination rates in Aster spp. The germination rates were decreased more at seeding in soil than at lab. test with Dicotyledon but no significant changes with Monocotyledon. Especially, the germination rates of fine seed on 5mm soil covering treatment were significantly decreased.

일당귀의 등숙에 따른 종자 발아 및 생육 특성 (Seed Germination Rate and Growth Characteristics according to Ripening Stages in Angelica acutiloba Kitagawa)

  • Background: Angelica acutiloba Kitagawa belongs to the flowering plant family Apiaceae. Its young leaves are consumed as a vegetable, and its roots have medicinal properties. This study was conducted to analyze the seed germination characteristics of A. acutiloba according to its ripening stages. Methods and Results: The seeds were harvested from a research farm managed by the Department of Herbal Crop Research, Rural Development Administration in 2018 and were divided into six groups according to their specific gravities. In particular, we studied the effect of harvesting periods and umbel orders on seed characteristics. The results showed that the rates of germination, emergence, and early growth improved as the specific gravity of the seeds increased. In addition, the germination rates of the seeds harvested in mid July and early August were significantly higher than those harvested in the other seasons, and the seeds obtained from the first floret had the greatest mass and weight. Conclusions: In this study, we demonstrated that it is possible to improve the germination rate by appropriate selection of seeds and harvesting period both of which are closely related to seed maturity.

Seed Germination Response to Temperature, Cold Stratification Period, and Gibberellin Treatment in Spiraea fritschiana

  • To improve the germination of Spiraea fritschiana seeds for mass propagation, we evaluated the effect of a range of temperatures, cold stratification periods, and gibberellic acid ($GA_3$) treatments on three germination characteristics. Final germination percentage (FGP) increased as the temperature for seed germination increased, up to $30^{\circ}C$, while the mean germination time (MGT) and the mean number of days to 30% germination ($T_{30}$) decreased when seeds were germinated at $25-30^{\circ}C$. The optimum germination temperature of S. fritschiana seeds is approximately $30^{\circ}C$ considering FGP, MGT, and $T_{30}$ together. FGP increased with the duration of cold stratification up to a period of 6 weeks, but declined after 8 weeks of cold stratification, as prolonged cold stratification can induce dormancy with a resultant decline in germination. Pretreatment with 6-8 weeks of cold stratification or soaking seeds in distilled water or $500mg{\cdot}L^{-1}$ $GA_3$ for 24 h accelerated and increased the germination of S. fritschiana seeds, regardless of temperature. However, further study might be required to evaluate the effect of $GA_3$ concentrations lower than $500mg{\cdot}L^{-1}$ on the promotion of germination in S. fritschiana seeds.

카멜리나 (Camelina sativa Crtz.) 발아 적온 및 발아초기 뿌리생육 특성 (Optimum germination temperature and seedling root growth characteristics of Camelina)

  • A genus Camelina has been attracted as a promising oil crop, especially available in drought and marginal conditions. Due to more demands on arable land for bioenergy crops, price of agricultural products has been a challengeable issue. In that respect, development of Camelina crop with higher germination rate and germination energy can be a strategy to secure seedling establishment, nutrient uptake and long vegetative period. In order to be easily available in the field and laboratory conditions, Camelina seed needs to be optimized for its germination temperature. Germination temperature regime was in a range of 8 to $32^{\circ}C$ initially, and consecutively narrowed down to 8 to $20^{\circ}C$. Based on the temperature range, Camelina germinated greater than 96% at $8-16^{\circ}C$ in two weeks after sowing, but germination rate started to decrease at the higher than $24^{\circ}C$ and was significantly low at higher than $32^{\circ}C$. In terms of rapid time to reach the maximum germination rate and greater germination energy, temperature ranged from 12 to $16^{\circ}C$ was found to be desirable for Camelina germination. Although germinationa rate was greater at $16^{\circ}C$, lower temperature close to $12^{\circ}C$ would be favored for the field conditions where greater root growth leading to healthier seedlings and better nutrient or water availability is considerably demanded.

  • This study evaluated the germination rate of $Rheum$ $palmatum$ L. in Korea as affected by storage temperature, germination temperature, and a soaking treatment. The germination rate of $R.$ $palmatum$ L. stored at various conditions for 8 weeks was > 60%. The highest germination rate occurred at $25^{\circ}C$. The germination point of $R.$ $palmatum$ L. with soaking was started 1 day after application, and non soaked seeds germinated in 2 days. When $R.$ $palmatum$ L. was soaked with water, the germination rate was lower, but mean germination velocity was the highest. These results may help our understanding of variations in germination characteristics for seeds treated under different germination conditions.

Physicochemical and Functional Properties of Germinated Glycine max Merr Soybeans

  • To investigate the applicability as the functional food materials of germinated Glycine max Merr soybeans, its biochemical characteristics and its abilities to inhibit platelet aggregation and hydrolyze alcohol were examined. With the progression of germination time, crude protein content gradually increased, and on the 5th day of germination it was 30.19%. However, crude fat content tended to decrease, and on the 5th day of germination it was 14.30%. Total amino acid content was highest on the 3rd day of germination at 80,875 mg%. The free amino acid content doubled from day 0 of germination (1,273.35 mg%) until the 5th day of germination (2,742.99 mg%). Fatty acid analysis revealed that linoleic acid was highest among all the samples, ranging from $53.55{\sim}56.00%$. Linolenic acid content slightly increased as the germination period was prolonged. The ability to inhibit platelet aggregation increased according to the germination period and then decreased again on the 5th day of germination; it was somewhat higher in the ethanol fraction. In measuring ADH, we found that the activity of the ethanol fraction increased with increasing days of germination. In the case of the water fraction, the activity decreased as germination was prolonged, and the ADH activity of the water fraction was higher than that of the ethanol fraction. Based on the above results, we deemed that the Glycine max Merr soybeans germinated for $2{\sim}3$ days were most pertinent for use as functional food materials.

채취 시기에 따른 느릅나무의 종자 형질 및 발아 특성 변화 (Change in the Seed Characteristics and Germination Properties of Ulmus davidiana var. Japonica According to Seed Collection Time)

  • 느릅나무는 개화 후 결실에 이르는 기간이 매우 짧으며, 또한 단시일에 결실에서 낙과(落果)에 이르기 때문에 적정 채취 시기를 구명하는데 어려움이 많아 이를 해결하고자 채취 시기별 종자 형질 및 발아 특성을 관찰하고 유묘의 생장 및 생리적 특성을 조사하였다. 종자는 강원 정선지역에서 낙화 후 1주일인 2005년 4월 21일부터 7일 간격으로 채취하였다. 채취 시기에 따른 종자 형질 및 발아 특성에는 차이가 있었다. 종자의 크기는 채취 시기에 따라 증가하였으나 수분 함량은 5월 5일까지 증가한 후 감소하였으며 실중은 5월 12일까지 증가한 후 감소하였다. 채취 시기는 종자 특성 중 종자 크기와 높은 정의 상관(r=0.790, p<0.01)을, 수분 함량과는 부(-)의 상관(r=-0.919, p<0.01)을 보여주었다. 발아율은 5월 5일 채취한 종자에서 가장 높게 나타났으며 이후 시간에 따라 감소하였다. 평균 발아 일수, 발아 속도 및 발아 균일 지수 역시 5월 5일 채취한 종자에서 가장 높았다. 발아율, 평균 발아 일수 및 발아 속도는 실중을 제외한 모든 종자 특성과 상관성을 나타냈다. 종자 채취 시기에 따른 유묘의 상대생장율 및 엽록소 함량에 있어 5월 5일 채취한 종자의 유묘에서 가장 높게 나타났다. 따라서 느릅나무 종자는 외형적으로 건조가 시작되기 전 단계인 개화 종료 후 약 20일 후에 채취하여 파종하면 활력이 좋은 유묘의 생산이 가능할 것으로 판단된다.

장기 저장과 저장 온도에 따른 소나무 종자 품질과 생리적 특성 (Quality and Physiological Characteristics of Pinus densiflora Seeds Under Different Storage Periods and Temperatures)

  • 연구는 장기 저장된 소나무 종자의 품질을 평가하고 발아 특성과 생리적 특성 간의 상관 관계를 검증하였으며, 이를 위해 저장 기간과 온도에 따른 종자 발아와 생리적 특성을 조사하였다. 그 결과, 소나무 종자는 4℃에 12년 간 저장되었을 때, 발아율 99%, T50 6.5일로 종자 활력 및 종자세 저하가 거의 일어나지 않았으며, -18℃에 저장할 경우, 저장기간이 더 길어졌다. 또한, 장기 저장에 따른 퇴화 과정 중 활력보다 종자세의 감소가 먼저 일어나는 것으로 나타났다. 종자 침출수의 전기전도도는 4℃에 19년 간 저장되어 활력을 완전히 소실한 종자에서 유의하게 높았으며, 발아율, T50, 평균발아 일수, 발아속도와의 상관관계가 있었다. 침출수에 누출된 무기질 중에서는 K의 농도가 가장 높았으며, 그 다음으로는 Na, Ca, Cu, Mg, Fe 순으로 평균 농도가 높았고, Mn과 Zn은 검출되지 않았다. K, Ca, Cu, Mg, Fe 농도는 처리별로 통계적인 차이가 있었으며, K, Ca, Cu, Na, Mg의 경우, 활력을 완전히 소실한 종자와 그렇지 않은 종자에서 차이를 보였고, Cu의 경우, 2003년에 채집한 종자와 나머지 종자와의 차이가 컸다. 발아율은 Ca, Cu, K, Mg, Na 농도와 부의 상관을 보였으며, 발아균일도는 Cu 농도와 부의 상관을 보였고, 다른 발아 특성은 무기질 농도와 상관이 없었다. 따라서 종자 침출수의 무기질 농도는 개체간의 품질 차이가 적은 경우, 경향이 뚜렷하지 않았으나, 전기전도도는 장기 저장 종자의 활력 뿐 아니라 종자세의 차이를 나타내는 지표가 될 수 있을 것으로 보인다.

군산 인근해역 준설토를 활용한 식생발아 특성 연구 (A Study of Germination Characteristics of Dredged Soil Collected in Gunsan)

  • 본 연구에서는 군산 인근해역의 준설토를 식생이 가능한 녹생토로써 토목현장의 사면안정화 및 해양공간개발에 사용이 가능한 생태 환경형 재료를 만들고자 한다. 식생발아 특성을 분석하기 위해서 군산 인근해역의 준설토에 Bio 개량제(0%, 6%), AC(아미노산) 개량제(0%, 6%), 톱밥비료(0%, 6%), 산 흙(0%, 50%)으로 처리한 준설토 개량제 실내 실험을 수행하였다. 실험 결과, 군산 새만금 준설토의 경우 SM시료로 분류되었으며, 수목생장조건이 불량하여 개량제(톱밥비료, Bio 개량제 등)의 사용이 필요한 것으로 판단된다.

Shattering-related Characteristics and Germination Speed Based on Grain Type and Esterase Isozyme Zymogram in Korean Off-rype Rice

  • This study was carried out to investigate the characteristics of shattering habit and germination of offtype rice plants collected in Korea which were classified into seven groups based on grain characteristics. In the long-grain red rice group, the short-grain red rice group, the long-grain normal rice group, and the long-grain waxy rice group, the tensile strength of grain and primary rachis branch was relatively low and the fiber cell of the primary rachis branch was short. Characteristics of shattering-related traits and germination in relation to grain and esterase isozyme zymogram type of off-type rice plants. In most lines of the extremely late and sterile rice group, the tensile strength of grain was low, while that of primary rachis branch was high, and the fiber cell was long. However, in the type 1 esterase isozyme zymogram(EIZ) lines among the long-grain normal rice group and the type 6 and 5+6 EIZ plants among the extremely late and sterile rice group, the tensile strength of grain was relatively higher. The long-grain waxy rice group and the type 3 EIZ lines among the long-grain red rice group, showed higher germinability than did cultivars which are known to germinate well even in low-temperature. The other off-type rice group, except for the short-grain waxy rice group and type 1 and 7 EIZ lines among the long-grain normal rice group, had higher germination speed than that of the cultivars.
