Effects of Gentiana scabra Bunge var.buergeri Max. Extract on Blood Glucose and Lipid Composition in Streptozotcin induced Hyperglycemic Rats (용담(Gentiana scabra Bunge var.buergeri Max.) 추출액이 Streptozotocin 유발 당뇨성 흰주의 혈당 및 지질 성분에 미치는 영향)

  • This study was designed to observe the effects of the feeding Gentiana scabra Bunge var.buergeri Max. extract on the improvement of the blood glucose, lipids in the serum of streptozotcin(55mg/kg of body weight, I.P. injection) induced hyperglycemic rats (S.D. strain, ) fed the experimental diets for 4 weeks. Concentrations of blood glucose were significantly higher in the streptozotocin administration(I.P.) groups (group 2(streptozotcin+water), 3(streptozocin+Gentiana scabra Bunge extract), 4(streptozotocin+cholesterol+water), 5(streptozotocin+cholesterol+Gentiana scabra Bunge extract) than those in the control group(group 1(water)). Bolld glucose concentrations were rather lower in the group 3(streptozotocin+Gentiana scabra Bunge extract)than in the group 2(streptozotocin+water), and then those of the group 5 were rather lower than in the group 4. There was almost little significance among the groups. concentrations of total cholesterol in serum were lower in the group 5 than those in the group 4, and the group 3 were rather lower than in the group 2. Concentrations of LDL-cholesterol in serum were significantly lower in the group 3 than those 2. In the ratio of HDL-cholesterol concentrations to total cholesterol concentrations, Gentiana scabra Bunge extract administration groups were higher percentage than the other goups. Concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3,5 than the other groups, and then those of the group were higher percentage than the other groups. concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3,5 than the groups, and then those of the group 3 were significantly lower than in the group 4. Concentrations of triglyceride and phospholipid in serum were decreased in the group 3 than in the other groups. The activities of aspartate aminotransferase and alanine aminotransferase in serum were increased in the all experimental groups than in the control group. From the above research, the Gentiana scabra Bunge var.buergeri Max. extracts were effective on the improvement of the lipid compositions in serum of streptozotcin induced hyperglycemic and hypercholosterolemic rats.

Effects of Gentiana scabra var. buergeri Extract on Toxoplasmastic Activity of Macrophages

  • Gentiana scabra var. buergeri (G. scabra) is a herb known to have therapeutic effect in infection diseases. We studied cellular activation and antitoxoplasmosis in macrophages after G. scabra stimulation. Macrophage activation was detected by nitrite production. Macrophages were treated with G. scabra extracted with water or methanol. Maximal nitrite production was detected in macrophages after stimulation of G. scabra extract 0.1 mg/ml. Maximal nitrite concentration was 23.22 0.003 uM/L in macrophages after water extract of G. scabra and was 24.07 1.41 uM/L after methanol extract of G. scabra. Effect of G. scabra in the phagocytic capacity of macrophages was monitored by using PI (percentage of macrophage infected by T gondii) method. The minimum PI (42.5 2.31) was detected in macrophages treated by water extract of G. scabra 0.1 mg/ml before infection of T gondii. We also examined toxoplasmastatic capacity of macrophage using FI (fold increase) method. The minimum FI (4.46 1.16) was shown in macrophages after water extract of G. scabra 0.1 mg/ml pretreatment before infection. Under electron microscope, proliferation of T gondii was inhibited by extract of G. scabra treatment in macrophages and the mitochondrion and lysosomal vacuoles within cells were increased. Taken together, G. scabra extract activates macrophages and induces toxopalsmastatic activity after T gondii infection. It is suggested that G. scabra may be used as a therapeutic drug against toxoplasmosis.

From the above research, the Gentiana scabra Bunge var.buergeri Max. exacts were effective on the improvement of the lipid composition in serum of dietary hyperlipidemic rats.

PAF Antagonistic Activity of 2-hydroxy-3-methoxybenzoic Acid Glucose Ester from Gentiana scabra

  • In order to find out anti-platelet activating factor (PAF) from natural resources, Korean medicinal plants used for the treatments of peripheral circulation disorders were tested for their possible protective effects on PAF-induced anaphylactic shock. From the above screening, the methanol extract of Gentiana scabra showed a potent antagonistic activity against PAF. Water suspension of the extract was partitioned with $CH_2$$CI_2$ and EtOAc, successively. The EtOAc fraction which showed the highest activity was chromatographed on silica gel to yield 6 fractions. From the fraction which showed higher PAF-antagonistic activity than the other fractions, compound 1 was isolated by recrystallization. On the basis of spectral data, compound 1 was identified as 2-hydroxy-3-methoxybenzoic acid glucose ester. The compound prevented the mice from the PAF-induced death at a dose of 300 $\mu\textrm{g}$/mouse.

  • Bioassay-guided fractionation of a $H_2O$ extract of the roots of Gentiana scabra has furnished 5-(hydroxymethyl)-2-furfural (1) as an inhibitory compound for nitric oxide (NO) production in murine macrophage RAW 264.7 cells stimulated with $interferon-{\gamma}$ plus lipopolysaccharide. Compound 1 showed the moderate inhibition of NO production with $IC_{50}$ value of $803\;{\mu}M$.

Effects of Dried Gentiana scabra Rhizomes and Roots on the Intestinal Transit Rate of Mice with Experimental Gastrointestinal Motility Dysfunctions (용담 열수 추출물이 위장관 운동 기능 저해 상황에서 위장관 이송률에 미치는 영향)

  • Our recent study has revealed that in vivo intestinal transit rate (ITR) in normal mice was significantly increased by the administration of an aqueous extract of dried Gentiana scabra rhizomes and roots (GS-W) in a dose-dependent manner. Following on from our previous study, the effect of GS-W on ITR was measured in mice with experimentally induced gastrointestinal motility dysfunctions (GMDs) in the present study. GS-W showed no significant acute toxicity even at an oral dose of 5 g/kg to mice. ITR was significantly retarded in the GMD mice compared with that in normal mice, and this retardation was significantly recovered by the oral administration of GS-W in a dose-dependent manner. Furthermore, the ITR value of GS-W at a dose of 1 g/kg appeared to be higher than that of cisapride, which was predominantly prescribed for human patients with various GMDs in the late 1900s but was withdrawn from the market in 2000 due to its fatal side effects. The current results suggest that GS-W is a potential substitute for cisapride to prevent or alleviate human GMDs.

Antiparasitic Effects of a Herb Extract from Gentiana scabra var buergeri on Trichomonas vaginalis

  • We studied antitrichomoniasis with the extract of Gentiana scabra val buergeri, which may be effective in treating infectious diseases. The growth inhibition against T. vaginalis became optimal when tile extract concentration was 0.7 mg/ml and the cells were seeded at a density of 3$\times$10$^5$ per well. After incubation for 12, 24, 36, and 48 hrs, respectively, the number of cells were each 5$\times$10$^5$, 1$\times$10$^5$, 1$\times$10$^5$, and none, respectively. Under the electron microscope, the experimental group showed that the nucleus, karyosomes, and chromatin were weaker than those in the control group. After incubating for 3 hrs, the cells were destroyed completely, and only a remnant remained. The hydrogenosomes disappeared almost. The vacuoles and autophagic vacuoles increased. The cells became regressive form.

Radical Scavenging Activities and Protective Effects against Oxidative Damage to DNA of Extracts from Medicinal Plants with known Osteoprotective Effects (골질 보호 작용이 있는 수종 생약 추출액의 래디칼 소거능 및 DNA 보호 효과)

  • Among aging diseases, the most basic problem is a decrease in bone mineral density. Patients with osteoporosis are steadily increasing in the world. This study was investigated to show the relationship between osteoporotic effects and oxidative damage. Water extracts of 15 medicinal plants and ethanol extracts of 14 medicinal plants with known anti-osteoporotic effects, were tested for their radical scavenging activity using DPPH, ABTS, SRSA and FRAP assay. Water extract of Cornus officinalis, Rubus coreanus and ethanol extract of Rubus coreanus, Viscium album var. coloratum, Cimicifuga heracleifolia showed about 15-20 mg/g of total phenolic contents. Water extract of Cornus officinalis, Rubus coreanus and Epimedium koreanum showed high radical scavenging activity. Ethanol extract of Drynaria fortunei, Cornus officinalis, Rubus coreanus, Gentiana scabra and Astragalus membranaceus showed high radical scavenging activity. Water extract of Drynaria fortunei, Cornus officinalis, Nelumbo nucifera, Epimedium koreanum, and Gentiana scabra showed very strong protective effect against oxidative damage to DNA. These results suggest the correlation between anti-osteoporotic effects and antioxidative effects.

A Study on the Whitening Activities of Gentiana scabra Extract (용담화 추출물의 미백 활성 연구)

  • In order to find new functional materials for the cosmetics application, we investigated the anti-oxidant and anti-melanogenic properties of Gentiana scabra extracts (GSE), which were prepared by the various extraction methods. Results showed that GSE had high DPPH radical scavenging activity in a dose-dependent manner. Also, GSE inhibited the prodution of melanin in B16F10 melanoma cell as well as tyrosinase activity. We also found that GSE inhibited mRNA expression of tyrosinase-related protein (TRP)-1 and 2. In conclusion we suggest that GSE is applicable to cosmetics as a potential ingredient for their anti-oxidant and whitening effects.

Survey for Approximate Composition and Mineral Content of Medicinal Herbs (약초중의 일반성분 및 무기질 함량조사)

  • Approximate composition and mineral content of eighty seven medicinal herbs marketed in Korea was analyzed on the basis of their dried weight. From the results anlyzed, it was known that Acanthopanax sessiliflorum Seem. (22.4%), Portulaca oleracea L. (20.6%), Torilis japonica DC. (15.0%) in crude ash, Ziziyphus spinosa Hu. (39.9%), Alisma orientale Juzep. (30.6%), Lepidium latifolium L. (28.8%), Cassia tora L. (27.5%) in crude protein, Ziziyphus spinosa Hu. (27.9%), Xantnium strumarium L. (24.0%), Perilla frutescens var. acuta Kude (22.8%), Benincasa hispida Cong. (22.4%) in crude lipid, Vitex rotundifolia L. (59.5%), Ganoderma lucidum Karst (53.9%), Pinus densifiora Sieb. et Zucc. (46.4%), Sambucus williamsii Hanse (41.2%) in crude fiber, and Poria cocos Wolf (91.2%), Gastrodiae elata Blume (87.9%), Gentiana scabra Bunge (86.3%), Rehnannia glutinosa Libosch (86.0%) in nitrogen free extract exhibited higher content than other kinds. Phosphrous contents were 1.5, 1.1 and 1.0% in order of Taxillus chinensis Danser, Malva verticillata L., Nelumbo nucifera Gaeten, and magnesium contents were 0.7, 0.5 and 0.5% in the order of Saururus chinesis Baill., Salivia milltriorrhiza Bunge and Pueraria thunberglans Benth. Calcium contents of Cimicifuge heradeifolia Komarov, Acanthopanax sessiliflorum Seem. and Rhus vericiflura Stokes were 8.5, 7.1 and 3.0%, and potassium contents of Tricholoma matsutake Sing., Houttuynia cordata Thunb., Achyranthes japonica Nakai were 5.6, 5,4 and 4.7%, respectively. Sodium contents were 2.1, 0.6 and 0.3% in the order of Boshinakia rossica Fedtsch., Eucommia ulmoides Oliv. and Prunus mume Seib., and iron contents were 0.3, 0.2 and 0.2% in the order of Cirsium japonicum var, ussuriense Kitamuraa, Gentiana scabra Bunge and Phlomis umbrosa Turcz.
