XRCC3 Thr241Met Gene Polymorphism and Risk of Colorectal Cancer in Kashmir: a Case Control Study

  • XRCC (X-ray cross-complementing group) genes contribute to important DNA repair mechanisms that play roles in the repair of single strand breaks (SSBs) induced by a variety of external and internal factors, including ionizing radiation, alkylating agents and reactive oxygen species. These repair genes have a pivotal role in maintaining genomic stability through different pathways of base excision repair (BER). The aim of this study was to investigate the XRCC3 Thr241Met gene polymorphism in colorectal cancer (CRC) in Kashmir. We investigated the genotype distribution of XRCC3 gene in 120 CRC cases in comparison with 150 healthy subjects and found a significant association between XRCC3 genotypes and CRC ($p{\leq}0.05$). Both heterozygous genotype (Thr/Met) as well as homozygous variant genotype (Met/Met) were moderately associated with elevated risk of CRC [OR=2.53; OR=2.29 respectively]. Also, Thr/Met and Met/Met genotypes demonstrated a significant association with the risk of CRC (p = 0.003). This study displayed a significantly elevated risk for CRC in individuals with XRCC3 Thr/Met and Met/Met Genotype of about 2.5 times that with the Thr/Thr wild genotype.

Study on Relationship between Tumor Necrosis $Factor-\alpha$ Gene Polymorphism and Obese Patients

  • Objective: A number of candidate genes have been in implicated in the pathogenesis of obesity in humans. Tumor necrosis factor-alpha $(TNF-{\alpha})$ is expressed primarily in adipocytes, and elevated levels of this cytokine have been linked to obesity and insulin resistance. Recently, the A allele of a polymorphism at position 308 in the promoter region of $TNF-{\alpha}$ (G-308A) has been shown to increase transcription of the gene in adipocytes. Therefore, we designed this study to test whether obese and non-obese subjects differ in $TNF-{\alpha}$ genotype distribution, and how the genotypes affect anthropometric parameters, including degrees of body mass index (BMI). Methods : The study included 153 obese but otherwise healthy women ($BMI{\geq}kg/m^2$, range 25-54.7, age range 15-40 years) and 82 non-obese healthy women ($BMI, age range 15-40 years). Total fat mass and percent body fat were determined by dual-energy X-ray absorptiometry. Genomic DNA was extracted and used for Ncol restriction fragment length polymorphism (RFLP) based genotyping of $TNF-{\alpha}$. Results: No differences were observed for allelic and genotype frequencies between the obese ($BMI{\geq}25$) and non-obese women. Also, no association of TNF-(l polymorphism was observed with body mass index (BMI) for genotype in obese women. In addition, age, pertent body fat, BMI, and cholesterol levels did not differ by $TNF-{\alpha}$ genotype. However, waist-to­hip ratio (WHR) was significantly lower in subjects with $TNF-{\alpha}$ GA or AA genotype (0.94 0.07 vs. 0.920.03, P<0.005). Conclusion: These results suggest that $TNF-{\alpha}$ promoter polymorphism at position-308 is not a significant factor for BMI, but affects the WHR in obese healthy women from Koreans.

Analysis of TP53 Polymorphisms in North Indian Sporadic Esophageal Cancer Patients

  • Background: To investigate the relationship of five TP53 polymorphisms (p.P47S, p.R72P, PIN3 ins16bp, p.R213R and r.13494g>a) with the esophageal cancer (EC) risk in North Indians. Materials and Methods: Genotyping of p.P47S, p.R72P, PIN3 ins16bp, p.R213R and r.13494g>a polymorphisms of TP53 in 136 sporadic EC patients and 136 controls using polymerase chain reaction and PCR-RFLP. Results: The frequencies of genotype RR, RP and PP of p.R72P polymorphism were 16.91 vs 26.47%, 58.82 vs 49.27% and 24.27 vs 24.27% among patients and controls respectively. We observed significantly increased frequency of RP genotype in cases as compared to controls (OR=1.87, 95% CI, 1.01-3.46, p=0.05). The frequencies of genotype A1A1, A1A2 and A2A2 of PIN3 ins16bp polymorphism were 69.12 vs 70.59%, 27.20 vs 25% and 3.68 vs 4.41% among patients and controls. There was no significant difference among genotype and allele distribution between patients and controls. The frequencies of genotype GG, GA and AA of r.13494g>a polymorphism were 62.50 vs 64.70%, 34.56 vs 30.15% and 2.94 vs 5.15% among patients and controls respectively. No significant difference between genotype and allele frequency was observed in the patients and controls. For p.P47S and p.R213R polymorphisms, all the cases and controls had homozygous wild type genotype. The RP-A1A1-GG genotype combination shows significant risk for EC (OR=2.01, 95%CI: 1.01-3.99, p=0.05). Conclusions: Among the five TP53 polymorphisms investigated, only p.R72P polymorphism may contributes to EC susceptibility.

Study on Body Composition, Biochemical Parameters, and Consumption of Convenience Foods According to ${\beta}$-3 Adrenergic Receptor Polymorphism in University Students (베타-3 아드레날린 수용체 유전자 다형성에 따른 대학생의 생화학 지표, 체성분과 편의식품 섭취실태에 관한 연구)

  • The purpose of this study was to investigate the body composition, biochemical parameters, and consumption of convenience foods according to ${\beta}$-3 adrenergic receptor polymorphism in university students. A survey was conducted on a total of 486 students - 189 males and 297 females. Based on a self-reporting method, questionnaires were administered for over 20 minutes, and ${\beta}$-3 adrenergic receptor and blood samples were also analyzed. The genotype frequencies of ${\beta}$-3 adrenergic receptor polymorphism were Trp/Trp homozygote (73.0%) and Trp/Arg heterozygote (27.0%) in male students. For the female students, the distribution of genotypes was Trp/Trp (71.0%) and Trp/Arg (29.0%). There were no differences according to biochemical parameters (ALT, cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol, and hemoglobin) or body composition. Males with TT genotype frequently ate Ramyon (2.40${\pm}$0.52), Cup Ramyon (2.37${\pm}$0.39), Kimchi (2.23${\pm}$0.61), and frozen meat (2.00${\pm}$0.44), whereas males with TA genotype ate Fries (frozen food) (1.90${\pm}$0.79), Smoked meat (1.67${\pm}$0.81), and Canned fruit (1.64${\pm}$0.81). Females with TT genotype frequently ate Frozen fries (2.21${\pm}$0.35), Kimbab (2.12${\pm}$0.44), and Ramyon (1.85${\pm}$0.40), whereas females with TA genotype frequently ate Kimchi (1.73${\pm}$0.98), Fries (frozen food) (1.46${\pm}$0.26), and Cup Ramyon (1.30${\pm}$0.34). When questioned about satisfaction about body shape, 22.8 and 60.8% of those with TT genotype answered that they were 'satisfied' or needed to 'lose weight', respectively, whereas 18.0 and 63.9% of those with TA genotype answered that they were 'satisfied' or needed to 'lose weight', respectively. In conclusion, this study found no significant effects in terms of ${\beta}$-3 adrenergic receptor polymorphism, which suggests that health-promoting education needs to be developed so that university students appropriately recognize their bodies and control their weight in desirable ways. Therefore, it is necessary to educate individuals with TT genotype how to buy reasonable foods by understanding the interrelationship between convenience foods and health care and by checking the nutrition index labels on convenience foods. Thus, it is recommended that a health-promoting program be developed for the promotion of healthy lifestyles.

Allele and Genotype Frequencies of the Polymorphic Methylenetetrahydrofolate Reductase and Lung Cancer in ther Jordanian Population: a Case Control Study

  • Background: Methylenetetrahydrofolate reductase (MTHFR) is involved in amino acid synthesis and DNA function. Two common polymorphisms are reported, C677T and A1298C, that are implicated in a number of human diseases, including cancer. Objective: The association between MTHFR C677T and A1298C genotype and haplotype frequencies in risk for lung cancer (LC) was investigated in the Jordanian population. Materials and Methods: A total of 98 LC cases were studied for MTHFR C677T and A1298C polymorphisms, compared to 89 controls taken from the general population, employing the PCR-RFLP technique. Results: The frequency of the genotypes of MTHFR C677T among Jordanians was: CC, 59.6%, CT, 33%; and TT, 7.4% among LC cases and 49.4%, 40.2% and 10.3% among controls. No significant association was detected between genetic polymorphism at this site and LC. At MTHFR A12987C, the genotype distribution was AA, 29.5%; AC, 45.3%, and CC 25.3% among LC cases and 36.8%, 50.6% and 12.6% among controls. Carriers of the CC genotype were more likely to have LC (OR=2.5; 95%CI: 1.04-6; p=0.039) as compared to AA carriers. Smokers and males with the CC genotype were 9.9 and 6.7 times more likely to have LC, respectively ($OR_{smokers}=9.9$; 95%CI: 1.2-84.5, p=0.018; $OR_{men}=6.6$; 95%CI: 1.7-26.2, p=0.005). Haplotype analysis of MTHFR polymorphism at the two loci showed differential distribution of the CC haplotype (677C-1298C) between cases and controls. The CC haplotype was associated with an increased risk for lung cancer (OR=1.6; 95% CI: 1.03-2.4, p=0.037). Conclusions: The genetic polymorphism of MTHFR at 1298 and the CC haplotype (risk is apparently lower with the C allele at position 677) may modulate the risk for LC development among the Jordanian population. Risk associated with the 1298C allele is increased in smokers and in males. The results indicate that a critical gene involved in folate metabolism plays a modifying role in lung cancer risk, at least in the Jordanian population.

IL-6 gene promoter polymorph isms in Korean generalized aggressive periodontitis (한국인 전반적 급진성 치주염 환자에서 IL-6 유전자 다변성에 관한 연구)

  • Purpose: The purpose of this study was to investigate the association of generalized aggressive periodontitis with IL-6 promoter gene single nucleotide polymorphisms(SNP). Material and Methods: The study population consisted of 52 generalized aggressive periodontitis patients(GAP) and 30 periodontally healthy control subjects, who were systemically healthy non-smokers. Genomic DNA was obtained from buccal swab. The IL-6 promotor SNP at the positions of -597, -572, and -174 were genotyped by amplifying the polymorphic region using polymerase chain reaction(PCR), restriction enzyme digestion and gel electrophoresis. Result: The genotype distributions for G/G, G/A and A/A genotypes of IL-6 -597 were 30.8%, 40.4%, and 28.8% in the GAP group and 53.3%, 40%, and 6.7% in the control group and were statistically different between 2 groups(p<0.05). Allele 2 frequency of IL-6 -597 were significantly higher in the GAP group than the control group(p<0.01). At the position of IL-6 -572, the distribution for C/C, C/G and G/G genotypes were 23.1%, 55.8% and 21.2% in the GAP group and 20%, 33.3%, and 46.7% in the control group. In female subjects, the genotype distribution were significantly different between 2 groups(p<0.01). In male subjects, allele 2 frequency of IL-6-572 was significantly lower in the GAP group than the control group(p<0.05). The genotype distribution of IL-6 -174 in the GAP group were 96.2%, 3.8% for G/G, G/C genotypes whereas only the G/G genotype was detected in the control group. Conclusion: In conclusion, significant associations were found in IL-6 gene promoter(-597, -572) polymorphisms and generalized aggressive periodontitis. Further cohort study will be necessary in larger population.

Human Papillomavirus Genotypes Associated with Mucopurulent Cervicitis and Cervical Cancer in Hangzhou, China

  • Background: To investigate the infection status and predominant genotype distribution of human papillomavirus (HPV) infection among Chinese patients with mucopurulent cervicitis (MPC) or cervical cancer (CC) in Hangzhou. Methods: Initially, 217 cases of healthy cervix controls (n=50), acute MPC (n=89), and CC (n=78) were included; samples were collected between January 1, 2010, and January 30, 2013. Cervical specimens were screened for HPV using a nested polymerase chain reaction assay and DNA sequencing. Results: Overall prevalence of HPV infection was 16.7% in the control group, 51.9% in the MPC group, and 84.4% in the CC group. The predominant genotype detected in all 3 groups was the oncogenic variant HPV 16 (55.8%, 17.3%, and 6.3% in the CC, MPC and control specimens, respectively), HPV58 was the second most predominant HPV type in CC (9.1%), MPC (8.6%), and control group (4.2%). Most or all of the genotypes were oncogenic in the three groups. Conclusions: Infection with HPV was found to be prevalent among Chinese women with MPC or CC and oncogenic variants were in the majority. Therefore, peoples who suffered MPC with HPV DNA positive should be regularly followed-up, for prevention and early treatment of cervical cancer.

Serotyping in Patients with Chronic Hepatitis C

  • To determine the clinical usefulness of Immuno Blot test, 160 samples from the patients with chronic HCV infection were analyzed. And serotyping and line probe assay were performed to evaluate the distribution of hepatitis C virus genotypes in Korean isolates. In this group, as a result of genotyping type 1 band 2a, the serotype I and II were the most common source of HCV infection. There were no significant difference in response to the alpha-interferon HCV infection treatment with the subtype 1 b or 2a. And the serotypes of NS4 peptides were compared with the genotypes to evaluate their clinical usefulness. Among 49 cases studied for genotypes and serotype, genotype 1 b, 1 b/2b, 2a, 2a/2c and 2b were 51.0%, 2.0%, 34.6%, 8.1% and 4.0%, respectively. The serotypes I and II were 57.1% and 42.8%, respectively; they were matched with genotypes in 85.7% and seemed to be easy to perform. To monitor their performing progress or treatment response, serotype test was made before the genotype test. The Result showed that there was no significant difference in response to the alpha-interferon HCV infection treatment with the subtype 1 b or 2a in Korea.

  • We investigated whether multiple infections can be used as predictors of progression to carcinogenesis in accordance with the cytological diagnosis in women receiving abnormal cytologic diagnosis as analysis genotype and compared to single infection. HPV prevalence is highest in the age of under 30 years old woman, HPV prevalence is started to lower after 30 years old and started to increase over 60 years old as like a U-shape. The specific HPV genotypes is an important factor because increased single infection and reduced multiple infections and appeared single infection with AC in progressing carcinogenesis. HPV 16 revealed the statistical significance at the single infection in squamous cell lesions, and HPV 18 revealed the statistical significance at the single infection in adenocarcinoma with showed HPV 16, 58, 18, 52-type distribution.

  • Hepatitis A virus (HAV) is a causative agent of triggering acute hepatitis which is transmitted by person-to-person contact and or fecal-oral route. In previous studies, most hepatitis A virus (HAV) isolates had been genotype IA in Korea. Recently, a small number of different genotypes were reported with an upsurge of acute hepatitis by HAV. Therefore, the distribution of HAV genotypes was investigated. RNA was extracted from anti-HAV IgM positive sera which were collected from February to August 2009, at a tertiary care hospital in eastern Jeonnam, Korea. Nested reverse transcription PCR and direct sequencing for VP1/P2A region of the HAV were performed. A total of 365 cases with suspected acute hepatitis were tested for anti-HAV IgM and positive results were obtained in 24 sera (9.0%), which were collected 2 to 15 days (median, 7 days) after the onset of symptoms. Of the 24 seropositive samples, 14 (58.3%) samples were positive for HAV RNA, among which 4 isolates (28.6%) were genotype IA and the other 10 (71.4%) were genotype IIIA. Both IA and IIIA genotypes were isolated from 5~6 neighboring administrative districts throughout the year without geographic or seasonal restrictions. HAV genotypes (IA and IIIA) were observed from the eastern Jeonnam for the studied.
