  • The clastogenecity and mutagenicity of antifungal 6-[(N-halophenyl)amino]-7-chloro-5, 8-quinolinedione (RCK 3, 7, 13, 14, and 15) had been evaluated. Salmonella typhimurium reversion assay (Ames test) was used to test the mutagenicity of RCKs. RCK14 was mutagenic in S. typhimurium(TA98 and TA100) with and without rat liver microsomal activation. Whereas RCK3, 7, 13 and 15 were negative in Ames test with Salmonella typhimurium(TA98 and TA100), The clastogenecity was tested on the RCKs with in vivo mouse micronucleus assay. All of RCKs tested did not show any clastogenic effect in mouse peripheral blood. Thus RCKs were not supposed to cause any chromosomal damage termed micronuclei. These results indicate that RCK 3, 7, 13 and 15 have no genotoxic potential under these experimental condition.

Influence of Food Ingredients on the Formation of Heterocyclic Aromatic Amine in Cooked Pork Patties

  • The effects of cooking method, cooking time and various food ingredients on the formation/ inhibition of heterocyclic aromatic amines (HAAs) in pork products were investigated. Three HAAs, 2-amino-3,8-dimethylimidazo [4,5-f] quinoxaline ($MeIQ_x$), 2-amino-3,4,8-trimethylimidazo [4,5-f] quinoxaline ($DiMeIQ_x$) and 2-amino-1-methyl-6-phenylimidazo [4,5-b] pyridine (PhIP) were measured in pork products using solid-phase extraction and HPLC. Pork patties were boiled, oven-broiled and pan-fried to internal temperatures of 71, 77 and $88^{\circ}C$. Generally, HAA concentrations increased with increasing internal temperature, and HAA formation was greatest with pan-fried. Selected food ingredients (vitamin E, sodium nitrite, sodium tripolyphosphate, sodium ascorbate, Nanking cherry tissue and cherry tissue extract) inhibited HAA formation in pork patties fried at $225^{\circ}C$ for 10 min/side, with the greater inhibition provided by cherry tissue and its methanolic extract.

  • 1,2-Dibromo-3-chloropropane (DBCP), a soil fumigant against nematodes, is a genotoxic carcinogen and also is classified by World Wildlife Fund as endocrine disruptors. DBCP has been extensively studied on genotoxicity, carcinogenicity, and damage in male reproductive-related organs. However, information on precise mechanism of mutagenesis and carcinogenesis of DBCP is yet unknown. Thus the mutation spectrum and mechanism of DBCP was determined in lacI transgenic Big Blu $e^{R}$ Rat2 fibroblast cell lines.(omitted)d)

  • This study was aimed to find out the comparative effects between non-irradiated, and 5kGy-10kGy of gamma-irradiated Panax Ginseng Radix powder on the genotoxicity for identification of possibility of DNA damage causing cancer. Four different short-term mutagenicity tests were used: (1) Salmonella typhimurium reversion assay (Ames test) (2) Chromosome aberration test in cultured Chinese hamster lung (CHL) fibroblast cells. (3) Micronucleus test in ddY mouse (4) Somatic mutation and recombination test in the wing cells of Drosophila melanogaster.Gamma-irradiated Panax Ginseng Radix powder revealed negative results in these four mutagenicity tests. This means gamma-irradiated ginseng could be safe on the genotoxic point of view.

  • Coumarin derivatives were shown to possess valuable pharmacological properties such as anticancer/anti carcinogenic, anti-inflammatory, anti helicobacter, anti genotoxic, neuroprotective and dietary effect. In this study, novel coumarin derivatives structurally related to 7-geranyloxycoumarin were effectively synthesised in good yields by $Cs_2CO_3$/acetonitrile in mild condition. The synthesis of geranyloxycoumarin derivatives in weak base($Na_2CO_3$, $K_2CO_3$, $Cs_2CO_3$ etc)/$CH_3CN$ at room temperature obtained in good yield. On the other hand, the reaction of geranyloxycoumarin formation in strong base(NaOH, KOH, CsOH etc)/$CH_3CN$ at reflux condition obtained in low yield.

Genotoxicity of the Herbicide 2,4-Dichlorophenoxyacetic acid (2,4-D): Higher Plants as Monitoring Systems

  • Higher plants provide valuable genetic assay systems for screening and monitoring environmental pollutants. They are now recognized as excellent indicators of mutagenic effects of environmental chemicals and are applicable for the detection of environmental mutagens both indoor and outdoor. 2,4-dichlorophenoxyacetic acid (2,4-D) is a herbicide commonly used in agriculture. The residues of 2,4-D are present in air, water, soil and edible plants. It constitutes a real hazard to the public health because it's wide spread use in agriculture. Genotoxic effects of 2,4-D on plant cells and potential of higher plants as a biomonitoring system for detecting chemical mutagens are evaluated. It is recommended that higher plant systems have been accepted by regulatory authorities as an alternative biomonitoring system for the detection of possible genetic damage resulting from pollution and the use of environmental chemicals.

  • The effectiveness of N-acetyl-L-cysteine (NAC) to protect blood cells against Mitomycin C (MMC) induced genotoxicity was investigated in human chronic myeloid leukemia cells (KCL22) using the alkaline comet assay. The comet assay was selected as sensitive and rapid method for analysis of DNA damage and repair in individual cells. NAC treatment alone did not produce any damage in KCL22 cell. But NAC was found to be effective in reducing genotoxic damage in KCL22 cells exposed to MMC. These results confirm the literature data that, given the safety and ability to reduce DNA damage. NAC may be useful to prevent drug-mediated genotoxicity.

  • The present study examined the effect of alcohol extract of Isaria sinclairii on blood pressure in spontaneously hypertensive rats (SHR). The blood pressure and heart rate were measured after treatment of alcohol extract of I. sinclairii by indirect tail cuff method and direct tn vivo model. Male SHR were treated with extracts for 2 or 4 weeks starting at 12 weeks of age. (omitted)

Application of the SCGE Assay for Detecting Induced DNA Damage in Plant Leaves

  • The possibility of using the alkaline protocol of the single cell gel electrophoresis (SCGE) assay as a method for detecting induced DNA damage has been studied for six major plants. The EMS was applied as a model genotoxic agent on young excised leaves of the tested crops for 18 h at 26$^{\circ}C$ in the dark. With increasing concentrations of 0 to 10 mM EMS, the DNA damage, expressed by the averaged median tail moment values, significantly increased in nuclei of all plants studied. As the results, no correlation between the diameter of nuclei and sensitivity to EMS treatment was observed. The data obtained demonstrate the feasibility of using the SCGE assay for detecting induced DNA damage in plants.
