Objectives: For our survey of the incidence of norovirus infections and the genogroup distribution of norovirus in Seoul, Republic of Korea, we evaluated through regular surveillance the prevalence of norovirus infections in patients with acute gastroenteritis occurring in Seoul from January 2007 to July 2011. Methods: For norovirus detection, we conducted epidemiological analyses on the basis of the junction of ORF1 and ORF2 (approximately 314 bp). 11,202 fecal specimens were collected from patients in Seoul with acute gastroenteritis between January 2007 and July 2011 and then tested for the presence of NoV via reverse transcription (RT) - polymerase chain reaction (PCR). Results: 16.6% (1,861/11,202) of the fecal specimens were determined to be positive for noroviruses. The incidences of norovirus infection in Seoul in the case of acute gastroenteritis with regular surveillance were 28.0% in 2007, 14.6% in 2008, 9.1% in 2009, 14.1% in 2010, and 12.9% in 2011, which shows that noroviruses constituted a major causative agent of acute gastroenteritis. Also, the incidence of noroviral infection in patients with acute gastroenteritis increased after the large-scale new influenza in 2009. Conclusions: The genetic characteristics of norovirus and the epidemiologic patterns of a viral pathogen in acute gastroenteritis patients may provide potentially effective data for epidemiological studies in Seoul, Korea.
RT-PCR method was applied to detect and clone the nucleocapsid protein (N) gene and glycoprotein (G) gene for sequencing 5 Korean VHSV isolates from cultured olive flounder, Paralichthys olivaceus. Phylogenetic analysis was performed to investigate their relationship with the VHSV strains described previously and isolated from different geographical area. Generally, VHSV strains were separated phylogenetically according to the major geographical area of isolation: Genogroup I (American type), Genogroup Il (British Isles) and Genogroup ill (European type). This study revealed that all 5 Korean VHSV isolates were belonged to Genogroup I and closely related to Japanese Obama25 type.
최근 어린 전복의 대량 폐사가 국내의 전복 종묘 양식장에서 발생하였다. 죽은 어린 전복의 마리 수는 전체 사육 마리 수의 약 50% 이상이었으며, 그 크기는 각장 3 cm 전후였다. 폐사의 현상은 어린 전복이 부착 기질로부터 탈락되어 힘이 없이 바닥에 깔려 있거나, 뒤집어진 상태에서 스스로 일어나지 못하였다. 이러한 개체들은 대부분 근육에 수포를 형성하고 있는 병리학적 특징을 나타내었다. 폐사 직전의 전복으로부터 3개의 균이 분리되었으며, 이들은 16S rDNA 염기서열 분석에 의해 모두 V. harveyi로 동정되었다. 또한 이들을 어류에서 분리된 V. harveyi 36개의 균들과 염기서열을 비교 분석한 결과, 어류에서 분리된 V. harveyi는 genogroup a와 b로 구별되며, 전복에서 분리된 3균주는 genogroup a와 새로운 genogroup c에 속하였다. 그중 WA AG-1과 WA CS-5 균을 건강한 전복에 인위 감염시킨 결과, 자연 감염된 개체와 같은 외부 및 병리조직학적 증상을 나타내면서 폐사하였으며, 그 반수 치사농도는 각각 $1.0{\times}10^3cfu\;animal^{-1}$ 및 $1.7{\times}10^4cfu\;animal^{-1}$이었다.
The cDNA of the aquabirnavirus, GC-1 isolated from rockfish Sebastes schlegeli in Korea, was synthesized using the reverse transcriptase-polymerase chain reaction. The nucleotide and deduced amino acid sequences were determined from cDNA of the VP2-NS-VP3 coding region of genome segment A. The nucleotide sequences of the segment A were 3,086 base pairs (bp) in length and contained large open reading frame (ORF) and terminal sequences. The large ORF was comprised of 2,916 bp nucleotides and composed of 972 deduced amino acid sequences. Pairwise comparisons were made with other aquabirnavirus sequences published previously. The study of genetic relationships between GC-1 and aquabirnaviruses in the large ORF and VP2 coding regions demonstrated that the GC-1 has the nearest genetic relationship with the marine birnaviruses (MABV strains), and the GC-1 and MABV strains can be clustered as the same genogroup. GC-1 can be included in MABV, which is the 7th genogroup of family Aquabirnaviridae.
노로바이러스는 국내에서도 지속적으로 유행성 바이러스성 위장염을 일으키고 있다. 노로바이러스 저감화사업단(NOROTECL)의 1중 2세부과제에서는 농산물의 노로바이러스 오염 원인을 추적조사하여 노로바이러스 전파를 예방하고 저감화하기 위한 과제를 수행하였다. 2015년 1월부터 11월 사이에 노로바이러스와 관련이 있을 것으로 추정되는 80개 농산물, 80개 토양, 78개 인체분변, 3개 가축분변, 80개 농업용수, 80개 하천수 시료를 대상으로 노로바이러스, Male specific coliphage (MSC), 위생지표세균(Coliform, E. coli) 세 가지 검사를 통해 오염현황을 조사하였다. Semi-nested PCR과 DNA sequencing을 통해 18개의 Genogroup I과 3개의 Genogroup II 노로바이러스가 총 18개의 시료에서 발견되었다. Genogroup이 확정된 노로바이러스에 대하여 RT-PCR을 진행하였다. 대장균군과 대장균은 농산물에서 68%와 1%, 토양에서 88%와 7.5%, 인체분변에서 44%와 12.8%, 가축 분변에서 67%와 67%, 농업용수에서 74%와 30%, 하천수에서 96%와 51%의 검출율을 각각 나타냈다. MSC 결과에서는 14개의 시료가 양성으로 나타났다.

Feline calicivirus (FCV) is a highly infectious pathogen in cats and widely distributed worldwide with high genetic variation. Full-length open reading frame 2 of 5 from recently isolated Korean FCV isolates were sequenced and compared with those of global isolates. The results of phylogenetic analysis supported dividing global FCV isolates into two genogroups (type I and II) and demonstrated the presence of genogroup II in Korea, indicating their geographic spread in East Asia. High sequence variations in region E of the FCV isolates emphasizes that a novel vaccine needs to be developed to induce protective immunity against various FCV strains.
Twenty isolates of Hantavirus were isolated from patients and reserovirs from 1988 to 1994 in Korea. Isolation rate was 1.9% (10/538) in patients, 6.2% (5/81) in Apodemus sp., 2.6% (1/38) in Rattus sp. and 0.6% (4/677) in bats. Reciprocal mean IFA titers ranged from 27.5 to 1,024 at the specimen collection. According to the growth rate and reaching peak titier of infectivity, the isolates were grouped as rapid, intermediate, and slow growing groups. All isolates were confirmed as Hantaan type by the nested RT-PCR on the G1 region of the M segment. Comparison of nucleotide sequence (Nt: 2101 - Nt: 2280) of the G2 region revealed that the sequence homology bewteen Hantaan 76/118 virus and the isolates was more than 90%. Several nucleotide positions of the isolates showed high variation. The variation rate of patientisolates was about one-half when compared with that of rodentisolates. On the basis of phylogenetic analysis Hantaan viruses isolated were divided into two genogroups. These results indicate that Hantaan virus is highly dominant serotype in Korea and the virologic property and genogroup are not correlated.

Low levels of virus contamination and naturally occurring reverse transcription-polymerase chain reaction (RT-PCR) inhibitors restrain virus detection in oysters. A rapid and efficient oyster-processing procedure that can be used for sensitive virus detection in oysters was developed. Poliovirus type 1 Sabin strain was used to evaluate the efficacy of virus recovery. The procedure included (a) acid-adsorption and elution with buffers (0.25M glycine-0.14 M NaCl, pH 7.5; 0.25M threonine-0.14M NaCl, pH 7.5); (b) polyethylene glycol (PEG) precipitation; (c) resuspension in Tween 80/Tris solution and chloroform extraction; (d) the second PEG precipitation; (e) viral RNA extraction with TRIzol and isopropanol precipitation; and (f) RT-PCR combined with semi-nested PCR. The overall recovery of elution/concentration was 19.5% with poliovirus. The whole procedure usually takes 19 hr. The overall detection sensitivity was 4 RT-PCR units of genogroup I norovirus (NoV) and 6.4 RT-PCR units of genogroup II Nov/25 g of oysters initially seeded. The virus-detecting method developed in this study should facilitate the detection of low levels of NoV in oysters.

Infectious haematopoietic necrosis virus (IHNV) is an important fish pathogen that infects both wild and cultured salmonids. Since the first isolation of IHNV from rainbow trout and masu salmon in 1991, a series of IHN disease outbreak has been reported in Korea. In 2011, we isolated two IHNV isolates from rainbow trout cultured in Korea. The full open-reading frame (ORF) encoding the glycoprotein (G) of them were sequenced and the amino acid sequences were phylogenetically analyzed. Phylogenetic analysis of the G revealed that both IHNV isolates were grouped into an Asian genogroup containing Korean IHNV isolates and Japanese IHNV isolates. However, based on their sequence variation, they were divided into different subgroup. While one isolate was similar to other Korean isolates, the other isolate showed a high level of similarity with Japanese isolates, suggesting the possibility of influx of new IHNV strain into Korea.
본 연구에서는 RT-nested PCR을 수행하여 부산 도심의 하천 중 동천에 존재하는 노로바이러스를 검출하고자 하였다. 기존에 보고되어진 노로바이러스의 capsid protein의 염기서열을 비교하여 노로바이러스 genogroup I,II (GI,II) 를 검출하기 위한 새로운 degenerated primer sets (PNK, KGIF/KGIR and KG2F/KG2R) 를 제작하였으며, 채수한 동천 시료를 초고속원심분리기를 통해 농축 후 물 속에 존재하는 노로바이러스의 검출을 시도하였다. 노로바이러스를 검출하기 위해 PCR primer를 비교한 결과 본 연구에서 제작한 capsid protein gene을 target으로 하는 primer set가 기존에 보고되어 있는 primer set보다 동일 시료에 대한 검출빈도가 우수하였다. 동천에 존재하는 노로바이러스의 오염 수준은 GI과 GII가 각각 76.47% (13/17), 70.59% (12/17) 로 나타났다. 그러나 기존에 알려진 primer와 본 연구에서 제작한 primer를 사용하였을 때 검출된 양성비율이 차이가 나지 않았다. 검출된 노로바이러스를 염기서열 비교를 통한 계통 발생학적 분석 결과, 동천에서 검출된 GI의 경우 1/2/4/5/9/10의 genotype이 GII의 경우 3/4/5/11/13의 genotype으로 분류되었다. 그리고 본 연구에서 검출된 major type 중 GII/4의 경우, 최근 아시아 각국에서 많은 문제를 일으키고 있는 major genotype으로 알려져 노로바이러스에 대한 위험성을 제고하게 하였다. 또한, 이러한 결과는 국내의 강, 호수, 하천 등이 비슷한 노로바이러스의 GI,II의 genotype으로 오염되어 있음을 암시하며 수계환경 중 미생물의 질을 개선하기 위한 지속적인 노로바이러스의 모니터링이 요구된다.
