Promoter Polymorphisms of ST3GAL4 and ST6GAL1 Genes and Associations with Risk of Premalignant and Malignant Lesions of the Cervix

  • Sialyltransferase gene expression is altered in several cancers, including examples in the cervix. Transcriptional regulation of the responsible genes depends on different promoters. We aimed to determine the association of single-nucleotide polymorphisms in the B3 promoter of the ST3GAL4 gene and the P1 promoter of the ST6GAL1 gene with cervical premalignant lesions or cervical cancer. A blood sample and/or cervical scrapes were obtained from 104 women with normal cytology, 154 with premalignant lesions and 100 with cervical cancer. We also included 119 blood samples of random donors. The polymorphisms were identified by sequencing from PCR products. For the B3 promoter, a fragment of 506 bp (from nucleotide -408 to +98) was analyzed, and for the P1 promoter a 490 bp (-326 to +164) fragment. The polymorphism analysis showed that at SNP rs10893506, genotypes CC and CT of the ST3GAL4 B3 promoter were associated with the presence of premalignant lesions (OR=2.89; 95%CI 1.72-4.85) and cervical cancer (OR=2.23; 95%CI 1.27-3.91). We detected only one allele of each polymorphism in the ST6GAL1 P1 promoter. We did not detect any genetic variability in the P1 promoter region in our study population. Our results suggest that the rs10893506 polymorphism -22C/T may increase susceptibility to premalignant and malignant lesions of the cervix.

Statistical Treatment on Amylose and Protein Contents in Rice Variety Germplasm Based on the Data Obtained from Analysis of Near-Infrared Reflectance Spectroscopy (NIRS) (NIRS 분석 Data에 의한 국내외 육성품종 벼 유전자원의 아밀로스 및 단백질 성분에 대한 통계분석)

  • A statistical analysis for 3651 genetic resources collected from China (1,542), Japan (1,409), Korea (413), and India (287) was conducted using normal distribution, variability index value (VIV), analysis of variation (ANOVA) and Ducan's multiple range test (DMRT) based on a data obtained from NIRS analysis. In normal distribution, the average protein content was 8.0%, whereas waxy type amylose and common rice amylose were found to be 8.7% and 22.7%, respectively. The protein contents ranged from 5.4 to 10.6% at the level of 95%. The waxy amylose and common rice amylose ranged from 5.9 to 11.5%, and from 16.9 to 28.5% at 95% confidence level, respectively. The VIV was 0.59 for protein, 0.64 for low amylose, and 0.81 for high amylose contents. The average amylose contents were 18.85% in Japanese, 19.99% in Korean, 20.27% in Chinese, and 25.46% in Indian resources, while the average protein contents were found to be 7.23% in Korean, 7.73% in Japanese, 8.01% in Chinese, and 8.17% in Indian resources. The ANOVA of amylose and protein content showed significant differences at the level of 0.01. The F-test for amylose content was 158.34, and for protein content 53.95 compared to critical value 3.78. The DMRT of amylose and protein content showed significant difference (p<0.01) between resources of different countries. Japanese resources had the lowest level of amylose contents, whereas, the lowest level of protein content was found in Korean resources compared to other origins. Indian resources showed the highest level of amylose and protein contents. It is recommended these results should be helpful to future breeding experiments.

Flowering Patterns of Miscanthus Germplasms in Korea (국내 수집 억새 유전자원의 출수 특성)

  • Miscanthus has been considered as the most promising bioenergy crop for lignocellulosic biomass production. In Korea, M. sacchariflorus and M. sinensis can be found easily in all regions. It is a great advantage to utilize as important species with respect to genetic and cross-breeding programs materials for creation of novel hybrids. For successful breeding programs, it is important to precisely understand the variability of flowering traits among Miscanthus species as breeding parents materials. In this study, flowering traits were observed daily in 960 germplasms of two Miscanthus species (M. sacchariflorus and M. sinensis) for growing seasons over 2 years. The flowering process was divided into three stages. ST (sprouting time) was recorded when first leaf of the plant emerged on soil. FS1 (flowering stage 1) and FS2 (flowering stage 2) were recorded when flag leaf was firstly observed, and 1 cm of panicle was showing on at least one stem, respectively. For 2013 and 2014, the latest germplasms exerted flag leaf, i.e. September 30 (DOY of FS1 164.1) and September 4 (DOY of FS1 141.0) occurred M. sacchariflorus cv. Geodae 1 and M. sacchariflorus cv. Uram collected from Southern Korea (Jeollanam-do), while Miscanthus germplasms collected from northern Korea (Gyeonggi-do) which emerged the earliest flag leaf in July and August, significantly decreased DOY. For DOY from ST to FS2, M. sacchriflorus germplasms ranged from 140 to 190 days, and 110 to 170 days for 2013 and 2014. The highest frequency showed to 160 days for 2013, and 150 days for 2014. In M. sinensis germplasms, the highest frequency showed to 180 days for 2013, and 170 days for 2014. In the results of correlation between the day of years from ST to FS2 for 2013 and 2014, M. sacchriflorus and M. sinensis showed high coefficient of correlation (0.70 and 0.89). It can be supposed that flowering characteristics of Miscanthus are largely affected by the unique phenotypic characteristic of native habitat than environmental factors of the current planted site. This study for flowering traits of Miscanthus may provides an important information in order to expedite the introduction as breeding materials for creation of new hybrid.
