Taxonomic Status of Acheilognathus sp. HR (Cypriniformes: Acheilognathidae) Found in the Dalcheon River, a Tributary of Hangang River, Korea (한강 지류 달천에서 발견된 Acheilognathus sp. HR (Cypriniformes: Acheilognathidae)의 분류학적 위치)

  • To investigate the taxonomic status of undescribed species Acheilognathus sp. HR from Korean Acheilognathidae discovered from the Dalcheon River, a tributary of the Hangang River, molecular phylogenetic and morphological characteristics were compared and analyzed with previous studies. As a result of molecular phylogenetic analysis, A. sp. HR formed the same genetic clade as the five subspecies of Acheilognathus tabira, but formed a separate monophyletic group based on the unique genotype, showing clear differences. As a result of morphological analyses, the dorsal fin color in males is grayish and the nuptial coloration of the outer edge of the anal fin is white. The outer edges of the dorsal and anal fins are convexly rounded. A black blotch is present on the dorsal fin of the juvenile, but there is a black blotch absent on the dorsal fin of the small adult female. In the counts, the number of branched dorsal rays is 12~13. In the measurements, the length of the barbels is short and the body depth is deep. Therefore, the A. sp. HR of Hangang River is considered at the level of a distinct species distinguished from each other by the five subspecies of A. tabira by molecular phylogenetic, morphological, and limited distributional characteristics.

Altitude training as a powerful corrective intervention in correctin insulin resistance

  • Oxygen is the final acceptor of electron transport from fat and carbohydrate oxidation, which is the rate-limiting factor for cellular ATP production. Under altitude hypoxia condition, energy reliance on anaerobic glycolysis increases to compensate for the shortfall caused by reduced fatty acid oxidation [1]. Therefore, training at altitude is expected to strongly influence the human metabolic system, and has the potential to be designed as a non-pharmacological or recreational intervention regimen for correcting diabetes or related metabolic problems. However, most people cannot accommodate high altitude exposure above 4500 M due to acute mountain sickness (AMS) and insulin resistance corresponding to a increased levels of the stress hormones cortisol and catecholamine [2]. Thus, less stringent conditions were evaluated to determine whether glucose tolerance and insulin sensitivity could be improved by moderate altitude exposure (below 4000 M). In 2003, we and another group in Austria reported that short-term moderate altitude exposure plus endurance-related physical activity significantly improves glucose tolerance (not fasting glucose) in humans [3,4], which is associated with the improvement in the whole-body insulin sensitivity [5]. With daily hiking at an altitude of approximately 4000 M, glucose tolerance can still be improved but fasting glucose was slightly elevated. Individuals vary widely in their response to altitude challenge. In particular, the improvement in glucose tolerance and insulin sensitivity by prolonged altitude hiking activity is not apparent in those individuals with low baseline DHEA-S concentration [6]. In addition, hematopoietic adaptation against altitude hypoxia can also be impaired in individuals with low DHEA-S. In short-lived mammals like rodents, the DHEA-S level is barely detectable since their adrenal cortex does not appear to produce this steroid [7]. In this model, exercise training recovery under prolonged hypoxia exposure (14-15% oxygen, 8 h per day for 6 weeks) can still improve insulin sensitivity, secondary to an effective suppression of adiposity [8]. Genetically obese rats exhibit hyperinsulinemia (sign of insulin resistance) with up-regulated baseline levels of AMP-activated protein kinase and AS160 phosphorylation in skeletal muscle compared to lean rats. After prolonged hypoxia training, this abnormality can be reversed concomitant with an approximately 50% increase in GLUT4 protein expression. Additionally, prolonged moderate hypoxia training results in decreased diffusion distance of muscle fiber (reduced cross-sectional area) without affecting muscle weight. In humans, moderate hypoxia increases postprandial blood distribution towards skeletal muscle during a training recovery. This physiological response plays a role in the redistribution of fuel storage among important energy storage sites and may explain its potent effect on changing body composition. Conclusion: Prolonged moderate altitude hypoxia (rangingfrom 1700 to 2400 M), but not acute high attitude hypoxia (above 4000 M), can effectively improve insulin sensitivity and glucose tolerance for humans and antagonizes the obese phenotype in animals with a genetic defect. In humans, the magnitude of the improvementvaries widely and correlates with baseline plasma DHEA-S levels. Compared to training at sea-level, training at altitude effectively decreases fat mass in parallel with increased muscle mass. This change may be associated with increased perfusion of insulin and fuel towards skeletal muscle that favors muscle competing postprandial fuel in circulation against adipose tissues.

Variations in Ecological Niche of Quercus variabilis and Quercus acutissima Leaf Morphological Characters in Response to Moisture and Nutrient Gradient Treatments under Climate Change Conditions (기후변화 조건에서 수분구배 및 영양소 구배에 따른 굴참나무와 상수리나무 잎 형태적 특성의 생태지위 변화)

  • This study attempted to elucidate the ecological niches and influencing environmental factors of Quercus variabilis and Quercus acutissima, which are representative deciduous broad-leaved trees in Korean forests, taxonomically close and genetically similar, under climate change conditions. Under climate change conditions induced by increased CO2 and temperature, soil moisture and nutrient environments were manipulated in four gradients. At the end of the growing, plants were harvested to measure growth responses, calculate ecological niches, and compare them with those of the control. Eperimental plants were grown for 180 days in a glass greenhouse designed with four gradients each for soil moisture and nutrient environments under climate change conditions induced by increased CO2 and temperature. After harvesting, growth responses of leaf traits were measured, ecological niches were calculated, and these were compared with those of the control groups. Furthermore, the responses of the two species' populations were interpreted using principal component analysis(PCA) based on leaf trait measurements. As a result, under climate change conditions, the ecological niche breadth for moisture environment was broader for Quercus variabilis than Quercus acutissima, whereas for the nutrient environment, Quercus acutissima exhibited a broader niche breadth than Quercus variabilis. And the rate of change in ecological niche breadth due to climate change decreased for Quercus variabilis in both moisture and nutrient environments, while for Quercus acutissima, it increased in the moisture environment but decreased in the nutrient environment. Additionally, in terms of group responses, both Quercus variabilis and Quercus acutissima expanded their ecological niches under climate change conditions in both soil moisture and nutrient conditions, with Quercus acutissima exhibiting a broader niche than Quercus variabilis under nutrient conditions. These results indicate that the changes in leaf morphological characteristics and the responses of individuals reflecting them vary not only under climate change conditions but also depending on environmental factors.

Age or Basal Serum FSH Levels; Which One is Better for Prediction of IVF Outcomes in Patients with Decreased Ovarian Reserve? (난소의 기능이 저하된 불임 환자에서 연령 및 기저 혈중 FSH 수치가 체외수정시술의 예후에 미치는 영향에 관한 연구)

  • Objectives: The purpose of this study is to investigate the clinical significance of age and basal serum FSH in predicting the outcomes of in vitro fertilization (IVF) in patients with poor-ovarian response. Methods: From January 2000 to December 2004, 85 second IVF cycles of 85 poor-ovarian response patients under the age of 42 with a back-ground of the first IVF cycles at our infertility center and 5 or less oocytes were retrieved and their basal serum FSH levels of 15$\sim$25 mIU/ml were enrolled in this study. Exclusion criteria were patients with a male factor for the etiology of infertility and undergoing genetic diagnosis of embryo such as PGD. Flare-up protocol was used for ovarian stimulation in all cases. Results: When we stratified the study groups by patient's age, the younger age group (age<35, n=35) showed significantly higher implantation rate (19.0% versus 4.0%, p<0.05) and higher ongoing pregnancy rate (100% versus 14.3%, p<0.05) than the older age group (age$\geq$35, n=50). And then, when we stratified the study populations by basal serum FSH level, the lower FSH group (basal serum FSH<20 mIU/ml, n=58) showed significantly higher number of retrieved oocytes (4.6$\pm$0.7 versus 2.2$\pm$0.5, p<0.05) and lower cancellation rate (19.0% versus 55.6%, p<0.05) than higher FSH group (basal serum FSH$\geq$20 mIU/ml, n=27). Conclusions: In conclusion, it was suggested that the patient's age could predict the IVF outcomes in respect to its potency of pregnancy and ongoing pregnancy. Serum basal FSH levels could predict more accurately the ovarian response of cycle, but not clinical outcomes.

Evaluation of Crop Characteristics of Sorghum (Sorghum bicolor L.) Germplasm for the Selection of Excellent Resources (우수자원 선발을 위한 수수(Sorghum bicolor L.) 유전자원의 특성평가)

  • The aim of this study is to select the superior resources of high yield, high content of functional material optimal to mechanical harvesting by the evaluation of crop growth and yield characteristics in sorghum germplasm. One hundred accessions of sorghum germplasm were used in this experiment. Days from seeding to heading date showed the range from 68 to 94 days with the highest frequency proportion was the group from 80 to 85 days, which occupied 34% (34 plant resources) of 100 germplasm. Ear types of 100 sorghum germplasm could be classified as 7 types of broom-tillering, half broom-tillering, extreme open-loose type, open-loose type, intermediate type, compact type, extreme-compact type of which intermediate type was the highest ratio of 28% (28 plant resources) of 100 germplasm. Yield showed the range from 106 to 365 ㎏/10a with the highest frequency proportion of it was the group from 150 to 200 ㎏/10a, which occupied 44% (44 plant resources) of 100 germplasm. Among 100 sorghum germplasm, 18 ideal resources of high yield and short plant height appropriate for mechanical harvesting were selected. In order to evaluate high content of functional substance, selected 18 resources were analyzed for total polyphenol content, DPPH radical scavenging activity and total anthocyanin content. Finally, we selected 5 resources of short plant height, high yield, high content of total polyphenol and high DPPH radical scavenging activity among 18 genetic resources.

Gene Expression Analysis of Inducible cAMP Early Repressor (ICER) Gene in Longissimus dorsi of High- and Low Marbled Hanwoo Steers (한우 등심부위 근육 내 조지방함량에 따른 inducible cAMP early repressor (ICER) 유전자발현 분석)

  • Marbling (intramuscular fat) is an important factor in determining meat quality in Korean beef market. A grain based finishing system for improving marbling leads to inefficient meat production due to an excessive fat production. Identification of intramuscular fat-specific gene might be achieved more targeted meat production through alternative genetic improvement program such as marker assisted selection (MAS). We carried out ddRT-PCR in 12 and 27 month old Hanwoo steers and detected 300 bp PCR product of the inducible cAMP early repressor (ICER) gene, showing highly gene expression in 27 months old. A 1.5 kb sequence was re-sequenced using primer designed base on the Hanwoo EST sequence. We then predicted the open reading frame (ORF) of ICER gene in ORF finder web program. Tissue distribution of ICER gene expression was analysed in eight Hanwoo tissue using realtime PCR analysis. The highest ICER gene expression showed in Small intestine followed by Longissimus dorsi. Interestingly, the ICER gene expressed 2.5 time higher in longissimus dorsi than in same muscle type, Rump. For gene expression analysis in high- and low marbled individuals, we selected 4 and 3 animal based on the muscle crude fat contents (high is 17-32%, low is 6-7% of crude fat contents). The ICER gene expression was analysed using ANOVA model. Marbling (muscle crude fat contents) was affected by ICER gene (P=0.012). Particularly, the ICER gene expression was 4 times higher in high group (n=4) than low group (n=3). Therefore, ICER gene might be a functional candidate gene related to marbling in Hanwoo.

Genetic and Agronomic Analysis of a Recombinant Inbred Line Population to Map Quantitative Trait Loci for Blast Resistance and Select Promising Lines in Rice (벼 RIL집단의 유전 분석과 농업형질 분석을 통한 도열병 저항성 QTL 탐색 및 유망계통 선발)

  • Koshihikari has been one of the most popular rice cultivars with good eating quality since the 1960s despite its susceptibility to blast disease and lodging. To map the genes controlling blast resistance and to develop promising blast-resistant breeding lines inheriting Koshihikari's high eating quality, a recombinant inbred line (RIL) population was developed from a cross between Koshihikari and a blast resistance donor with early maturity, Baegilmi. A total of 394 Koshihikari × Baegilmi RILs (KBRIL), and the two parents, were evaluated for blast resistance and major agronomic traits including heading date, culm length, panicle length, and tiller number. A linkage map encompassing 1,272.7 cM was constructed from a subset of the KBRIL (n = 142) using 130 single nucleotide polymorphisms. Two quantitative trait loci (QTL) for blast resistance, qBL1.1 harboring Pish/Pi35 and qBL2.1 harboring Pib, were mapped onto chromosomes 1 and 2, respectively. qBL1.1 was detected in both of the experimental sites, Namwon and Jeonju, while qBL2.1 was only detected in Namwon. qBL1.1 and qBL2.1 did not affect agronomic traits, including heading date, culm length, panicle length, and tiller number. From the 394 KBRILs, lines that were phenotypically similar to Koshihikari were selected according to heading date and culm length and were further divided into the following two groups based on blast resistance: Koshishikari-type blast resistant lines (KR, n = 15) and Koshishikari-type blast susceptible lines (KS, n = 15). Although no significant differences were observed in the major agronomic traits between the two groups, the KR group produced a greater mean head rice ratio than the KS group. The present study provides useful materials for developing blast-resistant cultivars that inherit both Koshihikari's high eating quality and Baegilmi's blast resistance.

Genotype analysis of genes involved in increasing grain number per panicle in rice germplasm (벼 유전자원의 수당립수 증진 유전자 유전형 분석)

  • ARice is an important staple food in the world and rice yield is one of the main traits for rice breeding. Several genes involved in increasing the yield have been identified through map-based gene cloning within natural variations in rice. These identified genes are good targets for introducing a genetic trait in molecular breeding. Here, we chose five genes reported to be involved in increasing grain number per panicle in rice; Gn1a, dep1, Apo1, Ghd7, and Nal1. We developed In/Del markers for Gn1a, and dep1, Apo1, and applied the reported SNP markers for Ghd7 and Nal1. We were easily able to examine the genotype of each gene on agarose gel. We tested the genotypes on 479 rice resources that we held with evaluated molecular markers. According to the genotype of each gene, rice resources were divided into 13 haplotypes, and most of the Indica and Japonica varieties were included in haplotypes 1 and 13, respectively. When we examined the effect of each gene on grain number per panicle and panicle number per plant, panicle number per plant in the yield negative allele group for each gene was reduced by approximately 0.3 to 0.8 compared to that in the yield positive allele group. However, the number of yield positive alleles for each gene was higher by about 21 to 27 grains per panicle than that of yield negative alleles. Although most of the varieties were grouped in haplotypes 1 and 13, we believe that this genotype information with evaluated molecular markers will be useful in rice breeding for increasing the yield with grain number per panicle.

Taxonomic Study of Korean Armillaria Species Based on Biological Characteristics and DNA Analyses (생물학적 특성과 DNA분석을 이용한 한국내 Armillaria속균의 분류)

  • From 1985 to 1993, we collected 20 isolates throughout Kangwon and obtained 6 isolates from other sources. A. mellea formed rhizomorph actively, and some of A. osroyae were poor in the formation of rhizomorph and some without formation of rhizomorph. A. tabescens was active in the growth of aerial mycelium and poor in the development of rhizomorph. In A. gallica, the mycelium development among the isolates were variable greatly, and especially in isolate A8(KNU-250), the mycelial development was similar to that of A. osroyae, but A8(KNU-250) showed the feature of A. gallica to change medium into brown color. In PCR-RFLP analysis of the IGS region in rDNA, the homology between each isolate in the A. mellea and A. ostoyae showed 100% homology. A. tabescens showed $0.919{\sim}0.974$ homology, and A. gallica showed $0.619{\sim}1.000$ homology. A19 and A12 showed 100% homology as the same group, but compared with other subgroups they showed less than 10% homology as $0.051{\sim}0.108$ value. In RAPD analyses, the isolates of A. mellea showed high homology among themselves as $0.983{\sim}1.000$, and A. ostoyae also showed high similarity. The homology between isolates of A. tabescens showed $0.594{\sim}0.953$ value because A. gallica showed $0.280{\sim}0.733$ value, and the variations between isolates were greater than other species. Especially, A19 and A22 were identified as new novel group which were remoted from other groups, and the homology between these two isolates showed 0.921 value, and the genetic similarity between these groups and other 4 groups showed less than 7% as $0.012{\sim}0.069$ value. Of 5 species identified in this study, 4 species were identified as A. mellea, A. ostoyae, A. tabescens, and A. gallica that were already reported ones and 1 species was suggested as a new specie in Korea.

The review on the identification of the gifted child in the sports (스포츠영재의 판별에 관한 고찰)

  • The purpose of this study was to indicate the ways to identify the gifted child in the sports, and the research direction in this area in the future. The most of researches related to the early development of the children who showed the gifted talents in the certain sports skills were published from the European area such as the old East Germany or Soviet Union. Those research papers or articles were main focus of this study to be analyzed. The analysis in this study will be assistant to provide the direction of the research in the future about the method to identify appropriately on the children with the gifted sports skills. -Related to the period of selection of the children with sports skill gifted, the divided selection procedure will be good such as the 1st selection and 2nd selection. In the 1st selection, the goal will be to test the potentials of the children with the sports talents in the public school with creating the special group, and the 2nd selection will be to examine the qualification of the admission to the special schools for the children with the sports talents. -Related to the method of selection of the children with sports talents, the special item on the developmental procedure should be considered on the each field as well as the potentials focused. And, the physical factors, the condition of growth and development, psychological factor, and environmental factor should be comprehensively considered. -The system of promotion in the period of the primary school will be appropriate in the special group for the children with the sports talents, and the special school will be good from the middle school. The system outlooks will be adequate if shaped as the hierarchical structure. -The curriculum for the children with the sports talents will be proper that the basic fitness for the earlier ages and the professional skills will be recommended if getting older. -The continuous research is strongly required for the judgement and promotion of the children with sports talents, therefore, the multiple intelligences approach will be helpful for the development of the bodily-kinesthetic intelligence, and in other words, the new era of the genetic method will be another prospective area such as the DNA analysis in the identification of the talented children for the sports filed.
