Genetic Diversity and Relationship of the Walleye Pollock, Theragra chalcogramma Based on Microsatellite Analysis (Microsatellite marker 분석을 이용한 명태(Theragra chalcogramma) 5 집단의 유전적 다양성 및 유연관계 분석)

  • A comprehensive analysis of the genetic diversity and relationship of the cold-water fishery walleye pollock (Theragra chalcogramma), the most abundant economically important fishery resource in the East sea of Korea, has not been carried out, despite its importance in Korea. The present study assessed the genetic diversity and relationship between five walleye pollock populations (Korean population, Russian population, USA population, and Japanese populations) of T. chalcogramma using eight microsatellite DNA (msDNA) markers to provide the scientific data for the preservation and management of the Pollock fishery resource. The results of the analysis of 186 individuals of the Pollock revealed a range of 7.13-10.63 numbers of alleles (mean number of alleles=9.05). The means of observed heterozygosity ($H_O$), expected heterozygosity ($H_E$) were 0.732 and 0.698, respectively. The results of genetic distance, Pairwise $F_{ST}$, UPGMA (UPGMA: un-weighted pair-group method with an arithmetical average) (the phylogenetic tree), PCA (PCA: Principal Coordinate analysis) analysis pointed to significant differences between the Korean population, Russian population, USA population, and Japanese populations, although small (p<0.05). These results shed light on the genetic diversity and relationships of T. chalcogramma and can be utilized for research on the evaluation and conservation of Korean T. chalcogramma as genetic resources.

Evaluation of Genetic Ability for Meat Quality in Hanwoo Cow (육질개량을 위한 한우번식우의 유전능력평가)

  • This study was carried out to estimate genetic parameters and breeding values of Hanwoo cows for carcass traits. Carcass records were collected from Korean steers raised at the private farms located in rural area of Gangwon-do and slaughtered from December 2004 to June 2008 at the three slaughter houses near fattening farm. The results obtained in this study were summarized as follows; The means and standard deviations of the carcass traits were $5.57{\pm}2.01$ for marbling score (MS), $10.87{\pm}4.06mm$ for backfat thickness (BFT), $87.87{\pm}9.07cm^2$ for eye muscle area (EMA), $426.75{\pm}49.27kg$ for carcass weight (CW), and $65.80{\pm}3.80$ for yield index (YI). Heritability estimates using single trait analyses were 0.36 for MS, 0.35 for BFT, 0.24 for EMA, 0.29 for CW, and 0.40 for YI, respectively. Genetic correlation coefficients of MS with BFT, EMA, CW, and YI were -0.21, 0.30, -0.21 and 0.30, and those of BFT with EMA, CW and YI were -0.12, 0.57 and -0.97, and those of EMA with CW and YI were 0.32 and 0.27, respectively. And genetic correlation of CW with YI was -0.62. Single trait selection for MS might lead to reducing BFT, but might be an obstacle to increase CW due to negative genetic correlations of MS with BFT and CW.

Analysis of the Genetic Diversity of Radish Germplasm through SSR Markers Derived from Chinese Cabbage (배추 SSR 마커를 이용한 무의 육성 계통 및 수집종의 유전적 다양성 분석)

  • Since the early 1980s, the National Institute of Horticultural & Herbal Sciences has been breeding and collecting diverse radish breeds to select those samples with better horticultural characteristics, to ultimately expand and develop as good radish produce. Genetic diversity is a crucial factor in crop improvement and therefore it is very important to obtain various variations through sample collection. The collected samples were compared with one another in order to assess the level of diversity among the collections, and this procedure allowed for increased application of the gathered resources and aided in determining the direction to secure further samples. Towards this end, this experiment was conducted in order to examine whether the SSR markers derived from Chinese cabbage samples could be transferred to the radish samples. Among the radish breeding lines and introduced resources, 44 lines were used as materials to analyze the genotype using 22 SSR markers selected. As a result, the analysis showed that among all the selected markers, 'cnu_m139' and 'cnu_m289' were the most useful markers for diversity evaluation. The genetic relationship of the radish genetic resources showed that the geographic origins affected the diversity. Furthermore, the different types of radish groups were also determined by the year they were bred. This result demonstrated that there are differences between the older radish breeds and the more recently developed radish breeds. Even though a relatively small number of markers were used in the analysis, it was possible to distinguish whether the radish was bred 30 years ago or in the 2000s, and that the similar physical shapes comprised a particular group, showed that the SSR markers can indeed be successfully applied to to study the diversity within radish breeding lines. Through the results of this study, it can be concluded that the SSR marker developed for the Chinese cabbage can be applied to examine the genetic diversity and analyze the relationship (genetic resource determination) of radish.

  • This paper considers two-sided assembly line balancing with preemptive multiple goals. In the problem, three goals are taken into account in the following priority order : minimizing the number of mated-stations, achieving the goal level of workload smoothness, and maximizing the work relatedness. An evolutionary algorithm is used to solve the multiple goal problems. A new structure is presented in the algorithm, which is helpful to searching the solution satisfying the goals in the order of the priority. The proper evolutionary components such as encoding and decoding method, evaluation scheme, and genetic operators, which are specific to the problem being solved, are designed in order to improve the algorithm's performance. The computational results show that the proposed algorithm is premising in the solution quality.

  • 일반적으로 컴퓨터를 이용한 디자인 지원 시스템은 많은 양의 수집된 데이터에 기반하기 때문에 계속해서 변화하는 유행에 대처할 수 없을 뿐만 아니라 개인의 취향을 반영하기도 어렵다. 이같은 문제점을 해결하기 위해 이 논문에서는 대화형 유전자 알고리즘(Interactive Genetic Algorithm : IGA)을 이용한 디자인 지원 시스템을 제안한다. IGA는 상호작용에서 얻어지는 사용자의 평가를 적합도 함수로 사용하는 유전자 알고리즘의 하나로, 일반적인 적합도 함수를 결정하기 어려운 디자인이나 예술 등의 문제 해결에 사용될 수 있다. 이때, 몇 개의 부분적인 곡선으로 의상의 디자인을 표현하였던 기존의 방법들과는 달리 전체 디자인을 목과 몸통, 팔과 소매, 치마와 허리선의 세 가지 부분 디자인으로 나누어 표현하는 문제영역 지식을 활용함으로써 좀 더 현실적인 여성복을 디자인할 수 있었다. 시스템의 인터페이스를 보완하기 위해 OpenGL이 사용되었으며, 실험 결과 지식기반 인코딩을 통한 IGA 의상디자인 시스템이 유용함을 알 수 있었다.

Evaluation of Nonanchored Inter Simple Sequence Repeat (ISSR) Marker to Detect DNA Damage in Common Bean (Phaseolus vulgaris L.) Exposed to Acrylamide

  • Acrylamide is present as a contaminant in heated food products, predominantly from the precursor asparagine. Nonanchored inter simple sequence repeats (ISSRs) are arbitrary multiloci markers produced by PCR amplification with a microsatellite primer. In order to assess the feasibility of microsatellite primers as markers for DNA damage, the study was conducted on common bean (Phaseolus vulgaris L.) exposed to different concentrations of acrylamide. Polymorphisms were abundant among plant samples treated with acrylamide in comparison to control (untreated one) tested with 4- tri-nucleotide, 2 tetra-nucleotide, and 3- dinucelotide primers. The primer (CCG)4 was the best tested primer to generate polymorphism between the DNA of plants treated or not by acrylamide. Polymorphisms became evident as the presence and absence of DNA fragments in treated samples compared with the untreated one. The highest number of DNA variation on ISSR patterns was observed at the micromollar concentrations of acrylamide. Acrylamide was able to induce DNA damage in non concentration-dependent manner with effectiveness at micromollar concentrations. This study demonstrated that ISSR markers can be highly reliable for identification of DNA damage induced by acrylamide.

Economic Analysis of Swimming Crab (Portunus trituberculatus) Fry Releasing Program (꽃게 방류사업의 경제성 분석)

  • In this study, we analyzed the economic feasibility of the swimming crab (Portunus trituberculatus) fry releasing program in the West Sea of Korea. The catch rate of released fry in 2010, measured by genetic markers, was applied to the economic surplus method to estimate benefits. As a result of our analysis, the B/C ratio, as an indicator of economic evaluation, was determined to be 2.168, which means that the releasing program was economically feasible. And it was shown that the benefits to consumers is six times greater than the benefits to producers, confirming the necessity of the releasing program as a public work.

  • Phylogenetic relationships of stereoid fungi were examined by comparing nuclear small subunit ribosomal RNA gene sequences. Stereoid taxa were scattered into several groups and the traditional Stereaceae proved to be polyphyletic. Srereum and Xylobolus were classified in the Stereaceae as the core group of stereoid fungi, and Amylostereum was grouped with Echinodontium of the Echinodontiaceae. Chondrostereum and Cystosrereum were clustered in the Stereaceae sensu Donk and Cymatoderma and Podoscypha in the Podoscyphaceae Reid. Columnocystis abietinum and C. ambigua were grouped with Meripilus giganteus and proved to be not included in the Chaetodermataceae sensu Nakasone. Lopharia cinerascens and L. mirabilis were grouped together but L. spadicea was unrelated to them, indicating that Lopharia is heterogeneous at a generic level.

Enumeration and Recovery of Bacterial Isolates from Ruminants Fed with Different Dietary Regimes and Their Antibacterial Activity

  • The study evaluated different synthetic and semisynthetic media for maximal recovery of rumen bacteria and expression of their antibacterial activity. Rumen Glucose Cellobiose Agar (RGCA) medium was found to be the best for recovery of rumen bacteria. However, L-10 medium was the best for expression of antibacterial activity of ruminal isolates followed by Easy, M-10, RGCA and M-98-5 medium. The present study recommends the use of L-10 medium as the medium of choice for screening of antibacterial activity of ruminal isolates. Comparative evaluation of bacterial counts on different dietary regimes indicated significant difference between different growth media on a specific diet and between diets on specific growth media within a species. However, there is no overall significant difference between total bacterial counts obtained from rumen liquor of cattle and buffalo with respect to either the feeding regime or growth media. Feeding straw based diet to the animal is the best for high recovery of rumen bacteria.
