Early detection continues to be the mainstay of breast cancer control as well as the improvement of its treatment. Even so, the absence of cancer symptoms at the onset has early detection quite challenging. Therefore, various researchers continue to focus on cancer as a topic of health to try and make improvements from the perspectives of diagnosis, prevention, and treatment. This research's chief goal is development of a system with deep learning for classification of the breast cancer as non-malignant and malignant using mammogram images. The following two distinct approaches: the first one with the utilization of patches of the Region of Interest (ROI), and the second one with the utilization of the overall images is used. The proposed system is composed of the following two distinct stages: the pre-processing stage and the Convolution Neural Network (CNN) building stage. Of late, the use of meta-heuristic optimization algorithms has accomplished a lot of progress in resolving these problems. Teaching-Learning Based Optimization algorithm (TIBO) meta-heuristic was originally employed for resolving problems of continuous optimization. This work has offered the proposals of novel methods for training the Residual Network (ResNet) as well as the CNN based on the TLBO and the Genetic Algorithm (GA). The classification of breast cancer can be enhanced with direct application of the hybrid TLBO- GA. For this hybrid algorithm, the TLBO, i.e., a core component, will combine the following three distinct operators of the GA: coding, crossover, and mutation. In the TLBO, there is a representation of the optimization solutions as students. On the other hand, the hybrid TLBO-GA will have further division of the students as follows: the top students, the ordinary students, and the poor students. The experiments demonstrated that the proposed hybrid TLBO-GA is more effective than TLBO and GA.
Background: There is an urgent need to find reliable and rapid bovine tuberculosis (bTB) diagnostics in response to the rising prevalence of bTB worldwide. Toll-like receptor 2 (TLR2) recognizes components of bTB and initiates antigen-presenting cells to mediate humoral immunity. Evaluating the affinity of antigens with TLR2 can form the basis of a new method for the diagnosis of bTB based on humoral immunity. Objectives: To develop a reliable and rapid strategy to improve diagnostic tools for bTB. Methods: In this study, we expressed and purified the sixteen bTB-specific recombinant proteins in Escherichia coli. The two antigenic proteins, MPT70 and MPT83, which were most valuable for serological diagnosis of bTB were screened. Molecular docking technology was used to analyze the affinity of MPT70, MPT83, dominant epitope peptide of MPT70 (M1), and dominant epitope peptide MPT83 (M2) with TLR2, combined with the detection results of enzyme-linked immunosorbent assay to evaluate the molecular docking effect. Results: The results showed that interaction surface Cα-atom root mean square deviation of proteins (M1, M2, MPT70, MPT83)-TLR2 protein are less than 2.5 A, showing a high affinity. It is verified by clinical serum samples that MPT70, MPT83, MPT70-MPT83 showed good diagnostic potential for the detection of anti-bTB IgG and M1, M2 can replace the whole protein as the detection antigen. Conclusions: Molecular docking to evaluate the affinity of bTB protein and TLR2 combined with ELISA provides new insights for the diagnosis of bTB.

As valuable antibiotics, microbial natural products have been in use for decades in various fields. Among them are polyene compounds including nystatin, amphotericin, and nystatin-like Pseudonocardia polyenes (NPPs). Polyene macrolides are known to possess various biological effects, such as antifungal and antiviral activities. NPP A1, which is produced by Pseudonocardia autotrophica, contains a unique disaccharide moiety in the tetraene macrolide backbone. NPP B1, with a heptane structure and improved antifungal activity, was then developed via genetic manipulation of the NPP A1 biosynthetic gene cluster (BGC). Here, we generated a Streptomyces artificial chromosomal DNA library to isolate a large-sized NPP B1 BGC. The NPP B1 BGC was successfully isolated from P. autotrophica chromosome through the construction and screening of a bacterial artificial chromosome (BAC) library, even though the isolated 140-kb BAC clone (named pNPPB1s) lacked approximately 8 kb of the right-end portion of the NPP B1 BGC. The additional introduction of the pNPPB1s as well as co-expression of the 32-kb portion including the missing 8 kb led to a 7.3-fold increase in the production level of NPP B1 in P. autotrophica. The qRT-PCR confirmed that the transcription level of NPP B1 BGC was significantly increased in the P. autotrophica strain containing two copies of the NPP B1 BGCs. Interestingly, the NPP B1 exhibited a previously unidentified SARS-CoV-2 RNA-dependent RNA polymerase (RdRp) inhibition activity in vitro. These results suggest that the Streptomyces BAC cloning of a large-sized, natural product BGC is a valuable approach for titer improvement and biological activity screening of natural products in actinomycetes.

In this study, the duck eggs were salted with none or 2.5% and 5.0% (v/v) of liquid smoke (LS), respectively. As a control, samples salted without LS were used. The 2-thiobarbituric acid (TBA) values, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging ability, and reducing power of the three groups were tested at 0, 7, 14, and 21 and 28 days to determine the effects of LS on the antioxidant activity of treated eggs. In addition, gas chromatography-mass spectrometry (GC-MS) and electronic nose (E-Nose) were used to analyze the volatile flavor components of fresh duck eggs, LS, control, and salted duck eggs enriched with 2.5% (v/v) LS after 28 days of salting. The TBA value considerably increased with an increase in salting period, and the treated egg's TBA value significantly associated with LS concentration. The TBA value decreased as the LS concentration increased. The amount of LS present was highly associated with their capacity to scavenge DPPH radicals. The reducing power of the samples was substantially correlated with the LS concentration, and the reducing power increased with increasing LS concentration. The GC-MS data revealed that phenols and ketones were the predominant chemicals present in the LS, and they were also found in the eggs added to the LS even though they were absent in the fresh eggs and control. The flavor of the control group and treated eggs with LS differed significantly, according to the principal component analysis and radar map of the E-nose. The texture study results revealed that the LS significantly impacted the hardness, cohesiveness, and chewiness of eggs.
Spring black stem and leaf spot, caused by Phoma medicaginis, is an issue in annual Medicago species. Therefore, in this study, we analyzed the response to P. medicaginis infection in a collection of 46 lines of three annual Medicago species (M. truncatula, M. ciliaris, and M. polymorpha) showing different geographic distribution in Tunisia. The reaction in the host to the disease is explained by the effects based on plant species, lines nested within species, treatment, the interaction of species × treatment, and the interaction of lines nested within species × treatment. Medicago ciliaris was the least affected for aerial growth under infection. Furthermore, the largest variation within species was found for M. truncatula under both conditions. Principal component analysis and hierarchical classification showed that M. ciliaris lines formed a separate group under control treatment and P. medicaginis infection and they are the most vigorous in growth. These results indicate that M. ciliaris is the least susceptible in response to P. medicaginis infection among the three Medicago species investigated here, which can be used as a good candidate in crop rotation to reduce disease pressure in the field and as a source of P. medicaginis resistance for the improvement of forage legumes.

The sufficient amount of protein supply is crucial for improving the growth performance of growing beef cattle. In addition, due to the improvement in the genetic potential of the carcass weight of Hanwoo steers, dietary protein requirements may be increased during the rapid growth period. Accordingly, the dietary crude protein (CP) level in growing Hanwoo steers has been increasing in the field. However, little scientific evidence is available in relation to this. Therefore, this study was conducted to test whether a higher dietary CP level than convention would improve the growth performance and body metabolism in growing Hanwoo steers. Fifty growing Hanwoo steers were randomly divided into two groups and fed either a commercial diet (CON) or a higher CP (HCP) concentrate mix, provided with a similar level of dietary energy. Tall fescue hay was provided ad libitum. The dietary CP level did not affect growth performance and blood metabolite. Nitrogen intake, predicted nitrogen excretion, and retained nitrogen were higher in the HCP group than in the CON group (p < 0.01). Although there was no difference in the nitrogen utilization efficiency, the growth efficiency per retained nitrogen decreased in the HCP group (p = 0.02). A higher dietary CP level may increase nitrogen retention in growing Hanwoo steers without improving growth performance, which leads to reduced growth efficiency per retained nitrogen. Furthermore, considering the high price of feed protein and increased nitrogen excretion to the environment, a further increase in the protein level may not be sustainable.

Recently, various machine learning algorithms have been actively applied to the field of livestock research, including genetic analysis, and have drawn noteworthy results. In this study, the statistical characteristics of meat color, hydrogen ion concentration, water holding capacity (WHC), shear force, and grilling loss that affect the quality of Hanwoo beef are examined using the Hanwoo beef data collected in various environments. Moreover, the prediction of meat quality is also investigated using the two machine learning algorithms, which are linear regression and regression tree. Analysis results show that meat color has the most significant effect on WHC, which determines the tenderness of beef, and hydrogen ion concentration significantly influences shear force and grilling loss. Through this study, we can confirm the applicability of machine learning algorithms in the research on the quality of Hanwoo beef. In addition, this study can also be applied to the prediction and improvement of the quality of Hanwoo beef.

Heterostyly continues to fascinate evolutionary biologists interested in heredity, evolution, breeding, and adaptive function. Polymorphism demonstrates how simply inherited developmental changes in the location of plant sexual associations can have important consequences for population pollination and mating biology. In contrast to homozygous self incompatibility, only a small number of mating phenotypes can be maintained in the population because insect pollinators have limitations in achieving multiple segregation sites for pollen deposition. Field studies of pollen tube growth have shown that reciprocal style-stamen polymorphisms function to increase the capacity of insect-mediated cross-pollination. The genetic pattern of style morphs is well established in various taxa, but despite recent advances, the identity, number, and structure of the genes controlling the heteromorphic syndrome have been poorly elucidated. The phenomenon of heterostyly in buckwheat has been controlled by gene complex concentrate to S-locus. Homomorphic autogamous buckwheat strains were established by the interspecific hybridization. Backcrossing of this line to the common buckwheat (pin) and selecting homostylar progenies made it possible to introduce the self-compatible gene into common buckwheat. In the result, we obtained the BC9F2 generation, and defined the strong linkage between flower type and self-incompatibility by microscopic observation of pollen tube growth. This finding suggests that self-incompatibility character is not controlled by one gene. Moreover, we defined the strong linkage between flower type and self-incompatibility. It strongly supports the S supergene theory. Therefore, we have plan to elucidate the heterostyly self-incompatibility by using molecular genetics, proteome analysis and apply to exploitation of buckwheat improvement. In near future, the expression of heterozygous syndromes in genus Fagopyrum with single isolated heterozygous species may provide clues to early stages of polymorphic assembly and shed light on evolutionary models of heterozygous strains.

Wheat (Triticum spp.) is an important source of food worldwide and the focus of considerable efforts to identify new combinations of genetic diversity for crop improvement. In particular, wheat starch composition is a major target for changes that could benefit human health. Starches with increased levels of amylose are of interest because of the correlation between high amylose content and elevated levels of resistant starch, which has been shown to have beneficial effects on health for combating obesity and diabetes. In this study, high amylose wheat germplasms from other countries were collected and cultivated in Korea, and then the content of amylose was evaluated, we examined amylose content in 614 wheat germplasm. Furthermore, amylose content was validated using several milling processes such as roller, hammer, and grinding mill. As a result, the amylose content distribution was divided into five groups. The range of the amylose levels in whole wheat flour was 18.3% to 29.6%. In addition, the mutant lines were screened for high amylose, and two mutant lines (WX-1046 and WX-1074) exhibited a comparable amylose content to Keumkang whole wheat (19.6%). It has been established that high amylose indicated SS IIa null and necessitate GBSS. Based on these findings, it may be helpful to develop high amylose wheat germplasm and production techniques, particularly in Korea.

Recently, stable crop production is threatened by the effects of climate change. In particular, it is difficult to consistently maintain agricultural policies due to large price fluctuations depending on the difference in total domestic rice production from year to year. For stable rice production amid changes in the crop growing environment, development of varieties with improved disease resistance and abiotic stress stability is becoming more important. In here, drought and cold tolerant trait have been studied. First, for the development of drought tolerant varieties, we analyzed which agricultural traits are mainly affected by domestic drought conditions. As a result, it was observed that drought caused by the lack of water during transplanting season inhibits the development of the number of tiller and reduces the yield. 'Samgang' was selected as a useful genetic resource with strong drought tolerant and stable tiller number development even under drought conditions by phenotype screening. Three of drought tolerant QTLs were identified using doubled haploid (DH) population derived from a cross between Nacdong and Samgang, a drought sensitive and a tolerant, respectively. Among these QTLs, when qVDT2 and qVDTl1 were integrated, it was investigated that the tiller number development was relatively stable in the rainfed paddy field conditions. It is known that the high-yielding Tongil-type cultivars are severely affected by cold stress throughout the entire growth stage. In this study, we established conditions that can test the cold tolerance phenotype with alternate temperature to treat low temperatures in indoor growth conditions similar to those in field conditions at seedling stage. Three cold tolerant QTLs were explored using population derived from a cross between Hanareum2 (cold sensitive variety, Tongil-type) and Unkwang (cold tolerant variety, Japonica). Among these QTLs, qSCT12 showed strong cold tolerant phenotype, and when all of three QTLs were integrated, it was investigated that cold tolerant score was relatively similar to its donor parent, Unkwang, in our experimental conditions. We are performing that development of new variety with improved cold tolerant through the introduction of these QTLs.
