Understanding the genetic variation among landrace collections is important for crop improvement and utilization of valuable genetic resources. The present study was carried out to analyse the genetic diversity and associated population structure of 621 foxtail millet accessions of Korean landraces using 22 EST-SSR markers. A total of 121 alleles were detected from all accessions with an average of 5.5 alleles per microsatellite locus. The average values of gene diversity, polymorphism information content, and expected heterozygosity were 0.518, 0.594, and 0.034, respectively. Following the unweighted neighbor-joining method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 3 clusters, and population structure analysis also separated into 3 populations. Principal coordinate analysis (PCoA) explained a variation of 13.88% and 10.99% by first and second coordinates, respectively. However, in PCoA analysis, clear population-level clusters could not be found. This pattern of distribution might be the result of gene flow via germplasm exchanges in nearby regions. The results indicate that these Korean landraces of foxtail millet exhibit a moderate level of diversity. This study demonstrated that molecular marker strategies could contribute to a better understanding of the genetic structure in foxtail millet germplasm, and provides potentially useful information for developing conservation and breeding strategies.
Biochemical characteristics of 24 Pongamia pinnata genotypes (candidate plus trees) from three agroclimatic zones were estimated and molecular characterization through RAPD markers was done. Various biochemical characters viz. seed oil, total carbohydrates, protein, acid value and Iodine number recorded significant variation among different genotypes. The highest seed oil content was 41.87% while seeds of 14 genotypes recorded above average (32.11%) for the trait. Seed oil and protein content exhibited a significant positive correlation and moderate heritability. Out of the initially selected twenty-five random primers, twenty-two RAPD primers were found to be highly reproducible and produced a total of 183 loci of which 147 (80.32%) loci were polymorphic. Percentage of polymorphism varied from 44% to 100% with an average of 80.62%. High level of genetic variation was found among different genotypes of P. pinnata. Both molecular and oil content (biochemical) markers appeared useful in analyzing the extent of genetic diversity in Pongamia and the result of these analyses will help to better understand the genetic diversity and relationship among populations. Overall, the Pongamia genotypes included in the study showed a correlation with their geographical origins such that genotypes from the same region tend to have higher genetic similarity as compared to those from different regions. However, in UPGMA based Nei's analysis, some genotypes were found not to be grouped based on geographical origins possibly due to the exchange of germplasm over time between farmers across the regions. The results from oil content analyses showed that several genotypes in 'Central and Western Plateau' agroclimatic zone of Jharkhand displayed a good potential for high oil content. The study provides insight about P. pinnata populations in Jharkhand (India) and constitutes a set of useful background information that can be used as a basis for future breeding strategy and improvement of the species.
The objective of this study was to estimate variance components and genetic parameters for growth curve parameters in Guilan sheep. Studied traits were parameters of Brody growth model which included A (asymptotic mature weight), B (initial animal weight) and K (maturation rate). The data set and pedigree information used in this study were obtained from the Agricultural Organization of Guilan province (Rasht, Iran) and comprised 8647 growth curve records of lambs from birth to 240 days of age during 1994 to 2014. Marginal posterior distributions of parameters and variance components were estimated using TM program. The Gibbs sampler was run 300000 rounds and the first 60000 rounds were discarded as a burn-in period. Posterior mean estimates of direct heritabilities for A, B and K were 0.39, 0.23 and 0.039, respectively. Estimates of direct genetic correlation between growth curve parameters were 0.57, 0.03 and -0.01 between A-B, A-K and B-K, respectively. Estimates of direct genetic trends for A, B and K were positive and their corresponding values were $0.014{\pm}0.003$ (P < 0.001), $0.0012{\pm}0.0009$ (P > 0.05) and $0.000002{\pm}0.0001$ (P > 0.05), respectively. Residual correlations between growth curve parameters varied form -0.52 (between A-K) to 0.48 (between A-B). Also, phenotypic correlations between growth curve parameters varied form -0.49 (between A-K) to 0.47 (between A-B). The results of this study indicated that improvement of growth curve parameters of Guilan sheep seems feasible in selection programs. It is worthwhile to develop a selection strategy to obtain an appropriate shape of growth curve through changing genetically the parameters of growth model.

Objective: This study was to estimate heritabilities, additive genetic correlations, and phenotypic correlations between number of piglets born alive (NBA), litter birth weight (LTBW), number of piglets weaned (NPW) and litter weaning weight (LTWW) in different parities of Landrace (L), Yorkshire (Y), Landrace×Yorkshire (LY), and Yorkshire×Landrace (YL) sows in a commercial swine operation in Northern Thailand. Methods: Two models were utilized, a single trait repeatability model (RM) and a multiple trait animal model (MTM). The RM assumed reproductive records from different parities to be repeated values of the same trait, whereas the MTM assumed these records to be different traits. The two models accounted for the fixed effects of farrowing year-season, genetic group of the sow, heterosis, and age at first farrowing, and the random effects of sow, boar, and residual. Results: Heritability estimates from RM were 0.02±0.01 for NBA, 0.10±0.01 for LTBW, 0.04±0.01 for NPW, and 0.11±0.01 for LTWW. Heritability estimates from MTM fluctuated across parities, ranging from 0.04±0.01 in parity 2 to 0.09±0.02 in parity 4 for NBA, 0.07±0.02 in parity 2 to 0.16±0.02 in parity 3 for LTBW, 0.04±0.02 in parity 4 to 0.08±0.01 in parity 1 for NPW, and 0.16±0.02 in parity 1 to 0.20±0.02 in parity 2 for LTWW. Additive genetic correlation estimates from MTM were also variable, ranging from 0.29±0.24 between NBA in parity 1 and NBA in parity 2 to 0.99±0.05 between LTWW in parity 3 and LTWW in parity 4. Conclusion: The findings of this study highlight the advantage of using MTM for the genetic improvement of reproductive traits in swine and contribute to the development of sustainable swine breeding programs in Thailand.

Genetic diversity serves as the basis for selecting and genetically enhancing any culturable species in aquaculture. Here, two different strains of wild (River Ravi and River Kabul) and six captive-bred strains of Labeo rohita from various provinces were se- lected, and genetic diversity among them was evaluated using three different microsatellite markers, i.e., Lr-28, Lr-29, and Lr-37, and one mitochondrial CO1 (Cytochrome c oxidase subunit 1) gene. Different strains of L. rohita were collected, and part of their caudal fin was cut and preserved in ethanol for DNA extraction and determination of genetic diversity among them. Results in- dicated that selected markers were polymorphic with polymorphic information content (PIC) content values above 0.5 with the highest in Lr-28 followed by Lr-29 and then Lr-37. The observed heterozygosity (Ho) of all strains was higher (Avg: 0.731) but less than the expected heterozygosity (He). Moreover, TMs and WRs showed the highest He, while TKs showed the lowest, He. Over- all, inbreeding coefficient (FIS) values observed for all strains with selected markers were positive. The DNA barcoding with the CO1 gene revealed genetic variation among various strains, as demonstrated by the clades in the phylogenetic tree separating the strains into two distinct clusters that then divided into sub-clusters. In conclusion, TMs showed the highest heterozygosity as compared to other strains. Overall results provide the baseline data for the initiation of the genetic improvement program.
The objective of the present study was to estimate genetic parameters for economic traits in Markhoz goats. Data collected from 1993 to 2006 by the Markhoz goat Performance Testing Station in Sanandaj, Iran, were analyzed. The traits recorded as body weight performance at birth (BW), weaning (WW), six month (6MW), nine month (9MW), yearling (YW) and yearling fleece weight (YFW) were investigated. Least square analyses were used for estimation of environmental effects. Genetic parameters were estimated with single and multi trait analysis using restricted maximum likelihood (REML) procedures, under animal models. By ignoring or including maternal additive genetic effects and maternal permanent environmental effects, five different models were fitted for each trait. The effects of sex, type of birth, age of dam and year of birth on the all body weights were significant (p<0.01), but had no effects on YFW except year of birth. Age of kids had significant influences on WW and 6MW (p<0.01). A log likelihood ratio test was carried out for choosing the most suitable model for each trait. Total heritability estimates for YFW and growth traits varied from 0.16 for YFW and WW to 0.41 for YW. For all traits, maternal heritability was lower than direct heritability, ranging from 0.06 for BW to 0.01 for 6MW and 9MW. The magnitude of $c^2$ was more substantial for BW than the others, and relative importance was reduced from 0.12 for BW to 0.04 for 9MW. The direct additive genetic correlations estimates were positive and varied from 0.21 between BW-YW to 0.96 between WW-6MW. Direct additive genetic correlations between YFW and body weight traits were positive and ranged from 0.14 between BW-YFW to 0.67 between 6MW-YFW. For all traits, the corresponding estimates for phenotypic correlation were positive and lower than genetic correlations. The maternal additive genetic correlations between various traits were varied and ranged from -0.19 between 9MW-YFW to 0.96 between 6MW-9MW. The estimates of the maternal permanent environmental correlations between various traits were positive and ranged from 0.33 between WW-YFW to 0.93 between WW-6MW. Also, the environmental correlations between various traits ranged from 0.01 between BW-YFW and WW-YFW to 0.70 between 9MW-YW. Estimates of genetic parameters for various traits in this study confirm that selection should be applied on WW for genetic improvement in Markhoz goats.

The ability to feed an expanding world population poses one of the greatest challenges to mankind in the future. Accompanying the increased demand for food by the expected nine billion inhabitants of Earth in 2050 will be a continual decrease in arable land area, together with a decline in crop yield due to a variety of stresses. For these formidable challenges to be met, future crops should not only by high-yielding, but also stress-tolerant and disease-resistant. In this review, we highlight the importance of genetic engineering as an indispensable tool to generate just such future crops. We briefly discuss strategies and available tools for biotechnological crop improvement and identify selected examples of candidate genes that may be manipulated so that current biological maxima in yield may be surpassed by comfortable margins. Future prospects and the necessity for basic research aimed at identifying novel target genes are also discussed.
TMC (Tautomycetin) is a liner polyketide immunosuppressive antifungal compound produced by Streptomyces spp. Inhibition of T cell proliferation with TMC was observed highly efficient at 100-fold lower than those needed to achieve maximal inhibition with cyclosporin A. To elucidate the biosynthetic pathway of TMC, a genomic DNA library was constructed using a E. coil-Streptomyces shuttle cosmid vector, pOJ446. The DNA libraries were screened by colony blot hybridization using several polyketide ${\beta}-ketosynthase$ (KS) probes amplified from TMC-producing Streptomyces genomic DNA using polymerase chain reaction (PCR), of which the degenerate primers were designed based on the highly conserved sequences present in KS domains of various type I polyketide synthase genes in Streptomyces species. This library construction and screening approach led to the isolation of several positive cosmid clones representing type I polyketide biosynthetic gene clusters. In addition, a Streptomyces regulatory gene called afsR2 (a global regulatory gene stimulating antibiotic production in both S. coelicolor and S. lividans) was successfully integrated into the TMC-producing Streptomyces chromosome via E. coil-Streptomyces heterologous conjugation mehtod. The more detailed results of production improvement and genetic characterization of TMC-producing Streptomyces spp. will be discussed.

To determine the level of heterosis, higher cocoon shell weight multivoltine congenic lines (Con. L) and bivoltine syngenic lines (Syn. L) of silkworm were used for crosses. First filial generations $(F_1s)$ expressed heterobeltiotic genetic interaction at significant magnitude (p < 0.01) for single cocoon shell weight (SCSW). The other linked characters viz., single cocoon weight (SCW) and yield by weight per 10, 000 larvae were also significantly higher (p < 0.01) than the better parental lines. All the hybrids showed significant improvement for these aforesaid characters over standard heterosis (Standard check). The reeling parameters viz., filament length, raw silk, neatness, cohesionstrokes etc, also showed improvement among the hybrids than check in congenial environment. Overall results suggested that the cross between congenic and syngenic lines provide better heterosis with good quality silk than conventional hybrids and may be used for commercial exploitation.
Population learning algorithm (PLA) is a population-based method that was inspired by the similarities to the phenomenon of social education process in which a diminishing number of individuals enter an increasing number of learning stages. The study aims to develop a framework that repeatedly applying the PLA to solve the discrete resource constrained project scheduling problem with two objectives: minimizing project makespan and renewable resource availability, which are two most common concerns of management when a project is being executed. The PLA looping framework will provide a number of near Pareto optimal schedules for the management to make a choice. Different improvement schemes and learning procedures are applied at different stages of the process. The process gradually becomes more and more sophisticated and time consuming as there are less and less individuals to be taught. An experiment with ProGen generated instances was conducted, and the results demonstrated that the looping framework using PLA outperforms those using genetic local search, particle swarm optimization with local search, scatter search, as well as biased sampling multi-pass algorithm, in terms of several performance measures of proximity. However, the diversity using spread metric does not reveal any significant difference between these five looping algorithms.
