Molecular Diagnosis for Personalized Target Therapy in Gastric Cancer

  • Gastric cancer is the second leading cause of cancer-related deaths worldwide. In advanced and metastatic gastric cancer, the conventional chemotherapy with limited efficacy shows an overall survival period of about 10 months. Patient specific and effective treatments known as personalized cancer therapy is of significant importance. Advances in high-throughput technologies such as microarray and next generation sequencing for genes, protein expression profiles and oncogenic signaling pathways have reinforced the discovery of treatment targets and personalized treatments. However, there are numerous challenges from cancer target discoveries to practical clinical benefits. Although there is a flood of biomarkers and target agents, only a minority of patients are tested and treated accordingly. Numerous molecular target agents have been under investigation for gastric cancer. Currently, targets for gastric cancer include the epidermal growth factor receptor family, mesenchymal-epithelial transition factor axis, and the phosphatidylinositol 3-kinase-AKT-mammalian target of rapamycin pathways. Deeper insights of molecular characteristics for gastric cancer has enabled the molecular classification of gastric cancer, the diagnosis of gastric cancer, the prediction of prognosis, the recognition of gastric cancer driver genes, and the discovery of potential therapeutic targets. Not only have we deeper insights for the molecular diversity of gastric cancer, but we have also prospected both affirmative potentials and hurdles to molecular diagnostics. New paradigm of transdisciplinary team science, which is composed of innovative explorations and clinical investigations of oncologists, geneticists, pathologists, biologists, and bio-informaticians, is mandatory to recognize personalized target therapy.

  • 차세대 생산 시스템(Next Generation Manufacturing System: NGMS)의 핵심 개념은 분산 생산 시스템과 다품종 소량의 유연 생산 시스템의 지원이다. 이러한 시스템의 구성을 위하여 실시간 데이터에 기반한 예측 모델이 필수적인데, 이러한 예측 기능을 통하여 생산공정의 관리와 운영, 특히 전체 공정관리를 효율적으로 수행할 수 있다. 한편, 공정으로부터 전송된 데이터는 특정한 형태의 지식으로 표현된다. 이러한 지식들은 시스템에 대한 다양한 정보를 가지고 있으므로 정보를 이용하여 시스템 상태를 빠르고 쉽게 진단할 수 있다. 공정 진단은 현재 공정 상태에서 생산되는 제품의 품질을 추정할 수 있는 정보로 활용된다. 본 논문에서는 이러한 개념이 바탕이 되어 공정관리 시스템을 설계하였다. 제안된 시스템의 적용 대상은 반도체 제조 공정의 단위 공정인 에칭 공정이다. 에칭 공정은 공정 중에 연속적인 검사가 수행되지 않고 최종 제품에 대한 검사가 수행되므로 불량 원인을 찾는 것이 쉽지 않다. 따라서 본 논문에서는 공정관리를 위한 의사지원시스템을 통해 공정의 연속적인 간접진단을 수행하고자 하였다. 본 연구에서 사용된 의사지원시스템은 각 공정에서 얻어지는 데이터와 경험적 지식을 토대로 공정시스템의 해석과 진단이 가능한 시스템이다.

Design and Experimental Validation of a Digital Predictive Controller for Variable-Speed Wind Turbine Systems

  • Advanced control algorithms must be used to make wind power generation truly cost effective and reliable. In this study, we develop a new and simple control scheme that employs model predictive control (MPC), which is used in permanent magnet synchronous generators and grid-connected inverters. The proposed control law is based on two points, namely, MPC-based torque-current control loop is used for the generator-side converter to reach the maximum power point of the wind turbine, and MPC-based direct power control loop is used for the grid-side converter to satisfy the grid code and help improve system stability. Moreover, a simple prediction scheme is developed for the direct-drive wind energy conversion system (WECS) to reduce the computation burden for real-time applications. A small-scale WECS laboratory prototype is built and evaluated to verify the validity of the developed control methods. Acceptable results are obtained from the real-time implementation of the proposed MPC methods for WECS.

  • Gestational diabetes mellitus (GDM) is a complex metabolic disorder of pregnancy that is suspected to have a strong genetic predisposition. It is associated with poor perinatal outcome, and both GDM women and their offspring are at increased risk of future development of type 2 diabetes mellitus (T2DM). During the past several years, there has been progress in finding the genetic risk factors of GDM in relation to T2DM. Some of the genetic variants that were proven to be significantly associated with T2DM are also genetic risk factors of GDM. Recently, a genome-wide association study of GDM was performed and reported that genetic variants in CDKAL1 and MTNR1B were associated with GDM at a genome-wide significance level. Current investigations using next-generation sequencing will improve our insight into the pathophysiology of GDM. It would be important to know whether genetic information revealed from these studies could improve our prediction of GDM and the future development of T2DM. We hope further research on the genetics of GDM would ultimately lead us to personalized genomic medicine and improved patient care.

  • In general wireless mobile channel is modeled as complex random processes having a narrowband spectrum. In this paper, a real-time feneration of fading signals using a DSP chip is described. Real-time simulator is designed so that simulation parameters such as mobile terminal speed, carrier frequency, power ratio of line-of-sight component versus multipath, and variance of received power can be chosen in the window. Design algorithms for the generation of ideal fading signals with a minimum DSP computation and trade-offs are investigated. The accuracy of the statistical characteristics is verified through the comparison of measured results with the theoretical prediction.

  • The radial turbine has been successfully applied to the systems which request relatively small output compared with the axial turbine, and has low manufacturing cost due to it's small size and simple structure. Recently, the researches on the development and the efficiency maximization of the radial turbine are in progress corresponding with the trend toward miniaturization in turbo machinery and the development of small dispersed power generation systems. The radial turbine is to be applied to our turbo refrigerator of which engine speed is 26,000 rpm and turbine efficiency is $88\%$. Also, as a heat exchanger is accepted instead of a combustor in our turbo refrigerator, the design of radial turbine has been performed to be appropriate to the circumstance of low temperature air, not high temperature combustor gas, into the turbine inlet . This radial turbine is being developed in consideration with not only the aero-dynamic performance but also the simplification of manufacturing and integration, and the durability at operating condition. This paper refer to the performance evaluation about the radial turbine design by comparison with consulting from Russia and the our evaluation about various design factors which are considered in aero-dynamic design process.

  • This paper presents the fatigue characteristics of LIPCA (LIghtweight Piezo-Composite Actuator) device system. The LIPCA device system is composed of a piezoelectric ceramic layer and fiber reinforced lightweight composite layers. Typically a PZT ceramic layer is sandwiched by a top fiber layer with low CTE (coefficient of thermal expansion) and base layers with high CTE. The advantages of the LIPCA design are weight reduction by using the lightweight fiber reinforced plastic layers without compromising the generation of high force and large displacement and design flexibility by selecting the fiber direction and the size of prepreg layers. To predict the degradation of actuation performance of LIPCA due to fatigue, the cyclic electric loading tests using PZT specimens were performed and the strain for a given excitation voltage was measured during the test. The results from the PZT fatigue test were implemented into CLPT (Classical Laminated Plate Theory) model to predict the degradation of LIPCA's actuation displacement. The fatigue characteristic of PZT was measured using a test system composed of a supporting jig, a high voltage power supplier, data acquisition board, PC, and evaluated.

Micromechanical Computational Analysis for the Prediction of Failure Strength of Porous Composites (다공성 복합재의 파손 강도 예측을 위한 미시역학 전산 해석)

  • Porosity in polymer matrix composites increases rapidly during thermochemical decomposition at high temperatures. The generation of pores reduces elastic moduli and failure strengths of composite materials, and gas pressures in internal pores influence thermomechanical behaviors. In this paper, micromechanical finite element analysis is carried out by using two-dimensional representative volume elements for unidirectionally fiber-reinforced composites with porous matrix. According to the state of the pores, effective elastic moduli, poroelastic parameters and failure strengths of the overall composites are investigated in detail. In particular, it is confirmed that the failure strengths in the transvers and through-thickness directions are predicted much more weakly than the strength of nonpored matrix, and decrease consistently as the porosity of matrix increases.

  • In gravity die casting, die cooling systems are frequently employed with water cooling to remove the heat of the solidifying metal. Thermal modeling is an important technique in mold design for improving the productivity of the process. Computer simulation system which consists of pre-processor, main solidification simulator and post-processor has been developed for three dimensional solidification analysis of cyclic gravity die casting. The pre-processor is used for mesh generation in a PC system. The modified finite difference method is adopted for the main solidification simulation algorithm during all the casting cycles. The post-processor graphically presents the simulation results. Several experiments in automotive cast piston were carried out. The temperature variations in casting and mold with time are measured experimentally, and the results are compared with calculation results. The effects of cycle number on solidification pattern are also studied. Several experimental results for the prediction of shrinkage defects are compared with calculated results.

Force Prediction and Stress Analysis of a Twist Drill from Tool Geometry and Cutting Conditions

  • Drilling process is one of the most common, yet complex operations among manufacturing processes. The performance of a drill is largely dependent upon drilling forces, Many researches focused on the effects of drill parameters on drilling forces. In this paper, an effective theoretical model to predict thrust and torque in drilling is presented. Also, with the predicted forces, the stress analysis of the drill tool is performed by the finite element method. The model uses the oblique cutting model for the cutting lips and the orthogonal cutting model for the chisel edge. Thrust and torque are calculated analytically without resorting to any drilling experiment, only by tool geometry, cutting conditions and material properties. The stress analysis is performed by the commercial FEM program ANSYS. The geometric modeling and the mesh generation of a twist drill are performed automatically. From the study, the effects of the variation of the geometric features of the drill and of the cutting conditions of the drilling on the drilling forces and the stress distributions in the tool are calculated analytically, which can be applicable for designing optimal drill geometry and for improving the drilling process.
