Effect of 5-aza-2'-deoxycytidine on Cell Proliferation of Non-small Cell Lung Cancer Cell Line A549 Cells and Expression of the TFPI-2 Gene

  • Objective: The present study employed 5-aza-2'-deoxycytidine (5-Aza-CdR) to treat non-small cell lung cancer (NSCLC) cell line A549 to investigate the effects on proliferation and expression of the TFPI-2 gene. Methods: Proliferation was assessed by MTT assay after A549 cells were treated with 0, 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR, a specific demethylating agent, for 24, 48 and 72h. At the last time point cells were also analyzed by flow cytometry (FCM) to identify any change in their cell cycle profiles. Methylation-specific polymerase chain reaction (MSPCR), real time polymerase chain reaction(real-time PCR) and western blotting were carried out to determine TFPI-2 gene methylation status, mRNA expression and protein expression. Results: MTT assay showed that the growth of A549 cells which were treated with 5-Aza-CdR was significantly suppressed as compared with the control group (0 ${\mu}mol/L$ 5-Aza-CdR). After treatment with 0, 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR for 72h, FCM showed their proportion in G0/G1 was $69.7{\pm}0.99%$, $76.1{\pm}0.83%$, $83.8{\pm}0.35%$, $95.5{\pm}0.55%$ respectively (P<0.05), and the proportion in S was $29.8{\pm}0.43%$, $23.7{\pm}0.96%$, $15.7{\pm}0.75%$, $1.73{\pm}0.45%$, respectively (P<0.05), suggesting 5-Aza-CdR treatment induced G0/G1 phase arrest. MSPCR showed that hypermethylation in the promoter region of TFPI-2 gene was detected in control group (0 ${\mu}mol/L$ 5-Aza-CdR), and demethylation appeared after treatment with 1, 5, 10 ${\mu}mol/L$ 5-Aza-CdR for 72h. Real-time PCR showed that the expression levels of TFPI-2 gene mRNA were $1{\pm}0$, $1.49{\pm}0.14$, $1.86{\pm}0.09$ and $5.80{\pm}0.15$ (P<0.05) respectively. Western blotting analysis showed the relative expression levels of TFPI-2 protein were $0.12{\pm}0.01$, $0.23{\pm}0.02$, $0.31{\pm}0.02$, $0.62{\pm}0.03$ (P<0.05). TFPI-2 protein expression in A549 cells was gradually increased significantly with increase in the 5-Aza-CdR concentration. Conclusions: TFPI-2 gene promoter methylation results in the loss of TFPI-2 mRNA and protein expression in the non-small cell lung cancer cell line A549, and 5-Aza-CdR treatment could induce the demethylation of TFPI-2 gene promoter and restore TFPI-2 gene expression. These findings provide theoretic evidence for clinical treatment of advanced non-small cell lung cancer with the demethylation agent 5-Aza-CdR. TFPI-2 may be one molecular marker for effective treatment of advanced non-small cell lung cancer with 5-Aza-CdR.

  • The identification of power flow in dynamically loaded structures Is essential in the analysis of structure-borne noise. However there are no general purpose tools to estimate powers flow. To make matters worse. It is very difficult to measure it. The power flow can be formulated in terms of balance forces(ELFORCE) at each element and velocities at the associated node obtained with MSC/NASTHAN. In this paper the procedure which is consist of the computations of the balance forces of al1 elements and the velocities at all nodes using MSC/NASTRAN. The calculations of the power f1ow at each element using PCL(PATRAN Command Language) and the Preparation of post -processes is set UP.

A practical application of cluster analysis using SPSS

  • Basic objective in cluster analysis is to discover natural groupings of items or variables. In general, clustering is conducted based on some similarity (or dissimilarity) matrix or the original input text data. Various measures of similarities (or dissimilarities) between objects (or variables) are developed. We introduce a real application problem of clustering procedure in SPSS when the distance matrix of the objects (or variables) is only given as an input data. It will be very helpful for the cluster analysis of huge data set which leads the size of the proximity matrix greater than 1000, particularly. Syntax command for matrix input data in SPSS for clustering is given with numerical examples.

  • In general, Automated Storage/Retrieval Systems (AS/RS ) have racks of equal sized cells to utilize the concept of unit-load. Most of the techniques for the performance estimation of a unit-load AS/RS are a static model or computer simulation. Especially, their models were developed under assumption that the Storage/Retrieval (S/R) machine performs only single command (SC) or dual command (DC). In reality, defending on the operating policy and the status of the system at a particular time, the S/B machine performs a SC or a DC, or becomes .: idle. In order to resolve these weak points, we propose a stochastic model for the performance estimation of unit-load AS/RS by using a single-server queueing model.   • This research extends a general theory of cognition to address cognitive constraints on complex command production and allows us to make system design recommendations. The research described in this paper addresses the cognitive origins of problems user have producing sequence-dependent command strings while interacting with the UNIX operating system. We describe an empirical and theoretical analysis of user difficulties, and then show how our analyses lead to design recommendations. In addition, we summarize results from testing the impact of our design recommendations on system usability

  • In the industrial motor drive systems, a torsional vibration is often generated because of the elastic elements in torque transmission. One of general methods for the system is H$\infty$ controller to suppress the torsional vibration and reject the torque disturbance. vibration and reject the torque disturbance. Moreover, the two-degrees-of-freedom controller, which includes the H$\infty$ controller, is designed in order to improve the command following property. In this paper, we propose a new H$\infty$ controller with partial state feedback. This method having simple structure satisfies with the fast command following property and the attenuation of disturbances and vibrations simultaneously, just like the complicated TDOF H$\infty$ controller

LOGO와 함께 곡선 만들기 - 다각형 패턴의 관점에서

  • Papert [17] introduced the LOGO environment in which we make a curve using LOGO commands (FORWARD, ROTATE). We call this geometry as turtle geometry. This environment has influenced many researchers and designers of computers and mathematics education. But the curve that we can make using LOGO command is elementary or too difficult. Polygon and circle is elementary and making other curves is difficult. In this paper, we introduce the method of drawing some other curves mediating new command. First, we study epicycloid and hypocycloid in the historical and the physical context. And we introduce the method of making epicycloid and hypocycloid using vector addition. Next we study the polygon patterns of this curve. Finally, we extend the method for making more general curve and we improve the computer environment using this metaphor.

  • In this paper, the optimal command input is considered in order to minimize the residual vibrations of a flexible cantilever beam when the beam simply changes its position by translation or rotation. Although a cantilever beam has many modes of vibration, it is shown that the consideration of the first mode is sufficient in this case. Thus, the problem becomes a single-degree-of-freedom system subjected to a ground excitation. Two simple methods are proposed to find the optimal command input based on the shock response spectrum (SRS). The first method is the simplest and can be applied to lightly damped cases, and the second method is applicable to more general problems. The second method gives almost the same results as the input shaping method. However the proposed method gives a easier and clearer control strategy.

Temporal Structure on Discourse bevel within the Controlled Information Packaging Theory

  • The temporal structure of events on the discourse level has long been of great interest in both theoretical and computational linguistics. In this paper, we offer a unified approach to the temporal relationships related to a hierarchical discourse structure. We apply the method of pronoun resolution to the interpretation of tense. It is based on an analysis within the framework of the controlled information packaging theory. A unique aspect of our account is that temporal interpretation across discourse segments in global discourse is subject to the same principles as the interpretation of global anaphora, and that there is thus no need to postulate independent principles to account for the discourse behaviour of tense. In this way, we can neatly explain the general view that tense parallels the anaphoric nature of pronouns.

한국군에서의 C4I체계와 M&S 상호운용을 위한 BML 적용에 관한 연구 (A Study on the Battle Management Language Application for the C4I and M&S Interoperation in ROK Forces)

  • 전투관리언어인 BML은 미군에서 가상 부대 및 실제 부대를 지휘 및 통제하기 제공하기 위한 명확한 언어라고 정의할 수 있다. BML은 C2 체계와 M&S를 연결하기 위해 미국과 NATO에서 개발되었다. BML이 개발된 목적은 전장정보를 통해 상황인식을 제공하고, C2 체계와 M&S간 상호운용을 위한 해법을 제시하는 것이라고 할 수 있다. 본 연구는 육군에서 시작하여 다국적국으로 구성된 환경으로 확장되고 있는 미국의 BML 개발과정에 대해서 설명하고, 한국군에서 C4I체계와 M&S간 상호운용을 위한 BML 적용 방안에 대해서 제안한다. 최근, 미국에서는 BML을 연합국으로 구성된 환경에서 적용하여 긍정적인 결과를 얻었는데, 이는 C2 체계와 M&S간 상호운용에 대한 가능성을 보여주었다. 마지막으로, 본 연구는 BML을 한국군에 적용하기 위한 방안으로 한국군 합동지휘통제체계를 중심으로 제안하였다. 이는 향후 한국군에서 BML 적용시 밑그림을 제공 할 것이라고 판단된다.
