Staphylococcus Species in the Dental and Medical Environment

  • Staphylococcus species are one of prevalent pathogens found in hospitals. Microbes that are a primary cause of nosocomial infection were isolated from a dental and medical environment it may assist the reader to explain what this is and how it differs from the 'dental health care providers and ward health care providers'. To investigate the distribution of staphylococcus species in this environment, we used vitek II to measure drug sensitivity, and further performed biochemical testing. The isolation rate of staphylococcus species from the dental and medical environment was 100% but from dental health care providers and ward health care providers were 44.4% and 33.3%, respectively. In the analyses, staphylococcus species showed resistance to diffusion of cefoxitin and oxacillin discs. These staphylococci may be sufficiently positive for the mecA gene. Our results suggest that staphylococci might be an important cause of nosocomial infection in the dental clinic.

Evaluation of the Genetic Toxicity of Synthetic Chemical (XVIII)-in vitro Mouse Lymphoma Assay and in vivo Supravital Micronucleus Assay with Butylated Hydroxytoluene (BHT)

  • Butylated hydroxytoluene (BHT) is widely used antioxidant food additives. It has been extensively studied for potential toxicities. BHT appears adverse effects in liver and thyroid. In this study, we evaluated the genetic toxicity of BHT with more advanced methods, in vitro mouse lymphoma assay $tk^{+/-}$ gene assay (MLA) and in vivo mouse supravital micronucleus (MN) assay. BHT did not appear the significantly results in the absence and presence of metabolic activation system with MLA. Also, in vivo testing of BHT yielded negative results with supravital MN assay. These results suggest that BHT itself was not generally considered genotoxic.

  • Coexistence of paroxysmal kinesigenic dyskinesia (PKD) with benign infantile convulsion (BIC) and centrotemporal spikes (CTS) is very rare. A 10-year-old girl presented with a 3-year history of frequent attacks of staggering while laughing and of suddenly collapsing while walking. Interictal electroencephalogram (EEG) revealed bilateral CTS, but no changes in EEG were observed during movement. The patient's medical history showed afebrile seizures 6 months after birth, while the family history showed that the patient's mother and relatives on the mother's side had similar dyskinesia. Genetic testing demonstrated that the patient had a heterozygous mutation, c.649_650insC, in the PRRT2 gene. To our knowledge, this constitutes only the second report of a patient with PKD, BIC, CTS, and a PRRT2 mutation.

  • Chlorobenzenes due to their acute toxicity and the capability of bioaccumulating are of great health and environmental concern. Especially, 1, 2-dichlorobenzene (CAS No. 95-50-1) is used for organic synthesis, dye manufacture, as a solvent and for other applications in chemical industry. Adverse effects of 1, 2-dichlorobenzene includes increases in liver and kidney weights and hepatotoxicity. In this study, we evaluated the genetic toxicity of 1, 2-dichlorobenzene with more advanced methods, in vitro mouse lymphoma assay $tk^{+/-}$ gene assay (MLA) and in vitro mouse supravital micronucleus (MN) assay. 1, 2-Dichlorobenzene appeared the significantly positive results and the induction of large mutant colonies only in the presence of metabolic activation system with MLA. But in vitro testing of 1, 2-dichlorobenzene yielded negative results with supravital MN assay. These results suggest that 1, 2-dichlorobenzene may play a mutagen rather than clastogen in vitro mammalian system.

The Effect of DanSamSaMul-Tang on Hematopoiesis (단삼사물탕이 조혈작용에 미치는 영향)

  • Objectives : Korean traditional medicinal herbs and some formulas have been used to improve the function of the hematopoietic system. The present research is to examine the effect of DanSamSaMul- Tang(DSSMT) as a biological response modifier related to hemato-potentiating function using bone marrow cell as a testing system. Methods : Mice with leukopenia and thrombocytopenia induced by CTX were treated with DSSMT Extracts. Then we observed cytokines mRNA expression level, the number of CFU-GEMM and BFU-E, and the expression of TPO and SCF in bone marrow cells of mice. Results : In the DSSMT group, the gene expressions of hematopoietic cytokines were increased significantly and the number of CFU-GEMM and BFU-E of this group was significantly increased compared to the group treated only with EPO and IL-3. The expressions of TPO and SCF in the group treated with DSSMT were higher than those of the control group. Conclusions : We showed that DSSMT has a good effect on the hematopoietic system. This could also be used as important research data for Korean oriental medicine about hemato-potentiating.

Colletotrichum cymbidiicola Causing Anthracnose on Cymbidium Orchids in Korea

  • A Colletotrichum species was isolated from leaves of Cymbidium exhibiting symptoms of anthracnose. In this study, the isolates obtained were identified based on recent taxonomic approaches for the genus Colletotrichum. The identity of the causal pathogen was confirmed using morphological data and phylogenetic analysis of combined multi-gene dataset (internal transcribed spacer, glyceraldehyde 3-phosphate dehydrogenase, chitin synthase-1, actin, histone3, beta-tubulin, and calmodulin). Pathogenicity testing revealed that the isolates were pathogenic to Cymbidium. Based on these results, the fungal pathogen occurring on Cymbidium orchids was identified as Colletotrichum cymbidiicola, which is a newly recorded species in Korea.

  • Analysis of intraepidermal nerve fibers using skin biopsy is a recently developed technique, providing diagnostic information on small fiber neuropathies. The specimens are obtained by 3 mm punch biopsy, which is safe and minimally invasive. Immunohistochemical staining by Protein gene product (PGP) 9.5 demonstrate not only intraepidermal nerve fibers but dermal structures, such as sweat gland and erector papillae. Up to now, many studies agree that intraepidermal nerve fiber density is dramatically reduced in various sensory neuropathies. The utility of density measure was confirmed with high sensitivity in the diagnosis of sensory neuropathy, comparable to sural nerve biopsy or quantitative sensory testing. Besides quantitative methods, morphological changes like axonal swelling and fragmentation can be used as predegenerative markers. This article reviews the technique of skin biopsy and clinical and experimental usefulness of skin biopsy in diagnosing and monitoring peripheral neuropathies.

Test of Degradation of Soman and Sarin Gas by Organophosphorus Acid Anhydrolase and Applicability of the Enzyme to the Development of Nerve Agent Decontaminant (신경작용제 분해효소의 Soman 및 Sarin Gas 분해 능력 측정 및 제독응용 가능성)

  • A gene expressing organophosphorus acid (OPA) anhydrolases have been cloned from Alteromonas haloplanktis strain and expressed in bacterial strain BL21. Crude extract was prepared from the transformed bacterial strain BL2l and used in testing its degrading capability of real nerve gas, soman and sarin. Within 1 minute after the start of the reaction, nearly 65% of the soman added to the reactant(3mM) was degraded by adding 1 mg of the crude extract enzyme(20.0 Unit $mg^-{1}$ crude protein). In 6 minutes, the reaction reached at its steady state, which indicates that soman was completely degraded by that time. In the case of sarin, the degradation efficiency was observed to be about 0.7 times of that of soman. If the specific activity of OPAA is enhanced by both increased expression efficiency and purification, OPAA seems to be applied for the development of decontaminant of skin, especially of eye.

Osmotic-inducible Gene Expression using MudJ(Km.lac) Operon Fusion in Salmonella typhimurium (Salmonella typhimurium에서 MudJ(Km.lac) 오페론 융합을 이용한 삼투유도유전자의 발현)

  • MudJ(Km.lac) operon fusions were used in the identification of osmotic-inducible genetic(osi) loci in Salmonella typhimurium. Expression of osi::lacZ(osi5001, 5027) genes were dramatically induced 39-189 fold when the osmolarity was increased. Seven osm::lacZ genes were constituvely expressed under both low and high osmotic strength. The osi5001::lacZ fusion strains showed the enhanced osmotolerance and the reduced expression of the osi5001::lacZ in the presence of 1mM proline or betaine as osmoprotectants. Four osmotic inducible genetic loci were mapped into 36 (YK531), 44 (YK504), 57 (YK501) and 84 (YK528) map unit by testing the cotransduction frequency.

Aural Abscess in a River Cooter (Pseudemys concinna)

  • A 3-year-old, captive female river cooter was presented with a 4-day history of progressive unilateral swelling of the right side of the head, lethargy, and anorexia. History, physical examination, and radiographic examination revealed an aural abscess. After administration of antibiotics and supportive care, surgical intervention was performed. Swab samples were collected from the tympanic cavity during surgery for cytology and antimicrobial susceptibility testing. Molecular analyses of 16S ribosomal RNA gene sequences identified Citrobacter spp. and Morganella morganii. The patient was treated with ciprofloxacin and meloxicam and recovered after 2 months. This report describes the successful correction of a unilateral aural abscess that responded well to surgical intervention and a properly selected antibiotic.
