Quality Characteristic of Beverage with Gastrodia elata Blume Extract (천마추출물을 이용한 음료의 품질특성)

  • This study was performed to evaluate the Quality characteristics of the beverage with Gastrodia elate Blume extract. In the sensory evaluation of Gastrodia elata Blume beverage, score of color, flavor, taste, and overall acceptability were the best for a mixing ratio of extract 50%, oligosaccharide 10%, citric acid 0.02%, apple juice 7%, and distilled water 32.98%. Brix and pH of Gastrodia elate Blume beverage were 16.97 and 4.27 Free sugars of Gastrodia elata Blume beverage were composed glucose (3417.72 mg/100g), fructose (2215.68 mg/100g), and sucrose (1615.68 mg/100g). Organic acids of Gastrodia elata Blume beverage were composed malic (472.60 mg/100g), citric (290.27 mg/100g), succinic (276.63 mg/100g), and fumaric acids (1.73 mg/100g).

  • In the course of continous work for the discovery of anti-thrombotic constituents from tubers of Gastrodia elata, citric acid 1,5-dimethyl ester was isolated from ethylacetate soluble fraction prepared from the methanol extract. The structure of the compound was determined by the spectroscopic data. The anti-thrombotic effect of this compound was observed with prolonging the bleeding time in thrombin-induced thrombosis model of mice.

  • In the course of the continual work on the fresh tubers of Gastrodia elata, a fructoside constituent was isolated from the n-butanol soluble fraction prepared from the methanol extract. The structure of this compound was identified as $n-butyl-{\beta}-D-fructopyranoside$ from various spectroscopic data including $^1H-\;and\;^{13}C-NMR$, HMQC, HMBC, and FABMS.

  • From the MeOH extract of the aerial part of Gastrodia elata Blume (GEB) (Family: Orchidaceae), eight compounds have been isolated on repeated column chromatography, and their structures were elucidated as dotriacontanoic acid (1), beta-sitosterol (2), 4-hydroxybenzaldehyde (3), docosanoic acid oxiranylmethyl ester (4), hentriacotanoic acid (5), octadecanoic acid (6), benzoic acid (7) and gastrodin (8) on the basis of their spectral evidences including EI-Mass and 2D-NMR spectrum. All of them were obtained from the aerial part of Gastrodia elata Blume for the first time, in which compound 4 is a novelty to our best knowledge. It is also known that a phenolic glucoside, gastrodin is a major constituent just like Gastrodia rhizome.

Studies on Antioxidant, Anti-inflammation and Moisturizing Activities of Gastrodia elata Flower Extract (천마꽃 추출물의 항산화, 항염, 보습 활성에 대한 연구)

  • Gastrodia elata has a very low pollination rate in natural state, and even in artificial cultivation, there are very few individuals that bloom due to the degeneration, so little studies have been conducted. This study confirmed that the potential as a cosmetic ingredient by evaluating the antioxidant activity through the evaluation of DPPH radical scavenging activity, anti-inflammatory activity through the inhibitory effect on nitric oxide production, and the moisturizing activity through the effect on promoting hyaluronic acid production by artificially flowering G. elata flower. It was also confirmed that the appearance rate and flowering rate of G. elata harvested in spring were high, and the content of gastrodin was 0.36%. The IC50 value of G. elata flower extract was 0.045% and it was confirmed that G. elata flower extract had higher radical scavenging activity than G. elata root extract. The NO production inhibitory activity against the flower extract showed a significant inhibitory effect from 1% to 83.2%. Hyaluronic acid production promotion efficacy was not confirmed in the G. elata root extract, but the production rate increased with concentration dependence in the flower extract, and it was the highest at 46.9% when 0.02% treatment was performed. Based on the above research results, it is judged that G. elata flower extract has high potential for use as an antioxidant, anti-inflammatory, and skin moisturizing cosmetic ingredient.

Deodorization of Gastrodia elata Blum Extracts using Diaion HP-20 Resin (Diaion HP-20 레진을 이용한 천마 추출물의 불쾌취 제거)

  • Background: Although Gastrodia elata contains the valuable, bio-active compounds gastrodin and 4-hydroxybenzyl alcohol (4-HBA), it also contain ρ-cresol, which is a major source of an objectionable pig slurry-like odor. Accordingly, this study was performed to determine the deodorization effect of Diaion HP-20 resin on a G. elata extract. Methods and Results: Using Diaion HP-20 with an increasing concentration of ethanol, an extract of G. elata was fractionated into 15 fractions, which were anlayzed to determine their concentrations of active compounds and ρ-cresol. The gastrodin, 4-HBA, total phenolics, and ρ-cresol concentrations of the G. elata extract were 403.13, 408.15, 2,476.09, and 11.77 ㎍/㎖, respectively. Among the different fractions, gastrodin and 4-HBA were detected in fractions 3 to- 6, and 4 to- 10, respectively whereas total phenolics were detected in all samples, with fractions 5 to- 7 being characterized by higher concentrations than were the other fractions. ρ-Cresol was detected in fractions 12 to- 15, at concentrations ranging from 0.39 ㎍/㎖ to 6.05 ㎍/㎖ with the highest concentration detected in fraction 14. The fractions containing ρ-cresol could be selectively separated from the G. elata extract without the loss of major bio-active compounds such as gastrodin and 4-HBA. Conclusions: The results of this study indicate that a deodorizing method using Diaion HP-20 resin can be considered for improving the sensory qualities of G. elata extract without a loss of the major bio-active compounds.

Anti-inflammatory Effects of Gastrodia elata Extract in Lipopolysaccharide-stimulated BV-2 Microglia Cell

  • Gastrodia elata Blume is used for a variety of purposes including treatment of inflammation in the Korean medicine. The present study investigated whether the G. elata extracts have the anti-inflammatory effect on lipopolysaccharide(LPS)-stimulated BV-2 microglia cells. G. elata extracts showed an anti-inflammatory effects in the morphological and nitric oxide(NO) analysis, especially in hexane extract. So we investigated the hexane extract from G. elata in the following experiments. Hexane extract significantly inhibited the secretion of NO with protein level of inducible nitric oxide synthase in LPS-stimulated BV2 microglia cells. Hexane extract also inhibited LPS-stimulated inflammatory responses involving the degradation of cytosolic inhibitory(I)-${\kappa}B{\alpha}$ and the translocation of nuclear factor(NF)-${\kappa}Bp65$ to nucleus in LPS-stimulated BV-2 microglia cells by morphological analysis. Western blot analyse confirmed that I-${\kappa}B{\alpha}$ and NF-${\kappa}Bp65$ showed a similar pattern as morphological analysis. Our results suggest that G. elata extracts, especially hexane extract, may act as a therapeutic agent for inflammatory disease in the central nervous system through a selective regulation of NO production and NF-${\kappa}B$ activation.

Antidepressant-like Effects of the Gastrodia elata Bl Extract in Mice

  • Objectives : A growing body of evidence has suggested that the dysfunction of glutamatergic systems plays a pivotal role in major depressive disorder (MDD). This study was performed to investigate the antidepressant-like effects of the ethanolic extract of Gastrodia elata Bl (GE) in mouse models and to investigate the role of ${\alpha}$-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) receptors in producing these antidepressant-like effects. Methods : The forced swim test (FST) and tail suspension test (TST) were used to investigate GE's behavioral effects in mice. Additional biochemical and behavioral experiments with NBQX, an AMPA receptor antagonist, were undertaken to determine whether the antidepressant-like properties of GE are involved in AMPA receptor throughput. Results : Oral administration of GE extract (1,600 mg/kg) 1h prior to testing significantly reduced the immobility times in the FST and TST. These antidepressant-like effects of GE extract were increased dose-dependently. Pre-treatment with NBQX significantly attenuated the reduction in immobility time induced by the GE extract in the FST and TST. Conclusions : The ethanolic extract of GE may exert antidepressant-like effects with involvement of AMPA receptor.

Change of Total Polyphenol Content of Fermented Gastrodia elata Blume and Radical Scavenging (발효천마의 총 폴리페놀 함량 변화 및 라디칼 소거능)

  • This study was conducted to compare and analyze change of content of polyphenol and p-hydroxybenzyl alcohol (HBA), which is an index-component, and antioxidant activity between fermented Gastrodia elata Blume and non-fermented G. elata Blume. The polyphenol contents before and after the fermentation were 108.65 and 389.99 mg/mL respectively, and the content of index-component HBA increased from 2.46 mg/g before fermentation to 7.94 mg/g after fermentation. In comparison between the non-fermented G. elata Blume extract (NFGE) and fermented G. elata Blume extract (FGE) in DPPH, ABTS, and superoxide radical scavenging assay, we found that FGE showed more activity than NFGE as the extract was more concentrated. Especially, the superoxide radical scavenging activity was increased more than twenty times in FGE. In conclusion, we confirmed increase in the electron donating ability and radical scavenging when the dried G. elata Blume is fermented, and its further feasibility as an antioxidant.
