A Study of Effects on Secretion of Gastric Juice, Movement of Intestine and Pulse Rate with Coptidis Rhizoma and Zingiberis Rhizoma (황연(黃連) 및 건강(乾薑)이 위액분비(胃液分泌).장관운동(腸管運動) 심장박동(心臟搏動)에 미치는 영향(影響)에 관(關)한 연구(硏究))

  • As a result of administration of Coptidis Rhizoma extract and Zingiberis Rhizoma extract to Rat and Rabbit, its effect to the Gastric Juice Volume, Acidity, Pepsin out put of Rat and the movement of Intestine, Pulse Rate of Rabbit was as follows; 1. In the secretion of Gastric Juice, the Juice Volume in Coptidis Rhizoma administered group in contrast to Control Group, has increased in 30mg the increasing rate 21.4%, and has decreased in 300mg the decreasing rate 113.9%. But the Juice Volume in Zingiberis Rhizoma administered group has all decreased in 12, 30, 60, 120mg, and has remarkably decreased in 60mg the decreasing rate 62.9%. 2. The Gastric Acidity in Coptidis Rhizome administered group, in contrast to Control Group, has increased in 30, and 75mg, but no significant value, and has decreased in 300mg the decreasing rate 104.7%. But the Gastric Acidity in Zingiberis Rhizoma administered group has all decreased in 12, 30, 60 120mg, and has remarkably decreased in 60mg the decreasing rate 105.6%. 3. The Pepsin out-put in Coptids Rhizoma administered group, in contrast to Control group, has increased in 30mg the increasing rate 30.3%, and has decreased in 150, 300mg, no significant value. But the Pepsin out-put in Zingiberis Rhizoma administered group has all decreased in 12, 30, 60, 120mg, and has remarkably decreased in 60mg the decreasing rate 86.4%. 4. In the movement of Intestine, the contraction Coptidis Rhizoma administered group, in contrast before the administration, has increased in 12mg, but has begun to decrease in 30mg, and has remarkably decreased in 70mg. The contraction in Zingiberis Rhizoma administered group, in contrast before the administration, has all increased in 30, 60, 120mg. 5. In Pulse Rate, the pulsation, in contrast to Control group after 60, 120 180, 240min. has decreased in Coptidis Rhizoma administered group, and has increased in Zingiberis Rhizoma administered group, but no significant value.

Characterization and Anti-Gastric Ulcer Activity of Bamboo Salt (죽염의 특성 분석과 항위궤양효과)

  • Bamboo salt has been used as a traditional remedy for gastric ulcer and gastro-intestinal disorders. It is produced by baking the salt packed in bamboo cylinder nine times under the fire of pine tree. Three of commercially available bamboo salt products (bamboo salt A, B, and C) were characterized by qualitative and quantitative analyses using inductively coupled plasma (ICP) spectrometer, ion chromatograph (IC), X-ray diffractometer (XRD), and electron microscope (EM). Compared with crude salt, the contents of iron, silicon, potassium, and phosphate in the bamboo salt products were higher whereas the sulfate content was lower. Water-insoluble fraction of bamboo salts contained the following compounds; MgO, $SiO_2,\;Mg_2Si0_4,\;and\;CaMgSi0_4$. The study on the microscopic structures of the bamboo salts were shown to have smooth surface and fused shape compared with crude salt. Among the three bamboo salt products, product A was used to test a possible inhibitory effect on gastric acid secretion. Each test material (bamboo salt A, crude salt, and reagent-grade NaCl) was given orally to Sprague-Dawley rats at doses of 0.2, 1.0, and 2.0 g/kg for 28 days before pyrolus ligation. Twenty four hours after the last administration of the test materials, volume, pH, total acidity, and pepsin activity of gastric juice were measured by the Shay-ligation method. No significant differences were observed in the secretion of gastric acid between treated groups (bamboo salt-, crude salt- and reagent-grade NaCI-treated groups) and control group (distilled water-treated group). This result demonstrated that bamboo salt did not exert anti-ulcer activities in experimental animals used in the present study.

  • In order to find out the effect of Anti-Histamine of the Sungmagalkuntang and Sungmagalkuntang-plus-samultang I've investigated the effect of the time to death, the peripheral vascular Pemeability, the amount of secretion of Gastric Juice, the total Acidity of Gastric Juice, the contraction of Intestine Tube by Histamine and the effect of the Skin-Reaction by the decrease of DNCB. These results are following ; 1. The effect of Mice's time to death by Histamine was that the injection group of sungmagalkungtang and the injection group of sungmagalkungtang-plus-samultang both have had the effect of the extention of the time to death, which indicated a statistical significance in comparison with comparative group. 2. The effect of the increased vascular permeability by Histamine was that the injection group of sungmagalkuntang and the injection group of sungmagalkuntang-plus-samultang both have had the effect of the restraint of the increased vascular permeability, which indicated a statistical significance in comparison with comparative group. 3. The effect of the the amount of the secretion of Gastric Juice by Histamine stimulus was that the injection group of sungmagalkuntang-plus-samultang only has had the effect of restraint of the increased of the secretion of Gastric Juice, which indicated a statistical significance in comparison with comparative group. 4. The effect of the change of total Acidity of Gastric Juice by Histamine's stimulus was that the injection group of sungmagalkuntang-plus-samultang only has had the effect of the restraint of the increase of total Acidity of Gastric Juice, which indicated a statistical significance in comparison with comparative group. 5. The effect of the contraction of Intestine Tube by Histamine was that sungmagalkuntang and sungmagalkuntang-plus-samultang both have had the effect of the contraction and the restraint. According to the increase of the density, the effect of the restraint was more clear. But the difference of the effects between two substances had little. 6. The effect of the reaction of skin by the decrease of DNCB was that the injection group of sungmagalkuntang and the injection group of sungmagalkuntang-plus-samultang have had the decreased effect of the Skin-Reaction in comparison with comparative group. sungmagalkuntang-plus-samultang aws more effective. In these results shown above, both sungmagalkuntang and sungmagalkuntang-plus-samultang was shown as more effective one.

Gastroprotective Activities of Sennoside A and Sennoside B via the Up-Regulation of Prostaglandin E2 and the Inhibition of H+/K+-ATPase

  • Sennoside A (erythro) and sennoside B (threo) are dianthrone glycosides and diastereomers. We investigated their abilities to prevent the gastric lesions associated with diseases, such as, gastritis and gastric ulcer. To elucidate their gastroprotective effects, the inhibitions of $HCl{\cdot}tOH$-induced gastritis and indomethacin-induced gastric ulcers were assessed in rats. It was observed that both sennoside A and sennoside B increased prostaglandin $E_2$ ($PGE_2$) levels and inhibited $H^+/K^+$-ATPase (proton pump). In a rat model, both compounds reduced gastric juice, total acidity and increased pH, indicating that proton pump inhibition reduces gastric acid secretion. Furthermore, sennoside A and B increased $PGE_2$ in a concentration-dependent manner. In a gastric emptying and intestinal transporting rate experiment, both sennoside A and sennoside B accelerated motility. Our results thus suggest that sennoside A and sennoside B possess significant gastroprotective activities and they might be useful for the treatment of gastric disease.

Gene Expression and Secretion of Human Epidermal Growth Factor in a Methylotrophic Yeast Hansenula polymorpha (메나놀 자화 효모 Hansenula polymorpha를 이용한 재조합 인체 표피 성장인자 유전자의 발현 및 분비)

  • Using a methylotrophic yeast Hansenula polymorpha, a heterologous gene expression and secretion system was developed for the production of hEGF(human Epidermal Growth Factor) which has been shown to promote epithelial cell proliferation and to inhibit gastric acid secretion. The hEGF gene was chemically synthesized according to the preferred codon usage in H. polymor- pha and expressed under the control of the strong and inducible methanol oxidase(MOX) promoter. The mating factor $\alpha$ pre-pro leader sequence of Saccharomyces cerevisiae was employed for hEGF to be secreted into the extracellular medium. This expression cassette was stably integrated into the host chromosomal DNA. Mature hEGF was efficiently expressed and secreted into the extracel- lular medium. About 24 mg/l of hEGF was detected in the cuture supernatant of a transformant with pA-EGF3 under the suboptimal culture conditions.

  • Gastric ulcer has multifactorial etiology, and the development of ulcer is known to be caused by gastric acidity, pepsin secretion, gastric motility and gastric mucosal blood flow. The ulcer results from the tissue necrosis and apoptotic cell death triggered by mucosal ischemia, free radical formation and cessation of nutrient delivery. The gastric mucosa is usually exposed to a wide range of aggressive insults, and has developed efficient mechanisms to repair tissue injury. The apoptotic process of gastric mucosa is triggered by the induction of such proapoptotic gene expression, such as BAX. The Bcl-2 family of proteins plays a pivotal role in the regulation of apoptosis. The maintenance of gastric mucosa integrity depends upon the ratio between cell proliferation and cell death. Stress-inducing factors may affect Bcl-2/BAX ratio and thus the rate of apoptosis through modulation of the expression of both proteins depends upon the experimental model. In addition to the regulation of apoptosis, new vessels have to be generated in order to ensure an adequate supply of oxygen and nutrients to the healing gastric mucosa. This events are regulated by several factors. Among them, such polypeptide growth factors, such as vascular endothelial growth factor (VEGF) regulates essential cell functions involved in tissue healing including cell proliferation and differentiation. The purpose of this study was carried to investigate whether Rhei Rhizoma administration might protect apoptotic cell death and promote angiogenesis in gastric mucosa. Sprague-Dawley rats were randomly divided into 4 groups; normal, saline, cimetidine and Rhei Rhizoma-treated group. The saline, cimetidine and Rhei Rhizoma extracts were orally administrated to each group and gastric ulcer was induced by HCl-EtOH solution. After 1 hour, the stomachs were collected for histological observation and immunohistochemistry. In results, Rhei Rhizoma proves to promote to heal wound in gastric ulcer in conclusion and the significant changes of BAX, Bcl-2 and VEGF quantity in gastric mucosa were observed. These results suggest that Rhei Rhizoma extract may promote incision wound healing and has protective effects on gastric ulcer in rats.

The Therapeutic Effects of Nizatidine in Gastroesophageal Disease with Laryngopharyngeal Reflux Symptoms: Observational Study (위식도 역류성 질환 관련 인후두성 역류(Laryngopharyngeal Reflux : LPR)증상을 호소하는 환자에서의 니자티딘의 치료효과 연구)

  • Larygopharyngeal reflux(LPR) is one form of the Gastroesophageal Reflux Diseases(GERD). It is known to cause various kinds of otolaryngologic symptoms such as hoarseness, globus sensation in throat, chronic throat clearing, and chronic cough, Disease entities diagnosed by otolaryngologists as posterior laryngitis, globus pharyngeus and reflux laryngitis should be suspected as LPR-related diseases. The nizatidine(AXID), as a Histamine H2-receptor antagonist, reduces gastric acid secretion and improves gastric motility function. Objectives : The effect of nizatidine using 150mg b.i.d was evaluated for symptom relief and improvement of laryngoscopic findings in patients with LPR. Materials and Methods : In 30 multicenter, observational trial performed nationalwidely in Korea. 308 patients with LPR symptom were observed to evaluate their symptoms and larygnoscopic findings after 4weeks, 8weeks, 12weeks of treatment with nizatidine. Results : The symptoms of LPR including globus sensation, chronic throat clearing and hoarseness, are reduced significantly after 4 weeks, 8weeks, and 12weeks of treatment(p<0.05). The laryngoscopic findings including diffuse erythema, edema and granulation are improved after nizatidine treatment(p<0.05). and the efficacy of nizatidine on LPR-related sympoms after 4 weeks is 88.6%, and those of after 8 weeks and 12weeks were 92.6%, and 99.1% in ITT(Intent To Treatment) group(p<0.05). And PPA(Per Protocol Analysis)group showed 93.7%, 97.3%, and 99.1% of efficacy after 4, 8, and 12 weeks of nizatidine treatment(p<0.05). Conclusion : These results indicate that in patient with LPR, nizatidine 150mg b.i.d treatment very effectively reduces LPR symptoms and improves laryngoscopic findings as well as reduces gastric acid secretion and improves gastric motility function.

Effects of hypertonic glucose solution on acid secretion of rat stomach (흰쥐의 위산(胃酸)분비에 대한 고장(高張)포도당용액의 영향)

  • Using modified technique of Schmidt et al, as described previously (Korean J. Pharmacol 9 : 17, 1973), the stomach of female rats were perfused with physiological saline under urethane anesthesia. The acid-secretory response of the perfused stomach to i.v. hypertonic glucose (50%), casein hydrolysate (20%) or saline (6%) solution were studied with or without histamine or methacholine stimulation. A significant decrease of acid secretion from the rat stomach was induced by i.v. hypertonic glucose or saline solution. The histamine-stimulated acid secretion was also decreased by simultaneous administration of the hypertonic glucose or saline. However, methacholine-stimulated acid response was not affected by the hypertonic glucose. Intravenous infusion of 20% casein hydrolysate solution resulted in an increase in acid output from the stomach under histamine stimuli. These results lead to the conclusion that the inhibitory responses of acid secretion due to i.v. hypertonic glucose solution are brought through the effect of histaminergic, not cholnergic mechanism(s) in the gastric secretion.

  • Effects of 50% glucose solution on pancreatic exocrine function were studied in rat, rabbit and cat. The alterations during the resting state, the continuous intravenous infusion of secretin and the infusion of secretin with CCK-PZ were determined. 1) No change of pancreatic secretion in rat was observed by intragastric administration of the hypertonic glucose solution. 2) Intragastric administration of the hypertonic glucose solution in rabbit produced the inhibitory effect on pancreatic secretion during secretion infusion. 3) While secretin with CCK-PZ were infused continuously, intragastric administration of the hypertonic glucose solution revealed the marked inhibitory effect on pancrcreatic secretion in cat. Oral administration of the hypertonic glucose solution produced no significant inhibition in the resting gland but markedly depressed the pancreatic flow and enzyme concentration in the secretin or CCK-PZ stimulated gland. It is felt that the inhibitory response of exocrine pancreas induced by intragastric hytertonic glucose solution is resulted in interaction between secretory hormone and gastric mucosal factor possibly enteroglucagon.

  • This research aims to study the changes In gastrointestinal function attributed to aging In human. The thresholds for recognition and detection of flavors became elevated and salivary gland acinar cells decreased in the old age. But most esophageal function remained relatively Intact. Although gastric emptying time has been slowed with aging, the total intestinal transit time did not differ. Atropic gastritis due to H. pylori in old man decreased secretion of acid and Intrinsic factor and absorbability of calcium and iron. Pancreatic secretion is droned in older persons. Prevalence of gallstones rised with age. Liver size and portal blood flow decreased significantly with age. Mucosal surface area has been reported to be slightly diminished in the aging man. Glucose transporters decreased and Insulin tolerance Increased. Absorption of aromatic amino acid is diminished with age. Dietary protein In that aging human increased fecal nitrogen excretion. Vitamin A tolerance increased. Vitamin D receptor concentration decreased and resistance to 1,25-(OH)2D3 action increased. Permeability of aging small Intestine Increased. Zinc balance dirt not differ Copper absorption appeared not to be significantly affected by age. Neurotensin secretion decreased thus slowed colonic peristaltic movements and Intestinal mucosal growth.
